2Enhanced RuBisCO activity and promoted dicotyledons growth with degradable carbon dots显示文摘The ~ 5 nm degradable carbon dots (CDs) were synthesized directly from carbon rod by a one-step electrochemical method at room temperature.The as-prepared CDs can effectively enhance the ribulose bisphosphate carboxylase oxygenase (RuBisCO) activity,and then promote the dicotyledons growth (soybean,tomato,eggplant and so on) and finally increase their yields.Here,we used Arabidopsis thaliana and Trifolium repens L.as model plants to systematically study the beneficial effects of CDs on plant growth.These include:(i) accelerating seed germination;(ii) enlarging root elongation;(iii) increasing metal ions absorption and delivery;(iv) improving enzymes activity;(v) enhancing the carbohydrate content;(vi) degradation into plant hormone analogues and CO2;and finally (vii) enhancing the grain production by about 20%.Hao Li Jian Huang Yang Liu Fang Lu Jun Zhong Yong Wang Shuiming Li Yeshayahu Lifshitz Shuit-Tong Lee Zhenhui Kang 2019Nano Research2019,12,7:8
3Genetic analysis of segregation distortion of mating-type factors in Lentinula edodes显示文摘Analyses of the mating-type ratio of protoplast monokaryons to spores in 28 strains of Lentinula edodes were performed through statistical methods. The results showed that the segregation distortion phenomenon commonly exists in both types of Lentinula edodes, 17 out of the 28 samples did not display the expected 1∶1 ratio of protoplast monokaryons; and among them 8 fruiting strains distorted 1∶1∶1∶1 segregation of spores. The degree of distortion in all tested wild strains was significantly lower than that of cultured strains. In fruiting test of positive-negative parent, predominant spores were those with the mating type identical to dikaryotic parent, indicating that the predominance of nuclei mainly depends on the mating type of dikaryotic parent. 4Rapid and accurate detection of African swine fever virus by DNA endonuclease-targeted CRISPR trans reporter assay显示文摘The first case of African swine fever(ASF)outbreak in China was reported in a suburban pig farm in Shenyang in 2018.Since then,the rapid spread and extension of ASF has become the most serious threat fo r the swine industry.Therefore,rapid and accurate detection of African swine fever virus(ASFV)is essential to provide effective strategies to control the disease.In this study,we developed a rapid and accurate ASFV-detection method based on the DNA endonuclease-targeted CRISPR trans reporter(DETECTR)assay.By com bining recombinase polymerase am plification w ith CRISPR-Cas12a proteins,the DETECTR assay dem onstrated a m inim um detection lim it of eight copies with no cross reactivity with other swine viruses.Clinical blood samples were detected by DETECTR assay and showed 100%(30/30)agreement w ith real-tim e polymerase chain reaction assay.The rapid and accurate detection of ASFV may facilitate tim ely eradication measures and strict sanitary procedures to control and prevent the spread of ASF.Zongjie Li Jianchao Wei Di Di Xin Wang Chenxi Li Beibei Li Yafeng Qiu Ke Liu Feng Gu Minglong Tong Shuiming Wang Xiaodong Wu Zhiyong Ma 2020Acta Biochimica et Biophysica Sinica2020,52,12:2
5C-X3-C motif chemokine ligand 1/receptor 1 regulates the M1 polarization and chemotaxis of macrophages after hypoxia/reoxygenation injury显示文摘Background:Macrophages play an important role in renal ischemia reperfusion injury,but the functional changes of macrophages under hypoxia/reoxygenation and the related mechanism are unclear and need to be further clarified.Methods:The effects of hypoxia/reoxygenation on functional characteristics of RAW264.7 macrophages were analyzed through the protein expression detection of pro-inflammatory factors TNF-αand CD80,anti-inflammatory factors ARG-1 and CD206.The functional implications of C-X3-C motif chemokine receptor 1(CX3CR1)down-regulation in hypoxic macrophages were explored using small interfering RNA technology.Significance was assessed by the parametrict-test or nonparametric Mann-Whitney test for two group comparisons,and a one-way ANOVA or the Kruskal-Wallis test for multiple group comparisons.Results:Hypoxia/reoxygenation significantly increased the protein expression of M1-related pro-inflammatory factors TNF-α,CD80 and chemokine C-X3-C motif chemokine ligand 1(CX3CL1)/CX3CR1 and inhibited the protein expression of M2-related anti-inflammatory factors ARG-1 and CD206 in a time-dependent manner in RAW264.7 cells.However,the silencing of CX3CR1 in RAW264.7 cells using specific CX3CR1-siRNA,significantly attenuated the increase in protein expression of TNF-α(P<0.05)and CD80(P<0.01)and the inhibition of ARG-1(P<0.01)and CD206(P<0.01)induced by hypoxia/reoxygenation.In addition,we also found that hypoxia/reoxygenation could significantly enhance the migration(2.2-fold,P<0.01)and adhesion capacity(1.5-fold,P<0.01)of RAW264.7 macrophages compared with the control group,and CX3CR1-siRNA had an inhibitory role(40%and 20%reduction,respectively).For elucidating the mechanism,we showed that the phosphorylation levels of ERK(P<0.01)and the p65 subunit of NF-κB(P<0.01)of the RAW264.7 cells in the hypoxic/reoxygenation group were significantly increased,which could be attenuated by down-regulation of CX3CR1 expression(P<0.01,both).ERK inhibitors also significantly blocked the effects of hypoxic/reoxygenation on the protein expression of M1-related pro-inflammatory factors TNF-α,CD80 and M2-related anti-inflammatory factors ARG-1 and CD206.Moreover,we found that conditioned medium from polarized M1 macrophages induced by hypoxia/reoxygenation,notably increased the degree of apoptosis of hypoxia/reoxygenation-induced TCMK-1 cells,and promoted the protein expression of pro-apoptotic proteins bax(P<0.01)and cleaved-caspase 3(P<0.01)and inhibited the expression of anti-apoptotic protein bcl-2(P<0.01),but silencing CX3CR1 in macrophages had a protective role.Finally,we also found that the secretion of soluble CX3CL1 in RAW264.7 macrophages under hypoxia/reoxygenation was significantly increased.Conclusions:The findings suggest that hypoxia/reoxygenation could promote M1 polarization,cell migration,and adhesion of macrophages,and that polarized macrophages induce further apoptosis of hypoxic renal tubular epithelial cells by regulating of CX3CL1/CX3CR1 signaling pathway.Shuiming Guo Lei Dong Junhua Li Yuetao Chen Ying Yao Rui Zeng Nelli Shushakova Hermann Haller Gang Xu Song Rong 2021Chronic Diseases and Translational Medicine2021,7,4:1
6Influence of flavone extract from cultivated saussurea on learning and memory in a mouse model of Alzheimer's disease A comparison with estradiol benzoate显示文摘The present study established a mouse model of Alzheimer's disease,and investigated the effects of treatment with flavone extract from artificially cultivated saussurea.A positive control group was treated with estradiol benzoate,and learning and memory ability were examined in the 8-arm radial maze.The learning and recognition ability of mice with Alzheimer's disease treated with flavone extract was significantly improved and the number of hippocampal neurons was significantly increased in the flavone-treated and positive control groups compared with the model group.The results indicate that flavone extract from artificially cultivated saussurea can improve learning and memory deficits in mice with Alzheimer's disease,exerting effects similar to those of estradiol benzoate.Weiqiang Chen Shuiming Gong Yan Li Ming Li Zemin Yang Lirong Zhanf 2011Neural Regeneration Research2011,6,30:1
