| 1 | Reduction of the incidence and mortality of rectal cancer bypolypectomy:a prospective cohort study in Haining County显示文摘AIM: To reduce the incidence and morlality of rectal cancerand address the hypothesis that colorectal cancer often arisefrom precursor lesion (s), either adenomas or non-alenomatous polyps, by conducting a population-basedmass screening for colorectal cancer in Haining County,Zhejiang, PRCMETHODS: From 1977 to 1980, physicians screened thepopulation of Haining County using 15 cm rigid endoscopy.Of over 240 000 participants, 4076 of them were diagnoselwith Precursor Iesions, eitler alenomas or non-adenomatous polyps, which were then removedl surgically.All individuals with precursor lesionswere followed up andreexamined by endoscopy every two to five years upto 1998.RESULTS: After the initial screening, 953 metactronousadenomas and 417 non-adenomatous polyps ware detected andremoved from the members of this cohort. Further, 27 cases ofcolorectal cancer were detected and treated. Log-rank tuestsshowed that the survival time among those cancer patients whounderwent mass screening increased significantly comparedtothat of other colorectal cancer patients (P < 0.0001). | 2 | Neuroprotective effect of exogenous vascular endothelial growth factor on rat spinal cord neurons in vitro hypoxia显示文摘Background Vascular endothelial growth factor (VEGF) is well known as a hypoxia-induced protein. That it markedly increased expression of VEGF and improvement of rat motor function after spinal cord injury suggested that VEGF could play a neuroprotective role in ischaemic tolerance. This study investigated whether vascular endothelial growth factor has direct neuroprotective effects on rat spinal cord neurons. Methods We employed primary cultures of embryonic rat spinal cord neurons, then administrated different concentrations of VEGF164 in the culture medium before hypoxia when the number of neurons was counted and the cell viability was detected by MTT. The neuronal apoptosis and expression of VEGF and its receptor genes were evaluated by terminal deoxynucleotidyl transferase mediated dUTP nick-end labelling (TUNEL) and immunohistochemistry. The VEGFR2/FLK-1 inhibitor, SU1498, was used to confirm whether the neuroprotective effect of VEGF was mediated through VEGFR2/Flk-1 receptors. Result In hypoxic conditions,the number and viability of neurons decreased progressively, while the number of TUNEL-positive cells increased along with the prolongation of hypoxic exposure. When the concentration of VEGF in cell culture medium reached 25 ng/ml, the cell viability increased 11% and neuronal apoptosis reduced to half, this effect was dose dependent and led to an approximately 25% increase in cell viability and about threefold decrease in TUNEL-positive cells at a maximally effective concentration of 100 ng/ml. In normal conditions, VEGF/Flk-1 but not VEGF/Flt-1 gene expressed at a low level: after hypoxia, the expression of VEGF/Flk-1, but not VEGF/Flt-1 was significantly increased. The protective effect of VEGF was blocked by the VEGFR2/Flk-1 receptor tyrosine kinase inhibitor, SU1498. | 4 | Apoptosis induction with polo-like kinase-1 antisense phosph-orothioate oligodeoxynucleotide of colon cancer cell line SW480显示文摘AIM: To investigate the effects of polo-like kinase-1 (PLK1) antisense phosphorothioate oligodeoxynucleotide (ASODN) on apoptosis and cell cycle of human colon cancer cell line 5W480.METHODS: After SW480 colon cancer cells were transfected with PLK1 ASODN, Northern and Western blot analyses were used to examine PLK1 gene expression in cancer cells. We studied apoptosis using terminal uridine deoxynucleotidyl nick end labeling. Apoptosis and cell cycle of SW480 cells were examined by fluorescence-activated cell sorter scan.RESULTS: The levels of PLK1 mRNA and protein were greatly inhibited by PLK1 ASODN in SW480 cancer cells transfected with PLK1 ASODN. | 6 | Sirtuin 3 deficiency exacerbates diabetic cardiomyopathy via necroptosis enhancement and NLRP3 activation显示文摘Sirtuin 3(SIRT3)is a potential therapeutic target for cardiovascular,metabolic,and other aging-related diseases.In this study,we investigated the role of SIRT3 in diabetic cardiomyopathy(DCM).Mice were injected with streptozotocin(STZ,60 mg/kg,ip)to induce diabetes mellitus.Our proteomics analysis revealed that SIRT3 expression in the myocardium of diabetic mice was lower than that of control mice,as subsequently confirmed by real-time PCR and Western blotting.To explore the role of SIRT3 in DCM,SIRT3-knockout mice and 129S1/SvImJ wild-type mice were injected with STZ.We found that diabetic mice with SIRT3 deficiency exhibited aggravated cardiac dysfunction,increased lactate dehydrogenase(LDH)level in the serum,decreased adenosine triphosphate(ATP)level in the myocardium,exacerbated myocardial injury,and promoted myocardial reactive oxygen species(ROS)accumulation.Neonatal rat cardiomyocytes were transfected with SIRT3 siRNA,then exposed to high glucose(HG,25.5 mM).We found that downregulation of SIRT3 further increased LDH release,decreased ATP level,suppressed the mitochondrial membrane potential,and elevated oxidative stress in HG-treated cardiomyocytes.SIRT3 deficiency further raised expression of necroptosis-related proteins including receptor-interacting protein kinase 1(RIPK1),RIPK3,and cleaved caspase 3,and upregulated the expression of inflammation-related proteins including NLR family pyrin domain-containing protein 3(NLRP3),caspase 1 p20,and interleukin-1βboth in vitro and in vivo.Collectively,SIRT3 deficiency aggravated hyperglycemia-induced mitochondrial damage,increased ROS accumulation,promoted necroptosis,possibly activated the NLRP3 inflammasome,and ultimately exacerbated DCM in the mice.These results suggest that SIRT3 can be a molecular intervention target for the prevention and treatment of DCM. | Shu Song Yue Ding Guo-liang Dai Yue Zhang Meng-ting Xu Jie-ru Shen Ting-ting Chen Yun Chen Guo-liang Meng | 2021 | Acta Pharmacologica Sinica2021,42,2: | 14 |

| 8 | Grape Seed Proanthocyanidin Extract Alleviates Arsenic-induced Oxidative Reproductive Toxicity in Male Mice显示文摘Objective To determine the ability of grape seed proanthocyanidin extract(GSPE) in alleviating arsenic-induced reproductive toxicity. Methods Sixty male Kunming mice received the following treatments by gavage: normal saline solution(control); arsenic trioxide(ATO; 4 mg/kg); GSPE(400 mg/kg); ATO+GSPE(100 mg/kg); ATO+GSPE(200 mg/kg) and ATO+GSPE(400 mg/kg). Thereafter, the mice were sacrificed and weighed, and the testis was examined for pathological changes. Nuclear factor(erythroid-derived 2)-like 2(Nrf2), heme oxygenase 1(HO1), glutathione S-transferase(GST), NAD(P)H dehydrogenase, and quinone 1(NQO1) expression in the testis was detected by real-time PCR. Superoxide dismutase(SOD), glutathione(GSH), total antioxidative capability(T-AOC), malondialdehyde(MDA), 8-hydroxydeoxyguanosine(8-OHd G), and reproductive indexes were analyzed. Results ATO-treated mice showed a significantly decreased sperm count and testis somatic index and activity levels of SOD, GSH, and T-AOC than control group. Compared to the ATO-treated group, ATO +GSPE group showed recovery of the measured parameters. Mice treated with ATO+high-dose GSPE showed the highest level of m RNA expression of Nrf2, HO, NQO1, and GST. Multiple models from different disciplines are usually integrated to solve those problems. Integrated modeling environment is an effective technical approach to model integration. Although a number of modeling environments worldwide are available, they cannot meet current challenges faced. Their old-fashion designs and original development purposes constrain their possible applications to the domain of hydrologic or land surface modeling. One of the challenges is that we intend to link knowledge database or ontology system to the modeling environment in order to make the modeling support more intelligent and powerful. In this paper, we designed and implemented an integrated modeling environment (HIME) for hydrological and land surface modeling purpose in a much extendable, efficient and easy use manner. With such design, a physical process was implemented as a module, or component. A new model can be generated in an intuitive way by linking module icons together and establishing their relationships. Following an introduction to the overall architecture, the designs for module linkage and data transfer between modules are described in details. Using XML based meta-information, modules in either source codes or binary form can be utilized by the environment. As a demonstration, with the help of HIME, we replaced the evaporation module of TOPMODEL with the evapotranspiration module from the Noah land surface model which explicitly accounts for vegetation transpiration. | 17 | The CRISPR/Cas9 induces large genomic fragment deletions of MSTN and phenotypic changes in sheep显示文摘The CRISPR/Cas9 system has been extensively used to engineer genetic loci for the generation of knockouts, insertions, and point mutations in animal models. However, many mutations that have been reported in animals are small insertions or deletions. This study used the CRISPR/Cas9 system to induce large DNA fragment deletions in MSTN via three guide RNAs in sheep. This successfully achieved the precise gene editing of the ovine MSTN gene by injecting both Cas9 m RNA and sg RNAs into embryos at the one-cell stage. Of 10 edited animals, 3 animals(30%) exhibited large genomic fragment deletions(~5 kb). Furthermore, the body weights of these 3 animals were significantly different(P0<0.0001, P15=0.001, P30=0.005, P60=0.027) between lambs with large deletions and wildtype lambs. In addition, the edited lambs were also significantly different(P0<0.0001, P15<0.0001, P30=0.002, P60=0.011) compared with wildtype. These results suggest that the generated MSTN knockout sheep is a reliable and effective animal model for further study. | 19 | Scalable volumetric imaging for ultrahigh-speed brain mapping at synaptic resolution显示文摘The speed of high-resolution optical imaging has been a rate-limiting factor for meso-scale mapping of brain structures and functional circuits,which is of fundamental importance for neuroscience research.Here,we describe a new microscopy method of Volumetric Imaging with Synchronized on-the-fly-scan and Readout(VISo R)for high-throughput,high-quality brain mapping.Combining synchronized scanning beam illumination and oblique imaging over cleared tissue sections in smooth motion,the VISo R system effectively eliminates motion blur to obtain undistorted images.By continuously imaging moving samples without stopping,the system achieves high-speed 3D image acquisition of an entire mouse brain within1.5 hours,at a resolution capable of visualizing synaptic spines.A pipeline is developed for sample preparation,imaging,3D image reconstruction and quantification.Our approach is compatible with immunofluorescence methods,enabling flexible cell-type specific brain mapping and is readily scalable for large biological samples such as primate brains.Using this system,we examined behaviorally relevant whole-brain neuronal activation in 16 c-Fos-sh EGFP mice under resting or forced swimming conditions.Our results indicate the involvement of multiple subcortical areas in stress response.Intriguingly,neuronal activation in these areas exhibits striking individual variability among different animals,suggesting the necessity of sufficient cohort size for such studies. | Hao Wang Qingyuan Zhu Lufeng Ding Yan Shen Chao-Yu Yang Fang Xu Chang Shu Yujie Guo Zhiwei Xiong Qinghong Shan Fan Jia Peng Su Qian-Ru Yang Bing Li Yuxiao Cheng Xiaobin He Xi Chen Feng Wu Jiang-Ning Zhou Fuqiang Xu Hua Han Pak-Ming Lau Guo-Qiang Bi | 2019 | National Science Review2019,6,5: | 7 |
