| 1 | Overexpression of the maize GRF10, an endogenous truncated growth-regulating factor protein, leads to reduction in leaf size and plant height显示文摘It has long been thought that growth‐regulating factors(GRFs) gene family members act as transcriptional activators to play important roles in multiple plant developmental processes. However, the recent characterization of Arabidopsis GRF7 showed that it functions as a transcriptional repressor of osmotic stress‐responsive genes. This highlights the complex and diverse mechanisms by which different GRF members use to take action. In this study, the maize(Zea mays L.) GRF10 was functionally characterized to improve this concept. The deduced ZmGRF10 protein retains the N‐terminal QLQ and WRC domains, the characteristic regions as protein‐interacting and DNA‐binding domains, respectively. However,it lacks nearly the entire C‐terminal domain, the regions executing transactivation activity. Consistently, ZmGRF10 protein maintains the ability to interact with GRF‐interacting factors(GIFs) proteins, but lacks transactivation activity.Overexpression of ZmGRF10 in maize led to a reduction in leaf size and plant height through decreasing cell proliferation,whereas the yield‐related traits were not affected. Transcriptome analysis revealed that multiple biological pathways were affected by ZmGRF10 overexpression, including a few transcriptional regulatory genes, which have been demonstrated to have important roles in controlling plant growth and development. | 2 | Construction of a genetic map and location of quantitative trait loci for dwarf trait in maize by RFLP markers显示文摘Using F2 population derived from the cross of tall inbred 7922 by dwarf inbred 5003, an RFLP linkage map of maize has been constructed, on which 85 markers are distributed among 10 linkage groups and span maize genome about 1827.8 cM with an average distance (24.4 cM) between markers. 106 F2:3 lines of the population were grown in a 10×11 simple rectangular lattice design of one-raw plots with two replications and evaluated for plant height (PH). With interval mapping procedure, 5 QTLs controlling plant height have been identified and their genetic effects and gene action determined. 2 major OTLs with opposite effect have been discovered. One for increasing plant height is ph 1 which is located at chromosome 2 and accounts for 51.8% of the total phenotypic variation; the other for decreasing plant height is ph3 which is located at chromosome 5 and accounts for 38.6% of the total phenotypic variation. | 3 | Cloning and characterization of a novel R1-MYB transcription factor in maize显示文摘R1-MYB transcription regulatory factors play vital roles in transcriptional modification during higher plant metabolism and development. In the present study, an R1-MYB gene was isolated from the maize (Zea mays L.) inbred line C8605-2 based on expressed sequence tags (ESTs), which expressed differentially between the hybrid C8605-2 × W1445 and its two parents in an oligonucleotide microarray. The full-length cDNA, designated as ZmMYBE1 (GeneBank Accession No. FJ024049), consists of 1992 nucleotides and contains a 1296-bp open reading frame. One conserved MYB domain was detected near the N-terminus of the deduced amino acid sequence, where an acidic Ser/Thr-rich area was also observed in the downstream region. The sequence of ZmMYBE1, mapped to chromosome 5 (bin 5.03), was revealed belonging to a multi-gene family in the maize genome. Subcellular localization analysis determined that ZmMYBE1 was located in the nucleus. Expression analysis showed that ZmMYBE1 transcripts accumulated in various tissues examined, with high expression levels in the immature ear and low levels in the tassel, root, and stem. The expression analysis for ZmMYBE1 in immature ears showed the highest expression level at the earliest vegetative developmental stage of the plant. Expression of ZmMYBE1 higher than that of the parents was observed in the hybrid C8605-2 × W1445. And also the expression level similar to the lowest of the parents in the hybrid C8605-2 × W245 was observed, revealing different expression patterns for the same gene in various hybrids combined from diversified inbred lines. | 4 | Fine Mapping of RppP25, a Southern Rust Resistance Gene in Maize显示文摘Southern rust (Puccinia polysora Underw.) is a major disease that can cause severe yield losses in maize (Zea mays L.). In our previous study, a major gene RppP25 that confers resistance to southern rust was identified in inbred line P25. Here, we report the fine mapping and candidate gene analysis of RppP25 from the near-isogenic line F939, which harbors RppP25 in the genetic background of the susceptible inbred line F349. The inheritance of resistance to southern rust was investigated in the BC1F1 and BC3F1 populations, which were derived from a cross between F939 and F349 (as the recurrent parent). The 1:1 segregation ratio of resistance to susceptible plants in these two populations indicated that the resistance is controlled by a single dominant gene. Ten markers, including three simple sequence repeat (SSR) markers and seven insertion/deletion (InDel) markers, were developed in the RppP25 region. RppP25 was delimited to an interval between P091 and M271, with an estimated length of 40 kb based on the physical map of B73. In this region, a candidate gene was identified that was predicted to encode a putative nucleotide-binding site leucine-rich repeat (NBS-LRR) protein. | 5 | Significance of neuroglobin in serum of acute atherosclerotic cerebral infarction patients显示文摘This study sought to examine neuroglobin (NGB) in the serum of acute cerebral infarction patients with double-antibody sandwich enzyme-linked immunosorbent assay to identify all risk factors, calculate infarct size, assess neurological impairment, and analyze the relation between NGB and each of these factors. The double-antibody sandwich assay indicated that levels of NGB in serum were unaltered within 6 hours following acute cerebral infarction compared with normal levels. NGB levels then underwent a distinct change, peaking at 24 hours then returning to normal levels in 72 hours. The results suggest that the level of NGB might be related to infarct size and low-density lipoprotein at 24 hours after acute cerebral infarction. There were no significant differences in neurological impairment scores and infarct size at different periods following infarction. | 6 | Chlorophyll Content and Photosynthetic Characteristics of Pepper Inbred Lines显示文摘This study aimed to investigate chlorophyll content and photosynthetic characteristics of 17 pepper inbred lines which were selected independently. The results showed that chlorophyll contents significantly varied among different inbred lines. To be specific,chlorophyll content of No. 68 inbred line was significantly higher than that of other 14 inbred lines; chlorophyll content of No. 55 inbred line was significantly lower than that of other 15 inbred lines. | 7 | Experimental and numerical interpretation on composite foundation consisting of soil-cement column within warm and ice-rich frozen soil显示文摘Affected by climate warming and anthropogenic disturbances, the thermo-mechanical stability of warm and ice-rich frozen ground along the Qinghai-Tibet engineering corridor(QTEC) is continuously decreased, which may delay the construction of major projects in the future. In this study, based on chemical stabilization of warm and icerich frozen ground, the soil-cement column(SCC) for ground improvement was recommended to reinforce the foundations in warm and ice-rich permafrost regions. To explore the validity of countermeasures mentioned above, both the original foundation and the composite foundation consisting of SCC with soil temperature of -1.0℃ were prepared in the laboratory, and then the plate loading tests were carried out. The laboratory investigations indicated that the bearing capacity of composite foundation consisting of SCC was higher than that of original foundation, and the total deformation of original foundation was greater than that of composite foundation, meaning that overall stability of foundation with warm and ice-rich frozen soil can be improved by SCC installation. Meanwhile, a numerical model considering the interface interaction between frozen soil and SCC was established for interpretating the bearing mechanism of composite foundation. The numerical investigations revealed that the SCC within composite foundation was responsible for the more applied load, and the applied load can be delivered to deeper zone in depth due to the SCC installation, which was favorable for improving the bearing characteristic of composite foundation. | 8 | Safety and efficacy of Endovascular Management of high-grade blunt renal injury显示文摘Objectives:To provide data on the safety and efficacy of renal arterial embolization(RAE)in patients with highgrade blunt renal injury.Materials and methods:Fifteen patients with high-grade blunt renal injury(AAST grades IV-V)admitted to our hospital from July 2014 to December 2019 were retrospectively reviewed in this study.Their clinical success rate and complications were investigated accordingly.Results:Fifteen patients with high-grade blunt renal injury,13 men and 2 women with an average age of 41.6 years,including 11 hemodynamically unstable patients and 4 stable patients,were treated with RAE.Among these patients,73.3%(11 of 15)had grade IV,and 26.7%(4 of 15)had grade V injuries,while 53.3%(8 of 15)patients had concomitant injuries.One patient received main RAE and 14 patients received selective RAE.The clinical success rate after the first embolization was 93.3%(14 of 15).RAE was repeated and was successfully performed in one patient with sustained hematuria.No significant difference in creatinine levels was found before and after embolization.During the follow-up period of 2–82 months,two patients required tube drainage due to urine leaks,one patient developed renal failure requiring renal replacement therapy,and one patient developed secondary hypertension.Conclusions:RAE can provide a high success rate of hemostasis for both hemodynamically stable and unstable patients with high-grade blunt renal injury,and only minor complications are observed with this procedure. | Bin Wang Chongpei Wen Songlin Song Guilian Li Yanggang Yan Shoucai Cheng Junmei Zeng Zhidong Lin Yong Wang | 2022 | Journal of Interventional Medicine2022,5,1: | 0 |
