1A case-control study about the association between vascular endothelial growth inhibitor gene polymorphisms and breast cancer risk in female patients in Northeast China显示文摘Objective: The inhibition of the neovascularization in tumors is a potential therapeutic target of cancer. Vascular endothelial growth inhibitor(VEGI) is a member of the TNF superfamily which has the ability to suppress the formation of new vessels in tumors. In order to study the association between VEGI gene polymorphisms and breast cancer risk, a case-control study was conducted in Chinese Han women in Northeast China.Methods: Our study involved 708 female breast cancer patients and 685 healthy volunteers. Four SNPs of VEGI gene were analyzed through the polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) method. The association between VEGI gene polymorphisms and breast cancer risk was analyzed in our study. The relation between VEGI gene variants and clinical features of breast cancer including lymph node(LN) metastasis, estrogen receptor(ER), progestrogen receptor(PR), tumor protein 53(p53), human epidermal growth factor receptor 2(Her-2) and triple negative(ER-/PR-/Her-2-) status was analyzed as well.Results: We found that the CT genotype and T allele of rs6478106 were more frequent in patients than in controls. There was also a statistical difference in the distribution of C_(rs6478106)G_(rs4263839) haplotype between patients and controls. 2Dark matter direct search sensitivity of the PandaX-4T experiment显示文摘The Panda X-4T experiment, a 4-ton scale dark matter direct detection experiment, is being planned at the China Jinping Underground Laboratory. In this paper we present a simulation study of the expected background in this experiment. In a 2.8-ton fiducial mass and the signal region between 1-10 keV electron equivalent energy, the total electron recoil background is found to be 4.9 × 10^(-5) kg^(-1) d^(-1) keV^(-1). The nuclear recoil background in the same region is 2.8 × 10^(-7) kg^(-1) d^(-1) keV^(-1). 3Development of a Widely Applicable and Simple Prognostic Score for Patients with Acute-on-chronic Liver Failure显示文摘Background and Aims:Acute-on-chronic liver failure(ACLF)tends to progress rapidly with high short-term mortality.We aimed to create a widely applicable,simple prognostic(WASP)score for ACLF patients.Methods:A retrospective cohort of ACLF cases recruited from three centers in China were divided into training and validation sets to develop the new score.A prospective longitudinal cohort was recruited for further validation.Results:A total of 541 cases were included in the training set,and seven independent ACLF prognostic factors were screened to construct a new quantitative WASP-ACLF table.In the validation set of 671 cases,WASP-ACLF showed better predictive ability for 28-day and 90-day mortality than the currently used prognostic scores at baseline,day 3,week 1,and week 2.The predictive efficacy and clinical validity of the model improved over time.Patients were assigned to low-,intermediate-,and high-risk groups by their WASP-ACLF scores.Compared with the other two groups,intermediate-risk patients had a more uncertain prognosis,with a 90-day mortality of 44.4–50.6%.Sequential assessments at weeks 1 and 2 found the 90-day mortality of intermediate-risk groups was<20%forpatients with a≥2 point decrease in WASP-ACLF and was up to 56%for patients with a≥2 points increase.Similar results were observed in prospective data.Conclusions:The new ACLF prognostic score was simple,widely applicable,and had good predictive efficacy.Continuous assessments and trend of change in WASP-ACLF need to be considered,especially for intermediate-risk patients.Zhenjun Yu Yu Zhang Yuhan Li Feng Zhou Manman Xu Shaoli You Yu Chen Bing Zhu Ming Kong Fangjiao Song Shaojie Xin Zhongping Duan Tao Han China Network for Severe Liver Diseases 2022Journal of Clinical and Translational Hepatology2022,10,5:1
4Cytokinin plays a critical role in bitter gourd rootstock-induced thermotolerance of cucumber显示文摘Plants,as sessile in nature,are constantly confronted with diverse biotic and abiotic stresses throughout their life cycle in the changing environment.As a result,plants evolved root-shoot communications to optimize plant growth and development,and regulate responses to environmental stresses.Here,we examined the roles of root-sourced cytokinin(CTK)response to heat stress in grafted cucumber seedlings.Cucumber plants grafted onto cucumber roots and bitter gourd(Momordica charantia)roots were exposed to heat to examine their heat tolerance by assessing the levels of photosynthetic capacity,CTK contents,chlorophyll-a/b-binding protein(Lhcb2),ribulose-1,5-bisphosphate carboxylase/oxygenase(Rubisco)and its activating enzyme(RCA)content,and the enzyme activity of Rubisco.Bitter gourd rootstock enhanced cucumber scions heat stress tolerance.This enhancement was positively correlated with a higher content of CTK in both leaf and root parts,chlorophyll contents,and Rubisco abundance and activity.In addition,the higher level of CTK and Rubisco content in bitter gourd grafted plants shoots than in cucumber self-gafted plants shoots were attributed to an increase in CTK transport from roots in grafted plants under hightemperature conditions.These results indicated that CTK transfer from bitter gourd rootstock to scion and triggered the accumulation of Rubisco in leaf,thus improving the heat resistance of bitter gourd-grafted plants.Shaoli Han Sheng Shu Yu Wang Mohammad Shah Jahan Jin Sun Shirong Guo 2022Vegetable Research2022,2,1:1
5Performance of^(18)F‑DCFPyL PET/CT in Primary Prostate Cancer Diagnosis,Gleason Grading and D'Amico Classifcation:A Radiomics‑Based Study显示文摘This study aimed to investigate the performance of^(18)F-DCFPyL positron emission tomography/computerized tomography(PET/CT)models for predicting benign-vs-malignancy,high pathological grade(Gleason score>7),and clinical D'Amico classifcation with machine learning.The study included 138 patients with treatment-naïve prostate cancer presenting positive^(18)F-DCFPyL scans.The primary lesions were delineated on PET images,followed by the extraction of tumor-to-backgroundbased general and higher-order textural features by applying fve diferent binning approaches.Three layer-machine learning approaches were used to identify relevant in vivo features and patient characteristics and their relative weights for predicting high-risk malignant disease.The weighted features were integrated and implemented to establish individual predictive models for malignancy(Mm),high path-risk lesions(by Gleason score)(Mgs),and high clinical risk disease(by amico)(Mamico).The established models were validated in a Monte Carlo cross-validation scheme.In patients with all primary prostate cancer,the highest areas under the curve for our models were calculated.The performance of established models as revealed by the Monte Carlo cross-validation presenting as the area under the receiver operator characteristic curve(AUC):0.97 for Mm,AUC:0.73 for Mgs,AUC:0.82 for Mamico.Our study demonstrated the clinical potential of^(18)F-DCFPyL PET/CT radiomics in distinguishing malignant from benign prostate tumors,and high-risk tumors,without biopsy sampling.And in vivo^(18)F-DCFPyL PET/CT can be considered a noninvasive tool for virtual biopsy for personalized treatment management.Yuekai Li Fengcai Li Shaoli Han Jing Ning Peng Su Jianfeng Liu Lili Qu Shuai Huang Shiwei Wang Xin Li Xiang Li 2023Phenomics2023,3,6:0

7Redesigning transcription factor Cre1 for alleviating carbon catabolite repression in Trichoderma reesei显示文摘Carbon catabolite repression(CCR),which is mainly mediated by Cre1 and triggered by glucose,leads to a decrease in cellulase production in Trichoderma reesei.Many studies have focused on modifying Cre1 for alleviating CCR.Based on the homologous alignment of CreA from wild-type Penicillium oxalicum 114–2(Po-0)and cellulase hyperproducer JUA10-1(Po-1),we constructed a C-terminus substitution strain—Po-2—with decreased transcriptional levels of cellulase and enhanced CCR.Results revealed that the C-terminal domain of CreAPo−1 plays an important role in alleviating CCR.Furthermore,we replaced the C-terminus of Cre1 with that of CreAPo−1 in T.reesei(Tr-0)and generated Tr-1.As a control,the C-terminus of Cre1 was truncated and Tr-2 was generated.The transcriptional profiles of these transformants revealed that the C-terminal chimera greatly improves cellulase transcription in the presence of glucose and thus upregulates cellulase in the presence of glucose and weakens CCR,consistent with truncating the C-terminus of Cre1 in Tr-0.Therefore,we propose constructing a C-terminal chimera as a new strategy to improve cellulase production and alleviate CCR in the presence of glucose.Lijuan Han Kuimei Liu Wei Ma Yi Jiang Shaoli Hou Yinshuang Tan Quanquan Yuan Kangle Niu Xu Fang 2020Synthetic and Systems Biotechnology2020,5,3:0
