1The intracellular mechanism of alpha-fetoprotein promoting the proliferation of NIH 3T3 cells显示文摘AIM The existence and properties of alpha-fetoprotein (AFP) receptor on the surface of NIH 3T3 cells and the effects of AFP on cellular signal transduction pathway were investigated. METHODS The effect of AFP on the proliferation of NIH 3T3 cells was measured by incorporation of 3H-TdR. Receptor-binding assay of 125I-AFP was performed to detect the properties of AFP receptor in NIH 3T3 cells. The influences of AFP on the [cAMP]i and the activities of protein kinase A (PKA) were determined. Western blot was used to detect the change of K-ras P21 protein expression. RESULTS The proliferation of NIH 3T3 cells treated with 0-80 mg/L of AFP was significantly enhanced. The Scatchard analysis indicated that there were two classes of binding sites with KD of 2.722×10-9M (Bmax=12810 sites per cell) and 8.931× 10-SM (Bmax=l19700 sites per cell) respectively. In the presence of AFP (20 mg/L), the content of cAMP and activities of PKA were significantly elevated . The level of K-ras P21 protein was upregulated by AFP at the concentration of 20 mg/L. The monoclonal antibody against AFP could reverse the effects of AFP on the cAMP content, PKA activity and the expression of K-ras p21 gene. 3CRISPR/Cas9-mediated PINK1 deletion leads to neurodegeneration in rhesus monkeys显示文摘Dear Editor,PINK1 mutations cause autosomal recessive and early-onset Parkinson’s disease(PD)with selective neurodegeneration.Unfortunately,current PINK1 knockout(KO)mouse1,2 and pig models3,4 are unable to recapitulate the selective and overt neurodegeneration seen in PD patients.Furthermore,endogenous Pink1 in the mouse brain is expressed at very low levels and can only be detected via immunoprecipitation,5 meaning that PINK1’s function in the mammalian brain needs to be assessed using larger animals that are closer to humans.We previously used CRISPR/Cas9 to target the monkey gene in one-cell stage embryos.6 Using the same approach,we designed two gRNAs to target exon 2(T1)and exon 4(T2),which encode a kinase domain in the PINK1 gene of rhesus monkeys(Fig.1a).CRISPR/Cas9 and PINK1 gRNAs were injected into one-cell stage rhesus monkey embryos.A T7E1 assay and sequencing of PCR products from the injected embryos showed high efficiency(61.5%)in targeting PINK1(Fig.1b and Supplementary information,Fig.S1).Transfer of 87 embryos to 28 surrogate rhesus monkeys resulted in 11 pregnancies(39.2%)(Fig.1b).Eleven fetuses developed to term and were born naturally.Of these live monkeys,eight carried PINK1 mutations(M),and three were wild type(WT).However,three mutant monkeys(M1,M3 and M4)were newborn triplets that struggled to survive and died 3-4 days after birth.One WT newborn monkey also died after a difficult labor.Another mutant monkey(M2)died 7 days after birth without noticeable warning signs or symptoms.The other three mutant monkeys(M6,M7 and M8)have lived for three years;M5,however,reduced its food intake and showed weakness at the age of 1.5 years,and died 30 days after anesthesia for MRI examination.Weili Yang Yunbo Liu Zhuchi Tu Chong Xiao Sen Yan Xishan Ma Xiangyu Guo Xiusheng Chen Peng Yin Zhengyi Yang Su Yang Tianzi Jiang Shihua Li Chuan Qin Xiao-Jiang Li 2019Cell Research2019,29,4:12
4Integration of USEPA WASP model in a GIS platform显示文摘The integration of water quality analysis simulation program (WASP) with a geographical information system (GIS) is presented.This integration was undertaken to enhance the data analysis and management ability of the widely used water quality model.Different types of data involved in WASP modeling were converted and integrated into GIS using a database method.The spatial data modeling and analysis capability of GIS were used in the operation of the model.The WASP water quality model was coupled with the environmental fluid dynamics code (EFDC) hydrodynamic model.A case study of the Lower Charles River Basin (Massachusetts,USA) water quality model system was conducted to demonstrate the integration process.The results showed that high efficiency of the data process and powerful function of data analysis could be achieved in the integrated model,which would significantly improve the application of WASP model in water quality management.Sen PENG George Yu-zhu FU Xin-hua ZHAO 2010Journal of Zhejiang University-Science A(Applied Physics & Engineering)2010,11,12:8
5Effects of insoluble dietary fiber from wheat bran on noodle quality显示文摘Benefiting from its favorable effects in promoting intestinal peristalsis,increasing satiety and reducing postprandial blood sugar,wheat bran insoluble dietary fiber(IDF)could be added to noodles to improve the nutrition value of noodle products.However,the addition of IDF usually results in an adverse effect on the quality characteristics of fiber fortified products,leading to unfavorable texture and consumption drop.It was found that IDF could weaken dough characteristics,thereby causing the quality deterioration of noodles,but the mechanism is unclear as yet.This study aimed to investigate the effects of different amounts of IDF on the texture quality and cooking quality of noodles.The water distribution,rheological properties,pasting properties and cooking properties of the noodles were determined by low-field nuclear magnetic resonance(LF-NMR),dynamic rheometer,and rapid visco-analyzer.The results of the LF-NMR showed that the increased IDF in the noodles resulted in a contraction of relaxation time and an increase in the proportion of loosely bound water.At a high amount of IDF,the water absorption and tensile distance of the noodles significantly decreased and the cooking loss rate increased,with a downward trend in peak viscosity,final viscosity and setback values.Moreover,IDF could improve the storage modulus and loss modulus of sheeted dough and enhance the texture of noodles.Furthermore,the correlations between IDF addition amounts and rheological properties as well as cooking properties were analyzed,and the results confirmed that noodles with the IDF amount of 2%–4%had higher nutrition and quality.This study provides the basis for future development and improvement of IDF-enriched health foods.Mengxu Lei Jihong Huang Xiaoling Tian Peng Zhou Qi Zhu Limin Li Li Li Sen Ma Xiaoxi Wang 2021Grain & Oil Science and Technology2021,4,1:8
6Current applications and future prospects of nanotechnology in cancer immunotherapy显示文摘Cancer immunotherapy is an artificial stimulation of the immune system to recognize cancer cells and activate specific immune cells to target and attack cancer cells.In clinical trials, immunotherapy has recently shown impressive results in the treatment of multiple cancers.Thus, cancer immunotherapy has gained a lot of attention for its unique advantages and promising future.With extensive research on cancer immunotherapy, its safety and effectiveness has gradually been revealed.However, it is still a huge challenge to expand and drive this therapy while maintaining low toxicity, high specificity, and long-lasting efficacy.As a unique technology, nanotechnology has been applied in many fields, the advantages of which will promote the development of cancer immunotherapies.Researchers have tried to apply nanomaterials to cancer immunotherapy due to their advantageous properties,such as large specific surface areas, effective drug delivery, and controlled surface chemistry, to improve treatment efficacy.Here,we briefly introduce the current applications of nanomaterials in cancer immunotherapy, including adoptive cell therapy(ACT),therapeutic cancer vaccines, and monoclonal antibodies, and throw light on future directions of nanotechnology-based cancer immunotherapy.Sen Yan Peng Zhao Tingting Yu Ning Gu 2019Cancer Biology & Medicine2019,16,3:6
7Association of colorectal cancer susceptibility variants with esophageal cancer in a Chinese population显示文摘AIM: To investigate the association between colorectal cancer(CRC) genetic susceptibility variants and esophageal cancer in a Chinese Han population.METHODS: A case-control study was conducted including 360 esophageal cancer patients and310 healthy controls. Thirty-one single-nucleotide polymorphisms(SNPs) associated with CRC risk from previous genome-wide association studies were analyzed. SNPs were genotyped using Sequenom Mass-ARRAY technology, and genotypic frequencies in controls were tested for departure from HardyWeinberg equilibrium using a Fisher's exact test. The allelic frequencies were compared between cases and controls using a χ 2 test. Associations between the SNPs and the risk of esophageal cancer were tested using various genetic models(codominant, dominant,recessive, overdominant, and additive). ORs and95%CIs were calculated by unconditional logistic regression with adjustments for age and sex.RESULTS: The minor alleles of rs1321311 and rs4444235 were associated with a 1.53-fold(95%CI:1.15-2.06; P = 0.004) and 1.28-fold(95%CI: 1.03-1.60;P = 0.028) increased risk of esophageal cancer in the allelic model analysis, respectively. In the genetic model analysis, the C/C genotype of rs3802842 was associated with a reduced risk of esophageal cancer in the codominant model(OR = 0.52, 95%CI:0.31-0.88; P = 0.033) and recessive model(OR =0.55, 95%CI: 0.34-0.87; P = 0.010). The rs4939827C/T-T/T genotype was associated with a 0.67-fold(95%CI: 0.46-0.98; P = 0.038) decreased esophageal cancer risk under the dominant model. In addition,rs6687758, rs1321311, and rs4444235 were associated with an increased risk. 8Novel magnetic silk fibroin scaffolds with delayed degradation for potential long-distance vascular repair显示文摘Although with the good biological properties,silk fibroin(SF)is immensely restrained in long-distance vascular defect repair due to its relatively fast degradation and inferior mechanical properties.It is necessary to construct a multifunctional composite scaffold based on SF.In this study,a novel magnetic SF scaffold(MSFCs)was prepared by an improved infiltration method.Compared with SF scaffold(SFC),MSFCs were found to have better crystallinity,magnetocaloric properties,and mechanical strength,which was ascribed to the rational introduction of iron-based magnetic nanoparticles(MNPs).Moreover,in vivo and in vitro experiments demonstrated that the degradation of MSFCs was significantly extended.The mechanism of delayed degradation was correlated with the dual effect that was the newly formed hydrogen bonds between SFC and MNPs and the complexing to tyrosine(Try)to inhibit hydrolase by internal iron atoms.Besides,theβ-crystallization of protein in MSFCs was increased with the rise of iron concentration,proving the beneficial effect after MNPS doped.Furthermore,although macrophages could phagocytose the released MNPs,it did not affect their function,and even a reasonable level might cause some cytokines to be upregulated.Finally,in vitro and in vivo studies demonstrated that MSFCs showed excellent biocompatibility and the growth promotion effect on CD34-labeled vascular endothelial cells(VECs).In conclusion,we confirm that the doping of MNPs can significantly reduce the degradation of SFC and thus provide an innovative perspective of multifunctional biocomposites for tissue engineering.Xin Liu Yuxiang Sun Bo Chen Yan Li Peng Zhu Peng Wang Sen Yan Yao Li Fang Yang Ning Gu 2022Bioactive Materials2022,7,1:3

10Heavy soil drying during mid-to-late grain filling stage of the main crop to reduce yield loss of the ratoon crop in a mechanized rice ratooning system显示文摘Yield loss(Y_(Loss)) in the ratoon crop due to crushing damage to left stubble from mechanical harvesting of the main crop is a constraint for wide adoption of mechanized rice ratooning technology.Soil drying before the harvest of the main crop has been proposed to overcome this problem.The objective of this study was to determine the effect of soil drying during the mid-to-late grain filling stage of the main crop on grain yield of the ratoon crop in a mechanized rice ratooning system.Field experiments were conducted to compare Y_(Loss) between light(LD) and heavy(HD) soil drying treatments in Hubei province,central China in 2017 and 2018.Y_(Loss) was calculated as the percentage of yield reduction in the ratoon crop with the main crop harvested mechanically,relative to the grain yield of the ratoon crop with the main crop harvested manually.In comparison with LD,soil hardness was increased by 42.8%-84.7% in HD at the 5-20 cm soil depth at maturity of the main crop.Soil hardness at 5 and 10 cm depths reached respectively 4.05 and 7.07 kg cm^(-2) in HD.Soil drying treatment did not significantly affect the grain yield of the main crop.Under mechanical harvesting of the main crop,HD increased the grain yield of the ratoon crop by 9.4% relative to LD.Consequently,Y_(Loss) was only 3.4% in HD,in contrast to 16.3% in LD.The differences in grain yield and Y_(Loos) between the two soil drying treatments were explained mainly by panicles m^(-2),which was increased significantly by HD in the track zone of the ratoon crop compared with LD.These results suggest that heavy soil drying practice during the mid-to-late grain filling stage of the main crop is effective for reducing Y_(Loss) of the ratoon crop in a mechanized rice ratooning system.Chang Zheng Yuechao Wang Shen Yuan Sen Xiao Yating Sun Jianliang Huang Shaobing Peng 2022The Crop Journal2022,10,1:3

12Development of a panel of unigene-derived polymorphic EST–SSR markers in lentil using public database information显示文摘Lentil(Lens culinaris Medik.), a diploid(2n = 14) with a genome size greater than 4000 Mbp, is an important cool season food legume grown worldwide. The availability of genomic resources is limited in this crop species. The objective of this study was to develop polymorphic markers in lentil using publicly available curated expressed sequence tag information(ESTs). In this study, 9513 ESTs were downloaded from the National Center for Biotechnology Information(NCBI) database to develop unigene-based simple sequence repeat(SSR) markers. The ESTs were assembled into 4053 unigenes and then analyzed to identify 374 SSRs using the MISA microsatellite identification tool. Among the 374 SSRs, 26 compound SSRs were observed.Primer pairs for these SSRs were designed using Primer3 version 1.14. To classify the functional annotation of ESTs and EST–SSRs, BLASTx searches(using E-value 1 × 10-5) against the public UniP rot(http://gffzzc0d2db89b939488bhkwnc0ufqf6vf6nku.ffgz.tsg.suse.edu.cn/) and NCBI(http://gffzz5b533c64939a43b3hkwnc0ufqf6vf6nku.ffgz.tsg.suse.edu.cn/) databases were performed. Further functional annotation was performed using PLAZA(version3.0) comparative genomics and GO annotation was summarized using the Plant GO slim category. Among the synthesized 312 primers, 219 successfully amplified Lens DNA. A diverse panel of 24 Lens genotypes was used to identify polymorphic markers. A polymorphic set of 57 markers successfully discriminated the test genotypes. 13Energy measurement system of a large-aperture high power laser experiment platform显示文摘An energy measurement system in a large-aperture high power laser experiment platform is introduced. The entire measurement system includes five calorimeters, which carry out the energy measurement of the fundamental frequency before the frequency conversion unit, remaining fundamental frequency, remaining second-harmonics, third-harmonics,as well as the energy balance measurement after the frequency conversion unit. Combinational indirect calibration and direct calibration are employed to calibrate the sampling coefficients of the calorimeters. The analysis of the data showed that, regarding the energy balance coefficients, combinational calibration approach gives a higher precision, and leads to an energy balance with 1%; and regarding the energy sampling coefficients for the various wavelengths after the frequency conversion, the results from direct and combinational calibration are consistent. 19Villin Family Members Associated with Multiple Stress Responses in Cotton显示文摘Villin(VLN)is considered to be one of the most important actin-binding proteins,participates in modulating the actin cytoskeleton dynamics,plays essential role in plant development and resisting adverse environments.However,systematic studies of the VLN gene family have not been reported in cotton(Gossypium).In this study,14 GhVLNs were identified in G.hirsutum.These GhVLN genes were distributed in 6 A-subgenome chromosomes and 6 D-subgenome chromosomes of the allotetraploid upland cotton and classified into three phylogenetical groups based on the classification model of AtVLNs.In addition,the 14 GhVLN genes have highly conserved gene structure and motif architecture.The number of introns was ranged from 18 to 22 and the length of protein sequences was varied from 901 to 1077.Six gelsolin homology domains,G1–G6,and villin headpiece domain,VHP,were existed in all GhVLNs with the exception of two VLNs(GhVLN6 and GhVLN13)which lacked VHP.Cis-elements analysis revealed that the promoter regions of GhVLNs contained various light related components and also elements responsible for phytohormones and stresses response,indicating that,when subjected to those adverse environments,cotton plants may activate the response system by targeting VLN genes to survive the crisis.Heatmaps showed that the GhVLN genes exhibited various expression patterns,some were accumulated in certain tissues,root,petal,stamen or elongating fibers,and some were obviously induced by environmental changes.Especially GhVLN3 and GhVLN10 were highly and preferentially expressed in elongating fibers and distinctly upregulated by abiotic(salt,PEG,cold and heat)and biotic(Verticillium dahliae V991)stresses.This study may provide useful information for biological function identification of GhVLN genes and gene resources for creating high-quality and various resistant cotton germplasms.Fenni Lv Sen Wang Ruiping Tian Peng Wang Kang Liu 2021Phyton-International Journal of Experimental Botany2021,90,6:1
