| 1 | Cholesterol transport through the peroxisome-ER membrane contacts tethered by PI(4,5)P2 and extended synaptotagmins显示文摘Most mammalian cells take up cholesterol from low-density lipoproteins(LDLs) via receptor-mediated endocytosis.After reaching lysosomes,LDL-derived cholesterol continues to transport to downstream organelles including the ER for specific structural and functional needs.Peroxisomes are recently found to receive cholesterol from lysosomes through lysosomeperoxisome membrane contacts.However,whether and how cholesterol is conveyed from peroxisomes to the ER remain unknown.Here,by combining high-resolution microscopic analyses and in vitro reconstitution of highly purified organelles or artificial liposomes,we demonstrate that peroxisomes form membrane contacts with the ER through the interaction between peroxisomal PI(4,5)P2 and ER-resident extended synaptotagmin-1,2 and 3(E-Syts).Depletion of peroxisomal PI(4,5)P2 or ESyts markedly decreases peroxisome-ER membrane contacts and induces cholesterol accumulation in lysosomes.Furthermore,we show that cholesterol is delivered from 3H-labeled peroxisomes or PI(4,5)P2-containing liposomes to the ER in vitro,and that the presence of peroxisomes augments cholesterol transfer from lysosomes to the ER.Together,our study reveals a new cholesterol transport pathway along the lysosome-peroxisome-ER membrane contacts in the cell. | Jian Xiao Jie Luo Ao Hu Ting Xiao Meixin Li Zekai Kong Luyi Jiang Zimu Zhou Yacheng Liao Chang Xie Beibei Chu Honghua Miao Boliang Li Xiongjie Shi Bao-Liang Song | 2019 | Science China(Life Sciences)2019,62,9: | 8 |
| 2 | Assessment and regulation of ocean health based on ecosystem services: Case study in the Laizhou Bay, China显示文摘The ecosystem-based management of nearshore waters requires integrated assessment of ocean health and scientific guidance on artificial regulations to promote sustainable development. Quantitative approaches were developed in this paper to assess present and near-term ocean health based on ecosystem services. Results of the case study in the Laizhou Bay of China showed that the index score of ocean health was 0.785 6 out of 1.0 at present and was expected to range from 0.555 1 to 0.804 1 in the near-term future depending on different intensities of artificial regulation of negative pressures. Specifically, the results of ocean health at present mainly indicated that cultural services and provisioning services performed essentially perfectly while supporting services and regulating services functioned less well. It can be concluded that this nearshore ecosystem would partially lose supporting and regulating services in the near-term future if the increasing pressures were not wellregulated but that all of these categories of ecosystem services could be slightly improved if the negative pressures were fully controlled. Additionally, it is recommended that publicity and education on ecosystem services especially on cultural services and regulating services should be further strengthened. | 3 | A model to assess fundamental and realized carrying capacities of island ecosystem:A case study in the southern Miaodao Archipelago of China显示文摘Research on carrying capacity, aiming at maintaining the limited ability of the Earth's life supporting system to sustain human development, requires a comprehensive and ecosystem-based approach to monitor and assess the localized sustainability of coupled social and ecological systems. A definition termed the ecosystem-based carrying capacity of island(EBCCI) was developed in this paper of which the indices of fundamental and realized carrying capacities of island(FCCI and RCCI) were highlighted to emphasize the inter-dependencies between social systems and ecological systems. In order to avoid the difficulties and uncertainties of direct assessment, the related assessment model was established on the basis of synthetic evaluation of inherent and external factors affecting the EBCCI. The southern Miaodao Archipelago(SMA) located in the intersection of the Bohai Sea and the Yellow Sea, China, was taken as a typical example to acquire integrated recognition of the island ecosystem and its carrying capacity so as to discuss its sustainable development. The index scores of FCCI and RCCI in the SMA were 0.818 5 and 0.712 9 out of 1.0, respectively, with acceptable uncertainties. The results showed a relatively well capacity to sustain progress and relatively well realization of the carrying capacity of island ecosystem, owing to a well capacity of ecologically regulating, general performance of both ecologically supporting and resource provisioning, and a relatively high level of social supporting system. The study implied that it was critical to optimize the inter-dependencies and to sustain the relative balance between social systems and ecological systems so as to improve the RCCI and further facilitate the sustainability of SMA. | 4 | Spatio-temporal variability of phytoplankton assemblages and its controlling factors in spring and summer in the Subei Shoal of Yellow Sea, China显示文摘The Subei Shoal is a special coastal area with complex physical oceanographic properties in the Yellow Sea.In the present study,the distribution of phytoplankton and its correlation with environmental factors were studied during spring and summer of 2012 in the Subei Shoal of the Yellow Sea.Phytoplankton species composition and abundance data were accomplished by Utermohl method.Diatoms represented the greatest cellular abundance during the study period.In spring,the phytoplankton cell abundance ranged from 1.59×10^3 to 269.78×10^3 cell/L with an average of 41.80×10^3 cell/L,and Skeletonema sp.and Paralia sulcata was the most dominant species.In summer,the average phytoplankton cell abundance was 72.59×10^3 cell/L with the range of 1.78×10^3 to 574.96×10^3 cell/L,and the main dominant species was Pseudo-nitzschia pungens,Skeletonema sp.,Dactyliosolen fragilissima and Chaetoceros curvisetus.The results of a redundancy analysis(RDA)showed that turbidity,temperature,salinity,pH,dissolved oxygen(DO),the ratio of dissolved inorganic nitrogen to silicate and SiO4-Si(DIN/SiO4-Si)were the most important environmental factors controlling phytoplankton assemblages in spring or summer in the Subei Shoal of the Yellow Sea. | Yuanzi Huo Honghua Shi Jianheng Zhang Qiao Liu Yuanliang Duan Qing He Kefeng Yu Hongsheng Bi Chunlei Fan Peimin He | 2019 | Acta Oceanologica Sinica2019,38,10: | 4 |
| 5 | Spatiotemporal heterogeneity of phytoplankton diversity and its relation to water environmental factors in the southern waters of Miaodao Archipelago,China显示文摘To study the water quality influenced by the anthropogenic activities and its impact on the phytoplankton diversity in the surface waters of Miaodao Archipelago, the spatiotemporal variations in phytoplankton communities and the environmental properties of the surface waters surrounding the Five Southern Islands of Miaodao Archipelago were investigated, based on seasonal field survey conducted from November 2012 to August 2013. During the survey, a total of 109 phytoplankton species from 3 groups were identified in the southern waters of Miaodao Archipelago, of which 77 were diatoms, 29 were dinoflagellates, and 3 were chrysophytes. Species number was higher in winter(73), moderate in autumn(70), but lower in summer(31) and spring(27). The species richness index in autumn(5.92) and winter(4.28) was higher than that in summer(2.83) and spring(1.41).The Shannon-Wiener diversity index was high in autumn(2.82), followed by winter(1.99) and summer(1.92), and low in spring(0.07). The species evenness index in autumn(0.46) and summer(0.39) was higher than that in winter(0.32) and spring(0.02). On the basis of principal component analysis(PCA) and redundancy analysis(RDA), we found that dissolved inorganic nitrogen(DIN) and chemical oxygen demand(COD) in spring, COD in summer, p H in autumn, and salinity and oil pollutant in winter, respectively, showed the strongest association with the distribution of phytoplankton diversity. | 7 | Sensitivity and Uncertainty Analysis of Regional Marine Ecosystem Services Value显示文摘Marine ecosystem services are the benefits which people obtain from the marine ecosystem,including provisioning services,regulating services,cultural services and supporting services. The human species,while buffered against environmental changes by culture and technology,is fundamentally dependent on the flow of ecosystem services. Marine ecosystem services become increasingly valuable as the terrestrial resources become scarce. The value of marine ecosystem services is the monetary flow of ecosystem services on specific temporal and spatial scales,which often changes due to the variation of the goods prices,yields and the status of marine exploitation. Sensitivity analysis is to study the relationship between the value of marine ecosystem services and the main factors which affect it. Uncertainty analysis based on varying prices,yields and status of marine exploitation was carried out. Through uncertainty analysis,a more credible value range instead of a fixed value of marine ecosystem services was obtained in this study. | 12 | Interaction between neutrophil extracellular traps and cardiomyocytes contributes to atrial fibrillation progression显示文摘Atrial fibrillation(AF)is a frequent arrhythmia associated with cardiovascular morbidity and mortality.Neutrophil extracellular traps(NETs)are DNA fragments with cytoplasm proteins released from neutrophils,which are involved in various cardiovascular diseases.To elucidate the role of NETs in AF,we investigated the effect of NETs on AF progression and the secretion of NETs in AF.Results showed that:NETs induced the autophagic apoptosis of cardiomyocytes,and NETs also led to mitochondrial injury by promoting mitochondrial depolarization and ROS production.Ongoing tachy-pacing led to the structural loss of cardiomyocytes and provided potent stimuli to induce NETs secretion from neutrophils.In the meanwhile,increased Ang II in AF facilitated NETs formation through the upregulation of AKT phosphorylation,while it could not directly initiate NETosis as the autophagy was not induced.In vivo,DNase I was administrated to abrogate NETs formation,and AF-related fibrosis was ameliorated as expected.Correspondingly,the duration of the induced AF was reduced.Our study addresses the formation mechanism of NETs in AF and demonstrates the lethal effects of NETs on cardiomyocytes through the induction of mitochondrial injury and autophagic cell death,which comprehensively describes the positive feedback comprised of NETs and stimuli secreted by cardiomyocytes that sustains the progression of AF and AF related fibrosis. | Li He Ruiqi Liu Honghua Yue Xiaoxin Zhang Xiaohui Pan Yutao Sun Jun Shi Guonian Zhu Chaoyi Qin Yingqiang Guo | 2023 | Signal Transduction and Targeted Therapy2023,8,8: | 1 |

| 14 | The Inhibitory Effects of PSS-Loaded Nanoparticles on the Dysfunction of Cardiac Microvascular Endothelia in Rats with Diabetic Cardiomyopathy显示文摘Propylene glycol alginate sodium sulfate-loaded nanoparticles(PSS-NP) has been shown potential to prevent the microvascular endothelial injuries caused by diabetic cardiomyopathy(DCM). In this study, we aimed to investigate the effects of PSS-NP on the dysfunction of cardiac microvascular endothelia in streptozotocin(STZ)-induced DCM rat model. Echocardiographic measurements showed a significant improvement of cardiac function in the PSS-NP-treated group. Our results revealed that the abnormalities of cardiac systolic and diastolic functions were suppressed by the treatments of prostaglandin E1(PGE1), low molecular weight heparin(LMWH), PSS and PSS-NP. Our comparison analysis indicated that PSS-NP showed the strongest inhibitory effects on microvascular endothelial injuries. Transmission electron microscopy analysis demonstrated that PSS-NP protected the cardiac microvascular endothelium and further improved endothelium dysfunction in DCM rats. ELISA and Western blot assays further showed a high efficiency of improving cardiac microvascular endothelial dysfunction with PSS-NP. Our results demonstrated that PSS-NP increased the protein expression of phosphatidylinositol 3-kinase(PI3K)-p85 and vascular endothelial growth factor(VEGF)-A, and the phosphorylation of protein kinase B(Akt) and endothelial nitric oxide synthase(eNOS), which were involved in the amelioration of cardiac microvascular endothelial dysfunction. | 17 | Degeneration of foundation cushion species induced by ecological constraints can cause massive changes in alpine plant communities显示文摘Foundational cushion plants can re-organize community structures and sustain a prominent proportion of alpine biodiversity,but they are sensitive to climate change.The loss of cushion species can have broad consequences for associated biota.The potential plant community changes with the population dynamics of cushion plants remain,however,unclear.Using eight plant communities along a climatic and community successional gradient,we assessed cushion population dynamics,the underlying ecological constraints and hence associated plant community changes in alpine communities dominated by the foundational cushion plant Arenaria polytrichoides.The population dynamics of Arenaria are attributed to ecological constraints at a series of life history stages.Reproductive functions are constrained by increasing associated beneficiary plants;subsequent seedling establishment is constrained by temperature,water and light availability,extreme climate events,and interspecific competition;strong competitive exclusion may accelerate mortality and degeneration of cushion populations.Along with cushion dynamics,species composition,abundance and community structure gradually change.Once cushion plants completely degenerate,previously cushion-dominated communities shift to relatively stable communities that are overwhelmingly dominated by sedges.Climate warming may accelerate the degeneration process of A.polytrichoides.Degeneration of this foundational cushion plant will possibly induce massive changes in alpine plant communities and hence ecosystem functions in alpine ecosystems.The assessment of the population dynamics of foundation species is critical for an effective conservation of alpine biodiversity. | Jianguo Chen Xufang Chen Lishen Qian Yazhou Zhang Bo Li Honghua Shi Lu Sun Christian Schöb Hang Sun | 2024 | Science China(Life Sciences)2024,67,4: | 0 |
| 18 | Research on stability and Hopf bifurcation of marine ecosystem dynamics models显示文摘The predictability of marine ecosystem dynamics models is one of the most vital factors to limit their practical applications, of which the stability is the fundamental condition. In order to discuss the stability and Hopf bifurcation of marine ecosystem dynamics models, an approach based on a theorem termed dimension reduction was proposed and further applied in the mass-conservative nutrient-phytoplankton-zooplankton-detritus(NPZD) model in this paper. Results showed that the nonsingular equilibrium point of NPZD model was analytically stable in use of the dimension reduction theorem and the Hopf bifurcation might occur when model parameters changed along the threshold values. The analytical results of the NPZD model were further verified by numerical simulation in this study.
