Intragenic and intergenic sequences regulating the expression of the 5'-to-5' linked adult α-and β-globin genes from large yellow croaker Pseudosciaena crocea显示文摘One adult α-globin gene and one β-globin gene have been cloned from the large yellow croaker Pseudosciaena crocea. Linkage analysis indicated that the α-and β-globin genes were oriented head-to-head relative to each other. To identify the regulatory elements present in the intergenic and intragenic regions of the globin complex, the intergenic region alone or together with the β-globin gene first intron was cloned into the luciferase-reporter vector pGL3-Basic respectively, and the chimeric constructs were transiently transfected into Vero cells and primary fish erythrocytes. The intergenic region cannot support the high-level expression of luciferase. However, the promoter activity of the intergenic region was strongly stimulated by the positive regulatory elements (PRE) located in the β-globin gene intron 1. Thus,it is proposed that the intergenic promoters and intragenic PRE were necessary for the effective expression of the linked α-and β-globin genes. | CHU Wuying YU Lian QIAN Ronghua MENG Tao ZHOU Ruixue FU Guihong CHEN Jia ZHANG Jianshe | 2008 | Acta Oceanologica Sinica2008,27,5: | 1
Atomically dispersed Fe sites on hierarchically porous carbon nanoplates for oxygen reduction reaction显示文摘Developing cost-effective,robust and stable non-precious metal catalysts for oxygen reduction reaction(ORR) is of paramount importance for electrochemical energy conversion devices such as fuel cells and metal-air batteries.Although Fe-N-C single atom catalysts(SACs) have been hailed as the most promising candidate due to the optimal binding strength of ORR intermediates on the Fe-N_(4) sites, they suffer from serious mass transport limitations as microporous templates/substrates,i.e.,zeolitic imidazolate frameworks(ZIFs),are usually employed to host the active sites.Motivated by this challenge,we herein develop a hydrogen-bonded organic framework(HOF)-assisted pyrolysis strategy to construct hierarchical micro/mesoporous carbon nanoplates for the deposition of atomically dispersed Fe-N_(4) sites.Such a design is accomplished by employing HOF nanoplates assembled from 2-aminoterephthalic acid(NH_(2)-BDC) and p-phenylenediamine(PDA) as both soft templates and C,N precursors.Benefitting from the structural merits inherited from HOF templates, the optimized catalyst(denoted as Fe-N-C SAC-950) displays outstanding ORR activity with a high half-wave potential of 0.895 V(vs.reversible hydrogen electrode(RHE)) and a small overpotential of 356 mV at 10 mA cm^(-2) for the oxygen evolution reaction(OER).More excitingly, its application potential is further verified by delivering superb rechargeability and cycling stability with a nearly unfading charge-discharge gap of 0.72 V after 160 h.Molecular dynamics(MD) simulations reveal that micro/mesoporous structure is conducive to the rapid mass transfer of O_(2),thus enhancing the ORR performance. In situ Raman results further indicate that the conversion of O_(2) to~*O_(2)-the rate-determining step(RDS) for Fe-N-C SAC-950.This work will provide a versatile strategy to construct single atom catalysts with desirable catalytic properties. | Ruixue Zheng Qinglei Meng Hao Zhang Teng Li Di Yang Li Zhang Xiaolong Jia Changpeng Liu Jianbing Zhu Xiaozheng Duan Meiling Xiao Wei Xing | 2024 | Journal of Energy Chemistry2024,90,3: | 0
Classification of ice and water in the Arctic using radar altimeter and microwave radiometer data from HY-2B satellite显示文摘Several Chinese marine satellites have been launched in recent years.Monitoring sea ice and the ocean in the Arctic is of great importance for climate research.Sea ice in the Arctic has changed rapidly during the past few decades with respect to the extent and thickness. In this study,we applied combined passive and active microwave data from the Chinese HaiYang-2B(HY-2B)satellite to classify ice and sea water in the Arctic. We use data from a radar altimeter(RA)and a calibration microwave radiometer(CMR)to discriminate between ice and water by applying several approaches(1)the single parameter threshold criteria,(2)the multi-parameters linear segmentations and(3)the K-means clustering. The results yielded by these methods were in good agreement(classification accuracy>95%)with the Satellite Application Facility on Ocean and Sea Ice products between November and April. For other months(May–October),however, the agreement was less good(lowest classification accuracy approximate 85%in summer).A hybrid approach combined with graphical ice edges detection and microwave radar waveform analysis is therefore developed.A visual comparison with SAR images suggested the hybrid approach results greatly improved the ice and water discrimination in summer. This study demonstrated that multi-sensors(RA and CMR)configurations from HY satellites can offer comparable polar earth observation to the European Space Agency and NOAA satellite products. | Chengfei Jiang Mingsen Lin Ruixue Cao Hao Wei Lijian Shi Bin Cheng Yongjun Jia Qimao Wang | 2023 | Acta Oceanologica Sinica2023,42,5: | 0
Metabolic profiles and morphological characteristics of leaf tips among different sweet potato(Ipomoea batatas Lam.)varieties显示文摘Sweet potato leaf tips have high nutritional value, and exploring the differences in the metabolic profiles of leaf tips among different sweet potato varieties can provide information to improve their qualities. In this study,a UPLC-Q-Exactive Orbitrap/MS-based untargeted metabolomics method was used to evaluate the metabolites in leaf tips of 32 sweet potato varieties.Three varieties with distinct overall metabolic profiles(A01,A02,and A03),two varieties with distinct profiles of phenolic acids(A20 and A18),and three varieties with distinct profiles of flavonoids(A05,A12,and A16)were identified. In addition,a total of 163 and 29 differentially expressed metabolites correlated with the color and leaf shape of sweet potato leaf tips, respectively, were identified through morphological characterization.Group comparison analysis of the phenotypic traits and a metabolite-phenotypic trait correlation analysis indicated that the color differences of sweet potato leaf tips were markedly associated with flavonoids. Also, the level of polyphenols was correlated with the leaf shape of sweet potato leaf tips, with lobed leaf types having higher levels of polyphenols than the entire leaf types. The findings on the metabolic profiles and differentially expressed metabolites associated with the morphology of sweet potato leaf tips can provide useful information for breeding sweet potato varieties with higher nutritional value. | Wenqing Tan Xinbo Guo Zhangying Wang Rong Zhang Chaochen Tang Bingzhi Jiang Ruixue Jia Yuanyuan Deng Shaohai Yang Jingyi Chen | 2024 | Journal of Integrative Agriculture2024,23,2: | 0
Preparation of Cerium Dioxide Functionalized Magnetic Layered Double Hydroxides for High-efficiency Phosphopeptide Enrichment显示文摘In this work,we prepared a material with magnetic nanoparticles(Fe_(3)O_(4))as core, layered double hydroxides(LDHs)as affinity shell, and cerium dioxide(CeO_(2))as functional molecules(denoted as Fe_(3)O_(4)@LDH-CeO_(2)).On the basis of combined immobilized metal ion affinity chromatography(IMAC)and metal oxide affinity chromatography(MOAC),Fe_(3)O_(4)@LDH-CeO_(2) was used to enrich phosphopeptides with high efficiency. The material exhibited high selectivity(α-casein:β-casein:BSA=1:1:5000,mass ratio),high recovery(95.87%),and good reusability of 10 times adsorption-desorption experiments. The feasibility of Fe_(3)O_(4)@LDH-CeO_(2) was further investigated by extracting phosphopeptides from biological samples(nonfat milk, serum, saliva, and A549 cell lysate). | JIANG Dandan QI Ruixue LYU Siqi WANG Wei LIU Jinghai JIA Qiong | 2023 | Chemical Research in Chinese Universities2023,39,2: | 0
Recent Advances in Stability Improvement Strategies of M-N_(x)/C Catalysts Towards Oxygen Reduction Reaction显示文摘Although fuel cells possess advantages of high energy conversion efficiency and zero-carbon emission, their large-scale commercialization is restricted by expensive and scarce platinum(Pt)catalysts.Metal-nitrogen-carbon(M-Nx/C)catalysts are hailed as the most promising candidates to replace Pt due to their considerable oxygen reduction reaction(ORR)activity and low cost.Despite tremendous progress in terms of active site identification and activity improvement being achieved in the past few decades, the M-Nx/C catalysts still suffer from insufficient durability, which drastically limits their practical application. In this regard, understanding degradation mechanisms and customizing stabilization strategies are of significant importance yet challengeable. In this review,we summarize the recent advances in the stability improvement of M-Nx/C catalysts. The stability test protocols of the M-Nx/C are firstly introduced.Subsequently, with the combination of advanced ex situ and in situ characterization techniques and density functional theory calculation,we present a comprehensive overview of the main degradation mechanisms during ORR process.Aiming at these deactivation issues,a variety of novel improvement strategies are developed to enhance the stability of M-Nx/C.Finally,the current challenges and prospects to design highly stable M-Nx/C catalysts are also proposed. | Xiaolong Jia Qinglei Meng Ruixue Zheng Xiaohui Liu Yaling Zhao Changpeng Liu Meiling Xiao Wei Xing | 2023 | Renewables2023,1,6: | 0
