1NLRP3 inflammasome activation and cell death显示文摘The NLRP3 inflammasome is a cytosolic multiprotein complex composed of the innate immune receptor protein NLRP3,adapter protein ASC,and inflammatory protease caspase-1 that responds to microbial infection,endogenous danger signals,and environmental stimuli.The assembled NLRP3 inflammasome can activate the protease caspase‐1 to induce gasdermin D-dependent pyroptosis and facilitate the release of IL-1β and IL-18,which contribute to innate immune defense and homeostatic maintenance.However,aberrant activation of the NLRP3 inflammasome is associated with the pathogenesis of various inflammatory diseases,such as diabetes,cancer,and Alzheimer’s disease.Recent studies have revealed that NLRP3 inflammasome activation contributes to not only pyroptosis but also other types of cell death,including apoptosis,necroptosis,and ferroptosis.In addition,various effectors of cell death have been reported to regulate NLRP3 inflammasome activation,suggesting that cell death is closely related to NLRP3 inflammasome activation.In this review,we summarize the inextricable link between NLRP3 inflammasome activation and cell death and discuss potential therapeutics that target cell death effectors in NLRP3 inflammasome-associated diseases.Yi Huang Wen Xu Rongbin Zhou 2021Cellular & Molecular Immunology2021,18,9:94
2CD205-TLR9-IL-12 axis contributes to CpG-induced oversensitive liver injury in HBsAg transgenic mice by promoting the interaction of NKT cells with Kupffer cells显示文摘Gut-derived bacterial products contribute to liver inflammation and injury during chronic hepatitis B virus infection;however,the underlying mechanisms remain obscure.In this study,hepatitis B surface antigen transgenic(HBs-Tg)mice and their wild-type(WT)control C57BL/6 mice were injected with CpG-oligodeoxynucleotides(ODNs)to mimic the translocation of gut microbial products into the systemic circulation.We found that,compared with the WT mice,the HBs-Tg mice were oversensitive to CpG-ODN-induced liver injury,which was dependent on natural killer T(NKT)cells.CpG-ODN injection enhanced the expression of Fas ligand(FasL)on NKT cells.In addition,hepatocytes from the HBs-Tg mice expressed higher levels of Fas than did those from the WT mice,which was further augmented by CpG-ODN.Interaction of Fas and FasL was involved in the cytotoxicity of NKT cells against hepatocytes in the HBs-Tg mice.Moreover,Kupffer cells in the HBs-Tg mice expressed higher levels of CD205 and produced greater amounts of interleukin(IL)-12 than did those in the WT mice.Finally,the depletion of Kupffer cells,neutralization of IL-12 or specific silencing of CD205 on Kupffer cells significantly inhibited CpG-ODN-induced liver injury and NKT activation in the HBs-Tg mice.Our data suggest that CD205-expressing Kupffer cells respond to CpG-ODNs and subsequently release IL-12 to promote NKT cell activation.Activated NKT cells induce liver damage through the Fas signaling pathway in HBs-Tg mice.Xin Hou Xiaolei Hao Meijuan Zheng Congfei Xu Jun Wang Rongbin Zhou Zhigang Tian 2017Cellular & Molecular Immunology2017,14,8:7

4Functional traits of poplar leaves and fine roots responses to ozone pollution under soil nitrogen addition显示文摘Concurrent ground-level ozone(O_(3))pollution and anthropogenic nitrogen(N)deposition can markedly influence dynamics and productivity in forests.Most studies evaluating the functional traits responses of rapid-turnover organs to O_(3) have specifically examined leaves,despite fine roots are another major source of soil carbon and nutrient input in forest ecosystems.How elevated O_(3) levels impact fine root biomass and biochemistry remains to be resolved.This study was to assess poplar leaf and fine root biomass and biochemistry responses to five different levels of O_(3) pollution,while additionally examining whether four levels of soil N supplementation were sufficient to alter the impact of O_(3) on these two organs.Elevated O_(3) resulted in a more substantial reduction in fine root biomass than leafbiomass;relative to leaves,more biochemically-resistant components were present within fine root litter,which contained high concentrations of lignin,condensed tannins,and elevated C:N and lignin:N ratios that were associated with slower rates of litter decomposition.In contrast,leaves contained more labile components,including nonstructural carbohydrates and N,as well as a higher N:P ratio.Elevated O_(3) significantly reduced labile components and increased biochemically-resistant components in leaves,whereas they had minimal impact on fine root biochemistry.This suggests that O_(3) pollution has the potential to delay leaf litter decomposition and associated nutrient cycling.N addition largely failed to affect the impact of elevated O_(3) levels on leaves or fine root chemistry,suggesting that soil N supplementation is not a suitable approach to combating the impact of O_(3) pollution on key functional traits of poplars.These results indicate that the significant differences in the responses of leaves and fine roots to O_(3) pollution will result in marked changes in the relative belowground roles of these two litter sources within forest ecosystems,and such changes will independently of nitrogen load.Pin Li Rongbin Yin Huimin Zhou Sheng Xu Zhaozhong Feng 2022Journal of Environmental Sciences2022,34,3:1

6Ecological wetland paradigm drives water source improvement in the stream network of Yangtze River Delta显示文摘Jiaxing created a precedent using bypass riparian marshes to purify micro-polluted water sources in China.Pond-wetland complex with constructed root channel technology becomes a paradigm which can be analogized as'human-body wetland model'based on bionics or biomimetics.Heterogeneous plant-bed/ditch system with highly active land/water ecotone interfaces,especially meandering boundaries,breeds many biochemical reactions'living areas'.Optimization of hydraulic regulation promotes redox environment alternations and wetland treatment efficiency.Here we reported a series of upgrades and performances in Guanjinggang wetland after the Shijiuyang prototype.Morphological reform of plant-bed/ditch system played a vital role.Spatially root channel zone was main force of wetland purification,and temporally the treatment effect was higher in low-temperature seasons indicating non-temperature dependent mechanisms worked.Water pollution comprehensive index improved steadily from IV to III,and comprehensive pollution load was reduced by ca.40%–60%.Comprehensive evaluation function value further showed the gradients purification effect of the upgraded wetland.Ecological wetlands ameliorated source water quality,and reduced drinking water treatment reagents,thereby bringing about economic benefits.Through wetlands operation,people can see how the micro-polluted surface water becomes clear and clean,so promoting a significant social benefit.As a viable component of urban green space,wetlands could beautify regional eco-environment,freshen the air,increase urban ecological taste,and enhance the eco-environmental protection publicity.Thus,the multifunctional service values and indirect benefits are substantial.Jiaxing ecological wetlands provide a typical paradigm for water pollution remediation in developing countries and plays a leading role in technology engineering radiation effect.Weidong Wang Ting Yang Weibing Guan Weixi Peng Ping Wu Bin Zhong Chundong Zhou Qinghua Chen Rongbin Zhang Kewen Xu Chengqing Yin 2021Journal of Environmental Sciences2021,33,12:1

8Cohort studies of long-term exposure to outdoor particulate matter and risks of cancer: A systematic review and meta-analysis显示文摘Robust evidence is needed for the hazardous effects of outdoor particulate matter(PM)on mortality and morbidity from all types of cancers.To summarize and meta-analyze the association between PM and cancer,published articles reporting associations between outdoor PM exposure and any type of cancer with individual outcome assessment that provided a risk estimate in cohort studies were identified via systematic searches.Of 3,256 records,47 studies covering 13 cancer sites(30 for lung cancer,12 for breast cancer,11 for other cancers)were included in the quantitative evaluation.The pooled relative risks(RRs)for lung cancer incidence or mortality associated with every 10-mg/m^(3) PM2.5 or PM10 were 1.16(95%confidence interval[CI],1.10–1.23;I2=81%)or 1.22(95%CI,1.02–1.45;I2=96%),respectively.Increased but non-significant risks were found for breast cancer.Other cancers were shown to be associated with PM exposure in some studies but not consistently and thus warrant further investigation.Pei Yu Suying Guo Rongbin Xu Tingting Ye Shanshan Li Malcolm R.Sim Michael J.A.bramson Yuming Guo 2021The Innovation2021,2,3:0
9A lncRNA from an inflammatory bowel disease risk locus maintains intestinal host-commensal homeostasis显示文摘Inflammatory bowel diseases(IBD)are known to have complex,genetically influenced etiologies,involving dysfunctional interactions between the intestinal immune system and the microbiome.Here,we characterized how the RNA transcript from an IBD-associated long non-coding RNA locus(“CARINH-Colitis Associated IRF1 antisense Regulator of Intestinal Homeostasis”)protects against IBD.We show that CARINH and its neighboring gene coding for the transcription factor IRF1 together form a feedforward loop in host myeloid cells.The loop activation is sustained by microbial factors,and functions to maintain the intestinal host-commensal homeostasis via the induction of the anti-inflammatory factor IL-18BP and anti-microbial factors called guanylate-binding proteins(GBPs).Extending these mechanistic insights back to humans,we demonstrate that the function of the CARINH/IRF1 loop is conserved between mice and humans.Genetically,the T allele of rs2188962,the most probable causal variant of IBD within the CARINH locus from the human genetics study,impairs the inducible expression of the CARINH/IRF1 loop and thus increases genetic predisposition to IBD.Our study thus illustrates how an IBD-associated lncRNA maintains intestinal homeostasis and protects the host against colitis.Hongdi Ma Taidou Hu Wanyin Tao Jiyu Tong Zili Han Dietmar Herndler-Brandstetter Zheng Wei Ruize Liu Tingyue Zhou Qiuyuan Liu Xuemei Xu Kaiguang Zhang Rongbin Zhou Judy H.Cho Hua-Bing Li Hailiang Huang Richard A.Flavell Shu Zhu 2023Cell Research2023,33,5:0
