3Detection of geosmin and 2-methylisoborneol by liquid-liquid extraction-gas chromatograph mass spectrum (LLE-GCMS) and solid phase extraction-gas chromatograph mass spectrum (SPE-GCMS)显示文摘Two sample preparation methods were introduced and compared in this paper to establish a simple,quick and exact analysis of geosmin and 2-methylisoborneol.LC-18 column was employed in solid phase extraction(SPE),1.0 mL of hexane was adopted in liquid-liquid extraction(LLE),and the extracts were analyzed by gas chromatograph mass spectrum(GCMS)in selected ion mode.Mean recoveries of SPE were low for 2-methylisoborneol(2-MIB)and geosmin(GSM)with values below 50%.For LLE,the recoveries were satisfyingly above 50%for 2-MIB and 80%for GSM.Detection limits of the LLE method were as low as 1.0 ng/L for GSM and 5.0 ng/L for 2-MIB.A year-long investigation on odor chemicals of drinking water in Shanghai demonstrated that in the summer,there was a serious odor problem induced by a high concentration of 2-MIB.The highest concentration of 152.82 ng/L appeared in July in raw water,while GSM flocculation was minimal with concentrations below odor threshold.MA Xiaoyan GAO Naiyun CHEN Beibei LI Qingsong ZHANG Qiaoli GU Guofen 2007Frontiers of Environmental Science & Engineering2007,1,3:7

7Substrate stiffness modulates bone marrow-derived macrophage polarization through NF-κB signaling pathway显示文摘The stiffness of the extracellular matrix(ECM)plays an important role in regulating the cellular programming.However,the mechanical characteristics of ECM affecting cell differentiation are still under investigated.Herein,we aimed to study the effect of ECM substrate stiffness on macrophage polarization.We prepared polyacrylamide hydrogels with different substrate stiffness,respectively.After the hydrogels were confirmed to have a good biocompatibility,the bone marrow-derived macrophages(BMMs)from mice were incubated on the hydrogels.With simulated by the low substrate stiffness,BMMs displayed an enhanced expression of CD86 on the cell surface and production of reactive oxygen species(ROS)in cells,and secreted more IL-1βand TNF-αin the supernatant.On the contrary,stressed by the medium stiffness,BMMs expressed more CD206,produced less ROS,and secreted more IL-4 and TGF-β.In vivo study by delivered the hydrogels subcutaneously in mice,more CD68+CD86+cells around the hydrogels with the low substrate stiffness were observed while more CD68+CD206+cells near by the middle stiffness hydrogels.In addition,the expressions of NIK,phosphorylated p65(pi-p65)and phosphorylated IκB(pi-IκB)were significantly increased after stimulation with low stiffness in BMMs.Taken together,these findings demonstrated that substrate stiffness could affect macrophages polarization.Low substrate stiffness promoted BMMs to shift to classically activated macrophages(M1)and the middle one to alternatively activated macrophages(M2),through modulating ROS-initiated NF-κB pathway.Therefore,we anticipated ECM-based substrate stiffness with immune modulation would be under consideration in the clinical applications if necessary.Mimi Chen Yu Zhang Pinghui Zhou Xingzhi Liu Huan Zhao Xichao Zhou Qiaoli Gu Bin Li Xuesong Zhu Qin Shi 2020Bioactive Materials2020,5,4:7
8Identification of QTL and candidate genes involved in early seedling growth in rice via high-density genetic mapping and RNA-seq显示文摘Early seedling vigor(ESV)is a major breeding target in rice,especially under direct seeding.To identify quantitative trait locus(QTL)affecting ESV,a recombinant inbred line population derived from a cross between 02428 and YZX,two cultivars differing in vigor during early seedling growth,was used for QTL analysis.Nine traits associated with ESV were examined using a high-density map.Of 16 additive loci identified,three were detected in two generations and thus considered stable.Four epistatic interactions were detected,one of which was repeated in two generations.Further analysis of the pyramiding effect of the three stable QTL showed that the phenotypic value could be effectively improved with an increasing number of QTL.These results were combined with results from our previous QTL analysis of the germination index.The lines G58 and G182 combined all the favourable alleles of all three stable QTL for ESV and three QTL for germination speed.These two lines showed rapid germination and strong ESV.A total of 37 candidate differentially expressed genes were obtained from the regions of the three stable QTL by analysis of the dynamic transcriptomic expression profile during the seedling growth period of the two parents.The QTL are targets for ESV breeding and the candidate genes await functional validation.This study provides a theoretical basis and a genetic resource for the breeding of directseeded rice.Jing Yang Zhenhua Guo Lixin Luo Qiaoli Gao Wuming Xiao Jiafeng Wang Hui Wang Zhiqiang Chen Tao Guo 2021The Crop Journal2021,9,2:6
9Effect of Er-Xian decoction(二仙汤) on autoimmune premature ovarian failure in mice显示文摘OBJECTIVE:To investigate the therapeutic effect of Er-Xian decoction(二仙汤,EXD)on the autoimmune premature ovarian failure(POF)in mice,and its regulatory mechanisms.METHODS:Female BALB mice were treated with a single intraperitoneal injection of zona pellucida 3(ZP3).One week later,mice received low(5 g/kg),moderate(10 g/kg)and high(20 g/kg)doses of EXD by gastrogavage once daily for 90 d.Premarin(0.03 mg/kg)served as the positive control group.Serum levels of estradiol(E2),follicle stimulating hormone(FSH)and luteinizing hormone(LH)were measured by enzyme-linked immunosorbent assay,lymphocyte subtypes were analyzed,and follicular structure differences were observed by hematoxylin andeosin staining.The main mechanisms of POF was investigated by immunohistochemical analysis.RESULTS:Serum E2 levels in POF model mice were decreased,whereas FSH and LH levels were dramatically increased.Serum levels of LH and FSH were reduced in POF model mice treated with EXD(moderate and high doses)and premarin,while serum level of E2 were increased after POF model mice had been treated with EXD and premarin.The CD3+T,CD4+T,CD4+T/CD8+T ratio of mice in the positive control group and high and medium dose groups of EXD increased(P<0.05),and the number of CD8+T cells decreased significantly(P<0.05)when compared with the model group.Bone morphogenetic protein 15(BMP15)and Akt were repressed in autoimmune POF model mice,whereas high expression was observed in control mice and those treated with EXD(moderate and high doses)and premarin.CONCLUSION:EXD is effective in treating ZP3-induced POF in mice and increased expression of BMP-15,and Akt is represented in the mechanism accounting for this therapeutic effect.ZHANG Xiaoli LI Jing ZHOU Xiaolin CHEN Qiaoli 2021Journal of Traditional Chinese Medicine2021,41,5:6
10All-fiber passively mode-locked laser using nonlinear multimode interference of step-index multimode fiber显示文摘We experimentally demonstrate for the first time, to the best of our knowledge, an all-fiber passively mode-locked laser operation based on the nonlinear multimode interference of step-index multimode fiber. Such a structure couples the light in and out of the multimode fiber via single-mode fibers, and its physical mechanisms for saturable absorption have been analyzed theoretically based on the third-order nonlinear Kerr effect of multimode fiber. Using the nonlinear multimode interference structure with 48.8 mm length step-index multimode fiber, the modulation depth has been measured to be ~5%. The passively mode-locked laser output pulses have a central wavelength of 1596.66 nm, bandwidth of 2.18 nm, pulsewidth of ~625 fs, and fundamental repetition rate of8.726 MHz. 1118beta-glycyrrhetinic acid induces ROS-mediated apoptosis to ameliorate hepatic fibrosis by targeting PRDX1/2 in activated HSCs显示文摘Hepatic stellate cells(HSCs)are essential drivers of fibrogenesis.Inducing activated-HSC apoptosis is a promising strategy for treating hepatic fibrosis.18beta-glycyrrhetinic acid(18b-GA)is a natural compound that exists widely in herbal medicines,such as Glycyrrhiza uralensis Fisch,which is used for treating multiple liver diseases,especially in Asia.In the present study,we demonstrated that 18b-GA decreased hepatic fibrosis by inducing the apoptosis in activated HSCs.18b-GA inhibited the expression of a-smooth muscle actin and collagen type Ⅰ alpha-1.Using a chemoproteomic approach derived from activity-based protein profiling,together with cellular thermal shift assay and surface plasmon resonance,we found that 18b-GA covalently targeted peroxiredoxin 1(PRDX1)and peroxiredoxin 2(PRDX2)proteins via binding to active cysteine residues and thereby inhibited their enzymatic activities.18b-GA induced the elevation of reactive oxygen species(ROS),resulting in the apoptosis of activated HSCs.PRDX1 knockdown also led to ROS-mediated apoptosis in activated HSCs.Collectively,our findings revealed the target proteins and molecular mechanisms of 18b-GA in ameliorating hepatic fibrosis,highlighting the future development of 18b-GA as a novel therapeutic drug for hepatic fibrosis.Qian Zhang Piao Luo Liuhai Zheng Jiayun Chen Junzhe Zhang Huan Tang Dandan Liu Xueling He Qiaoli Shi Liwei Gu Jiahao Li Qiuyan Guo Chuanbin Yang Yin Kwan Wong Fei Xia Jigang Wang 2022Journal of Pharmaceutical Analysis2022,12,4:3
12Surfactant dependent evolution of Au-Pd alloy nanocrystals from trisoctahedron to excavated rhombic dodecahedron and multipod: a matter of crystal growth kinetics显示文摘In wet chemical syntheses of noble metal nanocrystals,surfactants play crucial roles in regulating their morphology.To date,more attention has been paid to the effect of the surfactant on the surface energy of crystal facets,while less attention has been paid to its effect on the growth kinetics.In this paper,using the growth of Au-Pd alloy nanocrystals as an example,we demonstrate that different concentration of surfactant hexadecyltrimethyl ammonium chloride(CTAC)may cause the different packing density of CTA+bilayers on different sites(face,edge or vertex)of crystallite surface,which would change the crystal growth kinetics and result in preferential crystal growth along the edge or vertex of crystallites.The unique shape evolution from trisoctahedron to excavated rhombic dodecahedron and multipod structure for Au-Pd alloy nanocrystals was successfully achieved by simply adjusting the concentration of CTAC.These results help to understand the effect of surfactants on the shape evolution of nanocrystals and open up avenues to the rational synthesis of nanocrystals with the thermodynamically unfavorable morphologies.Qiaoli Chen Guifen Du Yongdi Dong Zhenming Cao Zhaoxiong Xie Lansun Zheng 2017Science Bulletin2017,62,20:2
13Relationship between corneal biomechanical parameters and corneal sublayer thickness measured by Corvis ST and UHR-OCT in keratoconus and normal eyes显示文摘Background:To explore the relationship between corneal biomechanical parameters and corneal sublayer thickness using corneal visualization Scheimpflug technology(Corvis ST)and ultrahigh-resolution optical coherence tomography(UHR-OCT)in clinical and suspected keratoconus and normal eyes.Methods:Cross-sectional prospective study.A total of 94 eyes of 70 participants were recruited.Twenty five eyes of 19 keratoconus patients,52 eyes of 34 patients showing high risk of developing keratoconus according to the Belin/Ambrosio Enhanced Ectasia Display,and each eye of 17 normal subjects were enrolled.All participants underwent Corvis ST,Pentacam,and UHR-OCT examinations at the same time.Stiffness parameter A1(SP-A1),deformation amplitude ratio(DA ratio),and other biomechanical parameters were recorded using Corvis ST.The vertical and horizontal thickness profiles of central 3mm corneal epithelium,Bowman’s layer,and stroma as measured by the perpendicular distance between the neighboring interfaces were generated using UHR-OCT.The flat keratometry and steep keratometry were obtained using Pentacam.Analysis of correlation was applied to explore the association between variables.Results:Most of the biomechanical parameters and corneal sublayer thickness profiles showed statistical differences among three groups.A statistically significant linear relationship was noted between SP-A1 and DA ratio in all three groups.SP-A1 was found to be positively correlated with epithelial and Bowman’s layer thickness in the keratoconus(KC)group,and with stromal thickness in all three groups.In the normal and suspected keratoconus(SKC)groups,only stromal thickness was included in the stepwise linear regression to predict SP-A1,whereas in the KC group,steep keratometry and Bowman’s layer thickness were included.Conclusions:Significant and different correlations were noted between corneal stiffness and corneal sublayer thickness in different groups,indicating that corneal sublayers may play different roles in maintaining corneal biomechanical stability between keratoconus and normal eyes.Yong Li Zhiqiang Xu Qiaoli Liu Yuzhou Wang Kan Lin Jiahui Xia Shihao Chen Liang Hu 2021Eye and Vision2021,8,1:2

15miR-101-3p induces vascular endothelial cell dysfunction by targeting tet methylcytosine dioxygenase 2显示文摘Endothelial cell(EC)dysfunction represents an early key event in atherosclerosis.Recently,MicroRNAs have been demonstrated to regulate EC function.miR-101-3p has been discovered to regulate cell apoptosis and proliferation in cardiovascular diseases.Therefore,the aim of the current study was to clarify whether miR-101-3p regulates the dysfunction of vascular endothelial cells.In this study,the transfection of human umbilical vein endothelial cells(HUVECs)with miR-101-3p mimic induced reactive oxygen species(ROS)production,EC dysfunction,and acti-vated nuclear factorB(NF-κB),whereas transfection with miR-101-3p inhibitor alleviated these events.The antioxidant N-acetylcysteine alleviated miR-101-3p-induced EC dysfunction.Moreover,we observed that miR-101-3p inhibited the expression of tet methylcytosine dioxygenase 2(TET2)at the posttranscriptional level,resulting in increased ROS production and activated NF-kB.TET2 overexpression inhibited ROS production,EC dysfunction,and NF-κB activation in miR-101-3p-transfected HUVECs.These results indicate that miR-101-3p induces EC dysfunction by targeting TET2,which regulates ROS production,EC dysfunction,and NF-κB activation.Taken together,our current study reveals a novel pathway associated with EC dysfunction.The modulation of miR-101-3p and TET2 expression levels may serve as a potential target for therapeutic strategies for atherosclerosis.Qiaoli Chen Xiaoye Li Lingjun Kong Qing Xu Zi Wang Qianzhou Lv 2020Acta Biochimica et Biophysica Sinica2020,52,2:2

17DBT parameters and dynamic monitoring during reservoir development, and distribution region prediction of remaining oil:A case study on the Sha-3~3 oil reservoir in the Liubei region, Nanpu sag显示文摘In this study, compositional characteristics of crude oil, including the variation of aliphatic, aromatic and pyrrolic nitrogen compounds, were systematically monitored and investigated in a high water-cut oil reservoir over a short time.The results showed that among the widely used parameters indicative of oil maturity and migration, tetramethyl/monomethyl DBT and tricyclic terpane/(tricyclic terpane+C30 hopanoid) varied remarkably, and a positive correlation was observed between these two parameters.The variation of each of these parameters during waterflooding development was correlated with the flow effect of crude promoted by the water drive in oil reservoirs.A solid consistency was observed among the results of numerical simulation and development; the direction and pathway of waterflooding crude was indicated by Tetramethyl/monomethyl DBT, and the distribution region prediction of remaining oil hereby obtained.Therefore, these two parameters could be used as molecular tracers for the oil during waterflooding.This study would be of practical significance for geochemical dynamic monitoring and reservoir development.XU YaoHui WANG TieGuan CHEN NengXue YANG CuiMin WANG QiaoLi 2012Science China Earth Sciences2012,55,12:1

19Low rate of pre-exposure prophylaxis and post-exposure prophylaxis uptake and high prevalence of transmitted drug resistance among newly diagnosed primary HIV infections in Shenzhen, China: a real-world retrospective study显示文摘Background: Understanding the characteristics of newly diagnosed primary human deficiency virus-1 (HIV-1) infection in the context of the post-antiretroviral therapy era and HIV drug prophylaxis is essential for achieving the new target of 95-95-95-95 by 2025. This study reported the characteristics of newly diagnosed primary HIV-1 infection in Shenzhen.Methods: This is a real-world retrospective study. Eighty-seven newly diagnosed primary HIV-1-infected patients were recruited from January 2021 to March 2022 at the Third People’s Hospital of Shenzhen. Demographic, epidemiological, diagnostic, drug resistance, and medical data were described and analyzed.Results: Overall, 96.6% (84/87) of the newly identified primary HIV-1-infected patients were male, including 88.5% (77/87) men have sex with men (MSM), with a median age of 29.0 years (Q_(1)-Q_(3): 24.0-34.0 years);of these, 85.1% (74/87) reported high-risk sexual behaviors with casual partners. The rate of condom usage was only 28.7% (25/87). The overall rate of pre-exposure prophylaxis (PrEP) and post-exposure prophylaxis (PEP) was 8.0% (7/87, including 4 PrEP and 3 PEP cases) around the potential exposure, although 41.4% of the patients had prior awareness of such interventions. Moreover, only 19.5% (17/87) had previously used PrEP or PEP. Of those, 58.8% (10/17) of the patients obtained drugs from the internet, and only 35.3% (6/17) reported good compliance. A total of 54.0% (47/87) of subjects were diagnosed by the HIV nucleic acid test. Acute retroviral syndrome appeared in 54.0% (47/87) of patients. The prevalence of transmitted drug resistance (TDR) mutation was 33.9% (19/56), including 6 (10.7%) against nucleoside reverse transcriptase inhibitor (NRTI) plus non-nucleoside reverse transcriptase inhibitor (NNRTI), 8 (14.3%) against NNRTI, and 5 (8.9%) against protease inhibitor (PI) only.Conclusions: Owing to the low utilization rate and incorrect usage of PrEP and PEP, massive efforts are needed to promote HIV-preventive strategies in the MSM population. 20Origin of symmetry breaking in the seed-mediated growth of bi-metal nano-heterostructures显示文摘Seed-mediated growth is the most general way to controllably synthesize bimetal nano-heterostructures.Despite successful instances through trial and error were reported, the way for second metal depositing on the seed, namely whether the symmetry of resulted nano-heterostructure follows the original crystal symmetry of seed metal, remains an unpredictable issue to date. In this work, we propose that the thermodynamic factor, i.e., the difference of equilibrium electrochemical potentials(corresponding to their Fermi levels) of two metals in the growth solution, plays a key role for the symmetry breaking of bimetal nano-heterostructures during the seed-mediated growth. As a proof-of-principle experiment, by reversing the relative position of Fermi levels of the Pd nanocube seeds and the second metal Au with changing the concentration of reductant(L-ascorbic acid) in the growth solution, the structure of as-prepared products successfully evolved from centrosymmetric Pd@Au core-shell trisoctahedra to asymmetric Pd-Au hetero-dimers. The idea was further demonstrated by the growth of Ag on the Pd seeds.
