| 1 | Biosynthesis of Triacylglycerol Molecules with a Tailored PUFA Profile in Industrial Microalgae显示文摘The composition of polyun saturated fatty acids (PUFAs) in triacylglycerols (TAGs) is key to health ben efits and for oil applications, yet the underlying genetic mechanism remains poorly understood. In this study, by in si Iico, ex vivo, and in vivo profili ng of type-2 diacylglycerol acyltransferases (DGAT2s) in Nannochloropsis oceanica we revealed two novel PUFA-preferring enzymes that discriminate individual PUFA species in TAG assembly, with NoDGAT2J for linoleic acid (LA) and NoDGAT2K for eicosapentaenoic acid (EPA). The LA and EPA composition of TAG molecules is mediated in vivo via the functional partitioning between NoDGAT2J and 2K, both of which are localized in the chloroplast envelope. By modulating transcript abundance of the DGAT2s, an N. oceanica strain bank was created, where proportions of LA and EPA in TAG vary by 18.7-fold (between 0.21% and 3.92% dry weight) and 34.7-fold (between 0.09% and 3.12% dry weight), respective!y. | 7 | Effect of Chronic Noise Exposure on Expression of N-Methyl-D-Aspartic Acid Receptor 2B and Tau Phosphorylation in Hippocampus of Rats显示文摘Objective To study the effect of chronic noise exposure on expression of N-methyl-D-aspartic acid receptor 2B (NR2B) and tau phosphorylation in hippocampus of rats. Methods Twenty-four male SD rats were divided in control group and chronic noise exposure group. NR2B expression and tau phosphorylation in hippocampus of rats were detected after chronic noise exposure (100 dB SPL white noise, 4 h/d×30d) and their mechanisms underlying neuronal apoptosis in hippocampus of rats with TUNEL staining. Results The NR2B expression decreased significantly after chronic noise exposure which resulted in tau hyperphosphorylation and neural apoptosis in hippocampus of rats. Immunohistochemistry showed that the tau hyperphosphorylation was most prominent in dentate gyrus (DG) and CA1 region of rat hippocampus. | 10 | Deformation features of garnet-bearing granites from Huwan,western Dabie Mountains显示文摘Foliated garnet-bearing granite,usually associated with high pressure and ultrahigh-pressure(UHP) metamophic rocks,is a particular rock-type extensively exposed in the Mesozoic Dabie-Sulu orogenic belt of China.This study focuses on deformation features and SHRIMP zircon dating of foliated garnet granite in a high-pressure metamorphic unit from Huwan,western Dabie Mountains in order to resolve discrepancies in current versions of its petrogenesis and structural evolution.SHRIMP dating reveals a zircon age of 762 ± 15 Ma(MSWD=1.7) for Huwan granites,representing the Middle to Late Neoproterozoic age of intrusion and crystallization.Field and microstructural studies show that the Huwan granite body underwent multiple-stage deformation.The deformation was manifested by an early stage of rootless folding and imposition of relict foliation(S1);an Indosinian main stage marked by imposition of north-dipping penetrative gneissosity(S2) and development of ductile shear zones under NNE-SSW directed compression;and a final Indosinian stage of southward thrusting of the Huwan high-pressure unit.Shallow level extension prevailed after the Late Triassic,giving rise to south-dipping thrust faults and north-dipping normal faults. | YANG KunGuang LIU Qiang XIE JianLei DAN Wei SHE ZhenBing MA ChangQian | 2009 | Science China Earth Sciences2009,52,1: | 2 |

| 17 | TNFα inhibitor C87 sensitizes EGFRvⅢ transfected glioblastoma cells to gefitinib by a concurrent blockade of TNFα signaling显示文摘Objective: More than half of human glioblastomas show EGFR gene amplification and mutation, but EGFR inhibitors have not been effective in treating EGFR-positive glioblastoma patients.The mechanism behind this type of primary resistance is not well understood.The aim of this study was to investigate gefitinib resistance in glioblastoma, and explore ways to circumvent this significant clinical problem.Methods: MTT method was used to test the cell viability after EGFR-positive glioblastoma cells were treated with indicated drugs;real-time quantitative PCR method was included to detect the TNFα mRNA levels in glioma tissues and cell lines.ELISA was introduced to measure the TNFα protein levels in cell culture supernatant of glioblastoma cells treated with gefitinib.Western blot was used to detect the activity change of intracellular kinases in drug-treated glioblastoma cells.Two mouse xenograft tumor models were carried out to evaluate the in vivo effects of a combination of EGFR and TNFα inhibitors.Results: We found that glioblastoma resistance to gefitinib may be mediated by an adaptive pro-survival TNFα-JNK-Axl signaling axis, and that high TNFα levels in the glioblastoma microenvironment may further intensify primary resistance.A combination of the TNFα-specific small-molecule inhibitor C87 and gefitinib significantly enhanced the sensitivity of glioblastoma cells to gefitinib in vitro and in vivo.Conclusions: Our findings provide a possible explanation for the primary resistance of glioblastoma to EGFR inhibitors and suggest that dual blockade of TNFα and EGFR may be a viable therapeutic strategy for the treatment of patients with chemotherapy-refractory advanced glioblastoma. | Li Ma Chunhua She Qian Shi Qiang Yin Xinxin Ji Yongrong Wang Yulong Fan Xinyao Kong Peng Li Zengfeng Sun Xiaohui Zhang Zhen Zhang Jian Wang Tong Wang Yuanfu Xu Wenliang Li | 2019 | Cancer Biology & Medicine2019,16,3: | 1 |
