1Overexpression of micro RNA_(40)8 enhances photosynthesis, growth, and seed yield in diverse plants显示文摘The ability of a plant to produce grain, fruit, or forage depends ultimately on photosynthesis. There have been few attempts, however, to study micro RNAs, which are a class of endogenous small RNAs post-transcriptionally programming gene expression, in relation to photosynthetic traits. We focused on miR_(40)8, one of the most conserved plant miRNAs, and overexpressed it in parallel in Arabidopsis, tobacco, and rice. The transgenic plants all exhibited increased copper content in the chloroplast, elevated abundance of plastocyanin,and an induction of photosynthetic genes. By means of gas exchange and optical spectroscopy analyses, we showed that higher expression of miR_(40)8 leads to enhanced photosynthesis through improving efficiency of irradiation utilization and the capacity for carbon dioxide fixation. Consequently, miR_(40)8 hyper-accumulating plants exhibited higher rate of vegetative growth. An enlargement of seed size was also observed in all three species overproducing miR_(40)8. Moreover, we conducted a 2-year-two-location field trial and observed miR_(40)8 overexpression in rice significantly increased yield, which was primarily attributed to an elevation in grain weight. 3LncDACH1 promotes mitochondrial oxidative stress of cardiomyocytes by interacting with sirtuin3 and aggravates diabetic cardiomyopathy显示文摘Diabetic cardiomyopathy(DCM)is a common complication in diabetic patients.The molecular mechanisms of DCM remain to be fully elucidated.The intronic long noncoding RNA of DACH1(lnc DACH1)has been demonstrated to be closely associated with heart failure and cardiac regeneration.In this study,we investigated the role of lnc DACH1 in DCM and the underlying molecular mechanisms.The expression of lnc DACH1 was increased in DCM hearts and in high glucose-treated cardiomyocytes.Knockout of lnc DACH1 reduced mitochondrial oxidative stress,cell apoptosis,cardiac fibrosis and hypertrophy,and improved cardiac function in DCM mice.Overexpression of lnc DACH1 exacerbated mitochondria-derived reactive oxygen species(ROS)level and apoptosis,decreased activity of manganese superoxide dismutase(Mn-SOD);while silencing of lnc DACH1 attenuated ROS production,mitochondrial dysfunction,cell apoptosis,and increased the activity of Mn-SOD in cardiomyocytes treated with high glucose.Lnc DACH1 directly bound to sirtuin3(SIRT3)and facilitated its degradation by ubiquitination,therefore promoting mitochondrial oxidative injury and cell apoptosis in mouse hearts.In addition,SIRT3 silencing abrogated the protective effects of lnc DACH1 deficiency in cardiomyocytes.In summary,lnc DACH1 aggravates DCM by promoting mitochondrial oxidative stress and cell apoptosis via increasing ubiquitination-mediated SIRT3 degradation in mouse hearts.Inhibition of lnc DACH1 represents a novel therapeutic strategy for the intervention of diabetic cardiomyopathy.Qi Zhang Danyang Li Xue Dong Xiaowen Zhang Junwu Liu Lili Peng Bo Meng Qi Hua Xinyu Pei Lu Zhao Xiaoxi Hu Yang Zhang Zhenwei Pan Yanjie Lu Baofeng Yang 2022Science China(Life Sciences)2022,65,6:4

5CSF-Based Analysis for Identification of Potential Serum Biomarkers of Neural Tube Defects显示文摘The protein composition of cerebrospinal fluid(CSF) in neural tube defects(NTDs) remains unknown. We investigated the protein composition of CSF from 9 infants with NTDs using isobaric tags for relative and absolute quantitation(iTRAQ). We identified 568 proteins in the CSF of infants with spina bifida, which is the most common type of NTD. Among these, 18 proteins were associated with neural tube closure in the CSF during human embryonic neurulation and 5 were involved in NTDs. Based on these results, an animal model was further utilized to investigate early serum biomarkers for NTDs.We found that the myristoylated alanine-rich C-kinase substrate, Kunitz-type protease inhibitor 2, andapolipoprotein B-100 protein levels were decreased in both embryos and the sera of pregnant Sprague-Dawley rats carrying embryos with NTDs. 7Dynamic analysis of a concentrating photovoltaic/concentrating solar power (CPV/CSP) hybrid system显示文摘Concentrating photovoltaic/concentrating solar power(CPV/CSP)systems suffer from varying irradiation and extreme working conditions.In this study,a dynamic physical model is developed for the CPV/CSP hybrid system to analyze the dynamic responses of several key parameters,such as the solar radiation saltation or linear variation to represent the typical weather variations.The results show that the hybrid system could rapidly reach the steady state in less than about 53 s after the solar radiation saltation increases or decreases by 10%.The response time reflects that the thermal hysteresis of the hybrid system is mainly determined by varying the outlet temperature of R134a from the solar thermal receiver.Meanwhile,when the solar radiation changes linearly,a lower gradient is beneficial to remit the thermal hysteresis of the hybrid system and improve the thermal stability,and the parameters could be treated as the steady state values with a gradient of less than 0.2 W m^-2 s^-1.Afterward,the quasi-steady state model was used to analyze the all-day dynamic performance of the hybrid system.It shows that the power output and the flow rate are directly related to the direct normal irradiance(DNI),while the outlet temperature of R134a vapor could be almost constant except for the starting and stopping periods.HAN Xue XU Chao PAN XinYu JU Xing DU XiaoZe 2019Science China(Technological Sciences)2019,62,11:2

9GOLM1 restricts colitis and colon tumorigenesis by ensuring Notch signaling equilibrium in intestinal homeostasis显示文摘Intestinal epithelium serves as the first barrier against the infections and injuries that mediate colonic inflammation.Colorectal cancer is often accompanied with chronic inflammation.Differed from its well-known oncogenic role in many malignancies,we present here that Golgi membrane protein 1(G0LM1,also referred to as GP73)suppresses colorectal tumorigenesis via maintenance of intestinal epithelial barrier.G0LM1 deficiency in mice conferred susceptibility to mucosal inflammation and colitis-induced epithelial damage,which consequently promoted colon cancer.Mechanistically,depletion of G0LM1 in intestinal epithelial cells(lECs)led to aberrant Notch activation that interfered with IEC differentiation,maturation,and lineage commitment in mice.Pharmacological inhibition of Notch pathway alleviated epithelial lesions and restrained pro-tumorigenic inflammation in G0LM1-deficient mice.Therefore,G0LM1 maintains IEC homeostasis and protects against colitis and colon tumorigenesis by modulating the equilibrium of Notch signaling pathway.Yang Pu Ya Song Mengdi Zhang Caifeng Long Jie Li Yanan Wang Yinzhe Xu Fei Pan Na Zhao Xinyu Zhang Yanan Xu Jianxin Cui Hongying Wang Yan Li Yong Zhao Di Jin Hongbing Zhang 2021Signal Transduction and Targeted Therapy2021,6,5:2
10Metal-organic frameworks as functional materials for implantable flexible biochemical sensors显示文摘Metal-organic frameworks(MOFs)exhibit attractive properties such as highly accessible surface area,large porosity,tunable pore size,and built-in redox-active metal sites.They may serve as excellent candidates to construct implantable flexible devices for biochemical sensing due to their high thermal and solution stability.However,MOFs-based sensors have only been mostly reported for in-vitro chemical sensing,their use in implantable chemical sensing and combination with flexible electronics to achieve excellent mechanical compatibility with tissues and organs has rarely been summarized.This paper systematically reviews the biochemical sensors based on MOFs and discusses the feasibility to achieve implantable biochemical sensing through MOFs-based flexible electronics.The properties of MOFs and underlying mechanisms have been intrcxluced,followed by a summarization of different biochemical sensing applications.Strategies to integrate MOFs with flexible devices have been supplied from the standpoints of matching mechanics and compatible fabrication processes.Issues that should be addressed in developing flexible MOFs sensors and potential solutions have also been provided,followed by the perspective for future applications of flexible MOFs sensors.This paper may serve as a reference to offer potential guidelines for the development of flexible MOFs-based biochemical sensors that may benefit future applications in personal healthcare,disease diagnosis and treatment,and fundamental study of various biological processes.Ya Li Wei Ling Xinyu Liu Xue Shang Pan Zhou Zhaorun Chen Hang Xu Xian Huang 2021Nano Research2021,14,9:2
11Chemotherapy initiation with single-course methotrexate alone or combined with dactinomycin versus multi-course methotrexate for low-risk gestational trophoblastic neoplasia: a multi-centric randomized clinical trial显示文摘We aimed to evaluate the effectiveness and safety of single-course initial regimens in patients with low-risk gestational trophoblastic neoplasia(GTN).In this trial (NCT01823315),276 patients were analyzed.Patients were allocated to three initiated regimens:single-course methotrexate(MTX),single-course MTX+dactinomycin(ACTD),and multi-course MTX(control arm).The primary endpoint was the complete remission(CR)rate by initial drug(s).The primary CR rate was 64.4%with multi-course MTX in the control arm.For the single-course MTX arm,the CR rate was 35.8%by one course;it increased to 59.3%after subsequent multi-course MTX,with non-inferiority to the control(difference-5.1%,95%confidence interval(CI)-19.4%to 9.2%,P=0.014).After further treatment with multi-course ACTD,the CR rate(93.3%)was similar to that of the control(95.2%,P=0.577).For the single-course MTX+ACTD arm,the CR rate was 46.7%by one course,which increased to 89.1%after subsequent multi-course,with non-inferiority(difference 24.7%,95%CI 12.8%-36.6%,P<0.001)to the control.It was similar to the CR rate by MTX and further ACTD in the control arm(89.1%vs.95.2%,P=0.135).Four patients experienced recurrence,with no death,during the 2-year follow-up.We demonstrated that chemotherapy initiation with single-course MTX may be an alternative regimen for patients with low-risk GTN.Lili Chen Ling Xi Jie Jiang Rutie Yin Pengpeng Qu Xiuqin Li Xiaoyun Wan Yaxia Chen Dongxiao Hu Yuyan Mao Zimin Pan Xiaodong Cheng Xinyu Wang Qingli Li Danhui Weng Xi Zhang Hong Zhang Quanhong Ping Xiaomei Liu Xing Xie Beihua Kong Ding Ma Weiguo Lu 2022Frontiers of Medicine2022,16,2:2
12A paper-based microfluidic platform with shapememory-polymer-actuated fluid valves for automated multi-step immunoassays显示文摘Smart fluid manipulation with automatically controlled paper valves will enable automated and multi-step immunoassays on paper-based microfluidic devices.In this work,we present an integrated paper-based microfluidic platform with shape-memory polymer(SMP)-actuated fluid valves capable of automated colorimetric enzyme-linked immunosorbent assays(ELISAs).A single-layer microfluidic paper-based analytical device(μPAD)was designed to store all the reagents on the chip,and sequentially transfer reagents to a paper test zone following a specific ELISA protocol through automatic fluidic flow control by the multiple SMP-actuated valves.The actuation of a paper valve was based on the thermally responsive,duel-state shape transformation of a SMP sheet attached to the root of a paper cantilever beam for driving a hydrophilic paper bridge to connect and disconnect two paper channels.A portable colorimetric reader was developed to control the on-chip valve operations,quantify the colorimetric signal output,display the assay result,and wirelessly transmit the data to a smart phone for the application of telemedicine.Reliable operations of the paper valve and the entireμPAD were demonstrated with success rates of 97%and 93%,respectively.A detection mechanism for valve malfunction was designed and confirmed effective to identify any mal-operation of individual valves,thus rendering our platform reliable in real assays.For device calibration,we conducted direct ELISAs of rabbit IgG in phosphate-buffered saline(PBS),and achieved a low limit of detection(LOD)of 27 pM(comparable to that of standard and paper-based ELISAs).In order to demonstrate the clinical application of our multi-step immunoassay platform,we also conducted sandwich ELISAs to quantify the protein level of an inflammatory cytokine,namely tumor necrosis factor(TNF)-α,in surgically injured laryngeal tissues of rats.The protein levels of TNF-αwere shown similar between the conventional andμPAD ELISAs.Hao Fu Pengfei Song Qiyang Wu Chen Zhao Peng Pan Xiao Li Nicole YKLi-Jessen Xinyu Liu 2019Microsystems & Nanoengineering2019,5,1:2
13A micro-nano optogenetic system based on probiotics for in situ host metabolism regulation显示文摘Genetically engineered bacteria have aroused attention as micro-nano drug delivery systems in situ.However,conventional designs of engineered bacteria usually function constantly or autonomously,which might be non-specific or imprecise.Therefore,designing and optimizing in situ control strategy are important methodological progress for therapeutic researches of intestinal engineered bacteria.Here,a micro-nano optogenetic system based on probiotic was developed combining microelectronics,nanotechnology,and synthetic biology to achieve in situ controllable drug delivery.Firstly,optogenetic engineered Lactococcus lactis was orally administrated in the intestinal tract.A wearable optical device was designed to control optical signals remotely.Then,L.lactis could be customized to secrete peptides according to optical signals.As an example,optogenetic L.lactis system can be constructed to secrete glucagon-like peptide-1(GLP-1)under the control of the wearable optical device to regulate metabolism.To improve the half-life of GLP-1 in vivo,Fc-domain fused GLP-1 was optimally used.Using this strategy,blood glucose,weight,and other features were well controlled in rats and mice models.Furthermore,upconversion microcapsules were introduced to increase the excitation wavelength of the optogenetic system for better penetrability.This strategy has biomedical potential to expand the toolbox for intestinal engineered bacteria.Xinyu Zhang Ning Ma Wei Ling Gaoju Pang Tao Sun Jing Liu Huizhuo Pan Meihui Cui Chunli Han Chun Yang Jin Chang Xian Huang Hanjie Wang 2023Nano Research2023,16,2:2
14Evaluation on the natural gas hydrate formation process显示文摘Gas hydrates have endowed with great potential in gas storage,and rapid formation of gas hydrates is critical to use this novel technology.This work evaluated the natural gas hydrate formation process,which was compared from six parameters,including conversion of water to hydrate,storage capacity,the rate of hydrate formation,space velocity(SV)of hydrate reaction,energy consumption and hydrate removal.The literature was selected by analyzing and comparing these six parameters mentioned above,meanwhile placing emphasis on the three parameters of storage capacity,the rate of hydrate formation and space velocity of hydrate reaction.Through analysis and comparison,four conclusions could be obtained as follows.Firstly,the overall performance of the stirring process and the spraying process were better than other processes after analyzing the six parameters.Secondly,the additive types,the reactor structure and the reactor size had influence on the natural gas hydrate formation process.Thirdly,the energy consumption via reciprocating impact in the hydrate formation process was higher than that via stirring,spraying and static higee.Finally,it was one key for hydrate removal to realize the hydrate industrial production.Shuqi Fang Xinyue Zhang Jingyi Zhang Chun Chang Pan Li Jing Bai 2020Chinese Journal of Chemical Engineering2020,28,3:2

17Progress and Understanding on Catalysts with Well-defined Interface for Boosting CO_(2)Conversion显示文摘Catalytic conversion of CO_(2)into valuable chemicals like CH;OH is the most promising way to alleviate CO_(2)emission for solving the serious climate change issue.Multi-component catalysts with well-defined interface show outstanding performance in CO_(2)conversion due to the synergistic effects and multifunctional properties caused by the well-defined interface.A discharge technique,named as cold plasma,has been recognized as an excellent strategy for tuning catalyst interface properties.The temperature of cold plasma is lower than 200℃,and can be further lowered to room temperature by simply changing the operation conditions of cold plasma.The lower temperature of cold plasma can well maintain the catalyst structures,especially the porous structures.When conducting cold plasma,in addition to nontoxic working gases like Ar and air,no harmful substances are used.Cold-plasma-prepared catalysts have unique interface properties,and thereby exhibit superior performance in CO_(2)conversion over the catalysts prepared by traditional methods.The present review summarizes the progress about the cold-plasma-prepared catalysts for CO_(2)conversion,discusses the origin for the outstanding catalytic performance,and proposes the challenges and opportunities for further studies.This will stimulate more deep insights into the cold-plasma-prepared catalysts with well-defined interface properties for achieving more efficient CO_(2)conversion.Binran Zhao Yiyi Zhao Peng Liu Yulong Men Xinyu Meng Yunxiang Pan 2022Chinese Journal of Structural Chemistry2022,41,4:1
