1Investigation of cell mechanics using single-beam acoustic tweezers as a versatile tool for the diagnosis and treatment of highly invasive breast cancer cell lines: an in vitro study显示文摘Advancements in diagnostic systems for metastatic cancer over the last few decades have played a significant role in providing patients with effective treatment by evaluating the characteristics of cancer cells.Despite the progress made in cancer prognosis,we still rely on the visual analysis of tissues or cells from histopathologists,where the subjectivity of traditional manual interpretation persists.This paper presents the development of a dual diagnosis and treatment tool using an in vitro acoustic tweezers platform with a 50MHz ultrasonic transducer for label-free trapping and bursting of human breast cancer cells.For cancer cell detection and classification,the mechanical properties of a single cancer cell were quantified by single-beam acoustic tweezers(SBAT),a noncontact assessment tool using a focused acoustic beam.Cell-mimicking phantoms and agarose hydrogel spheres(AHSs)served to standardize the biomechanical characteristics of the cells.Based on the analytical comparison of deformability levels between the cells and the AHSs,the mechanical properties of the cells could be indirectly measured by interpolating the Young’s moduli of the AHSs.As a result,the calculated Young’s moduli,i.e.,1.527kPa for MDA-MB-231(highly invasive breast cancer cells),2.650kPa for MCF-7(weakly invasive breast cancer cells),and 2.772 kPa for SKBR-3(weakly invasive breast cancer cells),indicate that highly invasive cancer cells exhibited a lower Young’s moduli than weakly invasive cells,which indicates a higher deformability of highly invasive cancer cells,leading to a higher metastasis rate.Single-cell treatment may also be carried out by bursting a highly invasive cell with high-intensity,focused ultrasound.Hae Gyun Lim Hsiao-Chuan Liu Chi Woo Yoon Hayong Jung Min Gon Kim Changhan Yoon Hyung Ham Kim KKirk Shung 2020Microsystems & Nanoengineering2020,6,1:3
2Function of flavonoids on different types of programmed cell death and its mechanism:a review显示文摘Cell death in the living system plays a vital role in maintaining the homeostasis and balancing the cell count in the body.Programmed cell death(PCD)is a crucial component of several development and defense mechanisms.PCD is also important in terms of aging which avoids the accumulation of cellular damage by maintaining cell division.Depending on the execution of cell death and its role in destruction,PCD is categorized into several subtypes.The major different forms of PCD in animals are apoptosis,autophagy and necrosis,which can be distinct in morphological terms.More intense investigations of cell death have given close insight showing other important types of cellular destruction and their pivotal roles in treating disease conditions like cancer.Flavonoids have been acquired a great interest for disease therapies and chemoprevention through activation of several PCD mechanisms.The significant potential of natural flavonoids in the induction of distinct signaling cascades is being a massive approach for targeting uncontrolled cell growth.For these reasons,understanding PCD mechanisms is a promising approach for the interventions in treating cancer.Thus,it is intriguing that understanding the different forms of PCD mechanism induced by flavonoids with more accurate descriptions on the biochemical and cellular processes are gaining more significance in cancer research.Here,we provide a brief overview on the different types of PCD and aim to discuss the functional role of flavonoids in promoting different types of cell death as well as an extensive brief review on their mechanism of action has been highlighted.Preethi Vetrivel Seong Min Kim Venu Venkatarame Gowda Saralamma Sang Eun Ha Eun Hee Kim Tae Sun Min Gon Sup Kim 2019The Journal of Biomedical Research2019,33,6:3

10Regeneration-associated macrophages: a novel approach to boost intrinsic regenerative capacity for axon regeneration显示文摘Axons in central nervous system(CNS) do not regenerate spontaneously after injuries such as stroke and traumatic spinal cord injury.Both intrinsic and extrinsic factors are responsible for the regeneration failure.Although intensive research efforts have been invested on extrinsic regeneration inhibitors,the extent to which glial inhibitors contribute to the regeneration failure in vivo still remains elusive.Recent experimental evidence has rekindled interests in intrinsic factors for the regulation of regeneration capacity in adult mammals.In this review,we propose that activating macrophages with pro-regenerative molecular signatures could be a novel approach for boosting intrinsic regenerative capacity of CNS neurons.Using a conditioning injury model in which regeneration of central branches of dorsal root ganglia sensory neurons is enhanced by a preceding injury to the peripheral branches,we have demonstrated that perineuronal macrophages surrounding dorsal root ganglia neurons are critically involved in the maintenance of enhanced regeneration capacity.Neuron-derived chemokine(C-C motif) ligand 2(CCL2) seems to mediate neuron-macrophage interactions conveying injury signals to perineuronal macrophages taking on a soley pro-regenerative phenotype,which we designate as regeneration-associated macrophages(RAMs).Manipulation of the CCL2 signaling could boost regeneration potential mimicking the conditioning injury,suggesting that the chemokine-mediated RAM activation could be utilized as a regenerative therapeutic strategy for CNS injuries.Min Jung Kwon Hyuk Jun Yoon Byung Gon Kim 2016Neural Regeneration Research2016,11,9:1
