| 1 | Dynamic expression of the LAP family of genes during early development of Xenopus tropicalis显示文摘The leucine-rich repeats and PDZ (LAP) family of genes are crucial for the maintenance of cell polarity as well as for epithelial homeostasis and tumor suppression in both vertebrates and invertebrates.Four members of this gene family are known:densin,erbin,scribble and lano.Here,we identified the four members of the LAP gene family in Xenopus tropicalis and studied their expression patterns during embryonic development.The Xenopus LAP proteins show a conserved domain structure that is similar to their homologs in other vertebrates.In Xenopus embryos,these genes were detected in animal cap cells at the early gastrula stage.At later stages of development,they were widely expressed in epithelial tissues that are highly polar in nature,including the neural epithelia,optic and otic vesicles,and in the pronephros.These data suggest that the roles of the Xenopus LAP genes in the control of cell polarity and morphogenesis are conserved during early development.Erbin and lano show similar expression patterns in the developing head,suggesting potential functional interactions between the two molecules in vivo. | YANG QiuTan LV XiaoYan KONG QingHua LI ChaoCui ZHOU Qin MAO BingYu | 2011 | Science China(Life Sciences)2011,54,10: | 3 |

| 5 | Immune‑Ageing Evaluation of Peripheral T and NK Lymphocyte Subsets in Chinese Healthy Adults显示文摘Ageing is often accompanied with a decline in immune system function,resulting in immune ageing.Numerous studies have focussed on the changes in different lymphocyte subsets in diseases and immunosenescence.The change in immune phenotype is a key indication of the diseased or healthy status.However,the changes in lymphocyte number and phenotype brought about by ageing have not been comprehensively analysed.Here,we analysed T and natural killer(NK)cell subsets,the phenotype and cell differentiation states in 43,096 healthy individuals,aged 20–88 years,without known diseases.Thirty-six immune parameters were analysed and the reference ranges of these subsets were established in different age groups divided into 5-year intervals.The data were subjected to random forest machine learning for immune-ageing modelling and confirmed using the neural network analysis.Our initial analysis and machine modelling prediction showed that na.ve T cells decreased with ageing,whereas central memory T cells(Tcm)and effector memory T cells(Tem)increased cluster of differentiation(CD)28-associated T cells.This is the largest study to investigate the correlation between age and immune cell function in a Chinese population,and provides insightful differences,suggesting that healthy adults might be considerably influenced by age and sex.The age of a person's immune system might be different from their chronological age.Our immune-ageing modelling study is one of the largest studies to provide insights into‘immune-age’rather than‘biological-age’.Through machine learning,we identified immune factors influencing the most through ageing and built a model for immune-ageing prediction.Our research not only reveals the impact of age on immune parameter differences within the Chinese population,but also provides new insights for monitoring and preventing some diseases in clinical practice. | Zhenghu Jia Zhiyao Ren Dongmei Ye Jiawei Li Yan Xu Hui Liu Ziyu Meng Chengmao Yang Xiaqi Chen Xinru Mao Xueli Luo Zhe Yang Lina Ma Anyi Deng Yafang Li Bingyu Han Junping Wei Chongcheng Huang Zheng Xiang Guobing Chen Peiling Li Juan Ouyang Peisong Chen Oscar Junhong Luo Yifang Gao Zhinan Yin | 2023 | Phenomics2023,3,4: | 1 |
| 6 | Embryonic expression and evolutionary analysis of the amphioxus Dickkopf and Kremen family genes显示文摘The secreted Wnt signaling inhibitor Dickkopf1 (Dkk1) plays key role in vertebrate head induction. Its receptor Kremen synergizes with Dkk1 in Wnt inhibition. Here we have carried out expression and functional studies of the Dkk and Kremen genes in amphioxus (Branchiostoma belcheri). During embryonic and larval development, BbDkk1/2/4 is expressed in the posterior mesoendoderm, anterior somatic mesoderm and the pharyngeal regions. Its expression becomes restricted to the pharyngeal region on the left side at larval stages. In 45 h larvae, BbDkk1/2/4 is expressed specifically in the cerebral vesicle. BbDkk3 was only detected at larval stages in the mid-intestine region. Seven Kremen related genes were identified in the genome of the Florida amphioxus (Branchiostoma floridae), clustered in 4 scaffolds, and are designated Kremen1-4 and Kremen-like 1-3, respectively. In B. belcheri, Kremen1 is strongly expressed in the mesoen-doderm during early development and Kremen3 is expressed asymmetrically in spots in the larval pharyngeal region. In luciferase reporter assays, BbDkk1/2/4 can strongly inhibit Wnt signaling, while BbDkk3, BbKremen1 and BbKremen3 can not. | 13 | Sequential stabilization of RNF220 by RLIM and ZC4H2 during cerebellum development and Shh-group medulloblastoma progression显示文摘Sonic hedgehog (Shh) signaling is essential for the proliferation of cerebellar granule neuron progenitors (CGNPs), and its misregulation is linked to various disorders, including cerebellar cancer medulloblastoma (MB). During vertebrate neural development, RNF220, a ubiquitin E3 ligase, is involved in spinal cord patterning by modulating the subcellular location of glioma-associated oncogene homologs (Glis) through ubiquitination. RNF220 is also required for full activation of Shh signaling during cerebellum development in an epigenetic manner through targeting embryonic ectoderm development. ZC4H2 was reported to be involved in spinal cord patterning by acting as an RNF220 stabilizer. Here, we provided evidence to show that ZC4H2 is also required for full activation of Shh signaling in CGNP and MB progression by stabilizing RNF220. In addition, we found that the ubiquitin E3 ligase RING finger LIM domain-binding protein (RLIM) is responsible for ZC4H2 stabilization via direct ubiquitination, through which RNF220 is also thus stabilized. RLIM is a direct target of Shh signaling and is also required for full activation of Shh signaling in CGNP and MB cell proliferation. We further provided clinical evidence to show that the RLIM‒ZC4H2‒RNF220 cascade is involved in Shh-group MB progression. Disease-causative human RLIM and ZC4H2 mutations affect their interaction and regulation.
