| 4 | Role of moxibustion in inflammatory responses during treatment of rat ulcerative colitis显示文摘AIM: To investigate the efficacy of moxibustion in ulcerative colitis(UC) rats from morphological, immunological and molecular biological perspectives.METHODS: Thirty-two Sprague-Dawley rats were randomly assigned to a blank control group(normal rats, n = 6) and a model replication(MR) group(UC rats, n = 26). A UC model was established by 2,4,6-trinitrobenzenesulfonic acid/dextran sulfate sodium enema. Rats in the MR group were further randomly assigned to a 9-min moxibustion(9M) group(9 moxa-cone, n = 6), 6-min moxibustion(6M) group(6 moxa-cone, n = 6), 3-min moxibustion(3M) group(3 moxa-cone, n = 6), and a waiting list control(WLC) group(no moxibustion treatment, n = 6). Rats in the moxibustion treatment group were treated in 14 sessions over 28 d. Disease activity, local tissue morphology, serum level of interleukin(IL)-8 and IL-10, and expression of Toll-like receptor(TLR)9 aswell as nuclear factor(NF)-κB p65 in colonic tissue were determined by disease activity index(DAI), hematoxylin and eosin staining, electron microscopy, enzyme-linked immunosorbent assay and Western blotting, respectively.RESULTS: DAI was lowest in the 9M group and highest in the WLC group. The differences in DAI between the moxibustion treatment(3M, 6M, 9M) and no treatment groups were significant for all one-to-one comparisons(0.60 ± 0.54 vs 1.20 ± 0.44, 0.60 ± 0.54 vs 1.80 ± 0.45, 0.60 ± 0.54 vs 3.0 ± 0.45, respectively, P < 0.05). Light and electron microscopy showed that the neatness of the glandular arrangement in colonic mucosal epithelia gradually increased in the WLC, 3M, 6M to 9M groups. IL-8 level successively decreased while IL-10 level increased from the WLC to 3M, 6M and 9M groups. The differences among these groups were significant for all comparisons(105.46 ± 8.75 vs 76.61 ± 3.58, 105.46 ± 8.75 vs 69.78 ± 1.87, 105.46 ± 8.75 vs 67.41 ± 1.84, respectively, P < 0.01 for IL-8; and 30.83 ± 1.29 vs 75.64 ± 1.90, 30.83 ± 1.29 vs 80.90 ± 3.16, 30.83 ± 1.29 vs 83.46 ± 2.37, respectively, P < 0.01 for IL-10), except comparison of 6M vs 9M. Expression of TLR9 and NF-κB p65 decreased in order: highest in the WLC group and lowest in the 9M group. | 5 | The influence of the Madden-Julian Oscillation activity anomalies on Yunnan's extreme drought of 2009-2010显示文摘Yunnan Province of China suffered a record-breaking drought that persisted from autumn 2009 into spring 2010. The present study examined the physical causes of this extreme drought event in terms of persistent anomalies of intraseasonal oscillation in the tropical atmosphere (the Madden-Julian Oscillation hereafter the MJO). The results show that the occurrence of severe drought in Yunnan was caused directly by deficient rainfall lasting from the summer of 2009 to the spring of 2010. Further exploration reveals a persistent positive variation of MJO index 1 from June to October. Accordingly, the convective activity over the Bay of Bengal (BOB) weakened continuously, and then an anomalous descending airflow was induced over the tropical Indian Ocean, resulted in the anomalous weakening of vertical Asian monsoon circulation in South Asia. Consequently, the transport of water vapor from the tropical Indian Ocean to Yunnan decreased abnormally, leading to persistent below-normal rainfall over Yunnan from summer to autumn in 2009. As a result, a severe drought began to appear in autumn. In the winter of 2009-2010, MJO index 1 remained persistently positive, indicating the continuous weakening of convective activity over the BOB. The atmospheric circulation associated with the persistent positive anomalies in the MJO also demonstrated anomalous patterns. Specifically, there was an anomalous high-pressure ridge stretching from South Asia through the Tibetan Plateau and into the western part of southwestern China. This indicates that the atmospheric circulation over Yunnan was dominated by vertical descending airflow in the high-pressure ridge. Simultaneously, the India-Burma trough was weakened, which resulted in unfavorable conditions for the transport of water vapor from the BOB to Yunnan, causing the observed persistent deficient precipitation in winter and the subsequently intensified drought. | 6 | Neuronal autophagy aggravates microglial inflammatory injury by downregulating CX3CL1/fractalkine after ischemic stroke显示文摘Ischemic stroke often induces excessive neuronal autophagy, resulting in brain damage; meanwhile, inflammatory responses stimulated by ischemia exacerbate neural injury. However, interactions between neuronal autophagy and microglial inflammation following ischemic stroke are poorly understood. CX3CL1/fractalkine, a membrane-bound chemokine expressed on neurons, can suppress microglial inflammation by binding to its receptor CX3CR1 on microglia. In the present study, to investigate whether autophagy could alter CX3CL1 expression on neurons and consequently change microglial inflammatory activity, middle cerebral artery occlusion(MCAO) was established in Sprague-Dawley rats to model ischemic stroke, and tissues from the ischemic penumbra were obtained to evaluate autophagy level and microglial inflammatory activity. MCAO rats were administered 3-methyladenine(autophagy inhibitor) or Tat-Beclin 1(autophagy inducer). Western blot assays were conducted to quantify expression of Beclin-1, nuclear factor kappa Bp65(NF-κB), light chain 3B(LC3B), and CX3CL1 in ischemic penumbra. Moreover, immunofluorescence staining was performed to quantify numbers of LC3B-, CX3CL1-, and Iba-1-positive cells in ischemic penumbra. In addition, enzyme linked immunosorbent assays were utilized to analyze concentrations of tumor necrosis factor alpha(TNF-α), interleukin 6(IL-6), interleukin 1 beta(IL-1β), and prostaglandin E2(PGE2). A dry/wet weight method was used to detect brain water content, while 2,3,5,-triphenyltetrazolium chloride staining was utilized to measure infarct volume. The results demonstrated that autophagy signaling(Beclin-1 and LC3B expression) in penumbra was prominently activated by MCAO, while CX3CL1 expression on autophagic neurons was significantly reduced and microglial inflammation was markedly activated. However, after inhibition of autophagy signaling with 3-methyladenine, CX3CL1 expression on neurons was obviously increased, whereas Iba-1 and NF-κB expression was downregulated; TNF-α, IL-6, IL-1β, and PGE2 levels were decreased; and cerebral edema was obviously mitigated. In contrast, after treatment with the autophagy inducer Tat-Beclin 1, CX3CL1 expression on neurons was further reduced; Iba-1 and NF-κB expression was increased; TNF-α, IL-6, IL-1β, and PGE2 levels were enhanced; and cerebral edema was aggravated. | 7 | Relative Contributions of Boundary-Layer Meteorological Factors to the Explosive Growth of PM2.5 during the Red-Alert Heavy Pollution Episodes in Beijing in December 2016显示文摘Based on observations of urban mass concentration of fine particulate matter smaller than 2.5 μm in diameter(PM_(2.5)), ground meteorological data, vertical measurements of winds, temperature, and relative humidity(RH), and ECMWF reanalysis data, the major changes in the vertical structures of meteorological factors in the boundary layer(BL) during the heavy aerosol pollution episodes(HPEs) that occurred in winter 2016 in the urban Beijing area were analyzed. The HPEs are divided into two stages: the transport of pollutants under prevailing southerly winds, known as the transport stage(TS), and the PM_(2.5) explosive growth and pollution accumulation period characterized by a temperature inversion with low winds and high RH in the lower BL, known as the cumulative stage(CS). During the TS, a surface high lies south of Beijing, and pollutants are transported northwards. During the CS, a stable BL forms and is characterized by weak winds, temperature inversion, and moisture accumulation. Stable atmospheric stratification featured with light/calm winds and accumulated moisture(RH > 80%) below 250 m at the beginning of the CS is closely associated with the inversion, which is strengthened by the considerable decrease in near-surface air temperature due to the interaction between aerosols and radiation after the aerosol pollution occurs. A significant increase in the PLAM(Parameter Linking Aerosol Pollution and Meteorological Elements) index is found, which is linearly related to PM mass change. During the first 10 h of the CS, the more stable BL contributes approximately 84% of the explosive growth of PM_(2.5) mass. Additional accumulated near-surface moisture caused by the ground temperature decrease, weak turbulent diffusion, low BL height, and inhibited vertical mixing of water vapor is conducive to the secondary aerosol formation through chemical reactions, including liquid phase and heterogeneous reactions, which further increases the PM_(2.5) concentration levels. The mechanism of the tension control system is analyzed and the numerical model is set up. The compensation technique of the radius of the scroll is analyzed. | 9 | Size distribution of chemical elements and their source apportionment in ambient coarse, fine, and ultrafine particles in Shanghai urban summer atmosphere显示文摘Ambient coarse particles (diameter 1.8-10 μm), fine particles (diameter 0.1-1.8 μm), and ultrafine particles (diameter < 0.1 μm) in the atmosphere of the city of Shanghai were sampled during the summer of 2008 (from Aug 27 to Sep 08). Microscopic characterization of the particles was investigated by scanning electron microscopy coupled with energy dispersive X-ray spectroscopy (SEM/EDX). Mass concentrations of Si, P, S, Cl, K, Ca, Ti, V, Cr, Mn, Fe, Ni, Cu, Zn, As, Se, Br, Rb, Sr, and Pb in the size-resolved particles were quantified by using synchrotron radiation X-ray fluorescence (SRXRF). Source apportionment of the chemical elements was analyzed by means of an enrichment factor method. Our results showed that the average mass concentrations of coarse particles, fine particles and ultrafine particles in the summer air were 9.38 ± 2.18, 8.82 ± 3.52, and 2.02 ± 0.41 μg/m3, respectively. The mass percentage of the fine particles accounted for 51.47% in the total mass of PM10, indicating that fine particles are the major component in the Shanghai ambient particles. SEM/EDX results showed that the coarse particles were dominated by minerals, fine particles by soot aggregates and fly ashes, and ultrafine particles by soot particles and unidentified particles. SRXRF results demonstrated that crustal elements were mainly distributed in the coarse particles, while heavy metals were in higher proportions in the fine particles. Source apportionment revealed that Si, K, Ca, Fe, Mn, Rb, and Sr were from crustal sources, and S, Cl, Cu, Zn, As, Se, Br, and Pb from anthropogenic sources. Here, we report a new single-phase bodycentered cubic(BCC) structured Ti_2 ZrHfV_(0.5)Mo_(0.2) HEA possessing excellent irradiation resistance, i.e.,scarcely irradiation hardening and abnormal lattice constant reduction after helium-ion irradiation,which is completely different from conventional alloys. This is the first time to report the abnormal XRD phenomenon of metallic alloys and almost no hardening after irradiation. These excellent properties make it to be a potential candidate material used as core components in next-generation nuclear reactors. | 11 | Climate and litter C/N ratio constrain soil organic carbon accumulation显示文摘Soil organic carbon(SOC) plays critical roles in stabilizing atmospheric CO2 concentration, but the mechanistic controls on the amount and distribution of SOC on global scales are not well understood. In turn, this has hampered the ability to model global C budgets and to find measures to mitigate climate change. Here, based on the data from a large field survey campaign with 2600 plots across China’s forest ecosystems and a global collection of published data from forested land, we find that a low litter carbon-to-nitrogen ratio(C/N) and high wetness index(P/PET, precipitation-to-potentialevapotranspiration ratio) are the two factors that promote SOC accumulation, with only minor contributions of litter quantity and soil texture. The field survey data demonstrated that high plant diversity decreased litter C/N and thus indirectly promoted SOC accumulation by increasing the litter quality. We conclude that any changes in plant-community composition, plant-species richness and environmental factors that can reduce the litter C/N ratio, or climatic changes that increase wetness index, may promote SOC accumulation. | 14 | Magnesium isoglycyrrhizinate inhibits inflammatory response through STAT3 pathway to protect remnant liver function显示文摘AIM: To investigate the protective effect of magnesium isoglycyrrhizinate(Mg IG) on excessive hepatectomy animal model and its possible mechanism.METHODS: We used the standard 90% hepatectomy model in Sprague-Dawley rats developed using the modified Emond's method,in which the left,middle,right upper,and right lower lobes of the liver were removed. Rats with 90% liver resection were divided into three groups,and were injected intraperitoneally with 3 m L saline(control group),30 mg/kg(low-dose group) and 60 mg/kg(high-dose group) of Mg IG,respectively. Animals were sacrificed at various time points and blood was drawn from the vena cava. Biochemical tests were performed with an automatic biochemical analyzer for the following items: serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),glutamyl endopeptidase,total bilirubin(TBil),direct bilirubin(DBil),total protein,albumin,blood glucose(Glu),hyper-sensitivity C-reactive protein,prothrombin time(PT),and thrombin time(TT). Postoperative survival time was observed hourly until death. Hepatocyte regeneration was analyzed by immunohistochemistry. Serum inflammatory cytokines(IL-1,IL-6,IL-10,and i NOS) was analyzed by ELISA. STAT3 protein and m RNA were analyzed by Western blot and quantitative reversetranscription PCR,respectively.RESULTS: The high-dose group demonstrated a significantly prolonged survival time,compared with both the control and the low-dose groups(22.0 ± 4.7 h vs 8.9 ± 2.0 vs 10.3 ± 3.3 h,P = 0.018). There were significant differences among the groups in ALT,Glu and PT levels starting from 6 h after surgery. The ALT levels were significantly lower in the Mg IG treated groups than in the control group. Both Glu and PT levels were significantly higher in the Mg IG treated groups than in the control group. At 12 h,ALT,AST,TBil,DBil and TT levels showed significant differences between the Mg IG treated groups and the control group. No significant differences in hepatocyte regeneration were found. Compared to the control group,the high-dose group showed a significantly increase in serum inflammatory cytokines IL-1 and IL-10,and a decrease in IL-6. Both STAT3 protein and m RNA levels were significantly lower in the Mg IG treated groups than in the control group at 6 h,12 h,and 18 h after surgery.CONCLUSION: High-dose Mg IG can extend survival time in rats after excessive hepatectomy. | 15 | Relationship between Two Blood Stasis Syndromes and Inflammatory Factors in Patients with Acute Coronary Syndrome显示文摘Objective: To investigate the relationship between inflammatory factors and two Chinese medicine(CM) syndrome types of qi stagnation and blood stasis(QSBS) and qi deficiency and blood stasis(QDBS) in patients with acute coronary syndrome(ACS). Methods: Sixty subjects with ACS, whose pathogenesis changes belongs to qi disturbance blood stasis syndrome, were divided into 2 groups: 30 in the QSBS group and 30 in the QDBS group. The comparative analysis on them was carried out through comparing general information, coronary angiography and inflammatory factors including intracellular adhesion molecule-1(ICAM-1), chitinase-3-like protein 1(YKL-40) and lipoprotein-associated phospholipase A2(Lp-PLA2). Results: Compared with the QSBS group, Lp-PLA2 and YKL-40 levels in the QDBS group showed no-significant difference(P>0.05); ICAM-1 was significantly higher in the QDBS group than in the QSBS group in the pathological processes of qi disturbance and blood stasis syndrome of ACS(P<0.05). | 16 | CFD-DEM simulation of spouting of corn-shaped particles显示文摘Three dimensionally coupled computational fluid dynamics (CFD) and discrete element method (DEM) were used to investigate the flow of corn-shaped particles in a cylindrical spouted bed with a conical base. The particle motion was modeled by the DEM, and the gas motion by the k-ε two-equation turbulent model. A two-way coupling numerical iterative scheme was used to incorporate the effects of gas-particle interactions in terms of momentum exchange. The corn-shaped particles were constructed by a multisphere method. Drag force, contact force, Saffman lift force, Magnus lift force, and gravitational force acting on each individual particle were considered in establishing the mathematical modeling. Calculations were carried out in a cylindrical spouted bed with an inside diameter of 200 mm, a height of 700 mm, and a conical base of 60 . Comparison of simulations with experiments showed the availability of the multi-sphere method in simulating spouting action with corn-shaped particles, but it depended strongly on the number and the arrangement of the spherical elements. Gas-solid flow patterns, pressure drop, particle velocity and particle concentration at various spouting gas velocity were discussed. The results showed that particle velocity reaches a maximum at the axis and then decreases gradually along the radial direction in the whole bed. Particle concentration increases along the radial direction in the spout region but decreases in the fountain region, while it is nearly constant in the annulus region. | 17 | BMAL1 regulates mitochondrial fission and mitophagy through mitochondrial protein BNIP3 and is critical in the development of dilated cardiomyopathy显示文摘Dysregulation of circadian rhythms associates with cardiovascular disorders.It is known that deletion of the core circadian gene Bma/1 in mice causes dilated car-diomyopathy.However,the biological rhythm regulation system in mouse is very different from that of humans.Whether BMAL1 plays a role in regulating human heart function remains unclear.Here we generated a BMAL1 knockout human embryonic stem cell(hESC)model and further derived human BMAL1 deficient cardiomy-ocytes.We show that BMAL1 deficient hESC-derived cardiomyocytes exhibited typical phenotypes of dilated cardiomyopathy including attenuated contractility,cal-cium dysregulation,and disorganized myofilaments.In addition,mitochondrial fission and mitophagy were suppressed in BMAL1 deficient hESC-cardiomyocytes,which resulted in significantly attenuated mitochondrial oxidative phosphorylation and compromised cardiomy-ocyte function.We also found that BMAL1 binds to the E-box element in the promoter region of BNIP3 gene and specifically controls BNIP3 protein expression.BMAL1 knockout directly reduced BNIP3 protein level,causing compromised mitophagy and mitochondria dysfunction and thereby leading to compromised cardiomyocyte function.Our data indicated that the core circadian gene S/VMLf is critical for normal mitochondria activities and cardiac function.Circadian rhythm disruption may directly link to compromised heart function and dilated cardiomyopathy in humans. | Ermin Li Xiuya Li Jie Huang Chen Xu Qianqian Liang Kehan Ren Aobing Bai Chao Lu Ruizhe Qian Ning Sun | 2020 | Protein & Cell2020,11,9: | 17 |
| 18 | Cancer incidence and mortality in Shandong province,2012显示文摘Objective: Population-based cancer registration data in 2012 from all available cancer registries in Shandong province were collected by Shandong Center for Disease Control and Prevention(SDCDC). SDCDC estimated the numbers of new cancer cases and cancer deaths in Shandong province with compiled cancer incidence and mortality rates.Methods: In 2015, there were 21 cancer registries submitted data of cancer incidence and deaths occurred in 2012. All the data were checked and evaluated based on the National Central Cancer Registry(NCCR) criteria of data quality. Qualified data from 15 registries were used for cancer statistics analysis as provincial estimation. The pooled data were stratified by area(urban/rural), gender, age group(0, 1-4, 5-9, 10-14, …, 85+ years) and cancer type. New cancer cases and deaths were estimated using age-specific rates and corresponding provincial population in 2012. The Chinese census data in 2000 and Segi's population were applied for age-standardized rates. All the rates were expressed per 100,000 person-year.Results: Qualified 15 cancer registries(4 urban and 11 rural registries) covered 17,189,988 populations(7,486,039 in urban and 9,703,949 in rural areas). The percentage of cases morphologically verified(MV%) and death certificate-only cases(DCO%) were 66.12% and 2.93%, respectively, and the mortality to incidence rate ratio(M/I) was 0.60. A total of 253,060 new cancer cases and 157,750 cancer deaths were estimated in Shandong province in 2012. The incidence rate was 263.86/100,000(303.29/100,000 in males, 223.23/100,000 in females), the age-standardized incidence rates by Chinese standard population(ASIRC) and by world standard population(ASIRW) were 192.42/100,000 and 189.50/100,000 with the cumulative incidence rate(0-74 years old) of 22.07%. The cancer incidence, ASIRC and ASIRW in urban areas were 267.64/100,000, 195.27/100,000 and 192.02/100,000 compared to 262.32/100,000, 191.26/100,000 and 188.48/100,000 in rural areas, respectively. The cancer mortality was 164.47/100,000(207.42/100,000 in males, 120.23/100,000 in females), the age-standardized incidence rates by Chinese standard population(ASMRC) and by world standard population(ASMRW) were 117.54/100,000 and 116.90/100,000, and the cumulative mortality rate(0-74 years old) was 13.53%. The cancer mortality, ASMRC and ASMRW were 141.59/100,000, 101.17/100,000 and 100.33/100,000 in urban areas, and 173.79/100,000, 124.20/100,000 and 123.64/100,000 in rural areas, respectively. Cancers of the lung, stomach, liver, esophagus, colorectum, female breast, brain, leukemia, bladder and pancreas were the most common cancers, accounting for about 82.12% of all cancer new cases. Lung cancer, stomach cancer, liver cancer, esophageal cancer, colorectal cancer, female breast cancer, pancreatic cancer, brain tumor, leukemia and lymphoma were the leading causes of cancer death, accounting for about 89.01% of all cancer deaths. The cancer spectrum showed difference between urban and rural, males and females both in incidence and mortality rates.Conclusions: Cancer surveillance information in Shandong province is making great progress with the increasing number of cancer registries, population coverage and the improving data quality. Cancer registration plays a fundamental role in cancer control by providing basic information on population-based cancer incidence, mortality, survival and time trend. | 19 | Identification Standard for ENSO Events and Its Application to Climate Monitoring and Prediction in China显示文摘The El Ni?o–Southern Oscillation(ENSO)reflects anomalous variations in the sea surface temperature(SST)and atmospheric circulation over the tropical central–eastern Pacific.It remarkably impacts on weather and climate worldwide,so monitoring and prediction of ENSO draw intensive research.However,there is not yet a unique standard internationally for identifying the timing,intensity,and type of ENSO events.The National Climate Center of China Meteorological Administration(NCC/CMA)has led the effort to establish a national identification standard of ENSO events,which was officially endorsed by the National Standardization Administration of China and implemented operationally in NCC/CMA in 2017.In this paper,two key aspects of this standard are introduced.First,the Ni?o3.4 SST anomaly index,which is well-recognized in the international ENSO research community and used operationally in the US,has replaced the previous Ni?o Z index and been used to identify the start,end,and peak times,and intensity of ENSO events.Second,two new indices—the eastern Pacific ENSO(EP)index and the central Pacific ENSO(CP)index,based on the SST conditions in Ni?o3 and Ni?o4 region respectively,are calculated to first determine the ENSO type before monitoring and assessing the impacts of ENSO on China’s climate.With this standard,all historical ENSO events since 1950 are consistently re-identified;their distinct properties are diagnosed and presented;and the impacts of ENSO events under different types on China’s climate are re-assessed.This standard is also employed to validate the intensity,grade,and type of the ENSO events predicted by the NCC/CMA operational ENSO prediction system.The new standard and the thus derived unified set of re-analyzed historical ENSO events and associated information provide a good reference for better monitoring and prediction of future ENSO events. | Hong-Li REN Bo LU Jianghua WAN Ben TIAN Peiqun ZHANG | 2018 | Journal of Meteorological Research2018,32,6: | 16 |
| 20 | Impact of hydrothermal process on the nutrient ingredients of restaurant garbage显示文摘In order to recover the nutrient resource from restaurant garbage, a complete trial with 2 factors on 5 levels of experiments was carried out. The temperature and heating time are the main factors influencing on hydrothermal process (HP) by which improves the degradability and digestibility of the restaurant garbage favorably to make animal feeds or fertilizer. The results showed the variation of protein, saccharide, and oil in the garbage. It showed that protein dissolved and liquefied during hydrothermal process, which made organic nitrogen in solid phase transfer to liquid phase. After heating at 180℃ for 60 min, organic nitrogen in liquid phase began to transform into ammonia. It also showed that hydrothermal process could promote the dextrinization, dissolution of the starch and its hydrolysis to reducing sugar, due to that starch in the restaurant garbage decreases and reducing sugar increases. When the temperature reached 140℃, the reducing sugar started to decrease due to chemical reactions. The cellulose was stable at 100—180℃. The floatable oil increased markedly in the hydrothermal process. The suitable condition for de-oil was observed at 160℃ heating for 80 min.
