1Gα_s Relays Sphingosine-1-Phosphate Receptor 1 Signaling to Stabilize Vascular Endothelial-Cadherin at Endothelial Junctions to Control Mouse Embryonic Vascular Integrity显示文摘Sphingosine-1-phosphate receptor 1(S1PR1),a G protein-coupled receptor(GPCR),controls vascular stability by stabilizing vascular endothelial(VE)-cadherin junctional localization and inhibiting vascular endothelial growth factor receptor 2(VEGFR2) signaling.However,the molecular mechanisms that link S 1PR1 signaling to intracellular effectors remain unknown,In this study,we demonstrate that the heterotrimeric G protein subfamily member Gα_s,encoded by GNAS,acts as a relay mediator of S1PR1 signaling to control vascular integrity by stabilizing VE-cadherin at endothelial junctions.The endothelial cell-specific deletion of Gαs in mice causes early embryonic lethality with massive hemorrhage and a disorganized vasculature.The immunostalning results revealed that Gα_s deletion remarkably reduces the junctional localization of VE-cadherin,whereas the mural cell coverage of the vessels is not impaired.In addition,we found that Gαs depletion blocks the S1PRl-activation induced VE-cadherin stabilization at junctions,supporting that Gα_s acts downstream of S1PR1 signaling,Thus,our results demonstrate that Gα_s is an essential mediator to relay S1PR1 signaling and maintain vascular integrity.Ximing Shao Ke Liu Yi Fan Zhihao Ding Min Chen Minyan Zhu Lee S.Weinstein Hongchang Li Huashun Li 2015Journal of Genetics and Genomics2015,42,11:8

4Practical high-performance lead-free piezoelectrics:structural flexibility beyond utilizing multiphase coexistence显示文摘Due to growing concern for the environment and human health, searching for high-performance lead-free piezoceramics has been a hot topic of scientific and industrial research. Despite the significant progress achieved toward enhancing piezoelectricity, further efforts should be devoted to the synergistic improvement of piezoelectricity and its thermal stability. This study provides new insight into these topics.A new KNN-based lead-free ceramic material is presented, which features a large piezoelectric coefficient(d33) exceeding 500 pC/N and a high Curie temperature(Tc) of ~200℃. The superior piezoelectric response strongly relies on the increased composition-induced structural flexibility due to lattice softening and decreased unit cell distortion. In contrast to piezoelectricity anomalies induced via polymorphic transition, this piezoelectricity enhancement is effective within a broad temperature range rather than a specific small range. 5Resistance mechanisms and reprogramming of microorganisms for efficient biorefinery under multiple environmental stresses显示文摘In the fermentation process of biorefinery,industrial strains are normally subjected to adverse environmental stresses,which leads to their slow growth,yield decline,a substantial increase in energy consumption,and other negative consequences,which ultimately seriously hamper the development of biorefinery.How to minimize the impact of stress on microorganisms is of great significance.This review not only reveals the damaging effects of different environmental stresses on microbial strains but also introduces commonly used strategies to improve microbial tolerance,including adaptive evolution,reprogramming of the industrial host based on genetic circuits,global transcription machinery engineering(gTME)and bioprocess integration.Furthermore,by integrating the advantages of these strategies and reducing the cost of system operation,the tolerance of industrial strains,combined with production efficiency and process stability,will be greatly improved,and the development prospects of biorefinery will be more widespread.Ke Xu Yun Seo Lee Jun Li Chun Li 2019Synthetic and Systems Biotechnology2019,4,2:4
6RELATIVE LEVELS OF INDOOR AND OUTDOOR PARTICLE NUMBER CONCENTRATIONS IN A RESIDENTIAL BUILDING IN XI'AN显示文摘The time series of indoor and outdoor particle number concentrations in a naturally ventilated residential building in Xi’an were tested simultaneously for 7 days in summer. The relationships between indoor and outdoor con-centrations were examined and discussed, and linear regression analysis was employed to correlate the indoor and outdoor concentrations. The diurnal cycles of indoor and outdoor particle concentrations of different particle size ranges all showed positive correlations between indoor and outdoor number concentrations. 7Critical size limit of biodegradable nanopartides for enhanced lymph node trafficking and paracortex penetration显示文摘Lymph node (LN) targeti ng through interstitial drain age of nan oparticles (NPs) is an attractive strategy to stimulate a pote nt immune respo nse, as LNs are the primary site for lymphocyte priming by antigen presenting cells (APCs) and triggering of an adaptive immune response. NP size has been shown to influence the efficiency of LN-targeting and retention after subcutaneous injection. For clinical translation, biodegradable NPs are preferred as carrier for vaccine delivery. However, the selective 'size gateM for effective LN-drainage, particularly the kinetics of LN trafficking, is less well defined. This is partly due to the challenge in generating size-controlled NPs from biodegradable polymers in the sub-100-nm range. Here, we report the preparation of three sets of poly(lactic-co-glycolic)-b-poly(ethylene-glycol)(PLGA-b-PEG) NPs with number average diameters of 20-, 40-, and 100-nm and narrow size distributions using flash nanoprecipitation. Using NPs labeled with a near-infrared dye, we showed that 20-nm NPs drain rapidly across proximal and distal LNs following subcutaneous inoculation in mice and are retai ned in LNs more effectively than NPs with a nu mber average diameter of 40-nm. The drain age of 100-nm NPs was n egligible. Furthermore, the 20-nm NPs showed the highest degree of penetration around the paracortex region and had enhanced access to dendritic cells in the LNs. Together, these data confirmed that small, size-controlled PLGA-b-PEG NPs at the lower threshold of about 30-nm are most effective for LN trafficking, retention, and APC uptake after s.c. administration.
