1Aeroservoelastic modeling and analysis of a canard-configured air-breathing hypersonic vehicles显示文摘Air-breathing hypersonic vehicles (HSVs) are typically characterized by interactions of elasticity, propulsion and rigid-body flight dynamics, which may result in intractable aeroservoelastic problem. When canard is added, this problem would be even intensified by the introduction of low-frequency canard pivot mode. This paper concerns how the aeroservoelastic stability of a canard-configured HSV is affected by the pivot stiffnesses of all-moveable horizontal tail (HT) and canard. A wing/pivot system model is developed by considering the pivot torsional flexibility, fuselage vibration, and control input. The governing equations of the aeroservoelastic system are established by combining the equations of rigid-body motion, elastic fuselage model, wing/pivot system models and actuator dynamics. An unsteady aerodynamic model is developed by steady Shock-Expansion theory with an unsteady correction using local piston theory. A baseline controller is given to provide approximate inflight characteristics of rigid-body modes. The vehicle is trimmed for equilibrium state, around which the linearized equations are derived for stability analysis. A comparative study of damping ratios, closed-loop poles and responses are conducted with varying controller gains and pivot stiffnesses. 5Association of systemic inflammation and body mass index with survival in patients with resectable gastric or gastroesophageal junction adenocarcinomas显示文摘Objective:The systemic inflammation index and body mass index(BMI)are easily accessible markers that can predict mortality.However,the prognostic value of the combined use of these two markers remains unclear.The goal of this study was therefore to evaluate the association of these markers with outcomes based on a large cohort of patients with gastric cancer.Methods:A total of 2,542 consecutive patients undergoing radical surgery for gastric or gastroesophageal junction adenocarcinoma between 2009 and 2014 were included.Systemic inflammation was quantified by the preoperative neutrophil-to-lymphocyte ratio(NLR).High systemic inflammation was defined as NLR≥3,and underweight was defined as BMI<18.5 kg/m2.Results:Among 2,542 patients,NLR≥3 and underweight were common[627(25%)and 349(14%),respectively].In the entire cohort,NLR≥3 or underweight independently predicted overall survival(OS)[hazard ratio(HR):1.236,95%confidence interval(95%CI):1.069–1.430;and HR:1.600,95%CI:1.350–1.897,respectively]and recurrence-free survival(RFS)(HR:1.230,95%CI:1.054–1.434;and HR:1.658,95%CI:1.389–1.979,respectively).Patients with both NLR≥3 and underweight(vs.neither)had much worse OS(HR:2.445,95%CI:1.853–3.225)and RFS(HR:2.405,95%CI:1.802–3.209).Furthermore,we observed similar results in subgroup analyses according to pathological stage,age,and postoperative chemotherapy.Conclusions:Our results showed that preoperative elevated NLR and decreased BMI had a significant negative effect on survival.Underweight combined with severe inflammation could enhance prognostication.Taking active therapeutic measures to reduce inflammation and increase nutrition may help improve outcomes.Xianchun Gao Yanan Pan Weili Han Caie Hu Chenchen Wang Ling Chen Yong Guo Yupeng Shi Yan Pan Huahong Xie Liping Yao Jianjun Yang Jianyong Zheng Xiaohua Li Xiaonan Liu Liu Hong Jipeng Li Mengbin Li Gang Ji Zengshan Li Jielai Xia Qingchuan Zhao Daiming Fan Kaichun Wu Yongzhan Nie 2021Cancer Biology & Medicine2021,18,1:2
6Gold nanoparticle-directed autophagy intervention for antitumor immunotherapy via inhibiting tumor-associated macrophage M2 polarization显示文摘Tumor-associated macrophages(TAMs),one of the dominating constituents of tumor microenvironment,are important contributors to cancer progression and treatment resistance.Therefore,regulation of TAMs polarization from M2 phenotype towards M1 phenotype has emerged as a new strategy for tumor immunotherapy.Herein,we successfully initiated antitumor immunotherapy by inhibiting TAMs M2 polarization via autophagy intervention with polyethylene glycol-conjugated gold nanoparticles(PEG-Au NPs).PEG-Au NPs suppressed TAMs M2 polarization in both in vitro and in vivo models,elicited antitumor immunotherapy and inhibited subcutaneous tumor growth in mice.As demonstrated by the m RFP-GFP-LC3 assay and analyzing the autophagy-related proteins(LC3,beclin1 and P62),PEGAu NPs induced autophagic flux inhibition in TAMs,which is attributed to the PEG-Au NPs induced lysosome alkalization and membrane permeabilization.Besides,TAMs were prone to polarize towards M2phenotype following autophagy activation,whereas inhibition of autophagic flux could reduce the M2polarization of TAMs.Our results revealed a mechanism underlying PEG-Au NPs induced antitumor immunotherapy,where PEG-Au NPs reduce TAMs M2 polarization via induction of lysosome dysfunction and autophagic flux inhibition.This study elucidated the biological effects of nanomaterials on TAMs polarization and provided insight into harnessing the intrinsic immunomodulation capacity of nanomaterials for effective cancer treatment.Siyue Zhang Fangyuan Xie Kaichun Li He Zhang You Yin Yuan Yu Guangzhao Lu Shihao Zhang Yan Wei Ke Xu Yan Wu Hong Jin Lan Xiao Leilei Bao Can Xu Yulin Li Ying Lu Jie Gao 2022Acta Pharmaceutica Sinica B2022,12,7:2

11Functional identification of lncRNAs in sweet cherry(Prunus avium)pollen tubes via transcriptome analysis using single-molecule long-read sequencing显示文摘Sweet cherry(Prunus avium)is a popular fruit with high nutritional value and excellent flavor.Although pollen plays an important role in the double fertilization and subsequent fruit production of this species,little is known about its pollen tube transcriptome.In this study,we identified 16,409 transcripts using single-molecule sequencing.After filtering 292 transposable elements,we conducted further analyses including mRNA classification,gene function prediction,alternative splicing(AS)analysis,and long noncoding RNA(lncRNA)identification to gain insight into the pollen transcriptome.The filtered transcripts could be matched with 3,438 coding region sequences from the sweet cherry genome.GO and KEGG analyses revealed complex biological processes during pollen tube elongation.A total of 2043 AS events were predicted,7 of which were identified in different organs,such as the leaf,pistil and pollen tube.Using BLASTnt and the Coding-Potential Assessment Tool(CPAT),we distinguished a total of 284 lncRNAs,among which 154 qualified as natural antisense transcripts(NATs).As the NATs could be the reverse complements of coding mRNA sequences,they might bind to coding sequences.Antisense transfection assays showed that the NATs could regulate the expression levels of their complementary sequences and even affect the growth conditions of pollen tubes.In summary,this research characterizes the transcripts of P.avium pollen and lays the foundation for elucidating the physiological and biochemical mechanisms underlying sexual reproduction in the male gametes of this species.Yang Li Chuanbao Wu Chunsheng Liu Jie Yu Xuwei Duan Wenqi Fan Jing Wang Xiaoming Zhang Guohua Yan Tianzhong Li Kaichun Zhang 2019Horticulture Research2019,6,1:1
