| 1 | A β-glucosidase-producing M-2 strain: Isolation from cow dung and fermentation parameter optimization for flaxseed cake显示文摘Flaxseed cake contains cyanogenic glucosides, which can be metabolized into hydrocyanic acid in an animal's body, leading to asphyxia poisoning in cells. Beta-glucosidase is highly efficient in degrading cyanogenic glucosides. The Cattle may have b-glucosidase-producing strains in the intestinal tract after eating small amounts of flaxseed cake for a long time. This study aimed to isolate of a strain from cow dung that produces b-glucosidase with high activity and can significantly reduce the amount of cyanogenic glucosides. We used cow dung as the microflora source and an esculin agar as the selective medium. After screening with 0.05% esculin and 0.01% ferric citrate, we isolated 5 strains producing high amounts of b-glucosidase. In vitro flaxseed cake fermentation was fermented by these 5 strains, in which the strain M-2 exerted the best effect(P < 0.05). The strain M-2 was identified as Lichtheimia ramosa and used as the fermentation strain to optimize the fermentation parameters by a single factor analysis and orthogonal experimental design. The optimum condition was as follows: inoculum size3%, water content 60%, time 144 h, and temperature 32℃. Under this condition, the removal rate of cyanogenic glucosides reached 89%, and crude protein increment reached 44%. | 2 | Porous Ruthenium Selenide Nanoparticle as a Peroxidase Mimic for Glucose Bioassay显示文摘Nanozyme is a promising field that offers the substitution for natural enzymes using various nanomaterials.Various nanoma-terials with peroxidase-like activity were investigated.Among them,transition metal chalcogenides were explored as promis-ing nanozymes due to their excellent enzyme-mimicking activities.However,ruthenium selenide has not been studied as a peroxidase mimic because of the difficulty for synthesis.Herein,we prepared ruthenium selenide nanomaterial with ordered mesoporous structure(P-RuSe_(2))employing KIT-6 silica as the template.The composition and structure of P-RuSe_(2) were fully characterized.Further,its peroxidase-like activity was investigated.P-RuSe_(2) possessed excellent peroxidase-mimicking activ-ity,which catalyzed the oxidation of peroxidase substrates,including 3,3′,5,5′-tetramethylbenzidine,o-phenylenediamine,and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)in the presence of H_(2)O_(2).Moreover,P-RuSe_(2) exhibited higher peroxidase-like activity when compared with several representative nanozymes as well as bulk RuSe_(2).To demonstrate its potential applications,the colorimetric detection systems for H_(2)O_(2) and glucose were successfully constructed based on P-RuSe_(2) nanozyme. | Wen Cao Junshu Lin Faheem Muhammad Quan Wang Xiaoyu Wang Zhangping Lou Hui Wei | 2019 | Journal of Analysis and Testing2019,3,3: | 1 |
| 3 | Intravitreal brimonidine inhibits formdeprivation myopia in guinea pigs显示文摘Background:The use of ocular hypotensive drugs has been reported to attenuate myopia progression.This study explores whether brimonidine can slow myopia progression in the guinea pig form-deprivation(FD)model.Methods:Three-week-old pigmented male guinea pigs(Cavia porcellus)underwent monocular FD and were treated with 3 different methods of brimonidine administration(eye drops,subconjunctival or intravitreal injections).Four different concentrations of brimonidine were tested for intravitreal injection(2μg/μL,4μg/μL,20μg/μL,40μg/μL).All treatments continued for a period of 21 days.Tonometry,retinoscopy,and A-scan ultrasonography were used to monitor intraocular pressure(IOP),refractive error and axial length(AL),respectively.On day 21,guinea pigs were sacrificed for RNA sequencing(RNA-seq)to screen for associated transcriptomic changes.Results:The myopia model was successfully established in FD animals(control eye vs.FD eye,respectively:refraction at day 20,0.97±0.18 D vs.−0.13±0.38 D,F=6.921,P=0.02;AL difference between day 0 and day 21,0.29±0.04mm vs.0.45±0.03 mm,F=11.655,P=0.004).Among the 3 different brimonidine administration methods,intravitreal injection was the most effective in slowing myopia progression,and 4μg/μL was the most effective among the four different concentrations of brimonidine intravitreal injection tested.The AL and the refraction of the brimonidine intravitreal injection group was significantly shorter or more hyperopic than those of other 2 groups.Fourμg/μL produced the smallest difference in AL and spherical equivalent difference values.FD treatment significantly increased the IOP.IOP was significantly lower at 1 day after intravitreal injections which was the lowest in FD eye of intravitreal injection of brimonidine.At day 21,gene expression analyses using RNA-seq showed upregulation of Col1a1 and Mmp2 expression levels by intravitreal brimonidine.Conclusions:Among the 3 different administration methods,intravitreal injection of brimonidine was the most effective in slowing myopia progression in the FD guinea pig model.Intravitreal brimonidine at 4μg/μL significantly reduced the development of FD myopia in guinea pigs.Expression levels of the Col1a1 and Mmp2 genes were significantly increased in the retinal tissues of the FD-Inj-Br group. | Yifang Yang Junshu Wu Defu Wu Qi Wei Tan Zhong Jun Yang Xiaowei Yang Meizhen Zeng Xingwu Zhong | 2023 | Eye and Vision2023,9,5: | 0 |

The polymers show broad absorption spectra and low-lying frontier energy levels due to both aromatic backbone and side units.
