1Six-year outcomes in neovascular age-related macular degeneration with ranibizumab显示文摘● AIM: To evaluate the outcomes of ≥6y ranibizumab therapy in neovascular age-related macular degeneration(AMD).● METHODS: HELIX was a retrospective, observational effeciveness study using medical records of patients treated in three clinics in Belgium. Patients had neovascular AMD and were initially treated with intravitreal ranibizumab(0.5 mg) between November 1, 2007 and October 31, 2008, had ≥6y of data available, and were treated on an ongoing, as-needed basis. Outcomes included best-corrected visual acuity(BCVA) and central retinal thickness(CRT).● RESULTS: The sample consisted of 88 eyes from 69 patients. Mean age was 76.4±6.5y, most patients were female(62.3%). Most eyes(62.5%) were treatment-naive, 33 previously treated eyes had received predominantly other anti-vascular endothelial growth factor agents and verteporfin. Mean baseline BCVA was 57.4±12.7 ETDRS letters and CRT was 292±86 μm. On average, patients received 20.6±11.9 ranibizumab injections over the ≥6y. Intervals between injections were on average 12.7±16.1wk. Mean change in BCVA from baseline to last observation for the sample was less than one letter(-0.9±17.3 letters), with an average loss of-3.2±15.6 letters in previously treated eyes versus a gain of 0.6±18.4 letters in treatment-na?ve eyes. When considering a loss of <15 letters over 6y as stabilization of disease, 75.9% of all eyes showed a positive(improvement or stabilization) outcome. Mean change in CRT from baseline to last observation for the sample was-26.9±148.4 μm with the greatest reduction observed in treatment-naive eyes.● CONCLUSION: This retrospective study of 69 neovascular AMD patients treated for ≥6y with ranibizumab demonstrates long-term visual stabilization. 2Aging and menopause reprogram osteoclast precursors for aggressive bone resorption显示文摘Women gradually lose bone from the age of〜35 years,but around menopause,the rate of bone loss escalates due to increasing bone resorption and decreasing bone formation levels,rendering these individuals more prone to developing osteoporosis.The increased osteoclast activity has been linked to a reduced estrogen level and other hormonal changes.However,it is unclear whether intrinsic changes in osteoclast precursors around menopause can also explain the increased osteoclast activity.Therefore,we set up a protocol in which CD14f blood monocytes were isolated from 49 female donors(40-66 years old).Cells were differentiated into osteoclasts,and data on differentiation and resorption activity were collected.Using multiple linear regression analyses combining in vitro and in vivo data,we found the following:(1)age and menopausal status correlate with aggressive osteoclastic bone resorption in vitro;(2)the type I procollagen N-terminal propeptide level in vivo inversely correlates with osteoclast resorption activity in vitro;(3)the protein level of mature cathepsin K in osteoclasts in vitro increases with age and menopause;and(4)the promoter of the gene encoding the dendritic cell-specific transmembrane protein is less methylated with age.We conclude that monocytes are'reprogrammed'in vivo,allowing them to'remember'age,the menopausal status,and the bone formation status in vitro,resulting in more aggressive osteoclasts.Our discovery suggests that this may be mediated through DNA methylation.We suggest that this may have clinical implications and could contribute to understanding individual differences in age-and menopause-induced bone loss.Anais Marie Julie Moller Jean-Marie Delaisse Jacob Bastholm Olesen Jonna Skov Madsen Luisa Matos Canto Troels Bechmann Silvia Regina Rogatto Kent Soe 2020Bone Research2020,8,3:4

13Cannabinoids function in defense against chewing herbivores in Cannabis sativa L.显示文摘In the decades since the first cannabinoids were identified by scientists,research has focused almost exclusively on the function and capacity of cannabinoids asmedicines and intoxicants for humans and other vertebrates.Very little is knownabout the adaptive value of cannabinoid production,though several hypotheses have been proposed including protection from ultraviolet radiation,pathogens,and herbivores.To test the prediction that genotypeswith greater concentrations of cannabinoidswill have reduced herbivory,a segregating F2 population of Cannabis sativa was leveraged to conduct lab-and field-based bioassays investigating the function of cannabinoids in mediating interactions with chewing herbivores.In the field,foliar cannabinoid concentration was inversely correlated with chewing herbivore damage.On detached leaves,Trichoplusia ni larvae consumed less leaf area and grew less when feeding on leaves with greater concentrations of cannabinoids.Scanning electron and light microscopy were used to characterize variation in glandular trichome morphology.Cannabinoid-free genotypes had trichomes that appeared collapsed.To isolate cannabinoids from confounding factors,artificial insect diet was amended with cannabinoids in a range of physiologically relevant concentrations.Larvae grew less and had lower rates of survival as cannabinoid concentration increased.These results support the hypothesis that cannabinoids function in defense against chewing herbivores.George M.Stack Stephen I.Snyder Jacob A.Toth Michael A.Quade Jamie L.Crawford John K.McKay John Nicholas Jackowetz Ping Wang Glenn Philippe Julie L.Hansen Virginia M.Moore Jocelyn K.C.Rose Lawrence B.Smart 2023Horticulture Research2023,10,11:0
