| 2 | Liraglutide alleviates cardiac fibrosis through inhibiting P4hα-1 expression in STZ-induced diabetic cardiomyopathy显示文摘Diabetic cardiomyopathy is an important contributor to morbidity and mortality of diabetic patients by causing heart failure.Interstitial and perivascular fibrosis plays a crucial role in diabetic cardiomyopathy.However,there is a lack of effective specific treatments available for diabetic cardiomyopathy.In the present study,we aim to explore the effects of Liraglutide,a GLP-1 analogue,on diabetic cardiomyopathy in STZ-induced diabetic rats fed with high-fat diet.A total of 60 male Wistar rats were randomly assigned to three groups,i.e.normal group,model group,and Liraglutide group,with 20 rats in each group.Serum levels of TC,TG,LDL-C,NEFA,and hydroxyproline were measured using commercial kits.Cardiac function was evaluated by QRS waves,LVEDd,LVESd,and LVEF.Myocardial fibrosis was measured by immunohistochemistry.Our results demonstrated that chronic administration of Liraglutide decreased the level of blood glucose and significantly alleviated lipid metabolic disturbance compared with the model group.Furthermore,Liraglutide was found to improve the damaged cardiac function.In line with this,we also found that the alleviation of cardiac dysfunction was associated with the decreased fibrosis in diabetic myocardial tissues,which was reflected by the decreased expressions of P4hα-1,COL-1,COL-3,MMP-1,and MMP-9.Our results thus suggest that Liraglutide might have a myocardial protective effect by inhibiting P4hα-1-mediated myocardial fibrosis. | Tong Zhao Huiqiang Chen Fei Xu Juan Wang Yusheng Liu Xiaowei Xing Linlin Guo Mingxiang Zhang Qinghua Lu | 2019 | Acta Biochimica et Biophysica Sinica2019,51,3: | 9 |
| 3 | Molecular pathogenesis of acetaminophen-induced liver injury and its treatment options显示文摘Acetaminophen,also known as N-acetyl-p-aminophenol(APAP),is commonly used as an antipyretic and analgesic agent.APAP overdose can induce hepatic toxicity,known as acetaminophen-induced liver injury(AILI).However,therapeutic doses of APAP can also induce AILI in patients with excessive alcohol intake or who are fasting.Hence,there is a need to understand the potential pathological mechanisms underlying AILI.In this review,we summarize three main mechanisms involved in the pathogenesis of AILI:hepatocyte necrosis,sterile inflammation,and hepatocyte regeneration.The relevant factors are elucidated and discussed.For instance,N-acetyl-p-benzoquinone imine(NAPQI)protein adducts trigger mitochondrial oxidative/nitrosative stress during hepatocyte necrosis,danger-associated molecular patterns(DAMPs)are released to elicit sterile inflammation,and certain growth factors contribute to liver regeneration.Finally,we describe the current potential treatment options for AILI patients and promising novel strategies available to researchers and pharmacists.This review provides a clearer understanding of AILI-related mechanisms to guide drug screening and selection for the clinical treatment of AILI patients in the future. | Xiaopeng CAI Huiqiang CAI Jing WANG Qin YANG Jun GUAN Jingwen DENG Zhi CHEN | 2022 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,4: | 7 |

X-ray diffraction (XRD) and high resolution transmission electron microscopy (HRTEM) of the sample do not show existence of any signal of second phase. The X-photoelectron spectroscopy (XPS) of the sample shows the chemical valence of Ni is +2 and the real Ni concentration is 2.19% determined by X-ray fluorescence spectra (XRFS). | 14 | Hexafluoropropylene oxide trimer acid,a perfluorooctanoic acid alternative,induces cardiovascular toxicity in zebrafish embryos显示文摘As an increasingly used alternative to perfluorooctanoic acid(PFOA),hexafluoropropylene oxide trimer acid(HFPO-TA)has been widely detected in global water environments.However,little is known regarding its toxic effects on cardiovascular development.Here,zebrafish embryos were treated with egg water containing 0,60,120,or 240 mg/L HFPO-TA.Results showed that HFPO-TA treatment led to a significant reduction in both larval survival percentage and heart rate.Furthermore,HFPO-TA exposure caused severe pericardial edema and elongation of the sinus venous to bulbus arteriosus distance(SV-BA)in Tg(myl7:GFP)transgenic larvae,disrupting the expression of genes involved in heart development and thus causing abnormal heart looping.Obvious sprouting angiogenesis was observed in the 120 and 240 mg/L exposed Tg(fli:GFP)transgenic larvae.HFPO-TA treatment also impacted the mRNA levels of genes involved in the vascular endothelial growth factor(VEGF)pathway and embryonic vascular development.HFPO-TA exposure significantly decreased erythrocyte number in Tg(gata1:DsRed)transgenic embryos and influenced gene expression associated with the heme metabolism pathway.HFPO-TA also induced oxidative stress and altered the transcriptional levels of genes related to cell cycle and apoptosis,inhibiting cell proliferation while promoting apoptosis.Therefore,HFPO-TA exposure may induce abnormal development of the cardiovascular and hematopoietic systems in zebrafish embryos,suggesting it may not be a suitable or safe alternative for PFOA. | Sujie Sun Li Zhang Xue Li Lu Zang Ling Huang Junquan Zeng Zigang Cao Xinjun Liao Zilin Zhong Huiqiang Lu Jianjun Chen | 2024 | Journal of Environmental Sciences2024,,5: | 0 |
| 15 | Targeting P21-activated kinase suppresses proliferation and enhances chemosensitivity in T-cell lymphoblastic lymphoma显示文摘T-cell lymphoblastic lymphoma(T-LBL)is a highly aggressive non-Hodgkin lymphoma with a poor prognosis.P21-activated kinase(PAK)is a component of the gene expression-based classifier that can predict the prognosis of T-LBL.However,the role of PAK in T-LBL progression and survival remains poorly understood.Herein,we found that the expression of PAK1 was significantly higher in T-LBL cell lines(Jurkat,SUP-T1,and CCRF-CEM)compared to the human T-lymphoid cell line.Moreover,PAK2 mRNA level of 32 relapsed T-LBL patients was significantly higher than that of 37 cases without relapse(P=.012).T-LBL patients with high PAK1 and PAK2 expression had significantly shorter median RFS than those with low PAK1 and PAK2 expression(PAK1,P=.028;PAK2,P=.027;PAK1/2,P=.032).PAK inhibitors,PF3758309(PF)and FRAX597,could suppress the proliferation of T-LBL cells by blocking the G1/S cell cycle phase transition.Besides,PF could enhance the chemosensitivity to doxorubicin in vitro and in vivo.Mechanistically,through western blotting and RNA sequencing,we identified that PF could inhibit the phosphorylation of PAK1/2 and downregulate the expression of cyclin D1,NF-κB and cell adhesion signaling pathways in T-LBL cell lines.These findings suggest that PAK might be associated with T-LBL recurrence and further found that PAK inhibitors could suppress proliferation and enhance chemosensitivity of T-LBL cells treated with doxorubicin.Collectively,our present study underscores the potential therapeutic effect of inhibiting PAK in T-LBL therapy. | Ning Su Yu Fang Xu Chen Xiaoqin Chen Zhongjun Xia Huiqiang Huang Yi Xia Panpan Liu Xiaopeng Tian Qingqing Cai | 2023 | Blood Science2023,5,4: | 0 |

| 17 | AtMCM10 promotes DNA replication-coupled nucleosome assembly in Arabidopsis显示文摘Minichromosome Maintenance protein 10(MCM10)is essential for DNA replication initiation and DNA elongation in yeasts and animals.Although the functions of MCM10 in DNA replication and repair have been well documented,the detailed mechanisms for MCM10 in these processes are not well known.Here,we identified AtMCM10 gene through a forward genetic screening for releasing a silenced marker gene.Although plant MCM10 possesses a similar crystal structure as animal MCM10,AtMCM10 is not essential for plant growth or development in Arabidopsis.AtMCM10 can directly bind to histone H3-H4 and promotes nucleosome assembly in vitro.The nucleosome density is decreased in Atmcm10,and most of the nucleosome density decreased regions in Atmcm10 are also regulated by newly synthesized histone chaperone Chromatin Assembly Factor-1(CAF-1).Loss of both AtMCM10 and CAF-1 is embryo lethal,indicating that AtM CM10 and CAF-1 are indispensable for replication-coupled nucleosome assembly.AtMCM10 interacts with both new and parental histones.Atmcm10 mutants have lower H3.1abundance and reduced H3K27me1/3 levels with releasing some silenced transposons.We propose that AtM CM10 deposits new and parental histones during nucleosome assembly,maintaining proper epigenetic modifications and genome stability during DNA replication. | Xinjie Zhao Jingyi Wang Dan Jin Jinkui Cheng Hui Chen Zhen Li Yu Wang Huiqiang Lou Jian-Kang Zhu Xuan Du Zhizhong Gong | 2023 | Journal of Integrative Plant Biology2023,65,1: | 0 |

| 19 | The growth temperatures dependence of optical and electrical properties of InN films显示文摘InN films grown on sapphire at different substrate temperatures from 550℃ to 700℃ by metalorganic chemical vapor deposition were investigated. The low-temperature GaN nucleation layer with high-temperature annealing (1100℃) was used as a buffer for main InN layer growth. X-ray diffraction and Raman scattering measurements reveal that the quality of InN films can be improved by increasing the growth temperature to 600℃. Further high substrate temperatures may promote the thermal decomposition of InN films and result in poor crystallinity and surface morphology. The photoluminescence and Hall measurements were employed to characterize the optical and electrical properties of InN films,which also indicates strong growth temperature dependence. The InN films grown at temperature of 600℃ show not only a high mobility with low carrier concentration,but also a strong infrared emission band located around 0.7 eV. | 20 | Multivessel coronary artery ectasia and severe calcification in a patient with pheochromocytoma:a case report显示文摘Multivessel coronary artery ectasia with severe calcification is rare among patients with coronary artery disease. A 74-year-old Chinese woman suffered from acute myocardial infarction on a background of 50 years of poorly controlled hypertension secondary to pheochromocytoma, which was surgically removed in June 2012 prior to the presentation. Coronary angiography revealed total occlusion of the proximal left anterior descending artery, and multiple ectasias with severe calcification in the left main, circumflex and right coronary artery. After an aspiration thrombectomy and balloon angioplasty, grade 3 coronary flow was restored in the left descending coronary artery. No cardiac events were found in the 12-month follow-up.
