1Effect of minocycline on cerebral ischemia-reperfusion injury显示文摘Minocylcine, a tetracycline derivate, has been shown to cross the blood-brain barrier and enter the central nervous system. In this study, cerebral ischemia-reperfusion injury models were established using the suture method, and minocycline was immediately injected intraperitoneally after cerebral ischemia-reperfusion (22.5 mg/kg, initially 45 mg/kg) at a 12-hour interval. Results showed that after minocycline treatment, the volume of cerebral infarction was significantly reduced, the number of surviving cell in the hippocampal CA1 region increased, the number of apoptotic cells decreased, the expression of caspase-3 and poly(adenosine diphosphate-ribose) polymerase-1 protein was down-regulated, and the escape latency in the water maze test was significantly shortened compared with the ischemia-reperfusion group. 2Chloride channel blocker 4,4-diisothiocyanatostilbene-2,2'-disulfonic acid inhibits nitric oxide-induced apoptosis in cultured rat hippocampal neurons显示文摘Apoptosis in cultured rat hippocampal neurons was induced using the nitric oxide donor 3-morpholinosydnonimine, and cells were treated with the chloride channel blocker, 4,4-diisothiocyanatostilbene-2,2'-disulfonic acid. Results showed that the survival rate of neurons was significantly increased after treatment with 4,4-diisothiocyanatostilbene-2,2'-disulfonic acid, and the rate of apoptosis decreased. In addition, the expression of the apoptosis-related proteins poly(adenosine diphosphate-ribose)polymerase-1 and apoptosis-inducing factor were significantly reduced. 3Targeting lactate dehydrogenase A (LDHA) exerts antileukemic effects on T-cell acute lymphoblastic leukemia显示文摘Background:T-cell acute lymphoblastic leukemia(T-ALL)is an uncommon and aggressive subtype of acute lymphoblastic leukemia(ALL).In the serum of T-ALL patients,the activity of lactate dehydrogenase A(LDHA)is increased.We proposed that targeting LDHA may be a potential strategy to improve T-ALL outcomes.The current study was conducted to investigate the antileukemic effect of LDHA gene-targeting treatment on T-ALL and the underlying molecular mechanism.Methods:Primary T-ALL cell lines Jurkat and DU528 were treated with the LDH inhibitor oxamate.MTT,colony formation,apoptosis,and cell cycle assays were performed to investigate the effects of oxamate on T-ALL cells.Quantitative real-time PCR(qPCR)and Western blotting analyses were applied to determine the related signaling pathways.A mitochondrial reactive oxygen species(ROS)assay was performed to evaluate ROS production after T-ALL cells were treated with oxamate.A T-ALL transgenic zebrafish model with LDHA gene knockdown was established using CRISPR/Cas9 gene-editing technology,and then TUNEL,Western blotting,and T-ALL tumor progression analyses were conducted to investigate the effects of LDHA gene knockdown on T-ALL transgenic zebrafish.Results:Oxamate significantly inhibited proliferation and induced apoptosis of Jurkat and DU528 cells.It also arrested Jurkat and DU528 cells in G0/G1 phase and stimulated ROS production(all P<0.001).Blocking LDHA significantly decreased the gene and protein expression of c-Myc,as well as the levels of phosphorylated serine/threonine kinase(AKT)and glycogen synthase kinase 3 beta(GSK-3β)in the phosphatidylinositol 3′-kinase(PI3K)signaling pathway.LDHA gene knockdown delayed disease progression and down-regulated c-Myc mRNA and protein expression in T-ALL transgenic zebrafish.Conclusion:Targeting LDHA exerted an antileukemic effect on T-ALL,representing a potential strategy for T-ALL treatment.Haizhi Yu Yafei Yin Yifang Yi Zhao Cheng Wenyong Kuang Ruijuan Li Haiying Zhong Yajuan Cui Lingli Yuan Fanjie Gong Zhihua Wang Heng Li Hongling Peng Guangsen Zhang 2020Cancer Communications2020,40,10:2

9Organophosphate esters in water,suspended particulate matter(SPM)and sediments of the Minjiang River,China显示文摘As organic pollutants of emerging concern,organophosphate esters(OPEs)have shown toxicity to organisms after entering the water environment.However,research on OPEs in freshwater in Southwest China is very limited.The levels,distribution and partitioning behavior of OPEs in the Minjiang River and their influencing factors is still unknown.In this study,six OPEs,tri-n-butyl phosphate(Tn BP),tri(2-chloroethyl)-phosphate(TCEP),trichloropropyl phosphate(TCPP),triphenyl phosphate(TPh P),tributoxyethyl phosphate(TBEP),and tris(2-ethylhexyl)-phosphate(TEHP),were determined in surface water,suspended particle matter(SPM)and sediments of the Minjiang River.The results showed that the average concentrations of∑_(6)OPEs in surface water,SPM and sediments of the Minjiang River were 199.32±124.95 ng/L,38463.79±45641.89 ng/g dry weight(dw)and 76.45±28.00 ng/g dw,respectively.High concentrations of OPEs were detected in SPM samples,indicating that more attention should be paid to pollution in SPM.It is worth noting that the variation trend of OPEs in SPM was almost opposite to that in water but basically similar to that in sediment.The proportions of alkyl OPEs in∑_(6)OPEs increased from surface water to SPM and sediments.Alkyl OPEs were the main pollutants in SPM(10.44%–80.88%of∑_(6)OPEs,mean of 54.52%)and sediments(59.08%–81.30%of∑_(6)OPEs,mean of 68.91%),whereas chlorinated OPEs were the most abundant components in surface water(43.16%–75.99%ofδ∑_(6)OPEs,mean of 55.50%).The water-sediment partition coefficient(logKOC)of OPEs was 4.97–7.58,while the water-SPM partition coefficient was 6.71–10.00.No significant correlations were found between log KOW and logKOC.KOW was not the main factor affecting the distribution of OPEs in the Minjiang River,China.Hongling Yin Qin Liu Xu Deng Xiaowen Liu Shuhong Fang Yuanming Xiong Jiaojiao Song 2021Chinese Chemical Letters2021,32,9:1

12Advancing the pressure sensing performance of conductive CNT/PDMS composite film by constructing a hierarchical-structured surface显示文摘Flexible pressure sensors have attracted wide attention due to their applications to electronic skin,health monitoring,and human-machine interaction.However,the tradeoff between their high sensitivity and wide response range remains a challenge.Inspired by human skin,we select commercial silicon carbide sandpaper as a template to fabricate carbon nanotube(CNT)/polydimethylsiloxane(PDMS)composite film with a hierarchical structured surface(h-CNT/PDMS)through solution blending and blade coating and then assemble the h-CNT/PDMS composite film with interdigitated electrodes and polyurethane(PU)scotch tape to obtain an h-CNT/PDMS-based flexible pressure sensor.Based on in-situ optical images and finite element analysis,the significant compressive contact effect between the hierarchical structured surface of h-CNT/PDMS and the interdigitated electrode leads to enhanced pressure sensitivity and a wider response range(0.1661 kPa^(-1),0.4574 kPa^(-1)and 0.0989 kPa^(-1)in the pressure range of 0–18 kPa,18–133 kPa and 133–300 kPa)compared with planar CNT/PDMS composite film(0.0066 kPa^(-1)in the pressure range of 0–240 kPa).The prepared pressure sensor displays rapid response/recovery time,excellent stability,durability,and stable response to different loading modes(bending and torsion).In addition,our pressure sensor can be utilized to accurately monitor and discriminate various stimuli ranging from human motions to pressure magnitude and spatial distribution.This study supplies important guidance for the fabrication of flexible pressure sensors with superior sensing performance in next-generation wearable electronic devices.Ye Zhao Taoyu Shen Minyue Zhang Rui Yin Yanjun Zheng Hu Liu Hongling Sun Chuntai Liu Changyu Shen 2023Nano Materials Science2023,5,4:1

16Fungal diversity and mycotoxins identification of mouldy rapeseed in China显示文摘Investigation on fungal populations and mycotoxins of mouldy rapeseed was conducted in this paper. A total of 578 rapeseed samples were collected from 11 Chinese provinces in 2015, and 141 samples were contaminted by mycete. Mouldy rapeseed samples with exceeding specified standard were found in all areas. The highest percentages of samples were from Hunan, Jiangxi and Hubei provinces with above standard rate of 31.91%, 24.54% and 20.45% respectively. Mycete in 29 severely mouldy samples were isolatedand determined by using dilution method. Eighty-three dominant isolates were found,among which fungal strain number ranged from 1 to 8. Aspergillus flavus, Penicillium steckii and Penicillium citrinum were dominant myco-populations identified by morphological and molecular biological methods. Rapeseed from the Southern provinces including Jiangxi,Hunan and Guangxi were contaminated by more isolates. Mycotoxins content were evaluated by LC–MS/MS. AFB1, BEA and FB1 were the top 3 prevalent mycotoxins. BEA and FB1 contaminations in rapeseed were relatively low. 17Rheological Simulating Study of Mechanical Conditions for Syn-extensional Basin and Mountain Coupling System显示文摘This article gives a mechanical model, in which the layers of lithosphere are assumed to be the creep materials, to study the coupling mechanism of a syn-basin-mountain system quantitatively by using the numerical simulating method. A geological dynamic extensional mode given by some geologists is theoretically discussed and verified. The study shows that lithosphere thickening or thinning is closely related to the thermal activity, or in other words, thermal convection beneath the lithosphere. It is one of the important factors affecting the formation of the basin-mountain coupling system. As an essential condition, only the upward buoyant force and the horizontal dragging force caused by the thermal convection jointly act on the bottom of the lithosphere, the stress and strain states in rock’s layers are advantageous to forming the tectonic-landforms of the basin-mountain coupling system. A study on the creep features of the lithosphere shows that the stress and strain in the rock’s layers vary with time when the lasting forces act on the boundary. They increase rapidly at initial stage and decrease steadily after reaching the peak value. 18Growth suppressor lingerer regulates bantam microRNA to restrict organ size显示文摘The evolutionarily conserved Hippo signaling pathway plays an important role in organ size control by regulating cell proliferation and apoptosis.Here,we identify Lingerer(Lig)as a growth suppressor using RNAi modifying screen in Drosophila melanogaster.Loss of lig increases organ size and upregulates bantam(ban)and the expression of the Hippo pathway target genes,while overexpression of lig results in diminished ban expression and organ size reduction.We demonstrate that Lig C-terminal exhibits dominant-negative function on growth and ban expression,and thus plays an important role in organ size control and ban regulation.In addition,we provide evidence that both Yki and Mad are essential for Lig-induced ban expression.We also show that Lig regulates the expression of the Hippo pathway target genes partially via Yorkie.Moreover,we find that Lig physically interacts with and requires Salvador to restrict cell growth.Taken together,we demonstrate that Lig functions as a critical growth suppressor to control organ size via ban and Hippo signaling.Liang Dong Jinhui Li Hongling Huang Meng-Xin Yin Jinjin Xu Peixue Li Yi Lu Wenqing Wu Hang Yang Yun Zhao Lei Zhang 2015Journal of Molecular Cell Biology2015,7,5:0
19Evolution of chemical composition of fogwater in winter in Chengdu, China显示文摘Two sampling sites representing the urban and suburban area of Chengdu, China were sampled and analyzed for selected chemicals to characterize the evolution of the chemical composition of fogwater. A trend of total organic carbon (TOC) > total nitrogen (TN) > total inorganic carbon (TIC) was observed for both sites. Variation of inorganic ions indicated that inorganic pollutants were not accumulated in the fog. Concentrations of n-alkanes (C11-C37) at the urban site ranged from 7.58 to 27.75 ng/mL while at the suburban site concentrations were 2.57-21.55 ng/mL. The highest concentration of n-alkanes was observed in the mature period of fog (393.12 ng/mL) which was more than ten times that in the fog formation period (27.83 ng/mL) and the fog dissipation period (14.87 ng/mL). Concentrations of Σ15PAHs were in the range of 7.27-38.52 ng/mL at the urban site and 2.59-22.69 ng/mL at the suburban site. Contents of PAHs in the mature period of fog (27.15 ng/mL) > fog dissipation period (11.59 ng/mL) > fog formation period (6.42 ng/mL). Concentrations of dicarboxylic acids (C5-C9) ranged from 10.92 to 40.78 ng/mL, with glutaric acid (C5) as the dominant dicarboxylic acid.
