| 3 | T cell–associated immunoregulation and antiviral effect of oxymatrine in hydrodynamic injection HBV mouse model显示文摘Although oxymatrine(OMT) has been shown to directly inhibit the replication of hepatitis B virus(HBV) in vitro, limited research has been done with this drug in vivo. In the present study, the antiviral effect of OMT was investigated in an immunocompetent mouse model of chronic HBV infection.The infection was achieved by tail vein injection of a large volume of DNA solution. OMT(2.2, 6.7 and20 mg/kg) was administered by daily intraperitoneal injection for 6 weeks. The efficacy of OMT was evaluated by the levels of HBV DNA, hepatitis B surface antigen(HBs Ag), hepatitis B e antigen(HBe Ag)and hepatitis B core antigen(HBc Ag). The immunoregulatory activity of OMT was evaluated by serum ELISA and flow cytometry. Results shows that OMT at 20 mg/kg inhibited HBV replication, and it was more efficient than entecavir(ETV) in the elimination of serum HBs Ag and intrahepatic HBc Ag. Inaddition, OMT accelerated the production of interferon-γ(IFN-γ) in a dose-dependent manner in CD4^+T cells. Our findings demonstrate the beneficial effects of OMT on the enhancement of immunological function and in the control of HBV antigens. | 5 | Cas9-NG Greatly Expands the Targeting Scope of the Genome-Editing Toolkit by Recognizing NG and Other Atypical PAMs in Rice显示文摘CRISPR technologies enabling precise genome manipulation are valuable for gene function studies and molecular crop breeding. However, the requirement of a protospacer adjacent motif (PAM)y such as NGG and TTN, for Cas protein recognition restricts the selection of targetable genomic loci in practical applications of CRISPR technologies. Recently Cas9-NG, which recognizes a minimal NG PAM, was reported to expand the targeting space of genome editing in human cells, but it remains unclear whether this Cas9 variant can be used in plants. In this study, we evaluated the nuclease activity of Cas9-NG toward various NGN PAMs by targeting endogenous genes in transgenic rice. We found that Cas9-NG edits all NGG, NGA, NGT, and NGC sites with impaired activity, while the gene-edited plants were dominated by monoallelic mutations. Cas9-NG-engineered base editors were then developed and used to generate O s B Z R I gainof- function plants that can not be created by other available Cas9-engineered base editors. Moreover, we showed that a Cas9-NG-based transcriptional activator efficiently upregulated the expression of endogenous target genes in rice. In addition, we discovered that Cas9-NG recognizes NAC, NTG, NTT, and NCG apart from NG PAM. | 9 | Effects of cooling rates on the mechanical properties of a Ti-based bulk metallic glass显示文摘Mechanical properties of the glassy specimens fabricated at different cooling rates with a composition of Ti40Zr25Cu12Ni3Be20 were systematically investigated. It was confirmed that faster cooling rates caused not only a larger amount of frozen-in free volume but also a higher glass transition temperature in the bulk glassy alloy. Increase in the free volume was found to favor plastic deformation and then to give rise to larger compressive plasticity, whilst the rise in the glass transition temperature seemed to be closely related to the higher yield strength. Moreover, the increase of yield strength and plasticity induced by fast cooling rates may also be associated with the residual stress generated during the fabrication process. | 11 | Migration patterns and habitat use of the tapertail anchovy Coilia mystus in the Oujiang River Estuary and the Zhujiang River Estuary, China显示文摘Habitat use of the tapertail anchovy(Coilia mystus Linnaeus, 1758) from the Oujiang River Estuary and the Zhujiang(Pearl) River Estuary was studied by examining the environmental signatures of Sr and Ca in otoliths using electron probe microanalysis. Individuals from the Oujiang River had higher and varied Sr:Ca ratios(expressed as(Sr:Ca)×1 000, 3.83–13.0 average) in the otolith core regions, suggesting that they were born in brackish or sea waters, and that a freshwater habitat might not be necessary for egg hatching and larval growth.While, individuals from the Zhujiang River had lower Sr:Ca ratios(0.39–2.51 average) in the core regions,suggesting a freshwater origin. After hatching, anchovies from the Zhujiang River migrate downstream to the river estuary close to brackish water. | 15 | Superior electrochemical performance of La-Mg-Ni-based alloys with novel A_(2)B_(7)-A_(7)B_(23) biphase superlattice structure显示文摘Nickel metal hydride(Ni-MH) rechargeable batteries hold an important position in the new-energy vehicle market owing to their key technology advantages. Their negative electrode materials—hydrogen storage alloys(HSAs) are always on the spotlight and are the key to compete with the burgeoning Li-ion batteries. Here, for the first time we report a series of biphase supperlattice HSAs with a(La,Mg)_(2)Ni_7 matrix phase and a novel(La,Mg)_(7)Ni_(23) secondary phase. The biphase alloys show discharge capacities of402–413 m Ahg^(-1) compared with 376–397 mAhg^(-1) of the other multi-or single-phase alloys. These values are among the highest for superlattice HSAs. In addition, the alloy with 15.4 wt.%(La,Mg)_(7)Ni_(23) phase exhibits good high rate dischargeability due to the proper compromise between the amount of crystal boundaries and equilibrium plateau voltage. The cycling stability of the biphase alloys is lower than that of the single-phase alloy but is till higher than the multiphase alloy. | 17 | Composition and Distribution of Planktonic Ciliates in the Southern South China Sea During Late Summer: Comparison Between Surface and 75 m Deep Layer显示文摘Ciliates are very important components in most marine ecosystem.They are trophic link between the microbial food web and grazing food chain.In this study,ciliates were collected from 11 sites in the southern South China Sea(SCS) during August 25 to September 28,2011.Their composition and distribution at the surface and 75 m deep depth of the ocean were studied.A total of 30 species belonging to 22 genera were identified,and 22 species of 15 genera were Tintinnids.Eutintinnus fraknoii and E.stramentus were the most common species.The other dominants were strombidiids ciliates including Strombidium conicum and S.globosaneum,which were followed by the tide form,Mesodinium pulex.Ciliates abundance ranged from 46 ind L^(-1) to 368 ind L^(-1) in the open sites,46–368 ind L^(-1) at surface and 73–198 ind L^(-1) at 75 m deep layer.In the Yongshu reef,ciliates abundance ranged from 167 ind L^(-1) to 365 ind L^(-1) in the water column,similar to that in Sanya coral reef waters.Ciliates composition showed obvious difference between surface and 75 m deep layer at station S2(P < 0.05),while no similar result was observed at other sites.At 75 m deep layer,salinity was negatively related to mixed layer depth(P < 0.05),but positively to chlorophyll a concentration(P < 0.05),indicating that the change of vertical mixing in water column influenced vertical distribution of ciliates in the southern SCS. | LIU Huaxue SHEN Pingping LI Chunhou CHEN Zuozhi QI Zhanhui HUANG Honghui | 2016 | Journal of Ocean University of China2016,15,1: | 3 |
| 18 | Effect of Huatan Jieyu granules in treatment of Parkinson’s disease patients with sleep disorder identified as symptom pattern of phlegma-heat-stirring wind显示文摘OBJECTIVE: To investigate the safety and efficacy of Huatan Jieyu granules in treatment of Parkinson’s disease(PD) patients with sleep disorder identified as symptom pattern of phlegma-heat-stirring wind in terms of the theory of Traditional Chinese Medicine.METHODS: In total, 107 Parkinson’s disease patients with sleep disorders identified as symptom pattern of phlegma-heat-stirring wind were selected and randomly divided into the experimental group(55 cases) and the control group(52 cases).Both groups were given basic treatment with prednisone. The experimental group of patients was treated with Huatan Jieyu granules and the control group of patents was treated with only the basic treatment. Treatment lasted for 4 weeks. | 19 | Influence of endogenous ciliary neurotrophic factor on neural differentiation of adult rat hippocampal progenitors显示文摘Ciliary neurotrophic factor is the only known neurotrophic factor that can promote differentiation of hippocampal neural progenitor cells to glial cells and neurons in adult rats.This process is similar to spontaneous differentiation.Therefore,ciliary neurotrophic factor may be involved in spontaneous differentiation of neural stem cells.To verify this hypothesis,the present study isolated neural progenitor cells from adult male rats and cultured them in vitro.Results showed that when neural progenitor cells were cultured in the absence of mitogen fibroblast growth factor-2 or epidermal growth factor,they underwent spontaneous differentiation into neurons and glial cells.Western blot and immunocytochemical staining showed that exogenous ciliary neurotrophic factor strongly induced adult hippocampal progenitor cells to differentiate into neurons and glial cells.Moreover,passage 4 adult hippocampal progenitor cells expressed high levels of endogenous ciliary neurotrophic factor,and a neutralizing antibody against ciliary neurotrophic factor prevented the spontaneous neuronal and glial differentiation of adult hippocampal progenitor cells.These results suggest that the spontaneous differentiation of adult hippocampal progenitor cells is mediated partially by endogenous ciliary neurotrophic factor. | Jun Ding Zhili He Juan Ruan Ying Liu Chengxin Gong Shenggang Sun Honghui Chen | 2013 | Neural Regeneration Research2013,8,4: | 2 |
