| 4 | DJ-1 is involved in the multidrug resistance of SGC7901 gastric cancer cells through PTEN/PI3K/Akt/Nrf2 pathway显示文摘Gastric cancer is a common malignancy worldwide.The occurrence of multidrug resistance(MDR)is the major obstacle for effective gastric cancer chemotherapy.In this study,the in-depth molec-ular mechanism of the DJ-1-induced MDR in SGC7901 gastric cancer cells was investigated.The results showed that DJ-1 expression level was higher in MDR variant SGC7901NCR cells than that in its parental SGC7901 cells.Moreover,DJ-1 overexpression conferred the MDR phenotype to SGC7901 cells,while DJ-1 knockdown in SGC7901NCR cells induced re-sensitization to adriamycin,vincristine,cisplatin,and 5-fluorouracil.These results suggested that DJ-1 mediated the develop-ment of MDR in SGC7901 gastric cancer cells.Importantly,further data revealed that the activation of PI3k/Akt and Nrf2 signaling pathway were required for the DJ-1-induced MDR phenotype in SGC7901 gastric cancer cells.Meanwhile,we found that Pl3k/Akt pathway was activated probably through DJ-1 directly binding to and negatively regulating PTEN,consequently resulting in Nrf2 phosphorylation and activation,and thereby inducing Nrf2-dependent P-glycoprotein(P-gp)and Bcl-2 expressions in the DJ-1-mediated MDR of SGC7901 gastric cancer cells.Overall,these results revealed that activating PTEN/PI3K/Akt/Nrf2 pathway and subsequently upregulating P-gp and Bcl-2 expression could be a critical mechanism by which DJ-1 mediates the development of MDR in SGC7901 gastric cancer cells.The new findings may be helpful for understanding the mechanisms of MDR in gastric cancer cells,prompting its further investigation as a molecular target to overcome MDR. | Lejia Qiu Zhaoxia Ma Xiaoran Li Yizhang Deng Guangling Duan Le Zhao Xingwang Xu Lin Xiao Haoyue Liu Zhengming Zhu Heping Chen | 2020 | Acta Biochimica et Biophysica Sinica2020,52,11: | 5 |

| 9 | Fructooligosaccharides enhance the therapeutic effect of xiao-zhi-fang on non-alcoholic fatty liver disease via regulating intestinal flora显示文摘Background:Non-alcoholic fatty liver disease(NAFLD)is a frequently occurring clinical disease,which could be treated by traditional Chinese medicine.However,the effects and mechanisms of xiao-zhi-fang(XZF)alone or in combination with fructooligosaccharides(FOS)on NAFLD remains unclear.Methods:This study established the NAFLD mouse model by feeding with a high-fat diet and by administering with XZF and/or FOS.Moreover,the fat content in the liver tissues was evaluated by oil red O and hematoxylin and eosin staining,and the liver function indices were examined using a biochemical analyzer.Changes in the intestinal flora were monitored using 16S rRNA gene sequencing.The results of the current study showed that the alanine aminotransferase,aspartate aminotransferase,total triglyc-eride,total cholesterol levels,and the amount of lipid accumulation were markedly elevated in NAFLD mice.Results:XZF alone or in combination with FOS significantly reduced lipid accumulation in the liver and blood lipid levels in NAFLD mice.Moreover,XZF alone or in combination with FOS notably altered the intestinal flora structure in NAFLD mice.Consequently,17 operational taxonomic units were decreased and nine were increased in the XZF and/or FOS groups.Conclusions:FOS may accelerate the intestinal microecological balance in NAFLD mice after treatment with XZF by promoting intestinal flora growth,thereby enhancing the therapeutic effect of XZF on NAFLD. | Jiongshan Zhang Yue Li Gemin Xiao Yongwei Li Heping Xie Yan Chen | 2021 | Liver Research2021,5,4: | 0 |
| 10 | Isolation by phage display and characterization of a single-chain antibody specific for O^6-methyldeoxyguanosine显示文摘New approaches of making single chain Fv antibodies against O6-methyl-2’ -deoxyguanosine (O6MdG) have been demonstrated by using the phage antibody display system. Using O6MdG as an antigen, 21 positive clones were identified by ELISA from this library, one of which, designated H3, specifically binds to O6MdG with high affinity. The H3 scFv antibody has an affinity constant (Kaff) of 5.94×1011(mol/L)-1. H3 scFv has been successfully used to detect O6MdG in DNA hydrolyses from yeast or E. coli cells treated with a DNA methylating agent. To our knowledge, this is the first report of the selection of a specific scFv against DNA adducts. | 14 | Full length cDNA cloning and expression analysis of annexinA2 gene from deer antler tissue显示文摘ANXA2(AnnexinA2), a calcium-dependent phospholipid binding protein, is involved in various Ca2+-related biological activities. In the present study, full-length cDNA of ANXA2 was isolated from the velvet antler tip tissue of sika deer(Cervus nippon hortulorum); the amino acid sequence and gene expression was analyzed by using bioinformatics and real-time reverse transcriptase polymerase chain reaction(RT-PCR) techniques. Nucleotide sequence analysis reveals that the full-length cDNA of the ANXA2 gene was 1372 bp, of which 1020 bp was in the open-reading frame(ORF) encoding 339 amino acids; its relative molecular weight was 38.3 kDa; and isoelectric point was 6.72. Sequence analysis indicates that the protein includes four conserved tandem-duplication ANX domains. The gene-accession nucleotide sequence number in GenBank is JX315571. | 16 | Dynamic mechanical characteristics of deep Jinping marble in complex stress environments显示文摘To reveal the dynamic mechanical characteristics of deep rocks,a series of impact tests under triaxial static stress states corresponding to depths of 300-2400 m were conducted.The results showed that both the strain rates and the stress environments in depth significantly affect the mechanical characteristics of rocks.The sensitivity of strain rate to the dynamic strength and deformation modulus shows a negative correlation with depth,indicating that producing penetrative cracks in deep environments is more difficult when damage occurs.The dynamic strength shows a tendency to decrease and then increase slightly,but decreases sharply finally.Transmissivity demonstrates a similar trend as that of strength,whereas reflectivity indicates the opposite trend.Furthermore,two critical depths with high dynamically induced hazard possibilities based on the China Jinping Underground Laboratory(CJPL)were proposed for deep engineering.The first critical depth is 600-900 m,beyond which the sensitivity of rock dynamic characteristics to the strain rate and restraint of circumferential stress decrease,causing instability of surrounding rocks under axial stress condition.The second one lies at 1500-1800 m,where the wave impedance and dynamic strength of deep surrounding rocks drop sharply,and the dissipation energy presents a negative value.It suggests that the dynamic instability of deep surrounding rocks can be divided into dynamic load dominant and dynamic load induced types,depending on the second critical depth. | Chendi Lou Heping Xie Ru Zhang Hai Ren Hao Luo Kun Xiao Yuan Peng Qiang Tan Li Ren | 2024 | Journal of Rock Mechanics and Geotechnical Engineering2024,16,2: | 0 |
