| 1 | Prioritizing natural-selection signals from the deep-sequencing genomic data suggests multi-variant adaptation in Tibetan highlanders显示文摘Human genetic adaptation to high altitudes(>2500 m)has been extensively studied over the last few years,but few functional adaptive genetic variants have been identified,largely owing to the lack of deep-genome sequencing data available to previous studies.Here,w e build a list of putative adaptive variants,including 63 missense,7 loss-of-function;1,298 evolutionarily conserved variants and 509 expression quantitative traits loci.Notably,the top signal of selection is located in TMEM 247,a transmembrane protein-coding gene.The Tibetan version of TMEM 247 harbors one high-frequency(76.3%)missense variant,rsl 16983452(c.248C>T;p.Ala83Val);with the T allele derived from archaic ancestry and carried by>94%of Tibetans but absent or in low frequencies(<3%)in non-Tibetan populations.The rsl 16983452-T is strongly and positively correlated with altitude and significantly associated with reduced hemoglobin concentration(p=5.78×10^-5),red blood cell count(p=5.72×10^-7)and hematocrit(p=2.57×10^-6).In particular,TMEM247-rs 116983452 shows greater effect size and better predicts the phenotypic outcome than any E P A S1 variants in association with adaptive traits in Tibetans.Modeling the interaction between TM£M247-rsl 16983452 and EPAS1 variants indicates weak but statistically significant epistatic effects.Our results support that multiple variants may jointly deliver the fitness of the Tibetans on the plateau,where a complex model is needed to elucidate the adaptive evolution mechanism. | Lian Deng Chao Zhang Kai Yuan Yang Gao Yuwen Pan Xueling Ge Yaoxi He Yuan Yuan Yan Lu Xiaoxi Zhang Hao Chen Haiyi Lou Xiaoji Wang Dongsheng Lu Jiaojiao Liu Lei Tian Qidi Feng Asifullah Khan Yajun Yang Zi-Bing Jin Jian Yang Fan Lu Jia Qu Longli Kang Bing Su Shuhua Xu | 2019 | National Science Review2019,6,6: | 9 |
| 2 | Genome editing in Drosophila melanogaster: from basic genome engineering to the multipurpose CRISPR-Cas9 system显示文摘Nowadays, genome editing tools are indispensable for studying gene function in order to increase our knowledge of biochemical processes and disease mechanisms. The extensive availability of mutagenesis and transgenesis tools make Drosophila melanogaster an excellent model organism for geneticists. Early mutagenesis tools relied on chemical or physical methods,ethyl methane sulfonate(EMS) and X-rays respectively, to randomly alter DNA at a nucleotide or chromosomal level. Since the discovery of transposable elements and the availability of the complete fly genome, specific genome editing tools, such as P-elements, zinc-finger nucleases(ZFNs) and transcription activator-like effector nucleases(TALENs), have undergone rapid development. Currently, one of the leading and most effective contemporary tools is the CRISPR-cas9 system made popular because of its low cost, effectiveness, specificity and simplicity of use. | 3 | De novo assembly of a Tibetan genome and identification of novel structural variants associated with high-altitude adaptation显示文摘Structural variants(SVs) may play important roles in human adaptation to extreme environments such as high altitude but have been under-investigated. Here, combining long-read sequencing with multiple scaffolding techniques, we assembled a high-quality Tibetan genome(ZF1), with a contig N50 length of 24.57 mega-base pairs(Mb) and a scaffold N50 length of 58.80 Mb. The ZF1 assembly filled 80 remaining N-gaps(0.25 Mb in total length) in the reference human genome(GRCh38). Markedly, we detected17 900 SVs, among which the ZF1-specific SVs are enriched in GTPase activity that is required for activation of the hypoxic pathway. Further population analysis uncovered a 163-bp intronic deletion in the MKL1 gene showing large divergence between highland Tibetans and lowland Han Chinese. This deletion is significantly associated with lower systolic pulmonary arterial pressure, one of the key adaptive physiological traits in Tibetans. Moreover, with the use of the high-quality de novo assembly, we observed a much higher rate of genome-wide archaic hominid(Altai Neanderthal and Denisovan) shared non-reference sequences in ZF1(1.32%–1.53%) compared to other East Asian genomes(0.70%–0.98%),reflecting a unique genomic composition of Tibetans. One such archaic hominid shared sequence-a662-bp intronic insertion in the SCUBE2 gene-is enriched and associated with better lung function(the FEV1/FVC ratio) in Tibetans. | 5 | Study on properties of ultra-low dielectric loss mPPO/MTCLT composites prepared by injection molding显示文摘A novel category of polyphenylene oxide/high-impact polystyrene(PPO/HIPS)alloy was used as the polymer matrix(abbreviated as mPPO)and loaded with different volume fractions(0,10,20,30,40,50 vol.%)of MgTiO_(3)-Ca_(0.7)La_(0.2)TiO_(3)(abbreviated as MTCLT)ceramics to prepare composites by injection molding.Its micromorphology,density,dielectric,thermal and mechanical properties were analyzed in detail.The experimental results show that the composites possess a compact microstructure because HIPS increases the fluidity of PPO.Due to the excellent dielectric properties of both mPPO and MTCLT,the composites have an extremely low dielectric loss.The realization of the high ceramic filler fraction greatly limits the thermal expansion of the polymer chain by introducing the interphase,so that the coefficient of thermal expansion of the composite material could be as low as 21.8 ppm/°C.At the same time,the presence of ceramic particles could reinforce the mechanical property of the compos-ites.When the ceramic filler fraction is higher than 20 vol.%,the bending strength of the composite material is around 110 MPa.When the ceramic filler fraction is 40 vol.%,the composite possesses the best comprehensive performance.The dielectric constant is 6.81,the dielectric loss is 0.00104,the thermal expansion coefficient is as low as 25.3 ppm/°C,and the bending strength is 110.4 MPa.Due to its excellent properties,this material can be a good candidate in the field of microwave communication. | Yahan Liu Haiyi Peng Xiaogang Yao Minmin Mao Kaixin Song Huixing Lin | 2022 | Journal of Advanced Dielectrics2022,12,3: | 1 |

| 9 | Spectroscopic analysis of high electric field enhanced ionization in laser filaments in air for corona guiding显示文摘We report on a systematic experimental study on the fluorescence spectra produced from a femtosecond laser filament in air under a high electric field. The electric field alone was strong enough to create corona discharge(CD). Fluorescence spectra from neutral and ionic air molecules were measured and compared with pure high-voltage CD and pure laser filamentation(FIL). Among them, high electric field assisted laser FIL produced nitrogen fluorescence more efficiently than either pure CD or pure FIL processes. The nonlinear enhancement of fluorescence from the interaction of the laser filament and corona discharging electric field resulted in a more efficient ionization along the laser filament zone, which was confirmed by the spectroscopic measurement of both ionization-induced fluorescence and plasma-scattered 800 nm laser pulses. | 14 | Pericytes may facilitate SARS-CoV-2 entry into the nervous system显示文摘Objective:Although the neurological and olfactory symptoms of coronavirus disease 2019 have been identified,the neurotropic properties of the causative virus,severe acute respiratory syndrome-associated coronavirus 2(SARS-CoV-2),remain unknown.We sought to identify the susceptible cell types and potential routes of SARS-CoV-2 entry into the central nervous system,olfactory system,and respiratory system.Methods:We collected single-cell RNA data from normal brain and nasal epithelium specimens,along with bronchial,tracheal,and lung specimens in public datasets.The susceptible cell types that express SARS-CoV-2 entry genes were identified using single-cell RNA sequencing and the expression of the key genes at protein levels was verified by immunohistochemistry.We compared the coexpression patterns of the entry receptor angiotensin-converting enzyme 2(ACE2)and the spike protein priming enzyme transmembrane serine protease(TMPRSS)/cathepsin L among the specimens.Results:The SARS-CoV-2 entry receptor ACE2 and the spike protein priming enzyme TMPRSS/cathepsin L were coexpressed by pericytes in brain tissue;this coexpression was confirmed by immunohistochemistry.In the nasal epithelium,ciliated cells and sustentacular cells exhibited strong coexpression of ACE2 and TMPRSS.Neurons and glia in the brain and nasal epithelium did not exhibit coexpression of ACE2 and TMPRSS.However,coexpression was present in ciliated cells,vascular smooth muscle cells,and fibroblasts in tracheal tissue;ciliated cells and goblet cells in bronchial tissue;and alveolar epithelium type 1 cells,AT2 cells,and ciliated cells in lung tissue.Conclusion:Neurological symptoms in patients with coronavirus disease 2019 could be associated with SARS-CoV-2 invasion across the blood-brain barrier via pericytes.Additionally,SARS-CoV-2-induced olfactory disorders could be the result of localized cell damage in the nasal epithelium. | Zijian Kang Jing Wang Tong Meng Hao Zhang Da Xu Haiyi Gong Zhenyan Chang Zifu Li Xingang Cui Jianru Xiao Adnan I.Qureshi Wang Zhou Jianmin Liu Huji Xua | 2023 | Journal of Bio-X Research2023,6,1: | 0 |
