1Effects of mixture ratio on anaerobic co-digestion with fruit and vegetable waste and food waste of China显示文摘The biochemical methane potentials for typical fruit and vegetable waste (FVW) and food waste (FW) from a northern China city were investigated,which were 0.30,0.56 m 3 CH 4/kgVS (volatile solids) with biodegradabilities of 59.3% and 83.6%,respectively.Individual anaerobic digestion testes of FVW and FW were conducted at the organic loading rate (OLR) of 3 kg VS/(m 3 ·day) using a lab-scale continuous stirred-tank reactor at 35℃.FVW could be digested stably with the biogas production rate of 2.17 m 3/(m 3 ·day) and methane production yield of 0.42 m 3 CH 4/kg VS.However,anaerobic digestion process for FW was failed due to acids accumulation.The effects of FVW:FW ratio on co-digestion stability and performance were further investigated at the same OLR.At FVW and FW mixing ratios of 2:1 and 1:1,the performance and operation of the digester were maintained stable,with no accumulation of volatile fatty acids (VFA) and ammonia.Changing the feed to a higher FW content in a ratio of FVW to FW 1:2,resulted in an increase in VFAs concentration to 1100-1200mg/L,and the methanogenesis was slightly inhibited.At the optimum mixture ratio 1:1 for co-digestion of FVW with FW,the methane production yield was 0.49 m 3 CH 4/kg VS,and the volatile solids and soluble chemical oxygen demand (sCOD) removal efficiencies were 74.9% and 96.1%,respectively.Jia Lin Jiane Zuo Lili Gan Peng Li Fenglin Liu Kaijun Wang Lei Chen Hainan Gan 2011Journal of Environmental Sciences2011,23,8:36
2Molecular modeling and spectroscopic studies on the binding of guaiacol to human immunoglobulin显示文摘The fluorogenic property of guaiacol was exploited for the first time to analyze the interaction with target protein as a probe by molecular modeling, fluorescence, circular dichroism (CD) and Fourier transform infrared (FT-IR) spectroscopy. Molecular docking was performed to reveal the possible binding mode or mechanism and suggested that guaiacol can strongly bind to human immu- noglobulin (HIgG). It is considered that guaiacol binds to HIgG mainly by a hydrophobic interaction and there are two hydrogen bond interactions between the drug and the residues LEU 80 and ASP 65, which is in good agreement with the results from the experimental thermodynamic parameters (the enthalpy change △H0 and the entropy change △S0 were calculated to be 65.55 kJ·mol-1 and 132.95 J·mol-1·K-1 according to the Vant’ Hoff equation). Data obtained by the fluorescence spectroscopy indicated that binding of guaiacol with HIgG leads to dramatic enhancement in the fluorescence emission intensity along with significant occurrence of efficient Frster resonance energy transfer (FRET) from the residue of HIgG to the protein bound guaiacol. From the low value of fluorescence anisotropy (r = 0.06), it is argued that the probe molecule is located in the motionally unrestricted environment of the protein. 4The involvement of PybZIPa in light-induced anthocyanin accumulation via the activation of PyUFGT through binding to tandem G-boxes in its promoter显示文摘To gain insight into how anthocyanin biosynthesis is controlled by light in fruit,transcriptome and metabolome analyses were performed in the Chinese sand pear cultivar“Mantianhong”(Pyrus pyrifolia)after bagging and bag removal.We investigated transcriptional and metabolic changes and gene-metabolite correlation networks.Correlation tests of anthocyanin content and transcriptional changes revealed that 1,530 transcripts were strongly correlated with 15 anthocyanin derivatives(R2>0.9,P-value<0.05),with the top 130 transcripts categorized as being associated with flavonoid metabolism,transcriptional regulation,and light signaling.The connection network revealed a new photosensitive transcription factor,PybZIPa,that might play an important role during light-induced anthocyanin accumulation.The overexpression of PybZIPa promoted anthocyanin accumulation in pear and strawberry fruit as well as tobacco leaves.Dual luciferase and Y1H assays further verified that PybZIPa directly activated the expression of PyUFGT by binding to tandem G-box motifs in the promoter,which was key to differential anthocyanin accumulation in debagged pear skin,and the number of G-box motifs affected the transcriptional activation of PyUFGT by PybZIPa.The results indicate that the light-induced anthocyanin biosynthesis regulatory mechanism in pear differs from that described in previous reports suggesting that a bZIP family member co-regulates anthocyanin biosynthesis with other transcription factors in apple and Arabidopsis.It was found that,in response to light,PybZIPa promoted anthocyanin biosynthesis by regulating important transcription factors(PyMYB114,PyMYB10,and PyBBX22)as well as structural genes(PyUFGT)via binding to G-boxes within promoters.This activation was amplified by the self-binding of PybZIPa to activate its own promoter.Overall,we demonstrate the utility of a multiomics integrative approach for discovering new functional genes and pathways underlying light-induced anthocyanin biosynthesis.Hainan Liu Jun Su Yangfan Zhu Gaifang Yao Andrew C.Allan Charles Ampomah-Dwamena Qun Shu Kui Lin-Wang Shaoling Zhang Jun Wu 2019Horticulture Research2019,6,1:3
5Effect of Nano Silica on Hydration and Microstructure Characteristics of Cement High Volume Fly Ash System Under Steam Curing显示文摘The influences of nano silica (NS) on the hydration and microstructure development of steam cured cement high volume fly ash (40 wt%, CHVFA) system were investigated. The compressive strength of mortars was tested with different NS dosage from 0 to 4%. Results show that the compressive strength is dramatically improved with the increase of NS content up to 3%, and decreases with further increase of NS content (e g, at 4%). Then X?ray diffraction (XRD), differential scanning calorimetry-thermogravimetry (DSCTG), scanning electron microscope (SEM), energy disperse spectroscopy (EDS), mercury intrusion porosimeter (MIP) and nuclear magnetic resonance (NMR) were used to analyze the mechanism. The results reveal that the addition of NS accelerates the hydration of cement and fly ash, decreases the porosity and the content of calcium hydroxide (CH) and increases the polymerization degree of C-S-H thus enhancing the compressive strength of mortars. 6Neuroprotective role of edaravone and the effects of endoplasmic reticulum stress in an adult rat model of focal cerebral ischemia/reperfusion显示文摘BACKGROUND: Endoplasmic reticulum (ER) stress impairs ER functions and leads to the accumulation of unfolded or misfolded proteins in the ER lumen. ER stress-induced cell death plays an important role in cerebral ischemia. Edaravon (3-methyl-1-phenyl-2-pyrazolin-5-one) is a potent and novel scavenger of free radicals that inhibit delayed neuronal death, as demonstrated by in vitro and animal studies. However, its effect on ER stress and induced neuronal apoptosis in a rat model of brief middle cerebral artery occlusion remains unclear. OBJECTIVE: To explore the effects of edaravone on the expression of ER stress-related factors and neuronal apoptosis, based on the hypothesis that edaravone influences ER stress in a rat model of cerebral ischemia/reperfusion. DESIGN, TIME AND SETTING: A randomized, controlled, animal study was performed at the Laboratory of Department of Neurology, Xiangya Hospital and the Department of Laboratory Animals, Xiangya Medical College, Central South University in China from June 2005 to May 2006. MATERIALS: Edaravone was purchased from Simcere Pharmaceutical Group, China. METHODS: A total of 216 adult, male, Sprague Dawley rats were randomly assigned to sham-surgery, model and edaravone groups, with 72 rats in each group. Brief middle cerebral artery occlusion was established in the model and edaravone groups. In addition, the edaravone group rats were injected with 3 mg/kg edaravone through the tail vein. MAIN OUTCOME MEASURES: RNA-dependent protein kinase-like endoplasmic reticulum eukaryotic translation initiation factor 2α kinase (PERK) and C/EBP homology protein (CHOP) mRNA expression in the ischemic parietal cortex was determined by reverse transcription- polymerase chain reaction; phosphorylated PERK and CHOP protein expression was detected by immunohistochemistry; neuronal apoptosis was detected by TdT-mediated-dUTP nick end labeling. RESULTS: Neurological deficit scores were significantly reduced in the edaravone group compared to the model group at 12, 24, and 72 hours following reperfusion (P < 0.05). In addition, PERK and CHOP mRNA as well as phosphorylated PERK and CHOP protein expression were significantly reduced in the edaravone group compared to the model group at 1, 3, and 6 hours following reperfusion (P < 0.05, P < 0.01). CHOP mRNA expression was decreased in the edaravone group compared to the model group at 3, 6, 12, and 24 hours following reperfusion (P < 0.01), while CHOP protein expression was less than the model group at 6, 12, and 24 hours following reperfusion (P < 0.05). CONCLUSION: Edaravone treatment resulted in decreased PERK and CHOP expression following ischemia/reperfusion, as well as reduced neuronal apoptosis. 8Single-event burnout hardening of planar power MOSFET with partially widened trench source显示文摘We present a single-event burnout(SEB) hardened planar power MOSFET with partially widened trench sources by three-dimensional(3 D) numerical simulation. The advantage of the proposed structure is that the work of the parasitic bipolar transistor inherited in the power MOSFET is suppressed effectively due to the elimination of the most sensitive region(P-well region below the N+ source). The simulation result shows that the proposed structure can enhance the SEB survivability significantly. The critical value of linear energy transfer(LET),which indicates the maximum deposited energy on the device without SEB behavior, increases from 0.06 to0.7 p C/μm. The SEB threshold voltage increases to 120 V, which is 80% of the rated breakdown voltage. Meanwhile, the main parameter characteristics of the proposed structure remain similar with those of the conventional planar structure. 11The PyPIF5-PymiR156a-PySPL9-PyMYB114/ MYB10 module regulates light-induced anthocyanin biosynthesis in red pear显示文摘Some cultivars of pear(Pyrus L.)show attractive red fruit skin due to anthocyanin accumulation.This pigmentation can be affected by environmental conditions,especially light.To explore the light-induced regulation network for anthocyanin biosynthesis and fruit coloration in pear,small RNA libraries and mRNA libraries from fruit skins of‘Yunhongyihao’pear were constructed to compare the difference between bagging and debagging treatments.Analysis of RNA-seq of fruit skins with limited light(bagged)and exposed to light(debagged),showed that PyPIF5 was down-regulated after bag removal.PymiR156a was also differentially expressed between bagged and debagged fruit skins.We found that PyPIF5 negatively regulated PymiR156a expression in bagged fruits by directly binding to the G-box motif in its promoter.In addition,PymiR156a overexpression promoted anthocyanin accumulation in both pear skin and apple calli.We confirmed that PymiR156a mediated the cleavage of PySPL9,and that the target PySPL9 protein could form heterodimers with two key anthocyanin regulators(PyMYB114/PyMYB10).We proposed a new module of PyPIF5-PymiR156a-PySPL9-PyMYB114/MYB10.When the bagged fruits were re-exposed to light,PyPIF5 was down-regulated and its inhibitory effect on PymiR156a was weakened,which leads to degradation of the target PySPL,thus eliminating the blocking effect of PySPL on the formation of the regulatory MYB complexes.Ultimately,this promotes anthocyanin biosynthesis in pear skin.Hainan Liu Qun Shu Kui Lin-Wang Andrew CAllan Richard VEspley Jun Su Maosong Pei Jun Wu 2021Molecular Horticulture2021,1,1:1

18Alternative splicing of the PECTINESTERASE gene encoding a cell wall-degrading enzyme affects postharvest softening in grape显示文摘The firmness of table grape berries is a crucial quality parameter. Despite extensive research on postharvest fruit softening, its precise molecular mechanisms remain elusive. To enhance our comprehension of the underlying molecular factors, we initially identified differentially expressed genes(DEGs) by comparing the transcriptomes of folic acid(FA)-treated and water-treated(CK) berries at different time points. We then analyzed the sequences to detect alternatively spliced(AS) genes associated with postharvest softening. A total of 2,559 DEGs were identified and categorized into four subclusters based on their expression patterns, with subcluster-4 genes exhibiting higher expression in the CK group compared with the FA treatment group. There were 1,045 AS-associated genes specific to FA-treated berries and 1,042 in the CK-treated berries, respectively. Gene Ontology(GO) annotation indicated that the AS-associated genes in CK-treated berries were predominantly enriched in cell wall metabolic processes,particularly cell wall degradation processes. Through a comparison between treatment-associated AS genes and subcluster-4 DEGs, we identified eight genes, including Pectinesterase 2(VvPE2, Vitvi15g00704), which encodes a cell wall-degrading enzyme and was predicted to undergo an A3SS event. 19Basic Properties of Concrete Incorporating Recycled Ceramic Aggregate and Ultra-fine Sand显示文摘Recycled ceramic mixed sand(RCMS) was obtained by partially replacing ultra-fine sand with recycled ceramic coarse sand(RCCS). The effects of RCCS replacement rate on the apparent density, workability, compressive strength and splitting tensile strength of recycled ceramic concrete(RCC) were investigated. In addition, the relationship between the water-cement ratio and compressive strength of RCC was also studied. The experimental results indicate that the reusing of recycled ceramic aggregate can improve the cohesiveness and water retentiveness of fresh concrete and benefit the mechanical properties development. When the RCCS replacement rate is not less than 40%, the mechanical properties of RCC are superior to those of the reference concrete. Moreover, when recycled ceramic medium sand was completely used as fine aggregate, the maximum increase in both compressive strength and splitting tensile strength were obtained, comparing with those of reference concrete, the increment ratio was 19.85% and 32.73%, respectively. The microscopic analysis shows that the using of recycled ceramic aggregate can meliorate distinctly the structure of the interfacial transition zone(ITZ) and increase the compaction degree of cement paste. Furthermore, an expression of the compressive strength of RCC and the cement-water ratio is regressed and gains a good linear relativity. 20Recent Advances in Understanding Multi-scale Climate Variability of the Asian Monsoon显示文摘Studies of the multi-scale climate variability of the Asian monsoon are essential to an advanced understanding of the physical processes of the global climate system.In this paper,the progress achieved in this field is systematically reviewed,with a focus on the past several years.The achievements are summarized into the following topics:(1)the onset of the South China Sea summer monsoon;(2)the East Asian summer monsoon;(3)the East Asian winter monsoon;and(4)the Indian summer monsoon.Specifically,new results are highlighted,including the advanced or delayed local monsoon onset tending to be synchronized over the Arabian Sea,Bay of Bengal,Indochina Peninsula,and South China Sea;the basic features of the record-breaking mei-yu in 2020,which have been extensively investigated with an emphasis on the role of multi-scale processes;the recovery of the East Asian winter monsoon intensity after the early 2000s in the presence of continuing greenhouse gas emissions,which is believed to have been dominated by internal climate variability(mostly the Arctic Oscillation);and the accelerated warming over South Asia,which exceeded the tropical Indian Ocean warming,is considered to be the main driver of the Indian summer monsoon rainfall recovery since 1999.A brief summary is provided in the final section along with some further discussion on future research directions regarding our understanding of the Asian monsoon variability.Wen CHEN Renhe ZHANG Renguang WU Zhiping WEN Liantong ZHOU Lin WANG Peng HU Tianjiao MA Jinling PIAO Lei SONG Zhibiao WANG Juncong LI Hainan GONG Jingliang HUANGFU Yong LIU 2023Advances in Atmospheric Sciences2023,40,8:0
