2Photostable and Biocompatible Fluorescent Silicon Nanoparticles for Imaging-Guided Co-Delivery of siRNA and Doxorubicin to Drug-Resistant Cancer Cells显示文摘The development of effective and safe vehicles to deliver small interfering RNA(siRNA) and chemotherapeutics remains a major challenge in RNA interference-based combination therapy with chemotherapeutics,which has emerged as a powerful platform to treat drug-resistant cancer cells.Herein,we describe the development of novel all-in-one fluorescent silicon nanoparticles(SiNPs)-based nanomedicine platform for imaging-guided co-delivery of siRNA and doxorubicin(DOX).This approach enhanced therapeutic efficacy in multidrug-resistant breast cancer cells(i.e.,MCF-7/ADR cells).Typically,the SiNP-based nanocarriers enhanced the stability of siRNA in a biological environment(i.e.,medium or RNase A) and imparted the responsive release behavior of siRNA,resulting in approximately 80% down-regulation of P-glycoprotein expression.Co-delivery of P-glycoprotein siRNA and DOX led to>35-fold decrease in the half maximal inhibitory concentration of DOX in comparison with free DOX,indicating the pronounced therapeutic efficiency of the resultant nanocomposites for drug-resistant breast cancer cells.The intracellular time-dependent release behaviors of siRNA and DOX were revealed through tracking the strong and stable fluorescence of SiNPs.These data provide valuable information for designing effective RNA interference-based co-delivery carriers.Daoxia Guo Xiaoyuan Ji Fei Peng Yiling Zhong Binbin Chu Yuanyuan Su Yao He 2019Nano-Micro Letters2019,11,2:5
3Short-working-distance optical imaging system and method for surface detection of underwater structures显示文摘In the surface imaging of underwater structures, long working distance will reduce image quality due to the turbidity of water. To acquire high definition and large field of view(FOV) images for surface detection, a short-working-distance underwater imaging system is proposed based on camera array. A multi-view calibration and rectification method is developed. A look-up table(LUT) method and a multi-resolution spline(MRS) method are applied to stitch array images real-time and seamlessly.Experiments both in the air and in the water are conducted. 4Interferon as a Mucosal Adjuvant for an Influenza Vaccine in Pigs显示文摘Interferon,a natural protein that is produced by a variety of cells during viral infection,activates the transcription of multiple functional genes in cells,regulates synergy among various signaling pathways,and mediates many biological functions such as antiviral activity,immune regulation,and cell growth.However,clinical research on interferon in livestock is lacking.In this study,recombinant porcine interferon(PoIFNα)was used as an adjuvant,in combination with inactivated influenza virus,to vaccinate 6-week-old pigs via nasal infusion.The transcription of target genes was then monitored and the functions of PoIFNαwere determined with respect to the activation of mucosal immunity.We found that a combination of low-dose PoIFNαand in activated influenza virus could significantly up-regulate the expression of immunoregulatory cytokines such as IL-2,IL-1β,IFN-γ,IL-6,and IL-10 by real-time PCR,suggesting the induction of a strong mucosal innate immune response after administration.In addition,low-dose PoIFNαcan significant enhancing the transcription of genes encoding homing factors including CCR9 and CCR10(P<0.001),thereby resulting in the induction of higher levels of HA-specific antibodies(P<0.05),which can be determined by ELISA and IFA.Post-immunization challenges with H1N1 virus demonstrated that PoIFNot,combined with inactivated influenza virus,could alleviate clinical signs in pigs during the early stages of viral infection.These studies reveal low-dose PoIFNαas a potential mucosal adjuvant for influenza virus in pigs.Lirong Liu Wenhui Fan He Zhang Shuang Zhang Liang Cui Meng Wang Xiaoyuan Bai Wenxian Yang Lei Sun Limin Yang Wenjun Liu Jing Li 2019Virologica Sinica2019,34,3:3
5The critical role of Indian summer monsoon on the remote forcing between Indian and Northwest Pacific during El Nio decaying year显示文摘Recent studies have found a connection between Indian Ocean Basin Warming and the anomalous Northwest Pacific Anticyclone(ANPWA) during El Ni?o decaying year.This study focuses on the necessary condition for this connection by using observation and numerical simulation.The seasonal transition of the Indian Ocean sea surface wind is critical to the climatic effect of Indian Ocean Basin Warming.When the South Asian Summer Monsoon reaches its peak,the background wind becomes desirable for basin warming,which then affects the climate in the Northwest Pacific.Via the Kelvin waves and Ekman divergence,the wind anomalies exist in the lower atmosphere east of the Indian Ocean warm Sea Surface Temperature(SST) anomalies,and intensify and sustain the ANWPA throughout the El Nio decaying summer.This impact plays an important role in the inter-annual variability of the East Asian Summer Monsoon.HU HaiBo HONG XiaoYuan ZHANG Yuan YANG XiuQun HE Jie 2013Science China Earth Sciences2013,56,3:3

8Radical recombination in a hydrocarbon-fueled scramjet nozzle显示文摘To reveal the radical recombination process in the scramjet nozzle flow and study the effects of various factors of the recombination, weighted essentially non-oscillatory(WENO)schemes are applied to solve the decoupled two-dimensional Euler equations with chemical reactions to simulate the hydrocarbon-fueled scramjet nozzle flow. The accuracy of the numerical method is verified with the measurements obtained by a shock tunnel experiment. The overall model length is nearly 0.5 m, with inlet static temperatures ranging from 2000 K to 3000 K, inlet static pressures ranging from 75 k Pa to 175 k Pa, and inlet Mach numbers of 2.0 ± 0.4 are involved.The fraction Damkohler number is defined as functions of static temperature and pressure to analyze the radical recombination progresses. Preliminary results indicate that the energy releasing process depends on different chemical reaction processes and species group contributions. In hydrocarbon-fueled scramjet nozzle flow, reactions with H have the greatest contribution during the chemical equilibrium shift. The contrast and analysis of the simulation results show that the radical recombination processes influenced by inflow conditions and nozzle scales are consistent with Damkohler numbers and potential dissociation energy release. The increase of inlet static temperature improves both of them, thus making the chemical non-equilibrium effects on the nozzle performance more significant. 9An oncolytic adenovirus delivering TSLC1 inhibits Wnt signaling pathway and tumor growth in SMMC-7721 xenograft mice model显示文摘Tumor suppressor in lung cancer-1(TSLC1)was first identified as a tumor suppressor for lung cancer,and frequently downregulated in various types of cancers including hepatocellular carcinoma(HCC).The Wnt pathway plays a critical role in tumorigenesis,migration,and invasion in HCC.However,the function of TSLC1 in modulating Wnt signaling in HCC is unclear.In this study,we evaluated the effect of TSLC1-armed oncolytic adenovirus(S24-TSLC1)on the Wnt/β-catenin pathway,cell viability,invasion and migration abilities of HCC in vitro and the growth of SMMC-7721-xenografted tumor in mice model.We detected the expression of TSLC1 in tumor samples and HCC cell lines.The results showed that TSLC1 expression was low in HCC,but high in pericarcinomatous tissue and normal cells,which implied that TSLC1 is a tumor suppressor of liver cancer.S24-TSLC1 exhibited an antitumor effect on HCC cell growth in vitro,but did little damage to normal liver cells.Overexpression of TSLC1 downregulated the transcriptional activity of TCF4/β-catenin and inhibited the mRNA or protein expression of Wnt target genes cyclinD1 and c-myc.S24-TSLC1 also inhibited the invasion and migration of HCC cells.Animal experiments further confirmed that S24-TSLC1 significantly inhibited tumor growth of the SMMC-7721-xenografted tumor.In conclusion,TSLC1 could downregulate the Wnt signal pathway and suppress HCC cell growth,migration and invasion,suggesting that S24-TSLC1 may be a potent antitumor agent for future clinical trials in liver cancer treatment.Yigang Wang Panpan Huang Yanping Hu Keni Guo Xiaoyuan Jia Biao Huang Xinyuan Liu Xianglei He Fang Huang 2021Acta Biochimica et Biophysica Sinica2021,53,6:2
10Integrated Co3O4/carbon fiber paper for high-performance anode of dual-ion battery显示文摘In dual-ion batteries (DIBs), energy storage is achieved by intercalation/de-intercalation of both cations and anions. Due to the mismatch between ion diameter and layer space of active materials, however, volume expansion and exfoliation always occur for electrode materials. Herein, an integrated electrode Co3O4/carbon fiber paper (CFP) is prepared as the anode of DIB. As the Co3O4 nanosheets grow on CFP substrate vertically, it promotes the immersion of electrolyte and shortens the pathway for ionic transport. Besides, the strong interaction between Co3O4 and CFP substrate reduces the possibility of sheet exfoliation. An integrated-electrode-based DIB is therefore packaged using Co3O4/CFP as anode and graphite as cathode. As a result, a high energy density of 72 Wh/kg is achieved at a power density of 150 W/kg. 11Fluorescent silicon nanoparticles-based nanotheranostic agents for rapid diagnosis and treatment of bacteria-induced keratitis显示文摘Recommended as a medical emergency,infectious keratitis with an acute and rapid disease progression can lead to serious damage of vision and even blindness.Herein,we present a kind of theranostic agents,which are made of vancomycin(Van)-modified fluorescent silicon nanoparticles(SiNPs-Van),enabling rapid and non-invasive diagnosis and treatment of Gram-positive bacteria-induced keratitis in a simultaneous manner.Typically,the resultant SiNPs-Van nanoagents have an ability of imaging bacteria in a short time both in vitro(5 min)and in vivo(10 min),making them an efficacious diagnostic agent for the detection of bacterial keratitis.In addition,the SiNPs-Van feature distinct antimicrobial activity,with superior activity of 92.5%at a concentration of 0.5 ng/mL against Staphylococcus aureus(S.aureus);comparatively,the antimicrobial rate of free vancomycin is 23.3%at the same concentration.We further explore the SiNPs-Van agents as eye drops for therapy of S.aureus-induced bacterial keratitis on rat model.Represented by slit-lamp scores,the keratitis severity of SiNPs-Van-treated corneas is 3.4,which is 59.6%and 77.3%slighter than vancomycin-(8.2 score)and PBS-treated corneas(15.0 score),respectively.The infected corneas recover to normal(1 score)after 7-d of SiNPs-Van treatment.Above results suggest that the SiNPs-Van could serve as a new kind of high-quality nanotheranostic agents,especially suitable for simultaneous diagnosis and therapy of Gram-positive bacteria keratitis.Lu Zhang Xiaoyuan Ji Yuanyuan Su Xia Zhai Hua Xu Bin Song Airui Jiang Daoxia Guo Yao He 2021Nano Research2021,14,1:2
12Two Ultraviolet Radiation Datasets that Cover China显示文摘Ultraviolet(UV) radiation has significant effects on ecosystems, environments, and human health, as well as atmospheric processes and climate change. Two ultraviolet radiation datasets are described in this paper. One contains hourly observations of UV radiation measured at 40 Chinese Ecosystem Research Network stations from 2005 to 2015. CUV3 broadband radiometers were used to observe the UV radiation, with an accuracy of 5%, which meets the World Meteorology Organization's measurement standards. The extremum method was used to control the quality of the measured datasets. The other dataset contains daily cumulative UV radiation estimates that were calculated using an all-sky estimation model combined with a hybrid model. The reconstructed daily UV radiation data span from 1961 to 2014. The mean absolute bias error and root-mean-square error are smaller than 30% at most stations, and most of the mean bias error values are negative, which indicates underestimation of the UV radiation intensity. These datasets can improve our basic knowledge of the spatial and temporal variations in UV radiation. 17Chain-shattering Pt(IV)-backboned polymeric nanoplatform for efficient CRISPR/Cas9 gene editing to enhance synergistic cancer therapy显示文摘CRISPR/Cas9 system has become a promising gene editing tool for cancer treatment.However,development of a simple and effective nanocarrier to incorporate CRISPR/Cas9 system and chemotherapeutic drugs to concurrently tackle the biological safety and packaging capacity of viral vectors and combine gene editing-chemo for cancer therapy still remains challenges.Herein,a chain-shattering Pt(IV)-backboned polymeric nanoplatform is developed for the delivery of EZH2-targeted CRISPR/Cas9 system(NPCSPt/pEZH2)and synergistic treatment of prostate cancer.The pEZH2/Pt(II)could be effectively triggered to unpack/release from NPCSPt/pEZH2 in a chain-shattering manner in cancer cells.The EZH2 gene disruption efficiency could be achieved up to 32.2%of PC-3 cells in vitro and 21.3%of tumor tissues in vivo,leading to effective suppression of EZH2 protein expression.Moreover,significant H3K27me3 downregulation could occur after EZH2 suppression,resulting in a more permissive chromatin structure that increases the accessibility of released Pt(II)to nuclear DNA for enhanced apoptosis.Taken together,substantial proliferation inhibition of prostate cancer cells and further 85.4%growth repression against subcutaneous xenograft tumor could be achieved.This chain-shattering Pt(IV)-backboned polymeric nanoplatform system not only provides a prospective nanocarrier for CRISPR/Cas9 system delivery,but also broadens the potential of combining gene editing-chemo synergistic cancer therapy.Qingfei Zhang Gaizhen Kuang Shasha He Sha Liu Hongtong Lu Xiaoyuan Li Dongfang Zhou Yubin Huang 2021Nano Research2021,14,3:1
