1Spatial and temporal variations of forest LAI in China during 2000-2010显示文摘Forests are crucial terrestrial ecosystems.Their leaf area index (LAI) is a key parameter determining the exchange of matter and energy between the atmosphere and the ground surface.In this study,MOD 09A1 and MCD 43A1 data were input into an inversion model based on the 4-scale geometric optical model to retrieve 8-d 500 m LAI products in China during the period 2000 to 2010.The resulting LAI product was validated using LAI measured in 6 typical areas.The spatial and temporal variations of forest LAI and its relationships with temperature and precipitation were analyzed.The results show that the accuracy of the 500 m LAI product was above 70% in the 6 typical areas,indicating the reliability of this product.From 2000 to 2010,forest LAI in northeast,north,and south central China showed increasing trends.However,forest LAI in southeast and parts of southwest China showed downward trends,mainly because of the significant decrease observed during the period 2008 to 2010.Annual mean forest LAI positively correlated with annual mean temperature (AMT) in northeastern China and negatively correlated with AMT in southwest China.It had positive correlations with annual total precipitation in central south and north China.The abnormal climate conditions in 2001 and 2009 caused forest LAI to be obviously lower than normal in regions south of the Qinling Mountains and Huaihe River.Annual mean LAI decreased by more than 1.0 in some areas.LIU YiBo JU WeiMin CHEN JingMing ZHU GaoLong XING BaiLing ZHU JingFang HE MingZhu 2012Chinese Science Bulletin2012,57,22:22
2Patterns of shrub species richness and abundance in relation to environmental factors on the Alxa Plateau:Prerequisites for conserving shrub diversity in extreme arid desert regions显示文摘Shrub species are considered the dominant plants in arid desert ecosystems,unlike in semiarid steppe zones or in grassland ecosystems.On the Alxa Plateau,northern China,sparse vegetation with cover ranging from 15% to 30% is characterized mainly by multifarious shrubs because herbaceous species are strongly restricted by the extreme drought climate,wind erosion,overgrazing and sand burial.Patterns in shrub species richness and species abundance in relation to environmental conditions were examined by DCA(detrended correspondence analysis) and interpreted by a biplot.The rela-tionships between species diversity and environmental factors were examined using regression analyses.Our results show that the distributions of the shrub species in response to environmental conditions can be grouped into four ecological types,corresponding with the biological traits of the shrubs and their responses to the gradients of soil texture and soil water content.Patterns in species richness and species abundance were mainly determined by the deeper soil water content,instead of the soil texture as hypothesized by numerous studies in semiarid grasslands.With exception of the deeper soil water content,soil organic matter and total N content were positively correlated with species abundance,while pH was negatively correlated with it.These findings imply that it is vital for cur-rent shrub diversity conservation to reduce agricultural water use in the middle reaches of the Heihe River,which supplies water for the lower reaches in the western parts of the plateau,and to reduce the amount of groundwater exploitation and urban and oasis water use,to increase the water supply from Helan Mountain to the eastern desert of the Alxa Plateau.XinRong Li HuiJuan Tan MingZhu He XinPing Wang XiaoJun Li 2009Science China Earth Sciences2009,52,5:17

4Improving Plant Genome Editing with High-Fidelity xCas9 and Non-canonical PAM-Targeting Cas9-NG显示文摘Two recently engineered SpCas9 variants, namely xCas9 and Cas9-NG, show promising potential in improving targeting specificity and broadening the targeting range. In this study, we evaluated these Cas9 variants in the model and crop plant, rice. We first tested xCas9-3.7, the most effective xCas9 variant in mammalian cells, for targeted mutagenesis at 16 possible NGN PAM (protospacer adjacent motif) combinations in duplicates. xCas9 exhibited nearly equivalent editing efficiency to wild-type Cas9 (Cas9-WT) at most canonical NGG PAM sites tested, whereas it showed limited activity at non-canonical NGH (H = A, C, T) PAM sites. High editing efficiency of xCas9 at NGG PAMs was further demonstrated with C to T base editing by both rAPOBECI and PmCDAI cytidine deaminases. With mismatched sgRNAs, we found that xCas9 had improved targeting specificity over the Cas9-WT. Furthermore, we tested two Cas9-NG variants, Cas9-NGv1 and Cas9-NG, for targeting NGN PAMs. Both Cas9-NG variants showed higher editing efficiency at most non-canonical NG PAM sites tested, and enabled much more efficient editing than xCas9 at AT-rich PAM sites such as GAT, GAA, and CAA. Nevertheless, we found that Cas9-NG variants showed significant reduced activity at the canonical NGG PAM sites. In stable transgenic rice lines, we demonstrated that Cas9-NG had much higher editing efficiency than Cas9-NGv1 and xCas9 at NG PAM sites. To expand the base-editing scope, we developed an efficient C to T base-editing system by making fusion of Cas9-NG nickase (D10A version), PmCDAI, and UGI. 6Enhanced Poly(ethylene terephthalate)Hydrolase Activity by Protein Engineering显示文摘Poly(ethylene terephthalate)hydrolase(PETase)from Ideonella sakaiensis exhibits a strong ability to degrade poly(ethylene terephthalate)(PET)at room temperature,and is thus regarded as a potential tool to solve the issue of polyester plastic pollution.Therefore,we explored the interaction between PETase and the substrate(a dimer of the PET monomer ethylene terephthalate,2PET),using a model of PETase and its substrate.In this study,we focused on six key residues around the substrate-binding groove in order to create novel high-efficiency PETase mutants through protein engineering.These PETase mutants were designed and tested.The enzymatic activities of the R61A,L88F,and I179F mutants,which were obtained with a rapid cell-free screening system,exhibited 1.4 fold,2.1 fold,and 2.5 fold increases,respectively,in comparison with wild-type PETase.The I179F mutant showed the highest activity,with the degradation rate of a PET film reaching 22.5 mg perμmol·L^-1 PETase per day.Thus,this study has created enhanced artificial PETase enzymes through the rational protein engineering of key hydrophobic sites,and has further illustrated the potential of biodegradable plastics.Yuan Ma Mingdong Yao Bingzhi Li Mingzhu Ding Bo He Si Chen Xiao Zhou Yingjin Yuan 2018Engineering2018,4,6:10
7Rainfall interception by sand-stabilizing shrubs related to crown structure显示文摘On the edge of the Tengger Desert in northern China,revegetation has changed the landscape from moving dunes to stabilized dunes covered by shrubs,which further modifies the pattern of rainfall redistribution.To study rainfall interception loss by shrubs and its relationship to rainfall properties and crown structure,throughfalls passing through crowns of Artemisia ordosica Krash.and Caragana korshinskii Kom.were measured using nine PVC cups under the canopy of each of the two shrubs during 73 rain events over a three-year period,with total rainfall of 260.9 mm.Interception losses of gross rainfall by A.ordosica and C.korshinskii account for 15% and 27% of the total on a crown area basis,and 6% and 11% on a ground area basis,respectively.Individual throughfall(T) and interception(I) were significantly related to rainfall amount(Pg),duration(D),and intensity(R).Ratios of throughfall to rainfall(T/Pg) and interception to rainfall(I/Pg) were not only significantly related to Pg,D,and R,but also to shrub species,and interactions of species with crown volume(CV) and leaf area index(LAI).Under most rain events,interceptions by C.korshinskii with greater CV and LAI were significantly higher than those by A.ordosica,and more rainfall interception occurred at locations closer to the stems of the two shrubs.For C.korshinskii,I/Pg had a significant positive linear relation with CV and LAI,while T/Pg had a significant negative linear relation with them.CV has a greater influence on T/Pg and I/Pg than does LAI.Using a regression method,canopy water storage capacities are estimated to be 0.52 and 0.68 mm,and free throughfall coefficient to be 0.62 and 0.47 for A.ordosica and C.korshinskii,respectively.ZhiShan Zhang 1*,XinRong Li 1,XueJun Dong 2,XiaoHong Jia 1,MingZhu He 1,HuiJuan Tan 1 1.Shapotou Desert Research and Experiment Station,Cold and Arid Regions Environmental and Engineering Research Institute,Chinese Academy of Sciences,Lanzhou,Gansu 730000,China.2.Central Grasslands Research Extension Center,North Dakota State University,Streeter,ND 58483,USA. 2009Research in Cold and Arid Regions2009,1,2:9
8SNP-based QTL mapping for panicle traits in the japonica super rice cultivar Liaoxing 1显示文摘Panicle traits directly associated with yield are a target of selection in rice breeding.Although abundant QTL for panicle traits have been identified,there is little information about the genetic basis of panicle traits in japonica super rice(JSR)cultivars.In this study,we identified QTL for panicle traits in three environments using a population of recombinant inbred lines(RILs)derived from the JSR cultivar Liaoxing 1.A total of 197 RILs were genotyped with 285 polymorphic SNP markers.Phenotypic data and best linear unbiased prediction(BLUP)value of primary branch number(BNP),secondary branch number(BNS),grain number on primary branch(GNP),grain number on secondary branch(GNS),grain number per panicle(GN),panicle length(PL)and grain density(GD)were used for QTL mapping.A total of 105 QTL for seven panicle traits were detected in single environments using their BLUP values.Individual QTL explained 0.51%–52.22%of the phenotypic variation.Of the 105,49 were also detected by joint multi-environment analyses.Five stable QTL:qGD9,qPL9,qGNP9,qGN6,and qBNS6.2 were identified in multiple environments.qGD9,qGNP9,and qPL9,co-localizing on chromosome 9,likely correspond to the known gene DEP1.Importantly,qGN6 and qBNS6.2 in a co-localization region were identified as novel QTL,and their Liaoxing 1 alleles had a positive effect.Several RILs with the QTL allele combinations qGD9/qPL9/qGNP9 and qGN6/qBNS6.2 showed greater GN.Further investigation of the putative gene underlying qGN6/qBNS6.2 would shed light on the molecular mechanism of JSR.Mingzhu Zhao Zuobin Ma Lili Wang Zhiqiang Tang Ting Mao Chuanbin Liang Hong Gao Liying Zhang Na He Liang Fu Changhua Wang Guomin Sui Wenjing Zheng 2020The Crop Journal2020,8,5:4
9Cultivation effects on soil texture and fertility in an arid desert region of northwestern China显示文摘In arid desert regions of northwestern China, reclamation and subsequent irrigated cultivation have become effective ways to prevent desertification, expand arable croplands, and develop sustainable agricultural production. Improvement in soil texture and fertility is crucial to high soil quality and stable crop yield. However, knowledge on the long-term effects of the conversion of desert lands into arable croplands is very limited. To address this problem, we conducted this study in an arid desert region of northwestern China to understand the changes in soil physical-chemical properties after 0, 2, 5, 10, 17, and 24 years of cultivation. Our results showed that silt and clay contents at the 17-year-old sites increased 17.5 and 152.3 folds, respectively, compared with that at the 0-year-old sites. The soil aggregate size fraction and its stability exhibited an exponential growth trend with increasing cultivation ages, but no significant change was found for the proportion of soil macroaggregates(>5.00 mm) during the 17 years of cultivation. The soil organic carbon(SOC) content at the 24-year-old sites was 6.86 g/kg and increased 8.8 folds compared with that at the 0-year-old sites. The total(or available) nitrogen, phosphorus, and potassium contents showed significant increasing trends and reached higher values after 17(or 24) years of cultivation. Changes in soil physical-chemical properties successively experienced slow, rapid, and stable development stages, but some key properties(such as soil aggregate stability and SOC) were still too low to meet the sustainable agricultural production. 10Mechanism of allosteric activation of SIRT6 revealed by the action of rationally designed activators显示文摘The recent discovery of activator compounds binding to an allosteric site on the NAD+-dependent protein lysine deacetylase,sirtuin 6(SIRT6)has attracted interest and presents a pharmaceutical target for aging-related and cancer diseases.However,the mechanism underlying allosteric activation of SIRT6 by the activator MDL-801 remains largely elusive because no major conformational changes are observed upon activator binding.By combining molecular dynamics simulations with biochemical and kinetic analyses of wild-type SIRT6 and its variant M136 A,we show that conformational rotation of 2-methyl-4-fluoro-5-bromo substituent on the right phenyl ring(R-ring)of MDL-801,which uncovers previously unseen hydrophobic interactions,contributes to increased activating deacetylation activity of SIRT6.This hypothesis is further supported by the two newly synthesized MDL-801 derivatives through the removal of the 5-Br atom on the R-ring(MDL-801-D1)or the restraint of the rotation of the R-ring(MDL-801-D2).We further propose that the 5-Br atom serves as an allosteric driver that controls the ligand allosteric efficacy.Our study highlights the effect of allosteric enzyme catalytic activity by activator binding and provides a rational approach for enhancing deacetylation activity.Shaoyong Lu Yingyi Chen Jiacheng Wei Mingzhu Zhao Duan Ni Xinheng He Jian Zhang 2021Acta Pharmaceutica Sinica B2021,11,5:2

12Effect of Dietary Lipid on the Growth, Fatty Acid Composition and Δ5 Fads Expression of Abalone(Haliotis discus hannai Ino) Hepatopancreas显示文摘This study investigated the effect of dietary lipid on the growth, fatty acid composition and Δ5 fatty acyl desaturase genes(Fads) expression of juvenile abalone(Haliotis discus hannai Ino) hepatopancreas. Six purified diets were formulated to contain tripalmitin(TP), olive oil(OO, 72.87% 18:1n-9), grape seed oil(GO, 68.67% 18:2n-6), linseed oil(LO, 70.48% 18:3n-3), ARA oil(AO, 41.81% ARA) or EPA oil(EO, 44.09% EPA and 23.67% DAH). No significant difference in survival rate was observed among abalone fed with different diets. Weight gain rate(WGR) and daily growth rate of shell length(DGRSL) were significantly increased in abalone fed with diets containing OO, AO and EO, but decreased in abalone fed with LO diet(P < 0.05) in comparison with those fed with TP. High level of dietary 18:2n-6 resulted in higher content of n-6 polyunsaturated fatty acids(PUFAs) in abalone fed with GO than those fed with TP, OO, LO and EO(P < 0.05). n-3 PUFAs in abalone fed with LO was significantly higher than those in abalone fed with TP, OO, GO and AO(P < 0.05). The highest contents of 20:1n-9 and 22:1n-9 were observed in abalone fed with OO. The expression of Δ5 Fads in hepatopancreas of abalone was enhanced by high concentration of 18:3n-3 and suppressed by dietary LC-PUFAs; however it was not affected by dietary high concentration of 18:1n-9 or 18:2n-6. 15Micro-interfaces modulation by UV-ozone substrate treatment for MPEA vapor fluorescence detection显示文摘Drug abuse directly endangers human health and social security,hence its sensitive and rapid detection is vitally important.In recent years,organic film-based fluorescent sensing technology has attracted more and more attention in the detection of drugs and explosives due to its advantages of simple operation and rapid detection.For film-based fluorescent sensors,in addition to sensitive materials,the surface morphology of the film is also an important factor affecting the performance.In previous studies,the regulation of surface morphology mainly depends on concentration changes or complex templates.Here,a novel fluorescent polymer probe was designed and synthesized,and a simple and efficient ultraviolet(UV)-ozone substrate treatment method is used to adjust their surface morphology.The results show that film has an excellent fluorescence enhancement effect upon exposure to methylphenethylamine(MPEA,a simulant of methamphetamine)vapor.The sensing effect of the film is significantly improved after UV-ozone substrate treatment,and the limit of detection was decreased by 10.4 times from 2.59 to 0.25 ppm.Further experiments show that the sensing performance of other fluorescent probe can also be improved by the UV-ozone substrate treatment.This convenient and general method may become a very effective approach to improve the performance of film-based fluorescent sensors.Bin Li Keke Li Wei Xu Mingzhu Yan Jianhao Zhao Wukun Zhang Mingshuai Yuan Yanyan Fu Qingguo He Jiangong Cheng 2023Nano Research2023,16,3:0
16Silicon-based nanoprobes cross the blood–brain barrier for photothermal therapy of glioblastoma显示文摘Traditional photothermal agents of indocyanine green(ICG)have poor stability,short circulation time,and poor brain permeability due to the blood–brain barrier(BBB),greatly impairing their therapeutic efficacy in glioblastoma(GBM).Herein,we develop a novel kind of SiNPs-based nanoprobes to bypass the BBB for photothermal therapy of GBM.Typically,the SiNPs-based nanoprobes are composed of the particle itself,the BBB-targeting ligand of glucosamine(G),and the therapeutic agent of ICG.We demonstrate that the as-synthesized nanoprobes could cross the BBB through glucose transporter-1(GLUT1)-mediated transcytosis,followed by accumulation at GBM tissues in mice.Compared with free ICG,G-ICG-SiNPs show stronger stability(for example,the fluorescence intensity of G-ICG-SiNPs loaded with the same dose of ICG decays by 34.6%after 25 days of storage,while the fluorescence intensity of ICG decays by 99.5%under the same conditions).Furthermore,the blood circulation time of G-ICG-SiNPs increases by about 17.3-fold compared with their ICG counterparts.After injection of the therapeutic agents into the GBM-bearing mice,GBM-surface temperature rises to 45.3℃in G-ICG-SiNPs group after 5-min 808 nm irradiation but climbs only to 36.1℃in equivalent ICG group under the identical conditions,indicating the superior photothermal effects of GICG-SiNPs in vivo.Rong Sun Mingzhu Liu Zhaojian Xu Bin Song Yao He Houyu Wang 2022Nano Research2022,15,8:0
17Nickel effects on growth and antioxidative enzymes activities in desert plant Zygophyllum xanthoxylon (Bunge) Maxim显示文摘One-month old Zygophyllum xanthoxylon seedlings were grown under five different Ni concentrations (0, 50, 150, 450 and 900 mg/kg). Growth parameters, tissues Ni accumulation, soluble protein content, photosynthetic pigment content, lipid peroxidation, antioxidative enzyme activities, and proline content were studied after one month of Ni exposure. The significant reduction in fresh biomass in the shoot and roots was detected at high Ni concentrations (≥450 mg/kg), whereas dry biomass was not significantly affected in both organs by any of the Ni doses tested. The Ni contents in the shoots and roots increased in a dose-dependent manner, and Ni contents in roots were higher than that in shoots at all treatments. The high Ni doses elevated soluble protein contents in leaves and roots. No visible symptoms of chlorosis were observed in leaves of plants grown in presence of Ni. Malondialdehyde (MDA) contents significantly increased in leaves and roots at high Ni treatments (≥450 mg/kg), indicating that Ni induces oxidative stress. Enzyme activities might play a central role in cellular protection against the Ni induced oxidative stress. Proline contents proportionally increased with the elevated Ni concentrations. 18Construction of a Stimuli-Responsive DNAzyme-Braked DNA Nanomachine for the Amplified Imaging of miRNAs in Living Cells and Mice显示文摘Synthetic DNA motors have spurred considerable research interest as a way to interrogate biochemical processes that can further facilitate disease diagnosis.However,current direct sense-on-demand DNA motors are hampered by undesired signal leakage originating from their nonspecific stimulation prior to their arrival at target sites.Herein,we propose a DNAzyme-guided DNA amplification motor through the facile built-in of a stimuli-responsive DNAzyme brake to facilitate the in vivo sensitive imaging of biomarkers by minimizing the off-site signal leakage in living systems.To realize the on-site activation,the inactive DNAzyme DNA motor was encapsulated into the folate(FA)-modified zeolitic imidazolate framework-8(ZIF-8)/poly(D,L-lactic-co-glycolic acid(PLGA)nanovesicles.The FA interaction-promoted tumor-cell-selective delivery and the pH-responsive disassembly of ZIF-8/PLGA permitted sufficient release of DNAzyme cofactors to achieve on-site stimulation of DNAzyme for restoring the temporally caged DNA amplification motor in tumor cells where the endogenous miRNA target was thus specifically and sensitively analyzed.By integration of the cellselective delivery and the site-specific stimulation,our DNAzyme-guided DNA motor can realize the reliable imaging of tumor cells and provide a new toolbox for disease diagnosis.Yingying Chen Mingzhu Yan Yushi Wang Jinhua Shang Shizhen He Yuhui Gao Chen Hong Xiaoqing Liu Fuan Wang 2023CCS Chemistry2023,5,7:0

20Suppression of Co(Ⅱ)ion deposition and hazards:Regulation of SEI film composition and structure显示文摘Despite the presence of Li F components in the solid electrolyte interphase(SEI)formed on the graphite anode surface by conventional electrolyte,these Li F components primarily exist in an amorphous state,rendering them incapable of effectively inhibiting the exchange reaction between lithium ions and transition metal ions in the electrolyte.Consequently,nearly all lithium ions within the SEI film are replaced by transition metal ions,resulting in an increase in interphacial impedance and a decrease in stability.Herein,we demonstrate that the SEI film,constructed by fluoroethylene carbonate(FEC)additive rich in crystalline Li F,effectively inhibits the undesired Li^(+)/Co^(2+)ion exchange reaction,thereby suppressing the deposition of cobalt compounds and metallic cobalt.Furthermore,the deposited cobalt compounds exhibit enhanced structural stability and reduced catalytic activity with minimal impact on the interphacial stability of the graphite anode.Our findings reveal the crucial influence of SEI film composition and structure on the deposition and hazards associated with transition metal ions,providing valuable guidance for designing next-generation electrolytes.Jiaqi Zhan Mingzhu Liu Yutian Xie Jiarong He Hebing Zhou Lidan Xing Weishan Li 2024Journal of Energy Chemistry2024,89,2:0
