2Status and development of high-power laser facilities at the NLHPLP显示文摘In this paper, we review the status of the multifunctional experimental platform at the National Laboratory of High Power Laser and Physics(NLHPLP). The platform, including the SG-II laser facility, SG-II 9th beam, SG-II upgrade(SG-II UP) facility, and SG-II 5 PW facility, is operational and available for interested scientists studying inertial confinement fusion(ICF) and a broad range of high-energy-density physics. These facilities can provide important experimental capabilities by combining different pulse widths of nanosecond, picosecond, and femtosecond scales. In addition, the SG-II UP facility, consisting of a single petawatt system and an eight-beam nanosecond system, is introduced including several laser technologies that have been developed to ensure the performance of the facility. 4Nano-sized Fe_2O_3/Fe_3O_4 facilitate anaerobic transformation of hexavalent chromium in soil–water systems显示文摘The purpose of this study is to investigate the effects of nano-sized or submicro Fe_2O_3/Fe_3O_4 on the bioreduction of hexavalent chromium(Cr(VI)) and to evaluate the effects of nano-sized Fe_2O_3/Fe_3O_4 on the microbial communities from the anaerobic flooding soil.The results indicated that the net decreases upon Cr(VI) concentration from biotic soil samples amended with nano-sized Fe_2O_3(317.1 ± 2.1 mg/L) and Fe_3O_4(324.0 ± 22.2 mg/L) within21 days,which were approximately 2-fold of Cr(VI) concentration released from blank control assays(117.1 ± 5.6 mg/L).Furthermore,the results of denaturing gradient gel electrophoresis(DGGE) and high-throughput sequencing indicated a greater variety of microbes within the microbial community in amendments with nano-sized Fe_2O_3/Fe_3O_4 than the control assays.Especially,Proteobacteria occupied a predominant status on the phylum level within the indigenous microbial communities from chromium-contaminated soils.Besides,some partial decrease of soluble Cr(VI) in abiotic nano-sized Fe_2O_3/Fe_3O_4 amendments was responsible for the adsorption of nano-sized Fe_2O_3/Fe_3O_4 to soluble Cr(VI).Hence,the presence of nano-sized Fe_2O_3/Fe_3O_4 could largely facilitate the mobilization and biotransformation of Cr(VI) from flooding soils by adsorption and bio-mediated processes.Yaxian Zhang Hua Li Libo Gong Guowen Dong Liang Shen Yuanpeng Wang Qingbiao Li 2017Journal of Environmental Sciences2017,29,7:4
5Roles of low?density lipoproteinreceptor?related protein 1 in tumors显示文摘Low-density lipoprotein receptor-related protein 1(LRP1,also known as CD91),a multifunctional endocytic and cell signaling receptor,is widely expressed on the surface of multiple cell types such as hepatocytes,fibroblasts,neurons,astrocytes,macrophages,smooth muscle cells,and malignant cells.Emerging in vitro and in vivo evidence demonstrates that LRP1 is critically involved in many processes that drive tumorigenesis and tumor progression.For example,LRP1 not only promotes tumor cell migration and invasion by regulating matrix metalloproteinase(MMP)-2and MMP-9 expression and functions but also inhibits cell apoptosis by regulating the insulin receptor,the serine/threonine protein kinase signaling pathway,and the expression of Caspase-3.LRPI-mediated phosphorylation of the extracellular signal-regulated kinase pathway and c-jun N-terminal kinase are also involved in tumor cell proliferation and invasion.In addition,LRP1 has been shown to be down-regulated by microRNA-205 and methylation of LRP1CpG islands.Furthermore,a novel fusion gene,LRP1-SNRNP25,promotes osteosarcoma cell invasion and migration.Only by understanding the mechanisms of these effects can we develop novel diagnostic and therapeutic strategies for cancers mediated by LRP1.Peipei Xing Zhichao Liao Zhiwu Ren Jun Zhao Fengju Song Guowen Wang Kexin Chen Jilong Yang 2016Chinese Journal of Cancer2016,35,1:4
6Total body irradiation of donors can alter the course of tolerance and induce acute rejection in a spontaneous tolerance rat liver transplantation model显示文摘Liver transplantation is an established therapy for end-stage liver diseases. Graft rejection occurs unless the recipient receives immunosuppression after transplantation. This study aimed to explore the mechanism of acute rejection of liver allografts in rats pre-treated with total body irradiation to eliminate passenger lymphocytes and to define the role of CD4 + CD25 + regulatory T cells in the induction of immunotolerance in the recipient. Male Lewis rats were used as donors and male DA rats were recipients. Rats were randomly assigned to the following four groups: control group, homogeneity liver transplantation group, idio-immunotolerance group and acute rejection group. After transplantation, the survival time of each group, serum alanine aminotransferase, total bilirubin levels, number of Foxp3 + CD4 + CD25 + regulatory T cells, expression of glucocorticoid-induced tumor necrosis factor receptor on T cell subgroups, histopathology of the hepatic graft and spleen cytotoxic T lymphocyte lytic activity were measured. In the acute rejection group, where donors were preconditioned with total body irradiation before liver transplantation, all recipients died between day 17 and day 21. On day 14, serum alanine aminotransferase increased signifi- cantly to (459.2±76.9) U L -1 , total bilirubin increased to (124.1±33.7) μmol L -1 (P<0.05) and the ratio of Foxp3 + CD4 + CD25 + regulatory T cells decreased significantly to 1.50%±0.50% (P<0.05) compared with the other groups. Analysis of the T cell subpopulations in the acute rejection group varied from the other groups. Histological analysis showed typical changes of acute rejection in the acute rejection group only. 8Plasmon-tunable Au@Ag core-shell spiky nanoparticles for surfaceenhanced Raman scattering显示文摘A facile synthetic approach has been developed to prepare uniform and size-tunable spiky Au@Ag core-shell nan oparticles (NPs) to tailor the localized surface plasm on res onance (LSPR) properties. The gradual assembly of small Au nano crystals allows the size of spiky Au NPs to be modulated from tens to several hundreds of nano meters by tuning the concentration of initial Au seeds and Au source;and the thick ness of the Ag shell can be adjusted with stepwise reduction of Ag(l)ions. The LSPR bands of such spiky Au@Ag core-shell NPs resemble those of pure spiky Au NP cores of similar sizes in near-infrared region, and increasing the Ag shell thickness results in a blue shift and broadening of the LSPR band in the n ear-i nfrared regi on. Additi on ally, the spiky Au@Ag core-shell NPs exhibit improved surface-e nhan ced Rama n scattering (SERS) activity as compared to the bare spiky Au NPs and spherical Ag@Au NPs. 9Investigation of the Interfacial Transition Zone between Aggregate-Cement Paste by AC Impedance Spectroscopy显示文摘Three different types and sizes of coarse aggregate were chosen,and the alternating current(AC) impedance of cement paste samples with and without aggregate was measured at different curing ages.Based on Song's equivalent circuit model,the electrical properties from the AC impedance results were obtained,and the resistance of connected pores RCCP was used to characterize the microstructure of the interfacial transition zone(ITZ).The results show that the RCCP of concrete sample with aggregate is lower than that of cement paste sample,which indicates that the introduction of aggregate in cement paste makes the ITZ porous.Furthermore,for the same type of aggregate,an increase in particle size leads to a more porous ITZ,which accounts for the 'water effect' and a larger aggregate would accumulate a thicker water film around it.In addition,for the same size of aggregate,the physical interaction between aggregate and cement paste is dominant in early age,and the microstructure of the ITZ around limestone aggregate is denser,which mainly depends on its rough surface and high water absorption.However,the microstructures of the ITZ around granite and basalt aggregates are denser in later age,which may be due to their higher chemical activity,and the chemical interaction between them and cement paste resulting in the generation of more hydrates.AC impedance spectroscopy thus proves to be powerful for evaluation of the microstructure of the ITZ.孔丽娟 hou lirong wang yuhua sun guowen 2016Journal of Wuhan University of Technology(Materials Science)2016,31,4:3
10Numerical calculation and influencing factors of the volume fraction of interfacial transition zone in concrete显示文摘The determination of volume fraction of interfacial transition zone(ITZ) is very important for investigating the quantitative relationship between the microstructure and macroscopical property of concrete.In this paper,based on Lu and Torquato's most nearest surface distribution function,a calculating process of volume fraction of ITZ is given in detail according to the actual sieve curve in concrete.Then,quantitative formulas are put forward to measure the influencing factors on the ITZ volume fraction.In order to validate the given model,the volume fractions of ITZ obtained by numerical calculation are compared with those by computer simulation.The results show that the two are in good agreement.The order of the factors influencing the ITZ volume fraction is the ITZ thickness,the volume fraction of aggregate and the maximum aggregate diameter for Fuller gradation in turn.The ITZ volume fraction obtained from the equal volume fraction(EVF) gradation is always larger than that from the Fuller gradation for a given volume fraction of aggregate.SUN GuoWen SUN Wei ZHANG YunSheng LIU ZhiYong 2012Science China(Technological Sciences)2012,55,6:3
11Effect of Friction Reclaimed Materials of Waste Brake-shoe on Basic Performance of Mortar显示文摘Discarded train brake shoes mainly consist of steel-backed friction material. To be better reutilized, its essential features and its interaction in cement-based material need to be studied. Consequently, particle size analysis, SEM, IR and TGA were used to investigate two types of waste brake shoes, i e, mechanical grinding friction reclaimed material of waste brake-shoe(G-FRMWBS) and pyrolysis-friction reclaimed materials of waste brake-shoe(P-FRMWBS). The latter exhibited less organic content, larger range of particle size distribution and smaller medium particle diameter. Both types contained inorganic particles of spherical and irregular shapes, striped with steel fiber. Upon isometric substituting fine aggregates, G-FRMWBS lifted the strength of mortar effectively that was increased by 16.6% and 17.5% when the replacing rate was 5%; the value went up to 19.2% and 19.2% when the replacing rate was 10%. 12Experimental study of separation of ammonia synthesis vent gas by hydrate formation显示文摘Termodynamic data on methane hydrate formation in the presence of ammonia are very important for upgrading of ammonia synthesis vent gas using hydrate formation. This paper is focused on the formation conditions of methane hydrate in the presence of ammonia and the effects of gas-liquid ratio and temperature on the separation of vent gas by hydrate formation. Equilibrium data for methane hydrate within an ammonia mole concentration range from 1% to 5 % were obtained. The experimental results indicated that ammonia has an inhibitive effect on hydrate formation. The higher the ammonia concentration, the higher is the pressure reguired for methane hydrate formation would be. 13Gene expression dynamics during the gonocyte to spermatogonia transition and spermatogenesis in the domestic yak显示文摘Background:Spermatogenesis is a cellular differentiation process that includes three major events:mitosis of spermatogonia,meiosis of spermatocytes and spermiogenesis.Steady-state spermatogenesis relies on functions of spermatogonial stem cells(SSCs).Establishing and maintaining a foundational SSC pool is essential for continued spermatogenesis in mammals.Currently,our knowledge about SSC and spermatogenesis is severely limited in domestic animals.Results:In the present study,we examined transcriptomes of testes from domestic yaks at four different stages(3,5,8 and 24 months of age)and attempted to identify genes that are associated with key developmental events of spermatogenesis.Histological analyses showed that the most advanced germ cells within seminiferous tubules of testes from 3,5,8 and 24 months old yaks were gonocytes,spermatogonia,spermatocytes and elongated spermatids,respectively.RNA-sequencing(RNA-seq)analyses revealed that 11904,4381 and 2459 genes were differentially expressed during the gonocyte to spermatogonia transition,the mitosis to meiosis transition and the meiosis to post-meiosis transition.Further analyses identified a list of candidate genes than may regulate these important cellular processes.CXCR4,a previously identified SSC niche factor in mouse,was one of the up-regulated genes in the 5 months old yak testis.Results of immunohistochemical staining confirmed that CXCR4 was exclusively expressed in gonocytes and a subpopulation of spermatogonia in the yak testis.Conclusions:Together,these findings demonstrated histological changes of postnatal testis development in the domestic yak.During development of spermatogonial lineage,meiotic and haploid germ cells are supported by dynamic transcriptional regulation of gene expression.Our transcriptomic analyses provided a list of candidate genes that potentially play crucial roles in directing the establishment of SSC and spermatogenesis in yak.Guowen Wang Yongchang Li Qilin Yang Shangrong Xu Shike Ma Rongge Yan Ruina Zhang Gongxue Jia Deqiang Ai Qi’en Yang 2019Journal of Animal Science and Biotechnology2019,10,4:3
14Hexagonally arranged arrays of urchin-like Ag hemispheres decorated with Ag nanoparticles for surface-enhanced Raman scattering substrates显示文摘The surface topography of noble metal particles is a significant factor in tailoring surface-enhanced Raman scattering (SERS) properties. Here, we present a simple fabrication route to hexagonally arranged arrays of surface-roughened urchin- like Ag hemispheres (Ag-HSs) decorated with Ag nanoparticles (Ag-NPs) for highly active and reproducible SERS substrates. The urchin-like Ag-HS arrays are achieved by sputtering Ag onto the top surface of a highly ordered porous anodic aluminum oxide (AAO) template to form ordered arrays of smooth Ag-HSs and then by electrodepositing Ag-NPs onto the surface of each Ag-HS. Owing to the ordered arrangement of the Ag-HSs and the improved surface roughness, the urchin-like hierarchical Ag-HS arrays can provide sufficient and uniform "hot spots" for reproducible and highly active SERS effects. 17Nuciferine protects against high-fat diet-induced hepatic steatosis and insulin resistance via activating TFEB-mediated autophagy——lysosomal pathway显示文摘Nonalcoholic fatty liver disease(NAFLD) is characterized by hepatic steatosis and insulin resistance and there are currently no approved drugs for its treatment.Hyperactivation of mTOR complex1(mTORCl) and subsequent impairment of the transcription factor EB(TFEB)-mediated autophagy-lysosomal pathway(ALP) are implicated in the development of NAFLD.Accordingly,agents that augment hepatic TFEB transcriptional activity may have therapeutic potential against NAFLD.The objective of this study was to investigate the effects of nuciferine,a major active component from lotus leaf,on NAFLD and its underlying mechanism of action.Here we show that nuciferine activated ALP and alleviated steatosis,insulin resistance in the livers of NAFLD mice and palmitic acid-challenged hepatocytes in a TFEB-dependent manner.Mechanistic investigation revealed that nuciferine interacts with the Ragulator subunit hepatitis B X-interacting protein and impairs the interaction of the Ragulator complex with Rag GTPases,thereby suppressing lysosomal localization and activity of mTORC1,which activates TFEB-mediated ALP and further ameliorates hepatic steatosis and insulin resistance.Our present results indicate that nuciferine may be a potential agent for treating NAFLD and that regulation of the mTORCl-TFEB-ALP axis could represent a novel pharmacological strategy to combat NAFLD.Xiliang Du Chiara Di Malta Zhiyuan Fang Taiyu Shen Xiaodi Niu Meng Chen Bo Jin Hao Yu Lin Lei Wenwen Gao Yuxiang Song Zhe Wang Chuang Xu Zhijun Cao Guowen Liu Xinwei Li 2022Acta Pharmaceutica Sinica B2022,12,6:2
