4Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29
5FINITE ELEMENT MODELLING OF COMPLEX 3D STATIC AND DYNAMIC CRACK PROPAGATION BY EMBEDDING COHESIVE ELEMENTS IN ABAQUS显示文摘This study proposes an algorithm of embedding cohesive elements in Abaqus and develops the computer code to model 3D complex crack propagation in quasi-brittle materials in a relatively easy and efficient manner.The cohesive elements with softening traction-separation relations and damage initiation and evolution laws are embedded between solid elements in regions of interest in the initial mesh to model potential cracks.The initial mesh can consist of tetrahedrons,wedges,bricks or a mixture of these elements.Neither remeshing nor objective crack propagation criteria are needed.Four examples of concrete specimens,including a wedge-splitting test,a notched beam under torsion,a pull-out test of an anchored cylinder and a notched beam under impact,were modelled and analysed.The simulated crack propagation processes and load-displacement curves agreed well with test results or other numerical simulations for all the examples using initial meshes with reasonable densities.Making use of Abaqus's rich pre/post-processing functionalities and powerful standard/explicit solvers,the developed method offers a practical tool for engineering analysts to model complex 3D fracture problems.Xiangting Su Zhenjun Yang Guohua Liu 2010Acta Mechanica Solida Sinica2010,23,3:23
6Movement and strain conditions of active blocks in the Chinese mainland显示文摘The definition of active block is given from the angles of crustal deformation and strain. The movement and strain parameters of active blocks are estimated according to the unified velocity field composed of the velocities at 1598 GPS stations obtained from GPS measurements carried out in the past years in the Chinese mainland and the surrounding areas. The movement and strain conditions of the blocks are analyzed. The active blocks in the Chinese mainland have a consistent E-trending movement component, but its N and S components are not consistent. The blocks in the western part have a consistent N-trending movement and the blocks in the eastern part have a consistent S-trending movement. In the area to the east of 90°E, that is the area from Himalayas block towards NE, the movement direction of the blocks rotates clockwisely and the movement rates of the blocks are different. Generally, the movement rate is large in the west and south and small in the east and north with a difference of 3 to 4 times between the rates in the west and east. The distributions of principal compressive strain directions of the blocks are also different. The principal strain of the blocks located to the west of 90oE is basically in the SN direction, the principal compressive strain of the blocks in the northeastern part of Qingzang plateau is roughly in the NE direction and the direction of principal compressive strain of the blocks in the southeastern part of Qingzang plateau rounds clockwisely the east end of Himalayas structure. In addition, the principal strain and shear strain rates of the blocks are also different. The Himalayas and Tianshan blocks have the largest principal compressive strain and the maximum shear strain rate. Then, Lhasa, Qiangtang, Southwest Yunnan (SW Yunnan), Qilian and Sichuan-Yunan (Chuan-Dian) blocks followed. The strain rate of the blocks in the eastern part is smaller. The estimation based on the stain condition indicates that Himalayas block is still the area with the most intensive tectonic activity and it shortens in the NS direction at the rate of 15.2±1.5 mm/a. Tianshan block ranks the second and it shortens in the NS direction at the rate of 10.1±0.9 mm/a. At present, the two blocks are still uprising. It can be seen from superficial strain that the Chinese mainland is predominated by superficial expansion. Almost the total area in the eastern part of the Chinese mainland is expanded, while in the western part, the superficial compression and expansion are alternatively distributed from the south to the north. In the Chinese mainland, most EW-trending or proximate EW-trending faults have the left-lateral or left-lateral strike-slip relative movements along both sides, and most NS-trending faults have the right-lateral or right-lateral strike-slip relative movements along both sides. According to the data from GPS measurements the left-lateral strike-slip rate is 4.8±1.3 mm/a in the central part of Altun fault and 9.8±2.2 mm/a on Xianshuihe fault. The movement of the fault along the block boundary has provided the condition for block movement, so the movements of the block and its boundary are consistent, but the movement levels of the blocks are different. The statistic results indicate that the relative movement between most blocks is quite significant, which proves that active blocks exist. Himalayas, Tianshan, Qiangtang and SW Yunnan blocks have the most intensive movement; China-Mongolia, China-Korea (China-Korea), Alxa and South China blocks are rather stable. The mutual action of India, Pacific and Philippine Sea plates versus Eurasia plate is the principal driving force to the block movement in the Chinese mainland. Under the NNE-trending intensive press from India plate, the crustal matter of Qingzang plateau moves to the NNE and NE directions, then is hindered by the blocks located in the northern, northeastern and eastern parts. 7Myocardial infarction secondary prevention study (MISPS)显示文摘Background Traditional Chinese medicine (TCM), especially herbal medicine, has been widely used in China and now is also being increasingly used in other countries for the treatment of cardiovascular diseases. Although many studies have demonstrated that certain Chinese herbal products are effective and safe for the treatment of cardiovascular diseases, most of these lack sufficient quality. Therefore, large randomized clinical trials and further scientific research to determine its safety, effectiveness are necessary.QiShen YiQi Dripping Pills (QSYQDP) is a herbal preparation clinically used in the treatment and prevention of coronary artery disease. Preliminary observations have shown its safety and effectiveness. Methods/Design This randomized, controlled trial will recruit 3600 patients with a history of myocardial infarction. Patients will be randomized into two groups by a Centr-Randomized System. One group receives QSYQDP, the other group receive aspirin. This trial protocol will describe eligibility criteria, detailed information on the treatment definition, blinding, endpoints, statistical methods, sample size determination, data management, legal aspects, and the current status of the trial. Discussion This trial is one of the first randomized, controlled clinical trial to evaluate the efficacy and safety of traditional Chinese herbal medicine in the treatment and secondary prevention of coronary artery disease. 8Chinese herbal medicine reduces mortality in patients with severe and critical coronavirus disease 2019: a retrospective cohort study显示文摘This study aimed to evaluate the efficacy of Chinese herbal medicine(CHM)in patients with severe/critical coronavirus disease 2019(COVID-19).In this retrospective study,data were collected from 662 patients with severe/critical COVID-19 who were admitted to a designated hospital to treat patients with severe COVID-19 in Wuhan before March 20,2020.All patients were divided into an exposed group(CHM users)and a control group(non-users).After propensity score matching in a 1:1 ratio,156 CHM users were matched by propensity score to 156 non-users.No significant differences in seven baseline clinical variables were found between the two groups of patients.All-cause mortality was reported in 13 CHM users who died and 36 non-users who died.After multivariate adjustment,the mortality risk of CHM users was reduced by 82.2%(odds ratio 0.178,95%CI 0.076–0.418;P<0.001)compared with the non-users.Secondly,age(odds ratio 1.053,95%CI 1.023–1.084;P<0.001)and the proportion of severe/critical patients(odds ratio 0.063,95%CI 0.028–0.143;P<0.001)were the risk factors of mortality.These results show that the use of CHM may reduce the mortality of patients with severe/critical COVID-19.Guohua Chen Wen Su Jiayao Yang Dan Luo Ping Xia Wen Jia Xiuyang Li Chuan Wang Suping Lang Qingbin Meng Ying Zhang Yuhe Ke An Fan Shuo Yang Yujiao Zheng Xuepeng Fan Jie Qiao Fengmei Lian Li Wei Xiaolin Tong 2020Frontiers of Medicine2020,14,6:10
9Recovery of rare earths from ion-absorbed rare earths ore with MgSO_4-ascorbic acid compound leaching agent显示文摘The magnesium sulfate leaching technology for the ion-absorbed rare earths ore can solve the ammonia pollution problem existing in ammonium sulfate leaching process. However, the leaching capacity of magnesium sulfate is slightly weaker than that of ammonium sulfate, resulting in a bigger consumption of magnesium sulfate. In this paper, the MgSO_4-ascorbic acid compound leaching agent had been demonstrated to deal with the ion-absorbed rare earths ore. The ascorbic acid could form a stable coordination with rare earth ions, so that it can strengthen the leaching of ion-exchangeable phase.Moreover, ascorbic acid has a strong reductive property, it can leach the colloidal sediment phase rare earth as well. The present study investigates the effect of the initial pH and the composition of leaching agent on the rare earth leaching. It is determined that the rare earth leaching efficiency is 107.5% under the condition of pH 2.00,0.15 mol/L magnesium sulfate and 1.0 g/L ascorbic acid in leaching agent. In this case, the content of the ion-exchangeable phase and colloidal sediment phase rare earth in the leaching residues are both only 0.02‰. The leaching efficiency of colloid sediment phase rare earth can be 85.7%,so that the Ce partition in the leaching liquor increases to be 5.77%. 10Whole genome sequencing of 10K patients with acute ischaemic stroke or transient ischaemic attack: design, methods and baseline patient characteristics显示文摘Background and purpose Stroke is the second leading cause of death worldwide and the leading cause of mortality and long-term disability in China,but its underlying risk genes and pathways are far from being comprehensively understood.We here describe the design and methods of whole genome sequencing(WGS)for 10914 patients with acute ischaemic stroke or transient ischaemic attack from the Third China National Stroke Registry(CNSR-III).Methods Baseline clinical characteristics of the included patients in this study were reported.DNA was extracted from white blood cells of participants.Libraries are constructed using qualified DNA,and WGS is conducted on BGISEQ-500 platform.The average depth is intended to be greater than 30×for each subject.Afterwards,Sentieon software is applied to process the sequencing data under the Genome Analysis Toolkit best practice guidance to call genotypes of single nucleotide variants(SNVs)and insertion-deletions.For each included subject,21 fingerprint SNVs are genotyped by MassARRAY assays to verify that DNA sample and sequencing data originate from the same individual.The copy number variations and structural variations are also called for each patient.All of the genetic variants are annotated and predicted by bioinformatics software or by reviewing public databases.Results The average age of the included 10914 patients was 62.2±11.3 years,and 31.4%patients were women.Most of the baseline clinical characteristics of the 10914 and the excluded patients were balanced.Conclusions The WGS data together with abundant clinical and imaging data of CNSR-III could provide opportunity to elucidate the molecular mechanisms and discover novel therapeutic targets for stroke.Si Cheng Zhe Xu Yang Liu Jinxi Lin Yong Jiang Yilong Wang Xia Meng Anxin Wang Xinying Huang Zhimin Wang Guohua Chen Songdi Wu Zhengchang Jia Yongming Chen Xuerong Qiu Jun Wu Binbin Song Weizhong Ji Zhongping An Wenjun Xue Lili Zhao Yu Geng Hongyan Li Hao Li Yongjun Wang 2021Stroke & Vascular Neurology2021,6,2:8
11Microstructure evolution of semi-solid Mg-10Gd-3Y-0.5Zr alloy during isothermal heat treatment显示文摘In this study,the microstructure evolution of semi-solid Mg-10Gd-3Y-0.5Zr alloy during isothermal heat treatment has been investigated.The results show that primary particles coarsen continuously during the holding.Coarsening rate decreases with the increase of isothermal temperature.When isothermal temperature increases from 600℃ to 620℃,the dominant mechanism for coarsening changes from particle coalescence to Ostwald ripening.Equiaxed as-cast microstructure is beneficial to the semi-solid microstructure after isothermal heat treatment,which brings about the refinement and spheroidization of primary particles,and shortening of holding time.Significant modification of second phases can also be achieved after isothermal heat treatment,due to its unique solidification process.The optimum processing parameters for Mg-10Gd-3Y-0.5Zr alloy in isothermal heat treatment are isothermal temperature of 610℃-620℃ and holding time of 20-40 min.Guohua Wu Yang Zhang Wencai Liu Wenjiang Ding 2013Journal of Magnesium and Alloys2013,1,1:8
12Effects of dietary leucine supplementation on the gene expression of mammalian target of rapamycin signaling pathway and intestinal development of broilers显示文摘This experiment was to investigate the effects of dietary leucine supplementation on the gene expression of mammalian target of rapamycin(mTOR) signaling pathway and intestinal development of broilers. A total of 384 one-day-old broilers were randomly assigned into 4 treatments with 6 replicates(16 broilers per replicate). Broilers in these treatment groups were offered the following diets with 1.37,1.77,2.17 and2.57% of leucine. These diet treatments were named 1.37 TM, 1.77 TM, 2.17 TM, and 2.57 TM. The experiment lasted 21 days and all birds had free access to feed and water. Results indicated that there was no significant difference in body weight, average daily gain and average feed intake among all treatments(P > 0.05). The broiler duodenal villus height in 2.57 TM was the lowest, but the highest occurred in1.37 TM on d 7 and 14(P < 0.05). The villus height in the jejunum and ileum increased along with leucine level from 1.37 to 2.17%. The villus height of jejunum was significantly higher in 2.17 TM than in 1.37 TM on d 7 and 14, and the ratio of villus height to crypt depth(V:C) in the duodenum, jejunum and ileum increased significantly(P < 0.05) on d 21. The gene expression level of mTOR in the duodenum decreased with increasing leucine level and was higher in 1.37 TM than in 2.57 TM on d 7 and 14(P < 0.05). On d 14 and 21 of the trial, the expression of S6 K1 in the duodenum was higher in 1.37 TM than in 2.57 TM(P < 0.05), and the expression of mTOR, S6 K1 in the jejunum and ileum increased with increasing leucine level form 1.37 to 2.17%, whereas a significant difference occurred between 1.37 TM and 2.17 TM(P< 0.05).In conclusion, the addition of leucine fails to enhance the growth performance of broilers. However,leucine can improve intestinal development by enhancing villus height and V:C ratio in the jejunum and ileum. 13An adaptive scaled boundary finite element method by subdividing subdomains for elastodynamic problems显示文摘The scaled boundary finite element method(SBFEM) is a semi-analytical numerical method,which models an analysis domain by a small number of large-sized subdomains and discretises subdomain boundaries only.In a subdomain,all fields of state variables including displacement,stress,velocity and acceleration are semi-analytical,and the kinetic energy,strain energy and energy error are all integrated semi-analytically.These advantages are taken in this study to develop a posteriori h-hierarchical adaptive SBFEM for transient elastodynamic problems using a mesh refinement procedure which subdivides subdomains.Because only a small number of subdomains are subdivided,mesh refinement is very simple and efficient,and mesh mapping to transfer state variables from an old mesh to a new one is also very simple but accurate.Two 2D examples with stress wave propagation were modelled.The results show that the developed method is capable of capturing propagation of steep stress regions and calculating accurate dynamic responses,using only a fraction of degrees of freedom required by adaptive finite element method.ZHANG ZiHua 1,2,YANG ZhenJun 2,LIU GuoHua 1 & HU YunJin 1 1 College of Civil Engineering and Architecture,Zhejiang University,Hangzhou 310058,China 2 School of Engineering,University of Liverpool,L69 3GQ,UK 2011Science China(Technological Sciences)2011,54,S1:4
14Ionic liquids containing the urea moiety for recognition of halides and complex anions显示文摘A series of novel imidazolium based ionic liquids containing the urea moiety were designed and synthesized for anion recognition. 1-Ethylurea-3-methylimidazolium acetate ([Eumim]OAc) was used as the receptor for the halides and complex anions (BF4-, PF6-, BPh4-). 1H NMR spectra showed that the urea protons and imidazolium C(2) proton of the receptor ([Eumim]OAc) moved upfield on addition of various anions. A Job plot showed that the [Eumim]OAc receptor formed a 1:1 complex with BPh4-. X-ray diffraction analysis and the molecular modeling study revealed that the conformations of [Eumim]OAc and [Eumim]BPh4 were different. 16Relation between the characteristics of strong earthquake activities in Chinese mainland and the Wenchuan earthquake显示文摘This paper studies the relations between the great Wenchuan earthquake and the active-quiet periodic characteristics of strong earthquakes,the rhythmic feature of great earthquakes,and the grouped spatial distribution of MS8.0 earthquakes in Chinese mainland.We also studied the relation between the Wenchuan earthquake and the stepwise migration characteristics of MS≥7.0 earthquakes on the North-South seismic belt,the features of the energy releasing acceleration in the active crustal blocks related to the Wenchuan earthquake and the relation between the Wenchuan earthquake and the so called secondarc fault zone.The results can be summarized as follows:1 the occurrence of the Wenchuan earthquake was consistent with the active-quiet periodic characteristics of strong earthquakes;2 its occurrence is consistent with the features of grouped occurrence of MS8.0 earthquakes and follows the 25 years rhythm(each circulation experiences the same time) of great earthquakes;3 the Wenchuan MS8.0 earthquake follows the well known stepwise migration feature of strong earthquakes on the North-South seismic belt;4 the location where the Wenchuan MS8.0 earthquake took place has an obvious consistency with the temporal and spatial characteristic of grouped activity of MS≥7.0 strong earthquakes on the secondarc fault zone;5 the second-arc fault zone is not only the lower boundary for earthquakes with more than 30 km focal depth,but also looks like a lower boundary for deep substance movement;and 6 there are obvious seismic accelerations nearby the Qaidam and Qiangtang active crustal blocks(the northern and southern neighbors of the Bayan Har active block,respectively),which agrees with the GPS observation data.Xiaodong Zhang Guohua Yang Xian Lu Mingxiao Li Zhigao Yang 2009Earthquake Science2009,22,5:3
17Enhancing phosphorus uptake efficiency through QTL-based selection for root system architecture in maize显示文摘Root system architecture(RSA) plays an important role in phosphorus(P) acquisition,but enhancing P use efficiency(PUE) in maize via genetic manipulation of RSA has not yet been reported.Here,using a maize recombinant inbred line(RIL) population,we investigated the genetic relationships between PUE and RSA,and developed P-efficient lines by selection of quantitative trait loci(QTLs) that coincide for both traits.In low-P(LP) fields,P uptake efficiency(Pup E) was more closely correlated with PUE(r =0.48-0.54),and RSA in hydroponics was significantly related to Pup E(r=0.25-0.30) but not to P utilization efficiency(Put E).QTL analysis detected a chromosome region where two QTLs for PUE,three for Pup E and three for RSA were assigned into two QTL clusters,Cl-bin3.04 a and Cl-bin3.04 b.These QTLs had favorable effects from alleles derived from the large-rooted and high-Pup E parent.Marker-assisted selection(MAS) identified nine advanced backcross-derived lines carrying Cl-bin3.04 a or Cl-bin3.04 b that displayed mean increases of 22%e26% in PUE in LP fields.Furthermore,a line L224 pyramiding Clbin3.04 a and Cl-bin3.04 b showed enhanced Pup E,relying mainly on changes in root morphology,rather than root physiology,under both hydroponic and field conditions.These results highlight the physiological and genetic contributions of RSA to maize Pup E,and provide a successful study case of developing P-efficient crops through QTL-based selection.Riliang Gu Fanjun Chen Lizhi Long Hongguang Cai Zhigang Liu Jiabo Yang Lifeng Wang Huiyong Li Junhui Li Wenxin Liu Guohua Mi Fusuo Zhang Lixing Yuan 2016Journal of Genetics and Genomics2016,43,11:3
18Assessing Adaptability of Planted Trees Using Leaf Traits: A Case Study with Robinia pseudoacacia L. in the Loess Plateau, China显示文摘Leaf trait patterns and their variations with climate are interpreted as an adaptive adjustment to environment.This study assessed the adaptability of planted black locust (Robinia pseudoacacia L.) based on the analysis of leaf traits and the comparison of its leaf traits with inter-specific ones existing in the same area.We measured some water and N use related leaf traits: leaf dry mass per unit area (LMA) and N,P and K concentrations based on both leaf area (Narea,Parea and Karea) and leaf mass (Nmass,Pmass and Kmass) of R.pseudoacacia at 31 sites along a water stress gradient in North Shaanxi Province,China.The results show that leaves of R.pseudoacacia have high Nmass and low LMA in the study area.High Nmass and low LMA are usually representative of luxurious resource use,and will advance plant resource competitiveness in high-resource conditions.As a whole,LMA-nutrient relationships of R.pseudoacacia display patterns that are fairly similar to the inter-specific relationships in both direction and intensity.The tendency for LMA and Narea to increase with decreasing water availability and the positive correlation between LMA and Narea reflect the trend for R.pseudoacacia to enhance water use efficiency (WUE) at the expense of down-regulated photosynthetic N use efficiency (PNUE) and high construction cost in dry conditions.However,the positive relationship between LMA and Narea in high mean annual precipitation (MAP) area is either unremarkable or reversed with decreasing water availability.This implies a lower photosynthetic capacity and a higher construction cost for high-LMA leaves.The inter-specific relationship between LMA and Narea is positive and does not change with water availability.This difference between inter-species and intra-species may be due to more diversified anatomies and more specialised structures for inter-species than intra-species.The failure of R.pseudoacacia adaption to dry conditions reflected by LMA-Narea relationship may be partially responsible for the emergence of rampike and dwarf forms found frequently in dry conditions.Incorporating intrinsic characteristics of planted trees into vegetation restoration project will be instructive and meaningful for species selection.JIN Tiantian LIU Guohua FU Bojie DING Xiaohui YANG Lei 2011Chinese Geographical Science2011,21,3:3
19Polyimide separators for rechargeable batteries显示文摘Separators are indispensable components of modern electrochemical energy storage devices such as lithium-ion batteries(LIBs).They perform the critical function of physically separating the electrodes to prevent short-circuits while permitting the ions to pass through.While conventional separators using polypropylene(PP) and polyethylene(PE) are prone to shrinkage and melting at relatively high temperatures(150℃ or above) causing short circuits and thermal runaway,separators made of thermally stable polyimides(PIs) are electrochemically stable and resistant to high temperatures,and possess good mechanical strength-making them a promising solution to the safety concerns of LIBs.In this review,the research progress on PI separators for use in LIBs is summarized with a special focus on molecular design and microstructural control.In view of the significant progress in advanced chemistries beyond LIBs,recent advances in PI-based membranes for applications in lithium-sulfur,lithium-metal,and solid-state batteries are also reviewed.Finally,practical issues are also discussed along with their prospects.Ziheng Lu Fan Sui Yue-E Miao Guohua Liu Cheng Li Wei Dong Jiang Cui Tianxi Liu Junxiong Wu Chunlei Yang 2021Journal of Energy Chemistry2021,30,7:3
