| 1 | The ex vivo and in vivo biological performances of graphene oxide and the impact of surfactant on graphene oxide's biocompatibility显示文摘Graphene oxide(GO)displays promising properties for biomedical applications including drug delivery and cancer therapeutics.However,GO exposure also raises safety concerns such as potential side effects on health.Here,the biological effects of GO suspended in phosphate buffered saline(PBS)with or without 1% nonionic surfactant Tween 80 were investigated.Based on the ex vivo experiments,Tween 80 significantly affected the interaction between GO and peripheral blood from mice.GO suspension in PBS tended to provoke the aggregation of diluted blood cells,which could be prevented by the addition of Tween 80.After intravenous administration,GO suspension with or without 1% Tween 80 was quickly eliminated by the mononuclear phagocyte system.Nevertheless,GO suspension without Tween 80 showed greater accumulation in lungs than that containing 1% Tween 80.In contrast,less GO was found in livers for GO suspension compared to Tween 80 assisted GO suspension.Organs including hearts,livers,lungs,spleens,kidneys,brains,and testes did not reveal histological alterations.The indexes of peripheral blood showed no change upon GO exposure.Our results together demonstrated that Tween 80 could greatly alter GO's biological performance and determine the pattern of its biodistribution in mice. | Guangbo Qu Xiaoyan Wang Qian Liu Rui Liu Nuoya Yin Juan Ma Liqun Chen Jiuyang He Sijin Liu Guibin Jiang | 2013 | Journal of Environmental Sciences2013,25,5: | 7 |
| 2 | Application of nanoparticle tracking analysis for characterising the fate of engineered nanoparticles in sediment-water systems显示文摘Novel applications of nanotechnology may lead to the release of engineered nanoparticles(ENPs), which result in concerns over their potential environmental hazardous impact. It is essential for the research workers to be able to quantitatively characterise ENPs in the environment and subsequently to assist the risk assessment of the ENPs. This study hence explored the application of nanoparticle tracking system(NTA) to quantitatively describe the behaviour of the ENPs in natural sediment-water systems. The NTA allows the measurement of both particle number concentration(PNC) and particle size distribution(PSD) of the ENPs. The developed NTA method was applied to a range of gold and magnetite ENPs with a selection of surface properties. The results showed that the positively-charged ENPs interacted more strongly with the sediment than neutral and negatively-charged ENPs. It was also found that the citrate coated Au ENPs had a higher distribution percentage(53%) than 11-mercaptoundecanoic acid coated Au ENPs(20%) and citrate coated magnetite ENPs(21%). | 8 | Presence of organophosphate flame retardants(OPEs)in different functional areas in residential homes in Beijing,China显示文摘The production and application of organophosphate esters(OPEs)have dramatically increased in recent years due to their use as a replacement for brominated flame retardants.In this study,13 OPEs(∑_(13)OPEs)were analyzed in indoor air samples from kitchens and living rooms in 14 residential homes in Beijing,China.The concentrations of ∑_(13)OPEs in kitchen air samples(mean:13 ng/m^(3))were significantly(p<0.05)higher than in living room air samples(5.0 ng/m^(3)).In addition,paired window surface organic film samples were collected and analyzed to investigate film-air partitioning,exhibiting a mean concentration of ∑_(13)OPEs of 4100 ng/m^(2).The congener profiles showed that tris(2-chloroisopropyl)phosphate(TCPP)was the predominant compound in both window film samples(48%)and the corresponding indoor air sample(56%).The estimated daily intakes(EDI)of OPEs via indoor air inhalation were 2.8 and 1.4 ng/kg/day for infants and adults,respectively,both of which are below the reference dose values(RfDs).Overall,these findings indicate that OPEs in the indoor air environment of residential homes in Beijing are not likely to pose a health risk to the general population. | Kun Lv Lu Bai Boyu Song Xindong Ma Minmin Hou Jie Fu Yali Shi Yawei Wang Guibin Jiang | 2022 | Journal of Environmental Sciences2022,34,5: | 1 |
| 9 | Effect of intrathymic injection of allogene antigen on immune response to sciatic nerve transplantation in allogenic mice显示文摘BACKGROUND: The latest researches demonstrate that intrathymic injection of MHC antigen which reaches a certain dosage (2 mg, i.e., 4 × 108 cell extraction) can induce immunologic tolerance under non-antilymphocyte serum condition. OBJECTIVE: To investigate the effect of intrathymic injection of allogene antigen on survival and function of sciatic nerve in allogenic mice. DESIGN: Randomized controlled animal study. SETTING: The 4th Affiliated Hosptial of Harbin Medical University. MATERIALS: A total of 32 male donor C57BL/6(H-2b) mice of 4-8 weeks old and weighing 18-22 g and 44 female receptor Balb/c(H-2d) mice of 4-8 weeks old and weighing 18-22 g were selected from Heilongjing Veterinary Institution. The animal experiment had got confirmed consent from local ethic committee. METHODS: The experiment was carried out in the Laboratory (Provincial Key Laboratory) of the Fourth Hospital, Harbin Medical University from June 2006 to May 2007. C57BL/6(H-2b) mice were anesthetized to extract MHC (H-2b) antigen from splenic cells and sciatic nerves. Allogenous nerve transplantation group: Mice were given intrathymic injection of 100 μL saline; two weeks later, frozen sciatic nerves of donor mice were transplanted. Immunosuppressive agent group: Mice were given intrathymic injection of 100 μL saline; two weeks later, fresh sciatic nerves of donor mice were transplanted. At three days before transplantation, 10 mg/kg per day cyclosporin A was intraperitoneally injected once a day till mice were sacrificed. MHC (H-2b) antigen injection group: Mice were given intrathymic injection of MHC (H-2b) antigen from C57BL/6(H-2b) donor mice; two weeks later, fresh sciatic nerves of donor mice were transplanted. Autogenous nerve transplantation group: Mice were given intrathymic injection of 100 μL saline; two weeks later, fresh sciatic nerves were transplanted. MAIN OUTCOME MEASURES: ① Three weeks later, transplanted part of exposured sciatic nerve was used to measure the motor nerve conduction velocity. ② Transplanted nerve was stained with histochemical staining and observed light microscope and electron microscope. ③ Mice received mixed lymphocyte culture and delayed-typed hypersensitiveness to observe absorbency and measure depth of soles. RESULTS: All 76 mice were involved in the final analysis. ① Motor nerve conduction velocity: The nerve recovery in MHC (H-2b) antigen injection group was higher than that in allogenous nerve transplantation group, equal to immunosuppressive agent group and lower than autogenous nerve transplantation group. There were significant differences among them (P < 0.05). ② Histological changes of transplanted nerve: Light and electron microscopes demonstrated that there were a lot of regenerative nerve fibers in autogenous nerve transplantation group, immunosuppressive agent group and MHC (H-2b) antigen injection group, and all nerve fibers passed grafts. ③ Immunological examination: There was no significant difference in mixed lymphocyte culture among allogenous nerve transplantation group, autogenous nerve transplantation group and MHC (H-2b) antigen injection group (P < 0.05). Depth of soles in other groups was deeper than that in the MHC (H-2b) antigen injection group, and there was significant difference (P < 0.05); that was to say, delayed-typed hypersensitiveness was remarkable. | 13 | CRL2^(APPBP2)-mediated TSPYL2 degradation counteracts human mesenchymal stem cell senescence显示文摘Cullin-RING E3 ubiquitin ligases(CRLs),the largest family of multi-subunit E3 ubiquitin ligases in eukaryotic cells,represent core cellular machinery for executing protein degradation and maintaining proteostasis.Here,we asked what roles Cullin proteins play in human mesenchymal stem cell(hMSC)homeostasis and senescence.To this end,we conducted a comparative aging phenotype analysis by individually knocking down Cullin members in three senescence models:replicative senescent hMSCs,Hutchinson-Gilford Progeria Syndrome hMSCs,and Werner syndrome hMSCs.Among all family members,we found that CUL2 deficiency rendered hMSCs the most susceptible to senescence.To investigate CUL2-specific underlying mechanisms,we then applied CRISPR/Cas9-mediated gene editing technology to generate CUL2-deficient human embryonic stem cells(hESCs).When we differentiated these into h MSCs,we found that CUL2 deletion markedly accelerates hMSC senescence.Importantly,we identified that CUL2 targets and promotes ubiquitin proteasome-mediated degradation of TSPYL2(a known negative regulator of proliferation)through the substrate receptor protein APPBP2,which in turn downregulates one of the canonical aging marker-P21^(waf1/cip1),and thereby delays senescence.Our work provides important insights into how CRL2^(APPBP2)-mediated TSPYL2 degradation counteracts hMSC senescence,providing a molecular basis for directing intervention strategies against aging and aging-related diseases. | Daoyuan Huang Qian Zhao Kuan Yang Jinghui Lei Ying Jing Hongyu Li Chen Zhang Shuai Ma Shuhui Sun Yusheng Cai Guibin Wang Jing Qu Weiqi Zhang Si Wang Guang-Hui Liu | 2024 | Science China(Life Sciences)2024,67,3: | 0 |
