1A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 2Toxicity of Graphene Quantum Dots in Zebrafish Embryo显示文摘Objective To evaluate the bio-safety of graphene quantum dots(GQDs), we studied its effects on the embryonic development of zebrafish. Methods In vivo, biodistribution and the developmental toxicity of GQDs were investigated in embryonic zebrafish at exposure concentrations ranging from 12.5-200 μg/m L for 4-96 h post-fertilization(hpf). The mortality, hatch rate, malformation, heart rate, GQDs uptake, spontaneous movement, and larval behavior were examined. Results The fluorescence of GQDs was mainly localized in the intestines and heart. As the exposure concentration increased, the hatch and heart rate decreased, accompanied by an increase in mortality. Exposure to a high level of GQDs(200 μg/mL) resulted in various embryonic malformations including pericardial edema, vitelline cyst, bent spine, and bent tail. The spontaneous movement significantly decreased after exposure to GQDs at concentrations of 50, 100, and 200 μg/mL. The larval behavior testing(visible light test) showed that the total swimming distance and speed decreased dose-dependently. Embryos exposed to 12.5 μg/mL showed hyperactivity while exposure to higher concentrations(25, 50, 100, and 200 μg/mL) caused remarkable hypoactivity in the light-dark test. Conclusion Low concentrations of GQDs were relatively non-toxic. 3Effect of tungsten oxide on ceria nanorods to support copper species as CO oxidation catalysts显示文摘In this work,tungsten oxide with different concentrations(0,0.4 at%,2.0 at%and 3.2 at%)was introduced to the ceria nanorods via a deposition-precipitation(DP)approach,and copper species of ca.10 at%were sequentially anchored onto the modified ceria support by a similar DP route.The aim of the study was to investigate the effect of the amount of tungsten oxide(0,0.4 at%,2.0 at%,and 3.2 at%)modifier on the copper-ceria catalysts for CO oxidation reaction and shed light on the structure-activity relationship.By the aids of multiple characterization techniques including N2 adsorption,high-resolution transmission electron microscopy(HRTEM),powder X-ray diffraction(XRD),X-ray absorption fine structure(XAFS),and temperature-programmed reduction by hydrogen(H2-TPR)in combination with the catalytic performance for CO oxidation reaction,it is found that the copper-ceria samples maintain the crystal structure of the fluorite fcc CeO2 phase with the same nanorod-like morphology with the introduction of tungsten oxide,while the textural properties(the surface area,pore volume and pore size)of ceria support and copper-ceria catalysts are changed,and the oxidation states of copper and tungsten are kept the same as Cu2+and W6+before and after the reaction,but the introduction of tungsten oxide(WO3)significantly changes the metal-support interaction(transfer the CuOx clusters to Cu-[Ox]-Ce species),which delivers to impair the catalytic activity of copper-ceria catalysts for CO oxidation reaction.Yu'nan Li Lin Gan Rui Si 2021Journal of Rare Earths2021,39,1:5

6Experimental Study on Analgesia and Hemostasis of Paris polyphylla,Scutellaria baicalensis and Their Compatibility显示文摘[Objectives]The analgesic and hemostatic effects of Paris polyphylla,Scutellaria baicalensis,and their compatibility were studied.[Methods](i)In the experiment on the writhing of mice induced by acetic acid,60 Kunming SPF mice were randomly selected,and were randomly divided into 6 groups according to the male-to-female ratio of 1∶1,including blank control group,P.polyphylla group,S.baicalensis group,and three P.polyphylla and S.baicalensis compatibility groups(with the ratio of 1∶2,1∶1 and 1∶2).There were 10 mice in each group.In the experimental groups,the mice were given 1 g/mL Chinese medicine extract according to a dose of 20 mL/kg.In the control group,the mice were given 0.9%normal saline in equal volumes.Gavage was performed one time every 24 h and lasted for 14 d.1 h after the gavage on the 14th day,they were given 0.5%glacial acetic acid solution via intraperitoneal injection.(ii)In the hot plate experiment,60 Kunming SPF mice were selected,and the ratio of male to female,grouping,administration and gavage were the same as those of the glacial acetic acid-induced writhing experiment.Gavage was lasted for 14 d.1 h after the gavage on the 14th day,the mice were placed on a hot plate apparatus at(55±0.5)℃,and the time of licking hindfoot was measured.(iii)The hemostatic effect was explored through three experimental methods of tail hemostasis,femoral artery hemostasis and liver hemostasis.60 Kunming SPF mice were taken in each of the three experiments,and the male-female ratio and grouping were the same as above.The powder was covered on the surface of the wound sites,and the bleeding was observed.The hemostatic time was recorded,and the hemostatic time was recorded as 3 min if it exceeded 3 min.[Results](i)In the acetic acid-induced writhing experiment,compared with the blank control group,the pain of mice in the experimental groups was inhibited.Among them,P.polyphylla group,S.baicalensis group,compatibility 1∶2 group,compatibility 1∶1 group,and compatibility 2∶1 group had significant effects(P<0.05),and the inhibition rate of writhing was 20.43%,28.32%,52.30%,32.79%,and 39.02%,respectively.The analgesic effect of the compatibility 1∶2 group was the most obvious.(ii)In the hot plate experiment,compared with the blank control group,the experimental groups had analgesic effect.Among them,P.polyphylla group,S.baicalensis group,compatibility 1∶2 group,compatibility 1∶1 group,and compatibility 2∶1 group had significant effects(P<0.05),and the increase rate of pain threshold was 29.17%,47.83%,61.54%,and 50.61%,and 53.83%,respectively.The analgesic effect of the compatibility 2∶1 group was the most significant.(iii)In the hemostatic experiment,there was a significant difference in hemostatic time between the compatibility 1∶2 group and the blank control group,P.polyphylla group,and S.baicalensis group(P<0.05).The hemostatic effect of the compatibility 1∶2 group was the best.[Conclusions]P.polyphylla,S.baicalensis,and its compatibility had good analgesic and hemostatic effects,and the best compatibility ratio was 1∶2.Shibo ZHAO Suoyi HUANG Zhenjing SI Lanlan GAN Shusheng HUANG Ye FU Jianjie LAN Xueying YU Yuekong CEN 2020Medicinal Plant2020,11,2:2

20PHISDetector:A Tool to Detect Diverse In Silico Phage–host Interaction Signals for Virome Studies显示文摘Phage-microbe interactions are appealing systems to study coevolution,and have also been increasingly emphasized due to their roles in human health,disease,and the development of novel therapeutics.Phage-microbe interactions leave diverse signals in bacterial and phage genomic sequences,defined as phage-host interaction signals(PHISs),which include clustered regularly interspaced short palindromic repeats(CRISPR)targeting,prophage,and protein-protein interaction signals.In the present study,we developed a novel tool phage-host interaction signal detector(PHISDetector)to predict phage-host interactions by detecting and integrating diverse in silico PHISs,and scoring the probability of phage-host interactions using machine learning models based on PHIS features.We evaluated the performance of PHISDetector on multiple benchmark datasets and application cases.When tested on a dataset of 758 annotated phage-host pairs,PHISDetector yields the prediction accuracies of 0.51 and 0.73 at the species and genus levels,respectively,outperforming other phage-host prediction tools.When applied to 125,842 metagenomic viral contigs(mVCs)derived from 3042 geographically diverse samples,a detection rate of 54.54% could be achieved.Furthermore,PHISDetector could predict infecting phages for 85.6% of 368 multidrug-resistant(MDR)bacteria and 30% of 454 human gut bacteria obtained from the National Institutes of Health(NIH)Human Microbiome Project(HMP).The PHISDetector can be run either as a web server(http://gffzz439de54f28c841f5h9n9w5ob0fvnu65u9.ffgz.tsg.suse.edu.cn/PHISDetector/)for general users to study individual inputs or as a stand-alone version(http://gffzz188fe103f8f1460as9n9w5ob0fvnu65u9.ffgz.tsg.suse.edu.cn/HITImmunologyLab/PHISDetector)to process massive phage contigs from virome studies.Fengxia Zhou Rui Gan Fan Zhang Chunyan Ren Ling Yu Yu Si Zhiwei Huang 2022Genomics, Proteomics & Bioinformatics2022,20,3:0
