| 2 | Genotypic Characterization of Methicillin-resistant Staphylococcus aureus Isolated from Pigs and Retail Foods in China显示文摘Objective To investigate the genotypic diversity of Methicillin-resistant Staphylococcus aureus(MRSA) isolated from pigs and retail foods from different geographical areas in China and further to study the routes and rates of transmission of this pathogen from animals to food. Methods Seventy-one MRSA isolates were obtained from pigs and retail foods and then characterized by multi-locus sequencing typing(MLST), spa typing, multiple-locus variable number of tandem repeat analysis(MLVA), pulsed-field gel electrophoresis(PFGE), and antimicrobial susceptibility testing. Results All isolated MRSA exhibited multi-drug resistance(MDR). Greater diversity was found in food-associated MRSA(7 STs, 8 spa types, and 10 MLVA patterns) compared to pig-associated MRSA(3 STs, 1 spa type, and 6 MLVA patterns). PFGE patterns were more diverse for pig-associated MRSA than those of food-associated isolates(40 vs. 11 pulse types). Among the pig-associated isolates, CC9-ST9-t899-MC2236 was the most prevalent clone(96.4%), and CC9-ST9-t437-MC621(20.0%) was the predominant clone among the food-associated isolates. The CC9-ST9 isolates showed significantly higher antimicrobial resistance than other clones. Interestingly, CC398-ST398-t034 clone was identified from both pig-and food-associated isolates. Of note, some community-and hospital-associated MRSA strains(t030, t172, t1244, and t4549) were also identified as food-associated isolates. Conclusion CC9-ST9-t899-MC2236-MDR was the most predominant clone in pigs, but significant genetic diversity was observed in food-associated MRSA. | 3 | A Phytophthora capsid Effector Targets ACD11 Binding Partners that Regulate ROS-Mediated Defense Response in Arabidopsis显示文摘Reactive oxygen species (ROS) play a vital role in plant immune response, but the genes involved in the regulation of ROS are scantily reported. Phytophthora pathogens produce a large number of effectors to promote infection, but the modes of action adopted are largely unknown. Here, we report that RxLR207 could activate ROS-mediated cell death in Nicotiana benthamiana and was essential for virulence of P. capsici. We found that this effector targeted BPA1 (binding partner of ACD11) and four members of BPLs (BPAl-Like proteins)in Arabidopsis, and the bpa1 and bpl mutants had enhanced ROS accumulation and cell death under biotic or abiotic stresses. Furthermore, we showed that BPA1 and several BPLs functioned redundantly in plant immunity to P. capsici. We discovered that BPA1 and all six BPLs interacted with ACD11, and stabilization of ACD11 was impaired in the bpa 1, bpl2, bpl3, and bpl4 mutants. RxLR207 could promote the degradation of BPA1, BPL1, BPL2, and BPL4 to disrupt ACD11 stabilization in a 26S proteasome-dependent manner. | 16 | Effectiveness of hospital disinfection and experience learnt from 11 years of surveillance显示文摘This survey was designed to assess the sanitation status of hospitals and the compliance of hospital staff to disinfection strategies within the past 11 years.A total of 199 provincial affiliated tertiary or secondary public hospitals from 2007 to 2017 were investigated and seven critical categories,namely indoor air,work surface,hand hygiene,ultraviolet(UV)irradiation intensity,use of disinfectants,sterilization of medical items,and effects of steam sterilizer,were monitored.The average qualified rates were(94.74±3.54)%(810/855),(97.25±1.65)%(1876/1929),(87.57±4.60)%(2508/2864),(95.00±4.50)%(1196/1259),and(98.76±1.14)%(1599/1619)for indoor air,work surface,hand hygiene,UV irradiation intensity,and sterilization of medical items,respectively.In terms of other categories,few samples were not qualified:3/1575 for use of disinfectants and 1/243 for effects of steam sterilizer.The hospital disinfection monitoring and supervision program effectively improved the effectiveness of disinfection.Routine monitoring and supervision must be conducted to ensure a safe hospital treatment environment. | Chun’ai Tao Yongxin Gan Weidong Su Zhutian Li Xiaolan Tang | 2019 | The Journal of Biomedical Research2019，33，6： | 1 |
