| 1 | MHD and kinetic analysis of flow bursts in the Earth's plasma sheet显示文摘In an attempt to study the flow bursts in the Earth’s plasma sheet we select an event that took place on August 7,2004 in the expansion phase of a substorm,using data from the geomagnetic index,solar wind data,plasma and magnetic field observations from C1 Cluster satellite(the Cluster mission has 4 satellites)and from Double Star TC-1 satellite.In MHD approach,TC-1 firstly observed the tailward flow,then the earthward,and finally the flow alternated in two directions.C1 firstly observed the earthward plasma flow,and then the tailward plasma flow.Before flow bursts are observed by TC-1 and C1,there are disturbances in local entropy with their tailward local entropy larger than those of the earthward.The kinetic features of the plasma flow observed by C1 are similar to those in MHD.However,kinetic characteristics of the plasma flow observed by TC-1 are far more than the description in MHD.The inadequacy mainly exists in two cases:(i)the firstly enhanced tailward flows given in MHD are found without significant increase of the energetic tailward flux;(ii)the almost stagnant flow in MHD is composed of the enhanced energetic ion flux in both earthward and tailward directions.The earthward flow burst observed by TC-1 might be multiple overshoots and rebounds.The earthward flow burst observed by C1 might be simply rebounded in the near-Earth.The pulsation observed by C1 is earlier than that observed by TC-1 with the former intensity less than that of the latter.After the energetic ion flux in the tailward direction is significantly enhanced,the power spectrum intensity of the ULF wave commences to increase obviously,which may suggest that the stream instability is closely correlated with ULF pulsations. | MA YuDuan CAO JinBin FU HuiShan REME H. | 2 | Mitotic motor CENP-E cooperates with PRC1 in temporal control of central spindle assembly显示文摘Error-free cell division depends on the accurate assembly of the spindle midzone from dynamic spindle microtubules to ensure chromatid segregation during metaphase-anaphase transition.However,the mechanism underlying the key transition from the mitotic spindle to central spindle before anaphase onset remains elusive.Given the prevalence of chromosome instability phenotype in gastric tumorigenesis,we developed a strategy to model context-dependent cell division using a combination of light sheet microscope and 3D gastric organoids.Light sheet microscopic image analyses of 3D organoids showed that CENP-E inhibited cells undergoing aberrant metaphase-anaphase transition and exhibiting chromosome segregation errors during mitosis.Highresolution real-time imaging analyses of 2D cell culture revealed that CENP-E inhibited cells undergoing central spindle splitting and chromosome instability phenotype.Using biotinylated syntelin as an affinity matrix,we found that CENP-E forms a complex with PRC1 in mitotic cells.Chemical inhibition of CENP-E in metaphase by syntelin prevented accurate central spindle assembly by perturbing temporal assembly of PRC1 to the midzone.Thus,CENP-E-mediated PRC1 assembly to the central spindle constitutes a temporal switch to organize dynamic kinetochore microtubules into stable midzone arrays.These findings reveal a previously uncharacterized role of CENP-E in temporal control of central spindle assembly.Since CENP-E is absent from yeast,we reasoned that metazoans evolved an elaborate central spindle organization machinery to ensure accurate sister chromatid segregation during anaphase and cytokinesis. | Xu Liu Leilei Xu Junying Li Phil Y.Yao Wanjuan Wang Hazrat Ismail Haowei Wang Bryce Liao Zhihong Yang Tarsha Ward Ke Ruan Jianchun Zhang Quan Wu Ping He Xia Ding Dongmei Wang Chuanhai Fu Zhen Dou Feng Yan Wenwen Wang Xing Liu Xuebiao Yao | 2020 | Journal of Molecular Cell Biology2020,12,8: | 3 |
| 3 | Kinectin 1 promotes the growth of triple-negative breast cancer via directly co-activating NF-kappaB/p65 and enhancing its transcriptional activity显示文摘Triple-negative breast cancer(TNBC)is the most challenging subtype of breast cancer.Various endeavor has been made to explore the molecular biology basis of TNBC.Herein,we reported a novel function of factor Kinectin 1(KTN1)as a carcinogenic promoter in TNBC.KTN1 expression in TNBC was increased compared with adjacent tissues or luminal or Her2 subtypes of breast cancer,and TNBC patients with high KTN1 expression have poor prognosis.In functional studies,knockdown of KTN1 inhibited the proliferation and invasiveness of TNBC both in vitro and in vivo,while overexpression of KTN1 promoted cancer cell proliferation and invasiveness.RNA-seq analysis revealed that the interaction of cytokine-cytokine receptor,particularly CXCL8 gene,was upregulated by KTN1,which was supported by the further experiments.CXCL8 depletion inhibited the tumorigenesis and progression of TNBC.Additionally,rescue experiments validated that KTN1-mediated cell growth acceleration in TNBC was dependent on CXCL8 both in vitro and in vivo.Furthermore,it was found that KTN1 enhanced the phosphorylation of NF-κB/p65 protein at Ser536 site,and specifically bound to NF-κB/p65 protein in the nucleus and cytoplasm of cells.Moreover,the transcription of CXCL8 gene was directly upregulated by the complex of KTN1 and NF-κB/p65 protein.Taken together,our results elucidated a novel mechanism of KTN1 gene in TNBC tumorigenesis and progression.KTN1 may be a potential molecular target for the development of TNBC treatment. | Lin Gao Shanze Chen Malin Hong Wenbin Zhou Bilan Wang Junying Qiu Jinquan Xia Pan Zhao Li Fu Jigang Wang Yong Dai Ni Xie Qinhe Yang Hsien-Da Huang Xiang Gao Chang Zou | 2021 | Signal Transduction and Targeted Therapy2021,6,8: | 2 |

| 7 | Bulk glass formation in ternary Cu-Zr-Ti system显示文摘The formation of bulk metallic glasses (BMGs) in ternary Cu-Zr-Ti system was investigated by a copper mold casting method. The nature of the amorphous phase was verified by X-ray diffraction (XRD) and differential scanning calorimetry (DSC). It was demonstrated that the BMGs could be formed in a broad composition range in this system. Cu50Zr42.5Ti7.5, Cu60Zr27.5Ti12.5, Cu60Zr30Ti10 and Cu60Zr32.5Ti7.5 alloys exhibit strong glass-forming ability (GFA), and fully glassy rods of 5 mm in diameter can be obtained. In the center region of the ternary diagram, however, the GFA of the alloys was degraded due to the presence of Laves phase. The addition of a small amount of Er (2at%) for replacing Zr effectively improves the glass-forming ability of Cu46.25Zr46.25Al7.5 alloy, and the glassy rod with a diameter of at least 12 mm can be formed. For further development, we introduced siloxane units into AIE molecules, designed and synthesized TPE derivatives with siloxane side chains via facile Piers-Rubinsztajn reaction. The obtained AIE molecular liquids exhibit unique photophysical properties. Compared with the obtained alkyl TPE-solids, siloxane TPE show liquid state, which proves that the siloxane units have stronger liquefaction effect than alkyl. Viscosity test shows that siloxane TPE-liquids has far more lower viscosity and better fluidity than the long-chain alkyl molecular liquids in previous research. All those properties are attributed to the weak interaction between flexible molecular chains of siloxane. Besides,fluorescence test shows temperature responsiveness of siloxane TPE-liquids.
