| 1 | EGFR and HER2 expression in advanced biliary tract cancer显示文摘AIM:To analyze the pathogenetic role and potential clinical usefulness of the epidermal growth factor receptor(EGFR)and the human epidermal growth factor receptor 2(HER2)in patients with advanced biliary tract cancer(BTC). METHODS:EGFR and HER2 expression was studied in biopsy samples from 124 patients(51%women; median age 64.8 years),with advanced BTC diagnosed between 1997 and 2004.Five micrometers sections of paraffin embedded tissue were examined by standard, FDA approved immunohistochemistry.Tumors with scores of 2+or 3+for HER2 expression on immunochemistry were additionally tested for HER2 gene amplification by fluorescence in situ hybridisation(FISH).RESULTS:34/124 patients(27.4%)had gallbladder cancer,47(37.9%)had intrahepatic BTC and 43(34.7%)had extrahepatic or perihilar BTC.EGFR expression was examined in a subset of 56 samples. EGFR expression was absent in 22/56 tumors(39.3%). Of the remaining samples expression was scored as 1+in 12(21.5%),2+in 13(23.2%)and 3+in 9(16%), respectively.HER2 expression was as follows:score 0 73/124(58.8%),score 1+27/124(21.8%),score 2+ 21/124(17%)and score 3+4/124(3.2%).HER2 gene amplification was present in 6/124,resulting in an overall amplification rate of 5%. | 3 | Anemia and long-term outcome in adjuvant and neoadjuvant radiochemotherapy of stage Ⅱ and Ⅲ rectal adenocarcinoma:The Freiburg experience(1989-2002)显示文摘AIM:To evaluate the long-term outcome of standard5-FU based adjuvant or neoadjuvant radiochemotherapyand to identify the predictive factors,especially anemiabefore and after radiotherapy as well as hemoglobinincrease or decrease during radiotherapy.METHODS:Two hundred and eighty-six patientswith Union International Contre Cancer(UICC)stageⅡ and Ⅲ rectal adenocarcinomas,who underwentresection by conventional surgical techniques(lowanterior or abdominoperineal resection),receivedeither postoperative(n=233)or preoperative(n=53)radiochemotherapy from January 1989 until July 2002.Overall survival(OAS),cancer-specific survival(CSS),disease-free survival(DFS),local-relapse-free(LRS)and distant-relapse-free survival(DRS)were evaluatedusing Kaplan-Meier,Log-rank test and Cox's proportionalhazards as statistical methods.Multivariate analysis wasused to identify prognostic factors.Median follow-uptime was 8 years.RESULTS:Anemia before radiochemotherapy was anindependent prognostic factor for improved DFS(riskratio 0.76,P=0.04)as well as stage,grading,R status(free radial margins),type of surgery,carcinoembryonicantigen(CEA)levels,and gender.The univariate analysisrevealed that anemia was associated with impaired LRS (better local control)but with improved DFS.In contrast,hemoglobin decrease during radiotherapy was anindependent risk factor for DFS(risk ratio 1.97,P=0.04).During radiotherapy,only 30.8% of RO-resected patientssuffered from hemoglobin decrease compared to 55.6%if R1/2 resection was performed(P=0.04).The 5-yearOAS,CSS,DFS,LRS and DRS were 47.0%,60.0%,41.4%,67.2%,and 84.3%,respectively.Significantdifferences between preoperative and postoperativeradiochemotherapy were not found.CONCLUSION:Anemia before radiochemotherapyand hemoglobin decrease during radiotherapy have nopredictive value for the outcome of rectal cancer.Stage,grading,R status(free radial margins),type of surgery,CEA levels,and gender have predictive value for theoutcome of rectal cancer. | Christian Weissenberger Geissler Michael Otto Florian Barke Annette Henne Karl von Plehn Georg Rein Alex Müller Christine Bartelt Susanne Henke Michael | 2006 | World Journal of Gastroenterology2006,12,12: | 4 |

| 9 | Forecasting the spatial and temporal charging demand of fully electrified urban private car transportation based on large-scale traffic simulation显示文摘To support power grid operators to detect and evaluate potential power grid congestions due to the electrification of urban private cars,accurate models are needed to determine the charging energy and power demand of battery electric vehicles(BEVs)with high spatial and temporal resolution.Typically,e-mobility traffic simulations are used for this purpose.In particular,activity-based mobility models are used because they individually model the activity and travel patterns of each person in the considered geographical area.In addition to inaccuracies in determining the spatial distribution of BEV charging demand,one main limitation of the activity-based models proposed in the literature is that they rely on data describing traffic flow in the considered area.However,these data are not available for most places in the world.Therefore,this paper proposes a novel approach to develop an activity-based model that overcomes the spatial limitations and does not require traffic flow data as an input parameter.Instead,a route assignment procedure assigns a destination to each BEV trip based on the evaluation of all possible destinations.The basis of this evaluation is the travel distance and speed between the origin of the trip and the destination,as well as the car-access attractiveness and the availability of parking spots at the destinations.The applicability of this model is demonstrated for the urban area of Berlin,Germany,and its 448 sub-districts.For each district in Berlin,both the required daily BEV charging energy demand and the power demand are determined.In addition,the load shifting potential is investigated for an exemplary district.The results show that peak power demand can be reduced by up to 31.7%in comparison to uncontrolled charging. | Florian Straub Otto Maier Dietmar Göohlich a Yuan Zou | 2023 | Green Energy and Intelligent Transportation2023,2,1: | 0 |
| 10 | Differential effect of ethanol intoxication on peripheral markers of cerebral injury in murine blunt traumatic brain injury显示文摘Background:Blood-based biomarkers have proven to be a reliable measure of the severity and outcome of traumatic brain injury(TBI)in both murine models and patients.In particular,neuronspecific enolase(NSE),neurofilament light(NFL)and S100 beta(S100B)have been investigated in the clinical setting post-injury.Ethanol intoxication(EI)remains a significant comorbidity in TBI,with 30–40%of patients having a positive blood alcohol concentration post-TBI.The effect of ethanol on blood-based biomarkers for the prognosis and diagnosis of TBI remains unclear.In this study,we investigated the effect of EI on NSE,NFL and S100B and their correlation with blood–brain barrier integrity in a murine model of TBI.Methods:We used ultra-sensitive single-molecule array technology and enzyme-linked immunosorbent assay methods to measure NFL,NSE,S100B and claudin-5 concentrations in plasma 3 hours post-TBI.Results:We showed that NFL,NSE and S100B were increased at 3 hours post-TBI.Interestingly,ethanol blood concentrations showed an inverse correlation with NSE but not with NFL or S100B.Claudin-5 levels were increased post-injury but no difference was detected compared to ethanol pretreatment.The increase in claudin-5 post-TBI was correlated with NFL but not with NSE or S100B.Conclusions:Ethanol induces an effect on biomarker release in the bloodstream that is different from TBI not influenced by alcohol.This could be the basis of investigations into humans. | Zhenghui Li Jin Zhang Steffen Halbgebauer Akila Chandrasekar Rida Rehman Albert Ludolph Tobias Boeckers Markus Huber-Lang Markus Otto Francesco Roselli Florian olde Heuvel | 2021 | Burns & Trauma2021,9,1: | 0 |
