| 1 | Climate and litter C/N ratio constrain soil organic carbon accumulation显示文摘Soil organic carbon(SOC) plays critical roles in stabilizing atmospheric CO2 concentration, but the mechanistic controls on the amount and distribution of SOC on global scales are not well understood. In turn, this has hampered the ability to model global C budgets and to find measures to mitigate climate change. Here, based on the data from a large field survey campaign with 2600 plots across China’s forest ecosystems and a global collection of published data from forested land, we find that a low litter carbon-to-nitrogen ratio(C/N) and high wetness index(P/PET, precipitation-to-potentialevapotranspiration ratio) are the two factors that promote SOC accumulation, with only minor contributions of litter quantity and soil texture. The field survey data demonstrated that high plant diversity decreased litter C/N and thus indirectly promoted SOC accumulation by increasing the litter quality. We conclude that any changes in plant-community composition, plant-species richness and environmental factors that can reduce the litter C/N ratio, or climatic changes that increase wetness index, may promote SOC accumulation. | 2 | In vitro culture and differentiation of rat embryonic midbrain-derived neural stem cells显示文摘BACKGROUND:Midbrain-derived neural stem cells (mNSCs) can differentiate into functional mature dopaminergic neurons. The mNSCs are considered the ideal choice for cell therapy of Parkinson’s disease. OBJECTIVE: To isolate rat embryonic mNSCs and to observe the differentiation characteristics of mNSCs induced by cell growth-promoting factors. DESIGN,TIME AND SETTING: An in vitro cell culture study based on the molecular biology of nerve cells was carried out at the Institute of Clinical Medicine,China-Japan Friendship Hospital (China) from March to November 2007. MATERIALS: Sprague Dawley rats at embryonic day 14 were used in this study. Nestin antibody,β-Ⅲ tubulin antibody,glial fibrillary acidic protein (GFAP) antibody and cyclic nucleotide 3’-phosphohydrolase (CNPase) antibody were provided by Abcam; DMEM/F12 medium and N2 supplement were provided by In-vitrogen; epidermal growth factor (EGF) and fibroblast growth factor-2 (FGF2) were provided by R&D Sys-tems. METHODS: The ventral mesencephalon was dissected from embryonic day 14 rat embryos. By trypsin di-gestion and mechanical separation,the brain tissue was triturated into a fine single-cell suspension. The cells were cultured in 5 mL serum-free medium containing DMEM/F12,1% N2 supplement,20 ng/mL EGF and FGF2. The mNSCs at the third generation were coated with 10 μg/mL polylysine and induced to differenti-ate in the DMEM/F12 supplemented with 1% fetal bovine serum and 1% N2. MAIN OUTCOME MEASURES: The neural spheres of the third passage were identified by nestin im-munofluorescence; at the same time,the cells were induced to differentiate,and the types of differentiated cell were identified by immunofluorescence for βⅢ tubulin,GFAP and CNPase. RESULTS: Seven days after primary culture,a great many neurospheres could be obtained by successive pasage. Immunofluorescence assays showed that the neurospheres were nestin positive,and after differentia-tion,the cells expressed GFAP,CNPase and β-Ⅲ-tubulin. | 3 | Effect of N and P addition on soil organic C potential mineralization in forest soils in South China显示文摘Atmospheric nitrogen deposition is at a high level in some forests of South China. The effects of addition of exogenous N and P on soil organic carbon mineralization were studied to address: (1) if the atmospheric N deposition promotes soil C storage through decreasing mineralization; (2) if the soil available P is a limitation to organic carbon mineralization. Soils (0-10 cm) was sampled from monsoon evergreen broad-leaved forest (MEBF), coniferous and broad-leaved mixed forest (CBMF), and Pinus massoniana forest (PMF) in Dinghushan Biosphere Reserve (located in Guangdong Province, China). The soils were incubated at 25°C for 45 weeks, with addition of N (NH4NO3 solution) or P (KH2PO4 solution). CO2-C emission and the inorganic N (NH4+-N and NO3?-N) of the soils were determined during the incubation. The results showed that CO2-C emission decreased with the N addition. The addition of P led to a short-term sharp increase in CO2 emission after P application, and the responses of CO2-C evolution to P addition in the later period of incubation related to forest types. Strong P inhibition to CO2 emission occurred in both PMF and CBMF soils in the later incubation. The two-pool kinetic model was fitted well to the data for C turnover in this experiment. The model analysis demonstrated that the addition of N and P changed the distribution of soil organic C between the labile and recalcitrant pool, as well as their mineralization rates. In our experiment, soil pH can not completely explain the negative effect of N addition on CO2-C emission. The changes of soil inorganic N during incubation seemed to support the hypothesis that the polymerization of added nitrogen with soil organic compound by abiotic reactions during incubation made the added nitrogen retard the soil organic carbon mineralization. | 4 | Primary structure of helix C to helix G of a new retinal protein in H.sp.xz515显示文摘The gene fragment encoding the retinal protein from helix C to helix G in a new strain of halobacteria, H.sp.xz515 has been amplified by PCR method. The nucleotide sequence of this fragment has been determined. The deduced amino acid sequence has been compared with halobium br and other two br-like proteins, ar-1 and ar-2. Results show that those amino acid residues in br, essential for proton pumping and binding to retinal, are conserved. | 7 | Gene expression dynamics during the gonocyte to spermatogonia transition and spermatogenesis in the domestic yak显示文摘Background:Spermatogenesis is a cellular differentiation process that includes three major events:mitosis of spermatogonia,meiosis of spermatocytes and spermiogenesis.Steady-state spermatogenesis relies on functions of spermatogonial stem cells(SSCs).Establishing and maintaining a foundational SSC pool is essential for continued spermatogenesis in mammals.Currently,our knowledge about SSC and spermatogenesis is severely limited in domestic animals.Results:In the present study,we examined transcriptomes of testes from domestic yaks at four different stages(3,5,8 and 24 months of age)and attempted to identify genes that are associated with key developmental events of spermatogenesis.Histological analyses showed that the most advanced germ cells within seminiferous tubules of testes from 3,5,8 and 24 months old yaks were gonocytes,spermatogonia,spermatocytes and elongated spermatids,respectively.RNA-sequencing(RNA-seq)analyses revealed that 11904,4381 and 2459 genes were differentially expressed during the gonocyte to spermatogonia transition,the mitosis to meiosis transition and the meiosis to post-meiosis transition.Further analyses identified a list of candidate genes than may regulate these important cellular processes.CXCR4,a previously identified SSC niche factor in mouse,was one of the up-regulated genes in the 5 months old yak testis.Results of immunohistochemical staining confirmed that CXCR4 was exclusively expressed in gonocytes and a subpopulation of spermatogonia in the yak testis.Conclusions:Together,these findings demonstrated histological changes of postnatal testis development in the domestic yak.During development of spermatogonial lineage,meiotic and haploid germ cells are supported by dynamic transcriptional regulation of gene expression.Our transcriptomic analyses provided a list of candidate genes that potentially play crucial roles in directing the establishment of SSC and spermatogenesis in yak. | Guowen Wang Yongchang Li Qilin Yang Shangrong Xu Shike Ma Rongge Yan Ruina Zhang Gongxue Jia Deqiang Ai Qi’en Yang | 2019 | Journal of Animal Science and Biotechnology2019,10,4: | 3 |
| 8 | Comparison of Nonlinear Local Lyapunov Vectors with Bred Vectors, Random Perturbations and Ensemble Transform Kalman Filter Strategies in a Barotropic Model显示文摘The breeding method has been widely used to generate ensemble perturbations in ensemble forecasting due to its simple concept and low computational cost. This method produces the fastest growing perturbation modes to catch the growing components in analysis errors. However, the bred vectors(BVs) are evolved on the same dynamical flow, which may increase the dependence of perturbations. In contrast, the nonlinear local Lyapunov vector(NLLV) scheme generates flow-dependent perturbations as in the breeding method, but regularly conducts the Gram–Schmidt reorthonormalization processes on the perturbations. The resulting NLLVs span the fast-growing perturbation subspace efficiently, and thus may grasp more components in analysis errors than the BVs.In this paper, the NLLVs are employed to generate initial ensemble perturbations in a barotropic quasi-geostrophic model.The performances of the ensemble forecasts of the NLLV method are systematically compared to those of the random perturbation(RP) technique, and the BV method, as well as its improved version—the ensemble transform Kalman filter(ETKF)method. The results demonstrate that the RP technique has the worst performance in ensemble forecasts, which indicates the importance of a flow-dependent initialization scheme. The ensemble perturbation subspaces of the NLLV and ETKF methods are preliminarily shown to catch similar components of analysis errors, which exceed that of the BVs. However, the NLLV scheme demonstrates slightly higher ensemble forecast skill than the ETKF scheme. | 9 | Study of Cracking Mechanism and Wear Resistance in Laser Cladding Coating of Ni-based Alloy显示文摘Nickel-based alloy coatings were widely used for the remanufacturing of dies and moulds by laser cladding,but the crack sensitivity would be increase due to the higher strength and hardness,which reduced the wear resistance of Ni-based alloys.In this paper,Ni-based coatings with the addition of a plastic phase(an austenitic stainless net)were prepared using laser cladding technology,and the CeO_(2)was added in cladding layers.The cracking mechanism,microhardness,microstructure,phase composition,and wear properties were investigated.The relationship between thermal stress and the elastic and plastic fracture had been developed from the standpoint of fracture mechan-ics and thermal elastic fracture mechanics.The fracture criterion of the nickel-based coating was obtained,and the study has shown that the crack sensitivity could be reduced by decreasing the thermal expansion coefficientΔα.Thus,a new method was proposed,which the stainless steel nets were prefabricated on the substrate.It was found that the number of cracks reduced significantly with the addition of stainless steel net.When the stainless steel net with 14 mesh was added in Ni-based coatings,the average microhardness of nickel composite coating was 565 HV_(0.2),which was 2.6 times higher than that of the 45 steel substrate.Although the rare earth oxide 4 wt.%CeO_(2)and stainless steel net were added in the Ni-based coating reducing the microhardness(the average microhardness is 425 HV_(0.2)),the wear resistance of it improved substantially.The wear volume of Ni-based composite coating was 0.56×10^(−5) mm^(3)·N^(−1)·m^(−1),which was 85.1%lower than that of 45 steel.The experiment results have shown that the Nickel-based composite coating is equipped with low crack sensitivity and high abrasive resistance with austenitic stainless net and the rare earth oxide 4 wt.%CeO2.This research offers an efficient solution to produce components with low crack susceptibility and high wear-resistance coatings fabricated by laser cladding. | Zhenglei Yu Lunxiang Li Deqiang Zhang Guangfeng Shi Guang Yang Zezhou Xu Zhihui Zhang | 2021 | Chinese Journal of Mechanical Engineering2021,34,5: | 2 |

| 20 | Experimental and theoretical study of the charge transport property of 4,4′-N,N′-dicarbazole-biphenyl显示文摘The hole and electron mobilities of the amorphous films of the organic semiconductor 4,4′-N,N′-dicarbazole-biphenyl (CBP) at different electric fields were measured through the time of flight (TOF) method. Based on its crystalline structure, the hole and electron mobilities of CBP were calculated. A detailed comparison between experimental and theoretical results is necessary for further understanding its charge transport properties. In order to do this, charge mobilities at zero electric field, μ(0), were deduced from experimental data as a link between experimental and theoretical data. It was found that the electron transport of CBP is less affected by traps compared with its hole transport. This unusual phenomenon can be understood through the distributions of frontier molecular orbitals.
