1China Antihypertensive Trial in Acute Ischemic Stroke II (CATIS-2): rationale and design显示文摘Backgrounds Increased blood pressure(BP)for patients who had an acute ischaemic stroke is associated with poor functional outcome,however the optimal timing of antihypertensive therapy is unknown.Aims We aim to compare early antihypertensive treatment to delayed antihypertensive treatment for reducing the risk of composite major disability and mortality at 3 months in acute ischaemic stroke.Design The China Antihypertensive Trial in Acute Ischemic Stroke II(CATIS-2)trial is a multicentre,randomised,open-label,blinded-endpoints trial that will be conducted in 100 hospitals in China.The primary outcome is the composite of death and major disability(modified Rankin Scale score≥3)at 3 months of randomisation.Antihypertensive treatment will be received immediately after randomisation in the early treatment group,aimed at average systolic BP by 10%-20%reduction within the first 24 hours,and achieving an average BP level of<140/90 mm Hg within 5 days.Patients in the delayed treatment group will discontinue any antihypertension medications for the first 7 days of randomisation,and will receive antihypertensive therapy achieving a BP goal of<140/90 mm Hg after 7 days.Conclusion The CATIS-2 trial will be testing the hypotheses that early BP lowering leads to improved functional outcome without any other harms,and developing clinical guidelines of the BP management for patients who had an acute ischaemic stroke.Liping Liu Yilong Wang Xuewei Xie Dacheng Liu Aili Wang Penglian Wang Suwen Shen Chongke Zhong Yufei Wei Tan Xu Yuesong Pan Yong Jiang Xia Meng David Wang Yonghong Zhang Jiang He Yongjun Wang 2021Stroke & Vascular Neurology2021,6,2:3
2Synthesis of Spirostanol Saponins via Gold(Ⅰ)-Catalyzed Glycosylation in the Presence of Ga(Otf)3, In(Otf)3, or HOTf显示文摘Summary of main observation and conclusion An effective approach relying on a Lewis acid-or Brφnsted acid-assisted gold(Ⅰ)-catalyzed glycosylation has been reported in the synthesis of a panel of the representative natural spirostanol saponins,namely polyphyllin D (1),polyphyllin V (2),dioscin (3),formosanin C (4),and a derivative of polyphyllin D bearing a terminal azide group (5).This approach highlights the engagement of low Ioadings of Ph3AuPOTf (≤ 0.5mol%) in the presence of Ga(OTf)3,In(OTf)3,or HOTf (~10 mol%) as a co-catalyst,at practical reaction rates as an alternative to the conventional ~10 mol%Ioadings of the gold(Ⅰ) catalyst in the glycosylation.Polyphyllin D (1) was obtained in 41% overall yield over six steps compared to the maximum 30% yield in previous syntheses,where conventional donors and promoters were used.By exploiting a regioselective rhamnosylation,a 'one-pot' approach was adopted to assemble 1 and 5,thus further strengthening the efficiency of the gold(Ⅰ)-catalyzed glycosylation.Teddy Stephen Ehianeta Dacheng Shen Peng Xu Biao Yu 2019Chinese Journal of Chemistry2019,37,8:2

4Total Synthesis of Starfish Cyclic Steroid Glycosides.Part 3,Formal Total Synthesis of Luzonicosides A and D显示文摘Comprehensive Summary Luzonicosides A(1)and D(2)are the major saponins isolated from pacific starfish Echinaster Luzonicus,featuring a sterically congested 16-membered ring which is formed by bridging the C3 and C6 of a 3β,6β-dihydroxy-Δ7-ene steroid with a 1,2-trans-linked linear trisaccharide via glycosidic and ether bonds,respectively.Here,we describe a formal total synthesis of starfish cyclic steroid glycosides 1 and 2.The synthetic route is highly modular and versatile,involving construction of the 16-membered macrocycles via Au(I)-catalyzed intramolecular glycosylation,highly efficient installation of unsaturated sugar moieties with ortho-hexynylbenzoates,elaboration of glucuronic acid unit via post-oxidation,and a de novo synthesis of the ether-linked pyranose unit.Dapeng Zhu Mingyu Geng Dacheng Shen Biao Yu 2023Chinese Journal of Chemistry2023,41,8:1
