1Oxidative stress, mitochondrial damage and neurodegenerative diseases显示文摘Oxidative stress and mitochondrial damage have been implicated in the pathogenesis of several neurodegenerative diseases, including Alzheimer's disease, Parkinson's disease and amyotrophic lateral sclerosis. Oxidative stress is characterized by the overproduction of reactive oxygen species, which can induce mitochondrial DNA mutations, damage the mitochondrial respiratory chain, alter membrane permeability, and influence Ca2+ homeostasis and mitochondrial defense systems. All these changes are implicated in the development of these neurodegenerative diseases, mediating or amplifying neuronal dysfunction and triggering neurodegeneration. 2Primary studies on integration optimization of differential steering of electric vehicle with motorized wheels based on quality engineering显示文摘The model of the differential steering system(DSS) of electric vehicle with motorized wheels and the three-degree-of-freedom dynamic model of vehicle are built.Based on these models,the concepts and quantitative expressions of steering road feel,steering portability and steering stability are proposed.Through integrating the Monte Carlo descriptive sampling,elitist non-dominated sorting genetic algorithm(NSGA-II) and Taguchi robust design method,the system parameters are optimized with steering road feel and steering portability as optimization targets,and steering stability and steering portability as constraints.The simulation results show that the system optimized based on quality engineering can improve the steering road feel,guarantee steering stability and steering portability and thus provide a theoretical basis for the design and optimization of the electric vehicle with motorized wheels system.ZHAO WanZhong WANG ChunYan SUN PeiKun LIU Shun 2011Science China(Technological Sciences)2011,54,11:15
3Key residues of the receptor binding motif in the spike protein of SARS-CoV-2 that interact with ACE2 and neutralizing antibodies显示文摘Coronavirus disease 2019(COVID-19),caused by the novel human coronavirus SARS-CoV-2,is currently a major threat to public health worldwide.The viral spike protein binds the host receptor angiotensin-converting enzyme 2(ACE2)via the receptor-binding domain(RBD),and thus is believed to be a major target to block viral entry.Both SARS-CoV-2 and SARS-CoV share this mechanism.Here we functionally analyzed the key amino acid residues located within receptor binding motif of RBD that may interact with human ACE2 and available neutralizing antibodies.The in vivo experiments showed that immunization with either the SARS-CoV RBD or SARS-CoV-2 RBD was able to induce strong clade-specific neutralizing antibodies in mice;however,the cross-neutralizing activity was much weaker,indicating that there are distinct antigenic features in the RBDs of the two viruses.This finding was confirmed with the available neutralizing monoclonal antibodies against SARS-CoV or SARS-CoV-2.It is worth noting that a newly developed SARS-CoV-2 human antibody,HA001,was able to neutralize SARS-CoV-2,but failed to recognize SARS-CoV.Moreover,the potential epitope residues of HA001 were identified as A475 and F486 in the SARS-CoV-2 RBD,representing new binding sites for neutralizing antibodies.Overall,our study has revealed the presence of different key epitopes between SARS-CoV and SARSCoV-2,which indicates the necessity to develop new prophylactic vaccine and antibody drugs for specific control of the COVID-19 pandemic although the available agents obtained from the SARS-CoV study are unneglectable.Chunyan Yi Xiaoyu Sun Jing Ye Longfei Ding Meiqin Liu Zhuo Yang Xiao Lu Yaguang Zhang Liyang Ma Wangpeng Gu Aidong Qu Jianqing Xu Zhengli Shi Zhiyang Ling Bing Sun 2020Cellular & Molecular Immunology2020,17,6:13

6A Multicenter Study of Viral Aetiology of Community-Acquired Pneumonia in Hospitalized Children in Chinese Mainland显示文摘Community-acquired pneumonia(CAP) is one of the leading causes of morbidity and mortality in children worldwide.In this study,we aimed to describe the aetiology of viral infection of pediatric CAP in Chinese mainland.During November2014 to June 2016,the prospective study was conducted in 13 hospitals.The hospitalized children under 18 years old who met the criteria for CAP were enrolled.The throat swabs or nasopharyngeal aspirates(NPAs) were collected which were then screened 18 respiratory viruses using multiplex PCR assay.Viral pathogens were present in 56.6%(1539/2721) of the enrolled cases,with the detection rate of single virus in 39.8% of the cases and multiple viruses in 16.8% of the cases.The most frequently detected virus was respiratory syncytial virus(RSV)(15.2%,414/2721).The highest detection rate of virus was in <6-month-age group(70.7%,292/413).RSV,human metapneumovirus(HMPV),human parainfluenza viruses(HPIVs) and influenza B virus(Flu B) showed the similar prevalence patterns both in north and south China,but HPIVs,Flu A,human bocavirus(HBoV),human adenovirus(HAdV) and human coronaviruses(HCoVs) showed the distinct circulating patterns in north and south China.Human enterovirus/human rhinovirus(HEV/HRV)(27.6%,27/98),HBoV(18.4%,18/98),RSV(16.3%,16/98) and HMPV(14.3%,14/98) were the most commonly detected viruses in severe pneumonia cases with single virus infection.In conclusion,viral pathogens are frequently detected in pediatric CAP cases and may therefore play a vital role in the aetiology of CAP.RSV was the most important virus in hospitalized children with CAP in Chinese mainland.Yun Zhu Baoping Xu Changchong Li Zhimin Chen Ling Cao Zhou Fu Yunxiao Shang Aihuan Chen Li Deng Yixiao Bao Yun Sun Limin Ning Shuilian Yu Fang Gu Chunyan Liu Ju Yin Adong Shen Zhengde Xie Kunling Shen 2021Virologica Sinica2021,36,6:7
7Mycorrhizal symbiosis modulates the rhizosphere microbiota to promote rhizobia–legume symbiosis显示文摘Plants establish symbioses with mutualistic fungi,such as arbuscular mycorrhizal(AM)fungi,and bacteria,such as rhizobia,to exchange key nutrients and thrive.Plants and symbionts have coevolved and represent vital components of terrestrial ecosystems.Plants employ an ancestral AM signaling pathway to establish intracellular symbioses,including the legume–rhizobia symbiosis,in their roots.Nevertheless,the relationship between the AM and rhizobial symbioses in native soil is poorly understood.Here,we examined how these distinct symbioses affect root-associated bacterial communities in Medicago truncatula by performing quantitative microbiota profiling(QMP)of 16S rRNA genes.We found that M.truncatula mutants that cannot establish AM or rhizobia symbiosis have an altered microbial load(quantitative abundance)in the rhizosphere and roots,and in particular that AM symbiosis is required to assemble a normal quantitative root-associated microbiota in native soil.Moreover,quantitative microbial co-abundance network analyses revealed that AM symbiosis affects Rhizobiales hubs among plant microbiota and benefits the plant holobiont.Through QMP of rhizobial rpoB and AM fungal SSU rRNA genes,we revealed a new layer of interaction whereby AM symbiosis promotes rhizobia accumulation in the rhizosphere of M.truncatula.We further showed that AM symbiosis-conditioned microbial communities within the M.truncatula rhizosphere could promote nodulation in different legume plants in native soil.Given that the AM and rhizobial symbioses are critical for crop growth,our findings might inform strategies to improve agricultural management.Moreover,our work sheds light on the co-evolution of these intracellular symbioses during plant adaptation to native soil conditions.Xiaolin Wang Huan Feng Yayu Wang Mingxing Wang Xingguang Xie Huizhong Chang Like Wang Jicheng Qu Kai Sun Wei He Chunyan Wang Chuanchao Dai Zhaohui Chu Changfu Tian Nan Yu Xuebin Zhang Huan Liu Ertao Wang 2021Molecular Plant2021,14,3:6

10Subthalamic nucleus deep brain stimulation for Parkinson’s disease: 8 years of follow-up显示文摘Objective:The short-term benefits of bilateral stimulation of the subthalamic nucleus(STN)in patients with advanced Parkinson’s disease(PD)are well documented,but long-term benefits are still uncertain.The aim of this study is to evaluate the outcome of 8 years of bilateral STN stimulation to PD patients.Methods:In this study,31 consecutive PD patients were treated with bilateral STN stimulation.Their functional status was measured using the Activities of Daily Living section of the Unified Parkinson’s Disease Rating Scale(UPDRS-ADL)at drug on(with medication)and drug off(without medication)states preoperatively and at 1,5,and 8 years postoperatively.In addition,Levodopa equivalent doses and stimulation parameters were also assessed.Results:After 8 years of STN stimulation,the UPDRS-ADL scores were improved by 4%at drug off status(P>0.05)and 22%at drug on status(P<0.05)compared with baseline;the levodopa daily doses were reduced by 28%(P<0.05)compared with baseline;the stimulation voltage and pulse width were not changed,but the stimulation frequency was decreased remarkably compared with the 5 years of follow-up.Adverse events were observed in 6 patients,including misplacement of the electrode and skin erosion requiring further surgery.All events were resolved without permanent sequelae.2 patients died of aspiration pneumonia 6 and 7 years after surgery.Conclusions:The marked improvement in UPDRS-ADL scores were still observed after 8 years of bilateral STN stimulation with medication.Dianyou Li Chunyan Cao Jing Zhang Shikun Zhan Shengdi Chen Bomin Sun 2013Translational Neurodegeneration2013,2,1:4
11Micelle or polymersome formation by PCL-PEG-PCL copolymers as drug delivery systems显示文摘Polye-caprolactone)-b-poly(ethylene glycol)-b-poly(e-caprolactone)(PCL-b-PEG-b-PCL,PCEC) triblock copolymers have been widely investigated in last several decades.Here,by altering the weight ratio of monomers in ring-opening polymerization,a series of PCEC triblock copolymers with varying hydrophobicity were synthesized,which were characterized by FTIR,1 H NMR,GPC and DSC.When PCEC copolymers with different weight ratios of PCL/PEG were dispersed in different aqueous solutions,they could self-assemble and form two distinctive nanoparticular structures:micelles or polymersomes.We then chose paclitaxel(PTX) as the model drug and encapsulate PTX into PCEC polymeric micelles and polymersomes.The physicochemical characterizations of the nanoparticles such as morphology,the size and distribution,zeta potential,drug loading content,and encapsulation efficiency were also performed.Our results showed that polymeric micelles or polymersomes from PCEC both displayed narrow size distributions and could achieve high drug loading efficiencies.In vitro cellular uptake results suggested that Nile Red loaded polymeric micelles or polymersomes displayed more internalization after 24 h incubation than those after 4 h incubation.These findings suggest that polymeric micelles and polymersomes based on PCL-b-PEG-b-PCL copolymers have great potential to effectively delivery hydrophobic drugs.Chunyan Hu Zhuo Chen Shengjie Wu Yanfeng Han Hai Wang Hongfan Sun Deling Kong Xigang Leng Chun Wang Linhua Zhang Dunwan Zhu 2017Chinese Chemical Letters2017,28,9:4
12Neuritin promotes angiogenesis through inhibition of DLL4/Notch signaling pathway显示文摘Neuritin is a member of the neurotrophic factor family,which plays an important role in the promotion and development of the nervous system.Neuritin is also involved in angiogenesis.Neuritin was recently found to be a negative regulatory factor of the Notch 1 signaling pathway.Notch signaling pathway is known as a regulatory pathway of angiogenesis.Thus,neuritin may play a role in angiogenesis through the Notch signaling pathway.In the present study,we investigated the expressions of neuritin and Notch signaling pathway factors in the pulmonary vascular tissue.The results showed that neuritin expression was increased in the paraneoplastic vascular tissue and decreased in the lung cancer vascular tissue.The neuritin expression was increased with the increase of vascular tissue density,and a negative correlation between neuritin expression and delta-like ligand 4(DLL4)was identified in vascular tissues of lung cancer.Overexpression of neuritin in human umbilical vein endothelial cells(HUVECs)inhibited the expressions of Notch signaling pathway-associated factors,including DLL4,NICD,and Hes-1,and promoted the migration and tubular formation of HUVECs.In conclusion,our results indicated that neuritin is involved in angiogenesis and may play a role in angiogenesis through the Notch signaling pathway.This study provides a theoretical basis for clinical anti-angiogenesis therapy.Li Yang Xuhui Wang Jiawei Sun Chunyan Liu Guoxiang Li Jingling Zhu Jin Huang 2021Acta Biochimica et Biophysica Sinica2021,53,6:4
13Enhanced production of seed oil with improved fatty acid composition by overexpressing NAD^(+)-dependent glycerol-3-phosphate dehydrogenase in soybean显示文摘There is growing interest in expanding the production of soybean oils(mainly triacylglycerol, or TAG) to meet rising feed demand and address global energy concerns. We report that a plastidlocalized glycerol-3-phosphate dehydrogenase(GPDH), encoded by GmGPDHp1 gene, catalyzes the formation of glycerol-3-phosphate(G3 P), an obligate substrate required for TAG biosynthesis.Overexpression of GmGPDHp1 increases soybean seed oil content with high levels of unsaturated fatty acids(FAs), especially oleic acid(C18:1), without detectably affecting growth or seed protein content or seed weight. Based on the lipidomic analyses, we found that the increase in G3 P content led to an elevated diacylglycerol(DAG) pool, in which the Kennedy pathwayderived DAG was mostly increased, followed by PC-derived DAG, thereby promoting the synthesis of TAG containing relatively high proportion of C18:1. The increased G3 P levels induced several transcriptional alterations of genes involved in the glycerolipid pathways. In particular, genes encoding the enzymes responsible for de novo glycerolipid synthesis were largely upregulated in the transgenic lines, in-line with the identified biochemical phenotype. 15Geniposide, the component of the Chinese herbal formula Tongluojiunao, protects amyloid-β peptide(1–42)-mediated death of hippocampal neurons via the non-classical estrogen signaling pathway显示文摘Tongluojiunao(TLJN) is an herbal medicine consisting of two main components, geniposide and ginsenoside Rg1. TLJN has been shown to protect primary cultured hippocampal neurons. However, its mechanism of action remains unclear. In the present study, primary cultured hippocampal neurons treated with Aβ1–42(10 μmol/L) significantly increased the release of lactate dehydrogenase, which was markedly reduced by TLJN(2 μL/mL), specifically by the component geniposide(26 μmol/L), but not ginsenoside Rg1(2.5 μmol/L). The estrogen receptor inhibitor, ICI182780(1 μmol/L), did not block TLJN- or geniposide-mediated decrease of lactate dehydrogenase under Aβ1–42-exposed conditions. However, the phosphatidyl inositol 3-kinase or mitogen-activated protein kinase pathway inhibitor, LY294002(50 μmol/L) or U0126(10 μmol/L), respectively blo cked the decrease of lactate dehydrogenase mediated by TLJN or geniposide. 16SHRIMP zircon dating and LA-ICPMS Hf analysis of early Precambrian rocks from drill holes into the basement beneath the Central Hebei Basin,North China Craton显示文摘The Central Hebei Basin(CHB) is one of the largest sedimentary basins in the North China Craton,extending in a northeast-southwest direction with an area of >350 km2.We carried out SHRIMP zircon dating,Hf-in-zircon isotopic analysis and a whole-rock geochemical study on igneous and metasedimentary rocks recovered from drill holes that penetrated into the basement of the CHB.Two samples of gneissic granodiorite(XG1-1) and gneissic quartz diorite(J48-1) have magmatic ages of 2500 and2496 Ma.respectively.Their zircons also record metamorphic ages of 2.41-2.51 and 2.5 Ga,respectively.Compared with the gneissic granodiorite,the gneissic quartz diorite has higher ∑REE contents and lower Eu/Eu* and(La/Yb)n values.Two metasedimentary samples(MG1,H5) mainly contain 2.5 Ga detrital zircons as well as late Paleoproterozoic metamorphic grains.The zircons of different origins haveεHf(2.5 Ga) values and Hf crustal model ages ranging from 0 to 5 and 2.7 to 2.9 Ga.respectively.Therefore,2.5 Ga magmatic and Paleoproterozoic metasedimentary rocks and late Neoarchean to early Paleoproterozoic and late Paleoproterozoic tectono-thermal events have been identified in the basement beneath the CHB.Based on regional comparisons,we conclude that the early Precambrian basement beneath the CHB is part of the North China Craton.Yusheng Wan Xianzheng Zhao Zejiu Wang Dunyi Liu Alfred Krner Chunyan Dong Hangqian Xie Yuansheng Geng Yuhai Zhang Runlong Fan Huiyi Sun 2014Geoscience Frontiers2014,5,4:3
17An Integrated Map of Soybean Physical Map and Genetic Map显示文摘Soybean is a major crop in the world, and it is a main source of plant proteins and oil. A lot of soybean genetic maps and physical maps have been constructed, but there are no integrated map between soybean physical map and genetic map. In this study, soybean genome sequence data, released by JGI (US Department of Energy's Joint Genome Institute), had been downloaded. With the software Blast 2.2.16, a total of 161 super sequences were mapped on the soybean public genetic map to construct an integrated map. The length of these super sequences accounted for 73.08% of all the genome sequence. 18Superhydrophobic fluoride conversion coating on bioresorbable magnesium alloy——fabrication,characterization,degradation and cytocompatibility with BMSCs显示文摘A micro-nano structure CaF_(2)chemical conversion layer was prepared on fluoride-treated AZ31 alloy,then the composite fluoride conversion film(CaF_(2)/MgF_(2))was modified by stearic acid(SA)and fabricated a superhydrophobic surface.The fluoride-treated magnesium,fluoride conversion film and superhydrophobic coating were characterized by SEM,EDS,XRD and FTIR.The properties of coatings1 adhesion and corrosion resistance were evaluated via tape test and electrochemical measurement.The cytocompatibility of the MgF_(2),CaF_(2)and superhydrophobic CaF_(2)/SA surface was investigated with bone marrow-derived mesenchymal stem cells(BMSCs)by direct culture for 24 h.The results showed that the superhydrophobic fluoride conversion coating composed of inner MgF_(2)layer and the outer CaF_(2)/SA composite layer had an average water contact angle of 152°.SA infiltrated into the micro-nano structure CaF_(2)layer and formed a strong adhesion with CaF_(2)layer.Furthermore,the super-hydrophobic coating showed higher barrier properties and corrosion resistance compared with the fluoride conversion film and fluoride-treated AZ31 alloy.The BMSC adhesion test results demonstrated MgF_(2)CaF_(2)and CaF_(2)/SA coatings were all nontoxic to BMSC.At the condition of in direct contact with cells,MgF_(2)showed higher cell density and enhanced the BMSCs proliferation,while CaF_(2)and CaF_(2)/SA coating showed no statistically difference in cell density compared with glass reference but the CaF_(2)and CaF_(2)/SA coating were not conducive to BMSCs adhesion.Chunyan Zhanga Shiyu Zhang Dongwei Sun Jiajia Lin Fancheng Meng Huinan Liu 2021Journal of Magnesium and Alloys2021,9,4:3
19Key technologies and practice for gas field storage facility construction of complex geological conditions in China显示文摘In view of complex geological characteristics and alternating loading conditions associated with cyclic large amount of gas injection and withdrawal in underground gas storage(UGS) of China, a series of key gas storage construction technologies were established, mainly including UGS site selection and evaluation, key index design, well drilling and completion, surface engineering and operational risk warning and assessment, etc. The effect of field application was discussed and summarized. Firstly, trap dynamic sealing capacity evaluation technology for conversion of UGS from the fault depleted or partially depleted gas reservoirs. A key index design method mainly based on the effective gas storage capacity design for water flooded heterogeneous gas reservoirs was proposed. To effectively guide the engineering construction of UGS, the safe well drilling, high quality cementing and high pressure and large flow surface injection and production engineering optimization suitable for long-term alternate loading condition and ultra-deep and ultra-low temperature formation were developed. The core surface equipment like high pressure gas injection compressor can be manufactured by our own. Last, the full-system operational risk warning and assessment technology for UGS was set up. The above 5 key technologies have been utilized in site selection, development scheme design, engineering construction and annual operations of 6 UGS groups, e.g. the Hutubi UGS in Xinjiang. 20Effect of Traditional Chinese Medicine plus narrow-band medium-wave ultraviolet B radiation on moderate-to-severe psoriasis vulgaris in a case series显示文摘OBJECTIVE: To investigate the efficacy and safety of Traditional Chinese Medicine (TCM) therapy combined with ultraviolet B light therapy in the treatment of moderate-to-severe psoriasis. METHODS: Patients with moderate-to-severe psoriasis (skin lesion area > 10% of the body surface area) for 2 consecutive years were treated with TCM (oral and external use of herbal medicines, acupuncture, and herbal bathing) and narrow-band medium-wave ultraviolet B light treatment for 12 weeks. The treatment effect was evaluated based on the Psoriasis Area Severity Index (PASI), the achievement of a 50% reduction in the PASI (PASI50), the achievement of a 75% reduction in the PASI (PASI75), pruritus score, Dermatology Life Quality Index, and safety. RESULTS: A total of 95 outpatients were enrolled, and 92 subjects (96.8%) completed the 12-week treatment course. At baseline, the average proportion of the body surface area covered by skin lesions was 12.4%, and the average PASI was 17.7. All patients had previously been treated with conventional medicine (89.1% of patients received ultraviolet light treatment, 50.0% received glucocorticoids, and 21.7% received acitretin). After the 12-week treatment course, 22 patients (23.9%) achieved PASI75, and 43 (46.7%) achieved PASI50. The post-treatment pruritus score and Dermatology Life Quality Index of all treated patients were significantly lower than the respective baseline values (P < 0.0001).
