| 2 | Vitamin B3 inhibits apoptosis and promotes autophagy of isletβcells under high glucose stress显示文摘Background:Hyperglycemia is a typical symptom of diabetes.High glucose induces apoptosis of isletβcells.While autophagy functions in cytoprotection and autophagic cell death.The interaction between autophagy and apoptosis is important in the modulation of the function of isletβcells.Vitamin B3 can induce autophagy and inhibit isletβapoptosis.Method:The mechanism of vitamin B3-mediated protective effect on the function of isletβcells was explored by the method of western blot,immunofluorescence and flow cytometry.Results:In the present study,high glucose stress increased the apoptosis rate,while vitamin B3 reduced the apoptosis rate.The effect of vitamin B3 on autophagy flux under normal and high glucose stress was also investigated.Vitamin B3 increased the number of autophagosomes and increased the light chain(LC)3-II/LC3-I ratio.In contrast,vitamin B3 decreased sequestosome 1(SQSTM1)/p62 protein expression and inhibited the phosphorylation of mammalian ribosomal protein S6 kinaseβ-1(p70S6K/S6K1),which was a substrate of mammalian target of rapamycin(mTOR)under normal and high glucose stress.To further verify the protective effect of vitamin B3 on apoptosis,we treated isletβcell RIN-m5F with autophagy inhibitor 3-methyladenine(3-MA).Vitamin B3 decreased the apoptosis rate under high glucose stress,while the inhibition of apoptosis by vitamin B3 was blocked after adding 3-MA.Conclusion:Our data suggested that vitamin B3 reduced the apoptosis rate ofβcells,possibly through inducing autophagy under high glucose stress. | YU ZHANG XI’AN ZHOU CHUNYAN ZHANG DENGNI LAI DONGBO LIU YANYANG WU | 2023 | BIOCELL2023,47,4: | 1 |
| 3 | 干扰COX-2基因表达对人肺腺癌A2细胞体外恶性增殖的影响(英文)显示文摘Objective:We explored the interfering effect of COX-2 gene expression and the influence on the malignant proliferation of A2 cells by RNAi after quenching COX-2 in vitro. Methods:COX-2 was selected as the subject. Three COX-2 siRNA expression vectors with human U6 promoter were constructed and three vectors and the vacant vector (pEGFP) were transfected into A2 cells with lipofectamine respectively. The cell strains transfected were constructed. The change of COX-2 expression levels was examined by Western blot and RT-PCR. The effects on the proliferation of A2 cells after silencing COX-2 were studied by cell growth curve and clonogenic assay in vitro. Results: The three siRNA and U6 promoter cloned into pEGFP plasmid were validated by PCR, restriction endonucleases identification, DNA sequencing and BLAST alignment. The cell strains transfected were named as A2-3, A2-7, A2-10 and A2-p respectively. Green fluorescence was seen in A2-p cells and not in A2-3, A2-7 and A2-10 cells in 24, 48 and 72 h after transfected. The results of RT-PCR and Western blot showed the three siRNA expression vectors produced effects and the expression of COX-2 was inhibited in different extent. In contrast to A2 cells, the levels of COX-2 mRNA of A2-3, A2-7 and A2-10 cells reduced 15.6%, 20.4% and 64.2% respectively; the levels of COX-2 protein of A2-3, A2-7 and A2-10 cells reduced 23.7%, 36.7% and 60.2% respectively. The results of cell growth curve and clonogenic assay showed the growth of A2-10 cell slowed and the colonial formation rate reduced but the growth of A2-3 and A2-7 cells had not obvious changes in contrast to the controls (A2 and A2-p). | 4 | Floating Escherichia coli by Expressing Cyanobacterial Gas Vesicle Genes显示文摘Gas vesicles are hollow, air-filled polyprotein structures that provide the buoyancy to cells. They are found in a variety of prokaryotes. In this study, we isolated a partial gas vesicle protein gene cluster containing gvp A and gvp C20Ψ from Planktothrix rubescens, and inserted it into an expression vector and expressed it in E. coli. The gas vesicle was developed in bacterial cells, which made bacterial cells to float on medium surface. We also amplified gvp A and gvp C20Ψ separately and synthesized an artificial operon by fusing these two genes with the standardized gene expression controlling elements of E. coli. The artificial operon was expressed in E. coli, forming gas vesicles and floating bacteria cells. Our findings verified that the whole set of genes and the overall structure of gas vesicle gene cluster are not necessary for developing gas vesicles in bacteria cells. Two genes, gvp A and gvp C20Ψ, of the gas vesicle gene cluster are sufficient for synthesizing an artificial operon that can develop gas vesicles in bacteria cells. | 7 | Permanent Magnet Temperature Estimation for PMSMs Using Virtual Position-offset Injection显示文摘This paper proposes a virtual position-offset injection based permanent magnet temperature estimation approach for permanent magnet synchronous machines(PMSMs). The concept of virtual position-offset injection is mathematically transforming the machine model to a virtual frame with a position-offset. The virtual frame temperature estimation model is derived to calculate the permanent magnet temperature(PMT) directly from the measurements with computation efficiency. The estimation model involves a combined inductance term, which can simplify the establishment of saturation compensation model with less measurements. Moreover, resistance and inverter distorted terms are cancelled in the estimation model, which can improve the robustness to the winding temperature rise and inverter distortion. The proposed approach can achieve simplified computation in temperature estimation and reduced memory usage in saturation compensation. While existing model-based approaches could be affected by either the need of resistance and inverter information or complex saturation compensation. | 9 | Infection Characteristics and Interaction of Three Species of Penicillium in the Postharvest Storage of‘Gonggan'Mandarin(Citrus reticulata)显示文摘[Objectives]This study was conducted to understand the infection characteristics of Penicillium on‘Gonggan’mandarin(Citrus reticulata)and its effects on postharvest storage quality.[Methods]Nine inoculation treatments were designed and tested for the three identified Penicillium species,namely:three single inoculation experiments,four compound inoculation experiments,and two controls without inoculation.After treatment,the fruit was put into plastic film fresh-keeping bags and stored at a natural room temperature.The determined and analyzed indexes included fruit disease incidence,disease index,disease symptoms,disease spot expansion speed and weight loss rate,soluble solid content and organic acid content in different inoculation treatments.[Results]All‘Gonggan’fruit inoculated with Penicillium were diseased 2 d after inoculation,and completely rotted 14 d later.Of the three Penicillium species isolated and identified,Penicillium digitorum and its combinations were the most harmful to‘Gonggan’fruit.The hazard of Penicillium italicum was equal to or even weaker than that of Penicillium expansum.Eventually,P.digitorum dominated when coexisting on the same fruit.Different species of Penicillium obviously showed competition and antagonism in the process of growth and infection,especially for P.digitorum which was stronger,but other two species of Penicillium also had obvious self-protection ability.Low-temperature storage played a crucial role in the preservation of‘Gonggan’fruit,because it could significantly inhibit Penicillium infestation,and suppress fruit weight loss and the reduction of the contents of soluble solids and organic acids.[Conclusions]The infection characteristics and interaction relationship of Penicillium in the postharvest storage of‘Gonggan’fruit were revealed. | Shaoping ZHANG Chengxiang XU Jinyan LAI Yuchan PENG Chunyan ZHONG Minzhi LIANG | 2022 | Agricultural Biotechnology2022,11,3: | 0 |
| 10 | In situ self-assembly assisted synthesis of N-doped mesoporous hierarchical carbon aerogels-wrapped Li_(2)ZnTi_(3)O_(8) composite for highrate lithium ion batteries显示文摘Reinforcing electronic and ionic conductivity is very important to realize the high-rate Li_(2)ZnTi_(3)O_(8)(LZTO)anode for Lithium-ion batteries(LIBs).Here,we reported a synthetic strategy toward in situ growth of homodispersed Li_(2)ZnTi_(3)O_(8)on ultralight 3D carbon aerogels(CAs)via facile heat treatment and activation process taking LiOH as activating agent.Such an optimized composite obtains a specific surface area of 154.427 m^(2) g^(-1) and a high pore volume.The strong interaction between the LZTO and N-doped CAs,a highly electrical conductivity and multidimensional ion transport channels of the hybrid result in a high capacity,outstanding high-rate performance and long cycle life.As a result,the NPC-LZTO electrode(NPC-LZTO-B)delivers a commendable reversible capacity at high-rate(e.g.162.4 mAh g^(-1) at 5 A g^(-1)(22 C)and 127.3 mAh g^(-1) at 10 A g^(-1))and durable long-term performance(capacity retention of 73% after 4500 cycles at 5 A g^(-1)).Moreover,the kinetic analysis confirms that the feature of pseudocapacitance boosts lithium-ion storage performance at high rate.Porous carbon aerogels frame constructed LZTO nanocomposite electrodes provide a facile way to design high durable LIBs anode. | Jingjing Wang Wenhan Zhang Jiawei Li Bin Wang Chao Xu Chunyan Lai | 2021 | Journal of Materiomics2021,7,5: | 0 |

| 12 | 同一靶点对不同Cyclooxygenase-2蛋白表达水平肺癌细胞恶性增殖的影响(英文)显示文摘Objective: The aim of the study was to explore the effects of the same target (si-10) on lung cancer cells with different expression levels of cyclooxygenase-2 (COX-2) protein by RNAi and malignant proliferation of these cells. Methods: COX-2 was selected as the target and one siRNA expression vector with the best effect was selected and thought as the subject from three COX-2 siRNA expression vectors with human U6 promoter. The siRNA expression vector (psi-10) and the vacant vector (pEGFP) were transfected into these cells with different COX-2 expression states (801D, A549 and LTEP-A2) with lipofectamine respectively and the transfected cell strains were constructed. The change of COX-2 expression levels was examined by Western blot and RT-PCR. The effects on the proliferation of lung cancer cells were studied by cell growth curve and clonogenic assay. Results: The siRNA and U6 promoter were validated by PCR, restriction endonucleases identification and DNA sequencing and BLAST alignment and cloned into the pEGFP vector. The cell strains transfected that 801D was used as maternal line were named as 801D-p and 801D-10 respectively. The cell strains transfected that A549 was used as maternal line were named as A549-p and A549-10 respectively. The cell strains transfected that LTEP-A2 was used as maternal line were named as LTEP-A2-p and LTEP-A2-10 respectively. These cells transfected pEGFP (801D-p, A549-p and LTEP-A2-p) had the expression of GFP and 801D-10, A549-10 and LTEP-A2-10 cells had not in 24, 48 and 72 hours after transfected. The results of RT-PCR and Western blot showed the siRNA expression vector produced marked effects in two cells (A549 and LTEP-A2) expressing COX-2 and the expression of COX-2 was inhibited. But the inhibited effects were differ- ent and the expression of COX-2 was more inhibited obviously in LTEP-A2 cells than in A549 cells though the expression of COX-2 was also inhibited obviously in A549 cells. In contract to their maternal line, the levels of COX-2 mRNA of LTEP-A2-10 and A549-10 cells reduced 64.2% and 61.2% respectively; the levels of COX-2 protein reduced 60.2% and 56.2% respectively. But the levels of COX-2 mRNA and protein had not change in 801D cells not expressing COX-2. The results of cell growth curve and clonogenic assay showed the growth of LTEP-A2-10 cells slowed and the clonal formation rate reduced and the size of the colonies became small; the growth of A549-10 cells showed slow and more obviously in the cell growth curve especially. But the growth of 801D-10 cells had not obvious change. | 13 | Transcriptome Profiling Reveals Candidate Genes Involved in Stem Swelling of Tumorous Stem Mustard显示文摘Tumorous stem mustard is well known for its swollen stem from which pickled“Fuling Mustard”is made.The molecular mechanisms governing the formation of the modified swollen stems are still poorly understood.This paper aims to identify candidate genes involved in the developmental regulation of the swollen stems.We sought to map previously published transcriptome datasets for Brassica juncea,including those derived from swollen stems at four different developmental stages and a mutant variety without swollen stems.Using pairwise comparisons of the five datasets,we identified 31368 differentially expressed genes(DEGs).A total of 55 DEGs related to plant hormone signal transduction and 259 continuously up-or downregulated transcription factors were identified during stem development using Gene Ontology(GO)analysis.Quantitative real-time PCR(qRT-PCR)results showed that the expressions of 12 important candidate DEGs were consistent with RNA-seq results.Our study provided digital gene expression profiling and a dynamic view of the swollen stem development process.Furthermore,we identified candidate genes for further studies on mechanisms of modified stem development in non-model species. | Lina Du Changman Li Rui Su Chunyan Tan Biao Lai | 2020 | Horticultural Plant Journal2020,6,3: | 0 |
