1Influence of ginsenoside Rg1, a panaxatriol saponin from Panax notoginseng, on renal fibrosis in rats with unilateral ureteral obstruction显示文摘Total saponins of Panax notoginseng (PNS) have been shown to ameliorate renal interstitial fibrosis. Ginsenoside Rg1, a panaxatriol saponin, is one of the major active molecules from PNS. The present study was undertaken to investigate the effect of ginsenoside Rg1 on renal fibrosis in rats with unilateral ureteral obstruction (UUO). The rats were randomly divided into 3 groups: sham-operation (n=15), UUO (n=15) and UUO with ginsenoside Rg1 treatment (n=15, 50 mg per kg body weight, intraperito- neally (i.p.) injected). The rats were sacrificed on Days 7 and 14 after the surgery. Histological examination demonstrated that ginsenoside Rg1 significantly inhibited interstitial fibrosis including tubular injury as well as collagen deposition. α-smooth muscle actin (α-SMA) and E-cadherin are two markers of tubular epithelial-myofibroblast transition (TEMT). Interestingly, ginsenoside Rg1 notably decreased α-SMA expression and simultaneously enhanced E-cadherin expression. The messenger RNA (mRNA) of transforming growth factor-β1 (TGF-β1), a key mediator to regulate TEMT, in the obstructed kidney increased dra- matically, but was found to decrease significantly after administration of ginsenoside Rg1. Further study showed that ginsenoside Rg1 considerably decreased the levels of both active TGF-β1 and phosphorylated Smad2 (pSmad2). Moreover, ginsenoside Rg1 substantially suppressed the expression of thrombospondin-1 (TSP-1), a cytokine which can promote the transcription of TGF-β1 mRNA and the activation of latent TGF-β1. These results suggest that ginsenoside Rg1 inhibits renal interstitial fibrosis in rats with UUO. 2Astragalus mongholicus ameliorates renal fibrosis by modulating HGF and TGF-β in rats with unilateral ureteral obstruction显示文摘Astragalus mongholicus(AM) derived from the dry root of Astragalus membranaceus Bge.var.mongolicus(Bge.) Hsiao is a widely used traditional Chinese medicine.The present study investigated the potential role of AM on renal fibrosis on a rat model of unilateral ureteral obstruction(UUO).We divided 48 Sprague-Dawley rats randomly into 4 groups:sham-operated group(Sham),untreated UUO group,AM-treated(10 g/(kg·d)) UUO group,and losartan-treated(20 mg/(kg·d)) UUO group as positive control.Haematoxylin & eosin(HE) and Masson staining were used to study the dynamic histological changes of the kidneys 7 and 14 d after operation.The expressions of fibronectin(FN),type I collagen(colI),hepatocyte growth factor(HGF),transforming growth factor-β1(TGF-β1),and α-smooth muscle actin(α-SMA) were analyzed by real-time polymerase chain reaction(PCR),immunohistochemistry staining,and Western blot.Results show that,similar to losartan,AM alleviated the renal damage and decreased the deposition of FN and colI from UUO by reducing the expressions of TGF-β1 and α-SMA(P<0.05),whereas HGF increased greatly with AM treatment(P<0.05).Our findings reveal that AM could retard the progression of renal fibrosis.The renoprotective effect of AM might be related to inhibition of myofibroblast activation,inducing of HGF and reducing of TGF-β1 expression.Chuan ZUO Xi-sheng XIE Hong-yu QIU Yao DENG Da ZHU Jun-ming FAN 2009Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2009,10,5:15
3Inhibition of pacemaker activity in interstitial cells of Cajal by LPS via NF-κB and MAP kinase显示文摘AIM:To investigate lipopolysaccharide(LPS) related signal transduction in interstitial cells of Cajal(ICCs) from mouse small intestine.METHODS:For this study,primary culture of ICCs was prepared from the small intestine of the mouse.LPS was treated to the cells prior to measurement of the membrane currents by using whole-cell patch clamp technique.Immunocytochemistry was used to examine the expression of the proteins in ICCs.RESULTS:LPS suppressed the pacemaker currents of ICCs and this could be blocked by AH6809,a prostaglandin E2-EP2 receptor antagonist or NG-Nitro-Larginine Methyl Ester,an inhibitor of nitric oxide(NO) synthase.Toll-like receptor 4,inducible NO synthase or cyclooxygenase-2 immunoreactivity by specific antibodies was detected on ICCs.Catalase(antioxidant agent) had no action on LPS-induced action in ICCs.LPS actions were blocked by nuclear factor kB(NF-kB) inhibitor,actinomycin D(a gene transcription inhibitor),PD 98059(a p42/44 mitogen-activated protein kinases inhibitor) or SB 203580 [a p38 mitogen-activated protein kinases(MAPK) inhibitor].SB 203580 also blocked the prostaglandin E2-induced action on pacemaker currents in ICCs but not NO.CONCLUSION:LPS inhibit the pacemaker currents in ICCs via prostaglandin E2-and NO-dependent mechanism through toll-like receptor 4 and suggest that MAPK and NF-kB are implicated in these actions.Dong Chuan Zuo Seok Choi Pawan Kumar Shahi Man Yoo Kim Chan Guk Park Young Dae Kim Jun Lee In Yeoup Chang Insuk So Jae Yeoul Jun 2013World Journal of Gastroenterology2013,19,8:9
4Knitted self-powered sensing textiles for machine learning-assisted sitting posture monitoring and correction显示文摘With increasing work pressure in modern society,prolonged sedentary positions with poor sitting postures can cause physical and psychological problems,including obesity,muscular disorders,and myopia.In this paper,we present a self-powered sitting position monitoring vest(SPMV)based on triboelectric nanogenerators(TENGs)to achieve accurate real-time posture recognition through an integrated machine learning algorithm.The SPMV achieves high sensitivity(0.16 mV/Pa),favorable stretchability(10%),good stability(12,000 cycles),and machine washability(10 h)by employing knitted double threads interlaced with conductive fiber and nylon yarn.Utilizing a knitted structure and sensor arrays that are stitched into different parts of the clothing,the SPMV offers a non-invasive method of recognizing different sitting postures,providing feedback,and warning users while enhancing long-term wearing comfortability.It achieves a posture recognition accuracy of 96.6%using the random forest classifier,which is higher than the logistic regression(95.5%)and decision tree(94.3%)classifiers.The TENG-based SPMV offers a reliable solution in the healthcare system for non-invasive and long-term monitoring,promoting the development of triboelectric-based wearable electronics.Yang Jiang Jie An Fei Liang Guoyu Zuo Jia Yi Chuan Ning Hong Zhang Kai Dong Zhong Lin Wang 2022Nano Research2022,15,9:4

subspecies,three new combinations and seven reference specimens.Awide geographic and taxonomic range of fungal taxa are detailed.In the Ascomycota the new genera 13A Proactive Selection Method for Dynamic Access Points Grouping in User-centric UDN显示文摘Network densification is a promising solution to fulfill network capacity requirement and transmission rate for beyond 5G and 6G wireless communications.Ultra-dense network(UDN)integrates heterogeneous network resources and coordinates technologies on quality of service controlling,to provide users with flexible service.However,dense deployment reduces coverage radius of the cell,resulting in an increase on handover frequency,which makes a serious impact on service continuity.In this paper,we propose a proactive selection method for dynamic access points grouping(DAPGing)in accordance with“user-centric”philosophy,which selects target Access Points(AP)and reduces handover times to ensure communication continuity.This method includes two criteria:1)the user’s sojourn time,which is determined by analyzing the AP coverage area;2)neighbor relationship between APs,which is determined by coverage area and signal strength characteristics between neighboring APs.Therefore,candidate APs become the proactive selected ones to update the AP group.Stochastic geometry is used to build system model and performance metrics are analyzed,including AP group coverage probability and average update frequency.Experimental analysis shows that the proposed proactive selection method brings similar coverage probability to traditional handover method,while average update frequency is reduced more than 20%selection criteria.Bo Hu Fangxia Zuo Chuan’anWang Shanzhi Chen 2021China Communications2021,18,4:0
14Assay development for determination of DZ2002, a new reversible SAHH inhibitor, and its acid metabolite DZA in blood and application to rat pharmacokinetic study显示文摘Methyl(S)-4-(6-amino-9 H-purin-9-yl)-2-hydroxybutanoate(DZ2002) is a potent reversible inhibitor of S-adenosyl-L-homocysteine hydrolase(SAHH). Due to its ester structure, DZ2002 is rapidly hydrolyzed in rat blood to 4-(6-amino-9 H-purin-9-yl)-2-hydroxybutyric acid(DZA) during and after blood sampling from rats; this hampers accurate determination of the circulating DZ2002 and its acid metabolite DZA in rats. To this end, a method for determining the blood concentrations of DZ2002 and DZA in rats was developed by using methanol to immediately deactivate blood carboxylesterases during sampling. The newly developed bioanalytical assay possessed favorable accuracy and precision with lower limit of quantification of 31 nM for DZ2002 and DZA. This validated assay was applied to a rat pharmacokinetic study of DZ2002. After oral administration, DZ2002 was found to be extensively converted into DZA. The level of systemic exposure to DZ2002 was significantly lower than that of DZA. The apparent oral bioavailability of DZ2002 was 90%–159%. The mean terminal half-lives of DZ2002 and DZA were 0.3–0.9 and 1.3–5.1 h, respectively. 16Supplementation with L-Carnitine Rescues Sperm Epigenetic Changes in Asthenospermic Male Semen with Altered Acetyl-L-Carnitine Levels显示文摘Objective:To investigate the relationship between the concentration of L-carnitine in semen and sperm parameters and investigate the epigenetic profile in sperm cell after L-carnitine usage.Methods:From February 2017 to February 2018,46 semen samples from asthenospermic males and 41 semen samples from healthy donors were acquired.Motility parameters were assessed using computer-assisted sperm analysis(CASA,n=78)and the DNA fragmentation index(DFI)was evaluated through flow cytometry(n=86),%DFI=%cells outside main population.Other oxidative stress markers,such as reactive oxygen species(ROS)levels(n=86)and the mitochondria DNA copy numbers,were detected(n=78).The concentration of L-carnitine and acetyl-L-carnitine was detected(n=82),and methylation was analyzed(n=30).After that,we collected 13 fresh semen samples from asthenospermic males and 23 fresh semen samples from healthy donors.These samples were used in a freeze-thaw model that was used to determine whether adding L-carnitine could change sperm progressive motility(n=23),apoptosis index(n=9),and methylation analysis(n=7).In total,we have done 13 asthenospermia samples for Western blot,and except for the poor Western result,we analyzed 6 samples for H3K9ac detection,7 samples for H3K9m3 and H3K27m3 detection,and immunofluorescence(n=3).Finally,we had recruited 30 volunteers,and they were given oral administration of L-carnitine for 3 months and then collected semen samples at different time points for methylation analysis.Results:The concentration of acetyl-L-carnitine is negatively correlated with the%DFI value(r^2=0.1090;P=0.0026),and the concentration of acetyl-L-carnitine is positively correlated with sperm forward motility(r^2=0.0543;P=0.0458)and ROS(r^2=0.1854;P<0.0001),and the acetyl-L-carnitine level is negatively correlated with%DFI in asthenospermia(r^2=0.1701;P=0.0066),and the level of acetyl-L-carnitine in asthenospermic semen is significantly lower than the normal group(P=0.0419).In addition,this study indicates that adding L-carnitine significantly improved sperm motility(P=0.0325)and reduced sperm apoptosis(P=0.0032).Importantly,Western blotting(P=0.0429)and immunofluorescence staining results showed that the addition of L-carnitine reduced H3K9Me3 methylation level in sperm,respectively.Furthermore,semen samples from asthenospermic patients had reduced methylation levels in a specific region(16^thP=0.0003;17^thP=0.0016)of the brain-derived neurotrophic factor(BDNF)promoter.The 16^th methylation decreased with age(r^2=0.1564;P=0.0306),and the 17^th methylation was decreased after treatment with L-carnitine for 28 days(P=0.0341).Conclusion:L-carnitine can reduce the%DFI and also affect the methylation of the histone modification marker in sperm as a possible epigenetic regulator.Xiao-Hui Jiang Chuan Jiang Lin Yu Xiao-Liang Li Tao Zuo Pei-Fei Gu Fu-Ping Li Wen-Ming Xu 2020Reproductive and Developmental Medicine2020,4,3:0
