1Chitosan-DNA microparticles as mucosal delivery system: synthesis, characterization and release in vitro显示文摘Background Mucosal immunity is important to defense against dental caries. To enhance mucosal immunity, a DNA vaccine mucosal delivery system was prepared by encapsulating anticaries DNA vaccine (plasmid pGJA-P/VAX) in chitosan under optimal conditions and the characteristics of the microparticles was investigated. Furthermore, the release properties and protective action of microparticles for plasmid were studied in vitro.Methods Plasmid loaded chitosan microparticles were prepared by complex coacervation. Three factors, concentration of DNA, sodium sulfate, and the chitosan/DNA ratios in complexes [better expressed as N/P ratio: the number of poly nitrogen (N) per DNA phosphate (P)] influencing preparation were optimized by orthogonal test. The characteristics of microparticles were evaluated by scanning electron microscopy (SEM) and confocal laser scanning microscopy (CLSM). DNA release rate of microparticles in similar gastro fluid (SGF) or similar intestinal fluid (SIF) at 37℃ was determined by ultraviolet spectrophotometry.Results High encapsulation efficiency (96.8%) was obtained with chitosan microparticles made under optimal conditions of 50 mmol/L Na2SO4, 200 μg/ml DNA and N/P ratio of 4. The size of particles was about 4 to 6 μm. The encapsulation process did not destroy the integrity of DNA. When incubated with SIL, after a release of about 10% in the first 60 minutes, no further DNA was released during the following 180 minutes. When incubated with SGL, the microparticles released a small burst (about 11%) in the first 60 minutes, and then slowly released at a constant, but different rate.Conclusions These chitosan microparticles showed suitable characteristics in vitro for mucosal vaccination and are therefore a promising carrier system for DNA vaccine mucosal delivery.LIYu-hong FANMin-wen BIANZhuan CHENZhi ZHANGQi YANGHai-rui 2005Chinese Medical Journal2005,,11:13
2Deletion analysis of oligomycin PKS genes (olmA) in Streptomyces avermitilis显示文摘Gene deletion vector pXL05(pKC1139∷△olmA1 +△olmA4) was used to disrupt oligomycin PKS en-coding genes (olmA) in Streptomyces avermitilis CZ8-73, the producer of anthelmintic avermectins B and the cell growth inhibitor oligomycin. olmA gene cluster in the chromosome was displaced by deletion allele on the plasmid via double crossover. Four of disruptants were confirmed by Southern blotting. Shaking flask experiments and HPLC analyses showed that the four mutants no longer produced the toxic oligomycin, but only made four components of avermectins B, which were avermectin B1a, B1b, B2a, B2b. The yields of avermectins B in these mutants were separately equal to those in CZ8-73. This revealed that olmA genes deletion did not affect the biosynthesis of avermectins. 3Automatic analytical approach for the determination of 12 illicit drugs and nicotine metabolites in wastewater using on-line SPE-UHPLC-MS/MS显示文摘In this study,we developed a novel on-line solid phase extraction(SPE)-ultra-high-performance liquid chromatography tandem mass spectrometry(UHPLC-MS/MS)-based analytical method for simultaneously quantifying 12 illicit drugs and metabolites(methamphetamine,amphetamine,morphine,codeine,6-monoacetylmorphine,benzoylecgonine,3,4-methylenedioxymethamphetamine,3,4-methylenedioxyamphetamine,cocaine,ketamine,norketamine,and methcathinone)and cotinine(COT)in wastewater samples.The analysis was performed by loading 2 m L of the sample onto an Oasis hydrophilic-lipophilic balance cartridge and using a cleanup step(5%methanol)to eliminate interference with a total run time of 13 min.The isotope-labeled internal standard method was used to quantify the target substances and correct for unavoidable losses and matrix effects during the on-line SPE process.Typical analytical characteristics used for method validation were sensitivity,linearity,precision,repeatability,recovery,and matrix effects.The limit of detection(LOD)and limit of quantification(LOQ)of each target were set at 0.20 ng/L and 0.50 ng/L,respectively.The linearity was between 0.5 ng/L and250 ng/L,except for that of COT.The intra-and inter-day precisions were<10.45%and 25.64%,respectively,and the relative recovery ranged from 83.74%to 162.26%.The method was used to analyze various wastewater samples from 33 cities in China,and the results were compared with the experimental results of identical samples analyzed using off-line SPE.The difference rate was between 19.91%and-20.44%,and the error range could be considered acceptable.These findings showed that on-line SPE is a suitable alternative to off-line SPE for the analysis of illicit drugs in samples.Jingyuan Wang Likai Qia Chenzhi Hou Tingting Zhang Mengyi Chen Haitao Meng Mengxiang Su Hui Xu Zhendong Hua Youmei Wang Bin Di 2021Journal of Pharmaceutical Analysis2021,11,6:5
4Hybrid waveguide scheme for silicon-based quantum photonic circuits with quantum light sources显示文摘We propose a hybrid silicon waveguide scheme to avoid the impact of noise photons induced by pump lights in application scenarios of quantum photonic circuits with quantum light sources.The scheme is composed of strip waveguide and shallow-ridge waveguide structures.It utilizes the difference of biphoton spectra generated by spontaneous four-wave mixing(SFWM)in these two waveguides.By proper pumping setting and signal/idler wavelength selection,the generation of desired photon pairs is confined in the strip waveguide.The impact of noise photons generated by SFWM in the shallow-ridge waveguide can be avoided.Hence,the shallowridge waveguide could be used to realize various linear operation devices for pump light and quantum state manipulations.The feasibility of this scheme is verified by theoretical analysis and a primary experiment.Two applications are proposed and analyzed,showing its great potential in silicon-based quantum photonic circuits.LINGJIE YU CHENZHI YUAN RENDUO QI YIDONG HUANG WEI ZHANG 2020Photonics Research2020,8,3:3
5Different Mortality Effects of Extreme Temperature Stress in Three Large City Clusters of Northern and Southern China显示文摘Extreme temperature events have affected Chinese city residents more frequently and intensively since the early 2000 s, but few studies have identified the impacts of extreme temperature on mortality in different city clusters of China. This study first used a distributed lag,nonlinear model to estimate the county/district-specific effects of extreme temperature on nonaccidental and cardiovascular mortality. The authors then applied a multivariate meta-analysis to pool the estimated effects in order to derive regional temperature–mortality relationship in three large city clusters—the Beijing-Tianjin-Hebei(BTH)region, the Yangtze River Delta(YRD), and the Pearl River Delta(PRD), which represent northern and southern regions. With 0–3 days' lag, the strongest heat-related mortality effect was observed in the BTH region(with relative risk(RR) of 1.29; 95% confidence interval(CI):1.13–1.47), followed by the YRD(RR = 1.25; 95% CI:1.13–1.35) and the PRD(RR = 1.14; 95% CI: 1.01–1.28)areas. With 0–21 days' lag, the cold effect was pronounced in all city clusters, with the highest extreme cold-related mortality risk found in the PRD area(RR = 2.27; 95% CI:1.63–3.16), followed by the YRD area(RR = 1.85; 95%CI: 1.56–2.20) and BTH region(RR = 1.33; 95% CI:0.96–1.83). People in the southern regions tended to be more vulnerable to cold stress, but the northern population was more sensitive to heat stress. 6Experimental observation of coherent interaction between laser and erbium ions ensemble doped in fiber at sub 10 mK显示文摘Rare-earth ions doped in solid-state materials are considered promising candidates for quantum information applications,especially for photonic quantum memory.Among them,erbium ions doped in an optical fiber have attracted a lot of attentions due to their ability to provide efficient photon-atom interaction on a telecom-C band compatible transition.The coherent photon-atom interaction,which is crucial for quantum memory,has not yet been investigated for erbium ions doped in fiber at the temperature of sub 10 m K with the magnetic environment.In this paper,we experimentally observe optical nutation,which results from the coherent interaction between laser and erbium ions ensemble,in a piece of 9.5-m-long fiber with the erbium concentration of 200 ppm.We also extract the transition dipole moment from the results of optical nutation and further investigate its dependence on laser wavelength and magnetic field.A transition dipole moment of(3.424±0.019)×10^-32 C·m is obtained at the wavelength of 1537 nm and magnetic field of 0.2 T.Our results could pave the way for realizing solid-state quantum networks at telecom-C band.Qi XI Shihai WEI Chenzhi YUAN Xueying ZHANG You WANG Haizhi SONG Guangwei DENG Bo JING Daniel OBLAK Qiang ZHOU 2020Science China(Information Sciences)2020,63,8:3
7The circFASN/miR-33a pathway participates in tacrolimusinduced dysregulation of hepatic triglyceride homeostasis显示文摘Dyslipidemia exhibits a high incidence after liver transplantation,in which tacrolimus,a widely used immunosuppressant,plays a fundamental role.MicroRNAs and related circRNAs represent a class of noncoding RNAs that have been recognized as important regulators of genes associated with lipid metabolism.However,their transcriptional activities and functional mechanisms in tacrolimus-related dyslipidemia remain unclear.In this study,we observed that tacrolimus could induce triglyceride accumulation in hepatocytes by stimulating sterol response element-binding proteins(SREBPs)and miR-33a.Our in silico and experimental analyses identified miR-33a as a direct target of circFASN.Tacrolimus could downregulate circFASN and result in elevated miR-33a in vivo and in vitro.Overexpression of circFASN or silencing of miR-33a decreased the promoting effects of tacrolimus on triglyceride accumulation.Clinically,the incidence of dyslipidemia in liver transplant recipients with elevated serum miR-33a after liver transplantation was higher than that in patients without elevated serum miR-33a(46.3%vs.18.8%p=0.012,n=73).Our results showed that the circFASN/miR-33a regulatory system plays a distinct role in tacrolimus-induced disruption of lipid homeostasis.MiR-33a is likely a risk factor for tacrolimus-related dyslipidemia,providing a potential therapeutic target to combat tacrolimus-induced dyslipidemia after liver transplantation.Chenzhi Zhang Kangchen Chen Rongli Wei Guanghan Fan Xuechun Cai Li Xu Beini Cen Jianguo Wang Haiyang Xie Shusen Zheng Xiao Xu 2020Signal Transduction and Targeted Therapy2020,5,1:2
8Crashworthiness analysis of a cylindrical auxetic structure under axial impact loading显示文摘Structures with negative Poisson’s ratio(NPR) have been widely used in engineering application due to its unusual properties. In this paper, crashworthiness of a novel cylindrical auxetic structure under axial impact loading is investigated by the numerical methods. The software LS-DYNA is adopted to analyze the effects of the geometry parameters on the force, energy absorption(EA) and specific energy absorption(SEA). It is found that an overlarge number of layers and cells will make NPR structure extend outward from the mid;NPR structures with small number of layers and cells will make some layers of structures rotate in final crushing state. If the thickness of the long-inclined bean(L-beam) is larger than that of short-inclined beam(S-beam), there will be a smoother transition from the lower to a higher value in crushing force. Conversely, the force will maintain a relatively steady value, which determined by the sum of the thickness of L-and S-beams. In addition, there is also a critical inner circle radius which distinguishes different deformation modes.
