| 1 | Effect of sodium arsenite on spermatogenesis, plasma gonadotrophins and testosterone in rats显示文摘Aim:To investigate the effect of arsenic on spermatogenesis.Methods:Mature(4 months old)Wistar rats were intraperitoneally administered sodium arsenite at doses of 4,5 or 6mg·kg^-1·day^-1 for 26 days.Different varieties of germ cells at stage Ⅶ seminiferous epithelium cycle,namely,type A spermatogonia(ASg),preleptotene spermatocytes(pLSc),midpachytene spermatocytes(mPSc) and step 7 spermatids(7Sd) were quantitatively evaluated, along with radioimmunoassay of plasma follicle-stimulating hormone(FSH),lutuneizing hormone(LH),testosterone and assessment of the epididymal sperm count.Results:In the 5 and 6 mg/kg groups,there were significant dosedependent decreases in the accessory sex organ weights,epididymal sperm count and plasma concentrations of LH,FSH and testosterone with massive degeneration of all the germ cells at stage Ⅶ,The changes were insignificant in the 4 mg/kg group.Conclusion:Arsenite has a suppressive influence on spermatogenesis and gonadotrophin and testosterone release in rats. | Mahitosh Sarkar Gargi Ray Chaudhuri Aloke Chattopadhyay Narendra Mohan Biswas | 2003 | Asian Journal of Andrology2003,5,1: | 24 |
| 2 | Outcome of gall bladder polypoidal lesions detected by transabdominal ultrasound scanning: A nine year experience显示文摘AIM: To determine the outcome of polypoidal lesions within the gall bladder (PLG) diagnosed by trans-abdominal scanning.METHODS: A nine-year (1993-2002) retrospective casenote review of all patients who underwent ultrasound scanning after referral to a single Upper GI Surgeon at a District General Hospital was conducted. Patients who were diagnosed with a PLG were included in our study. A database was constructed and patient details, investigations including ultrasound scan (USS) findings, treatment and histology and final diagnosis were recorded. RESULTS: Twenty-three (out of 651) patients were diagnosed pre-operatively by USS to have a polyp-likegall bladder lesion (PLG). Post cholecystectomy histological examination revealed 12 gallstones, 7 cholesterol polyps, 3 adenocarcinomas within polyps and 1 normal gall bladder. The specificity of USS in the diagnosis of PLG was 92.3%. All the true polyps were malignant. Overall USS had 66.66% sensitivity and 100% specificity in the pre-operative suspicion of malignancy. Using size greater than 10 mm as measured on USS as a cut-off, we find 100% sensitivity and 86.95% specificity with a positive predictive value of 50% in the diagnosis of malignancy in PLG.CONCLUSION: A large number of PLG are in fact calculi within diseased gall bladder. In cases of gall bladder polyps more then 10 mm in size on USS further imaging (crosssectional and/or EUS) is indicated prior to surgery. | 5 | Portal biliopathy显示文摘Biliary ductal changes are a common radiological finding in patients with portal hypertension, however only a small percentage of patients (5%-30%) develop symptomatic bile duct obstruction. The exact pathogenesis is not clear, but an involvement of factors such as bile duct compression by venous collaterals, ischemia, and infection is accepted by most authors. Although endoscopic retrograde cholangiopancrea- tography was used to define and diagnose this condition, magnetic resonance cholangiopancreatography is currently the investigation of choice for diagnosing this condition. Treatment is indicated only for symptomatic cases. Portosystemic shunts are the treatment of choice for symptomatic portal biliopathy. In the majority of patients, the changes caused by biliopathy resolve after shunt surgery, however, 15%-20% patients require a subsequent bilio-enteric bypass or endoscopic management for persistent biliopathy. Generation of globular equi-axed grains during solidification of rheocast components, compared to the columnar dendritic structure of conventional casting routes, facilitates the manufacturing of components with improved mechanical properties and structural integrity. In the present investigation, a cooling slope has been designed and indigenously fabricated to produce semi solid slurry of Al-Si-Mg (A356) alloy and successively cast in a metallic mould. The scope of the present work discusses about development of a numerical model to simulate the liquid metal flow through cooling slope using Eulerian two-phase flow approach and to investigate the effect of pouring temperature on cooling slope semi-solid slurry generation process. The two phases considered in the present model are liquid metal and air. Solid fraction evolution of the solidifying melt is tracked at different locations of the cooling slope, following Schiel s equation. The continuity equation, momentum equation and energy equation are solved considering thin wall boundary condition approach. During solidification of the liquid metal, a modified temperature recovery scheme has been employed taking care of the latent heat release and change of fraction of liquid. | 16 | Physiological Basis of Stagnant Flooding Tolerance in Rice显示文摘Stagnant flooding(SF) is an important constraint which prolonges partial submergence damages of rice plants and reduces grain yield. Due to the heterogeneity in flood-prone ecosystem, many different types of traditional rice varieties are being grown by the farmers. The local landraces adapted to extreme in water availability could be the sources of new gene(s) which would be utilized to improve the adaptability of rice to SF with high yield. The main goal of this study is to identify new genetic resources tolerant to SF based on morpho-physiological traits. A total of 16 rice varieties were selected after initial screening from more than 400 rice varieties which were collected from eastern states of India. The increase rate of plant height was higher under SF compared to control, whereas no such trend was observed in the increment rate of aboveground total dry weight and culm dry weight. Area of aerenchyma gas spaces per tiller increased whereas root oxidase activity decreased under SF. The reduction of root oxidase activity, leaf area, and leaf dry weight was higher in susceptible varieties under SF compared to control. Stability index for different grain yield and yield attributes revealed that the impact of SF differed among different varieties. Correlation coefficient studies among different parameters taking stability index showed significant association with the grain yield. | 17 | Healing property of the Piper betel phenol, allylpyrocatechol against indomethacin-induced stomach ulceration and mechanism of action显示文摘AIM: To evaluate the protective activity of allylpyrocatechol (APC), the major antioxidant constituent of Piper betel, against the indomethacin-induced stomach ulceration in the rat model and correlates with its antioxidative and mucin protecting properties. METHODS: Male Sprague-Dawley rats were divided into five groups. Normal control rats (group Ⅰ) were given the vehicle oral dose of gum acacia in distilled water (1 mL per rat); ulcerated control and treated rats (groups Ⅱ-Ⅴ) were given a single dose of indomethacin (30 mg/kg body wt.); group Ⅱ rats were sacrificed 4 h after indomethacin administration; groups Ⅲ-Ⅴ rats were given the vehicle (1 mL per rat) or APC (2 mg/kg body wt.) or misoprostol (1.43 μg/kg body wt.) once daily by oral intubation for 7 d starting from 4 h after the indomethacin administration. After 7 d, the stomach tissues were excised for histological examination and biochemical analysis. RESULTS: Treatment with APC (2 mg/kg body wt per day) and misoprostol (1.43 μg/kg body wt per day) for 7 d could effectively heal the stomach ulceration as revealed from the ulcer index and histopathological studies. Compared to the zero day ulcerated group, treatment with APC and misoprostol reduced the ulcer index by 93.4% and 85.4% respectively (P < 0.05). Both APC and misoprostol accelerated ulcer healing observed in natural recovery (P < 0.05), their respective healing capacities not being significantly different. The healing capacities of APC and misoprostol could be attributed to their antioxidant activity as well as the ability to enhancethe mucin content of the gastric tissues. Compared to the ulcerated untreated rats, those treated with APC and misoprostol showed near normal MDA levels, while the protein levels were 86% and 78% of the normal value respectively (P < 0.05). Likewise, both APC and misoprostol increased the SOD, catalase, and mucin levels significantly (P < 0.05), the effect of APC being better. Methods:Among cases that underwent appendectomy,the appendicitis cases having fruit seeds and undigested plant residuals in their etiology were examined retrospectively.Also, histopathological features,age,sex,and parameters of morbidity and mortality were used. | 20 | Gene expression profile of esophageal cancer in North East India by cDNA microarray analysis显示文摘AIM: To identify alterations in genes and molecular functional pathways in esophageal cancer in a high incidence region of India where there is a widespread use of tobacco and betel quid with fermented areca nuts. METHODS: Total RNA was isolated from tumor and matched normal tissue of 16 patients with esophageal squamous cell carcinoma. Pooled tumor tissue RNA was labeled with Cy3-dUTP and pooled normal tissue RNA was labeled with Cy5-dUTP by direct labeling method. The labeled probes were hybridized with human 10K cDNA chip and expression profiles were analyzed by Genespring GX V 7.3 (Silicon Genetics). RESULTS: Nine hundred twenty three genes were differentially expressed. Of these, 611 genes were upregulated and 312 genes were downregulated. Using stringent criteria (P ≤ 0.05 and ≥ 1.5 fold change), 127 differentially expressed genes (87 upregulated and 40 downregulated) were identified in tumor tissue. On the basis of Gene Ontology, four different molecular functional pathways (MAPK pathway, G-protein coupled receptor family, ion transport activity, and serine or threonine kinase activity) were most significantly upregulated and six different molecular functional pathways (structural constituent of ribosome, endopeptidase inhibitor activity, structural constituent of cytoskeleton, antioxidant activity, acyl group transferase activity, eukaryotic translation elongation factor activity) were most significantly downregulated. CONCLUSION: Several genes that showed alterations in our study have also been reported from a high incidence area of esophageal cancer in China.
