| 2 | Feasibility of establishing cerebral ischemia models by using aerocyst-blocking bilateral ascending pharyngeal artery of piglets Imaging assessment显示文摘BACKGROUND: The cerebral ischemia and ischemia/reperfusion animal models are used to simulate the human cerebrovascular diseases is one of the popular topics of neurological science recently. To study the pathophysiology, pathogenesis, prophylaxis and treatment of ischemic cerebrovascular diseases and to establish the ideal animal model that is the most similar to the human cerebral ischemia, are the topics that the people generally cared about. OBJECTIVE: To evaluate the effects of aerocyst-blocking bilateral ascending pharyngeal artery on the establishment of cerebral ischemia models by using digital subtraction angiography (DSA), magnetic resonance diffusion-weighted imaging (DWI) and magnetic resonance perfusion-weighted imaging (PWI). DESIGN: Repetitive measure animal experiment. SETTING: Zhongshan Hospital Affiliated to Dalian University. MATERIALS: The experiment was carried out in the Animal Laboratory (Provincial Laboratory), Zhongshan Hospital of Dalian Univeristy from January to May 2006. A total of 14 domestic piglets, of 6 months old, weighing 12-15 kg, of either gender, were selected from Animal Experimental Center, Dalian University. Multistar T.O.P digital subtraction angiography machine was provided by Siemens Company, German. METHODS: Aerocyst-blocking bilateral ascending pharyngeal artery was used to establish cerebral ischemia models. And then, Multistar T.O.P. DSA was used for imaging of cerebral vessels before blocking, during blocking and at 0.5 and 2 hours after ischemia perfusion. GE Signa 1.5 T supraconduction magnetic resonance imaging was used for DWI examination; in addition, PWI was used based on focal sites and areas. Otherwise, magnetic resonance imaging (MRI) was used to detect signal changes of T1WI and T2WI in ischemic areas. MAIN OUTCOME MEASURES: Analytic results of DSA, DWI, PWI and MRI. RESULTS: All 14 experimental piglets were involved in the final analysis. ① DSA: The blood flow of bilateral ascending pharyngeal arteries and its branch were blocked at blocking phase, which restored 0.5 and 2 hours after reperfusion. ② DWI and PWI: There were no observable abnormalities in PWI and DWI at pre-blocking. Abnormal increased signals were found on both DWI and PWI at during and post-blocking. There were reduction in ADC and rCBF and delay in rTTP at all time points except pre-blocking. ③ MRI: There were no abnormal signals observable at any time of pre- and post-blocking in T1WI and T2WI. | 6 | Quantitative analysis of glutamate compounds in the swine brain following central analgesics nasal spray using proton magnetic resonance spectroscopy and linear combination model显示文摘<正>BACKGROUND:In localized brain proton magnetic resonance spectroscopy(~1H-MRS),metabolite levels are often expressed as ratios,rather than absolute concentrations.Frequently,the denominator is creatine,which is assumed to be stable in normal,as well as many pathological, states.However,in vivo creatine levels do not remain constant.Therefore,absolute metabolite measurements,which provide the precise concentrations of certain chemical compounds,are superior to metabolite ratios for determining pathological and evolutional changes. OBJECTIVE:To investigate the feasibility of quantification analysis of brain metabolite changes caused by central analgesics nasal spray using the ~1H-MRS and linear combination model(LCModel) methods. DESIGN,TIME AND SETTING:This neuroimaging,observational,animal study was performed at the Laboratory of the Department of Medical Imaging,Second Affiliated Hospital,Medical College, Shantou University,China from July to December 2007. MATERIALS:Butorphanol tartrate nasal spray,as a mixed agonist-antagonist opioid analgesic,was purchased from Shanghai Hengrui Pharmacy,China.A General Electric Signa 1.5T System (General Electric Medical Systems,Milwaukee,Wl,USA) and LCModel software(Stephen Provencher,Oakville,Ontario,Canada) were used in this study. METHODS:MRS images were acquired in ten healthy swine aged 2 weeks using single-voxel point-resolved spectroscopic sequence.A region of interest(2 cm×2 cm×2 cm) was placed in the image centers of maximum brain parenchyma.Repeated MRS scanning was performed 15-20 minutes after intranasal administration of 1 mg of butorphanol tartrate.Three settings of repetition time/echo time were selected before and after nasal spray administration 3000 ms/30 ms, 1500 ms/30 ms,and 3000 ms/50 ms.Metabolite concentrations were estimated by LCModel software. MAIN OUTCOME MEASURES:~1H-MRS spectra was obtained using various repetition time/echo time settings.Concentrations of glutamate compounds(glutamate+glutamine),N-acetyl aspartate, and choline were detected in swine brain prior to and following nasal spray treatment. RESULTS:The glutamate compounds curve was consistent with original spectra,when a repetition time/echo time of 3 000 ms/30 ms was adopted.Concentrations of glutamate compounds,N-acetyl aspartate,and choline decreased following administration.The most significant reduction was observed in glutamate compound concentrations from(9.28±0.54) mmol/kg to(7.28±0.54) mmol/kg (P<0.05). | 7 | Suppression of transmembrane sodium currents on the freshly isolated hippocampal neuron cell with continuous infrared light显示文摘Physiotherapeutic effects of infrared lasers have been proved in clinic.These infrared-based regulations of the bioelectrical activities can roughly be classied into enhancement and suppression of action potential(AP),which are described by sodium(Na)and potassium(K)transmembrane current equations,named as Hodgkin and Huxley(HH)-model.The enhancement effect is able to evoke or strengthen the AP when infrared light is applied.Its corresponding mechanism is commonly ascribed to the changes of the cell membrane capacitance,which is transiently increased in response to the infrared radiation.The distinctive feature of the suppression effect is to inhibit or reduce the AP by the designed protocols of infrared radiation.However,its mechanism presents more complexity than that in enhancement cases.HH-model describes how the Na current determines the initial phase of AP.So,the enhancement and suppression of AP can be also ascribed to the regulations of the corresponding Na currents.Here,a continuous infrared light at the wavelength of 980 nm(CIS-980)was employed to stimulate a freshly isolated hippocampal neuron in vitro and a suppression effect on the Na currents of the neuron cell was observed.Both Na and K currents,which are named as whole cell currents,were simultaneously recorded with the cell membrane capacitance current by using a patch clamp combined with infrared irradiation.The results demonstrated that the CIS-980 was able to reversibly increase the capacitance currents,completely suppressed Na currents,but little changed K currents,which forms the steady outward whole cell currents and plays a major role on the AP repolarization.A conrmation experiment was designed and carried out by synchronizing tens of milliseconds of infrared stimulation on the same kinds of hippocampal neuron cells.After the blocked K channel,a reduction of Na current amplitude was still recorded.This proved that infrared suppression of Na current was irrelevant to K channel.A membrane capacitance mediation process was preliminarily proposed to explain the Na channel suppression process. | Fanyi Kong Xinyu Li Ruonan Jiao Kun Liu Xue Han Changkai Sun Changsen Sun | 2023 | Journal of Innovative Optical Health Sciences2023,16,2: | 0 |
| 8 | Discovery of novel exceptionally potent and orally active c-MET PROTACs for the treatment of tumors with MET alterations显示文摘Various c-mesenchymal-to-epithelial transition(c-MET) inhibitors are effective in the treatment of non-small cell lung cancer;however, the inevitable drug resistance remains a challenge, limiting their clinical efficacy. Therefore, novel strategies targeting c-MET are urgently required. Herein, through rational structure optimization, we obtained novel exceptionally potent and orally active c-MET proteolysis targeting chimeras(PROTACs) namely D10 and D15 based on thalidomide and tepotinib. D10 and D15 inhibited cell growth with low nanomolar IC_(50) values and achieved picomolar DC_(50) values and>99% of maximum degradation(D_(max)) in EBC-1 and Hs746T cells. Mechanistically, D10 and D15dramatically induced cell apoptosis, G1 cell cycle arrest and inhibited cell migration and invasion.Notably, intraperitoneal administration of D10 and D15 significantly inhibited tumor growth in the EBC-1 xenograft model and oral administration of D15 induced approximately complete tumor suppression in the Hs746T xenograft model with well-tolerated dose-schedules. Furthermore, D10 and D15 exerted significant anti-tumor effect in cells with c-MET^(Y1230H) and c-MET^(D1228N) mutations, which are resistant to tepotinib in clinic. | 9 | Screening of Drought Resistance Identification Indexes for Rice(Oryza sativa L.) Cultivars in Ningxia显示文摘The morphological characters, physiological characters, yield traits and yield per plant of total 49 rice cultivars from Ningxia were investigated under conditions of water stress and non-water stress so as to determine the relationship between each trait and yield per plant under water stress and the relationship between each relative character and drought resistance coefficient under water stress and non-water stress. The correlation, grey correlation, stepwise regression and path analyses showed that the tiller number per plant, plant height, grain density, effective panicle number per plant and grain number per panicle, total 5 traits, were significantly correlated with the drought resistance of rice, and they could be used to identify the drought resistance of rice in Ningxia. In addition, the drought resistance of rice was graded qualitatively according to the subordinate function value of corresponding drought resistance coefficient. | 10 | Wearable Fiber‑Based Supercapacitors Enabled by Additive‑Free Aqueous MXene Inks for Self‑Powering Healthcare Sensors显示文摘Wearable fiber-based electronics have found diverse applications including energy storage,healthcare or thermal management,etc.In particular,additive-free aqueous inks play significant roles in fabrication of wearable fiber-based devices,owning to their nontoxic nature and ease of manufacturing.Herein,wearable carbon fiber-based asymmetric supercapacitors(WASSC)are developed based on additive-free aqueous MXene inks,for self-powering healthcare sensors.The sediments of MXene without further modification are used as inks.Furthermore,combined with additive-free aqueous MXene/polyaniline(MP)inks,WASSC,with a wide voltage window and high capacitance is developed for practical energy supply.Impressively,WASSC has been successfully utilized to power wearable pressure sensors that could monitor motions and pulse signals.This wearable self-powered monitoring system on can accurately monitor the human motions,pronunciation,swallow or wrist pulse,without using the rigid batteries.This advantage realizes a great potential in simple and cost effective monitoring of human health and activities.Besides,self-powered system enables waste recycling of MXene and provides an effective approach for designing wearable and fiber-based self-powered sensors. | Junlin Ma Zewei Cui Yuhang Du Jianxin Zhang Changkai Sun Chengyi Hou Nan Zhu | 2022 | Advanced Fiber Materials2022,4,6: | 0 |
