| 7 | Calcium pyrophosphate dihydrate crystals in a 9-year-old with osteomyelitis of the knee:A case report显示文摘BACKGROUND Calcium pyrophosphate dihydrate deposition disease(CPPD),or pseudogout,is an inflammatory arthritis common among elderly patients,but rarely seen in patients under the age of 40.In the rare cases presented of young patients with CPPD,genetic predisposition or related metabolic conditions were almost always identified.CASE SUMMARY The authors report the case of a 9-year-old boy with no past medical history who presented with acute knee pain and swelling after a cat scratch injury 5 d prior.Synovial fluid analysis identified calcium pyrophosphate dihydrate crystals.Further MRI analysis identified osteomyelitis and a small soft tissue abscess.CONCLUSION This case presents the extremely rare diagnostic finding of calcium pyrophosphate dihydrate crystals in a previously healthy pediatric patient.The presence of osteomyelitis presents a unique insight into the pathogenesis of these crystals in pediatric patients.More research needs to be done on the role of CPPD in pediatric arthritis and joint infection. | William Pavlis David S Constantinescu Rajan Murgai Spencer Barnhill Brian Black | 2022 | World Journal of Orthopedics2022,13,9: | 1 |
| 8 | Upregulation of RIN3 induces endosomal dysfunction in Alzheimer’s disease显示文摘Background In Alzheimer’s Disease(AD),about one-third of the risk genes identified by GWAS encode proteins that function predominantly in the endocytic pathways.Among them,the Ras and Rab Interactor 3(RIN3)is a guanine nucleotide exchange factor(GEF)for the Rab5 small GTPase family and has been implicated to be a risk factor for both late onset AD(LOAD)and sporadic early onset AD(sEOAD).However,how RIN3 is linked to AD pathogenesis is currently undefined.Methods Quantitative PCR and immunoblotting were used to measure the RIN3 expression level in mouse brain tissues and cultured basal forebrain cholinergic neuron(BFCNs).Immunostaining was used to define subcellular localization of RIN3 and to visualize endosomal changes in cultured primary BFCNs and PC12 cells.Recombinant flag-tagged RIN3 protein was purified from HEK293T cells and was used to define RIN3-interactomes by mass spectrometry.RIN3-interacting partners were validated by co-immunoprecipitation,immunofluorescence and yeast two hybrid assays.Live imaging of primary neurons was used to examine axonal transport of amyloid precursor protein(APP)andβ-secretase 1(BACE1).Immunoblotting was used to detect protein expression,processing of APP and phosphorylated forms of Tau.Results We have shown that RIN3 mRNA level was significantly increased in the hippocampus and cortex of APP/PS1 mouse brain.Basal forebrain cholinergic neurons(BFCNs)cultured from E18 APP/PS1 mouse embryos also showed increased RIN3 expression accompanied by early endosome enlargement.In addition,via its proline rich domain,RIN3 recruited BIN1(bridging integrator 1)and CD2AP(CD2 associated protein),two other AD risk factors,to early endosomes.Interestingly,overexpression of RIN3 or CD2AP promoted APP cleavage to increase its carboxyl terminal fragments(CTFs)in PC12 cells.Upregulation of RIN3 or the neuronal isoform of BIN1 increased phosphorylated Tau level.Therefore,upregulation of RIN3 expression promoted accumulation of APP CTFs and increased phosphorylated Tau.These effects by RIN3 was rescued by the expression of a dominant negative Rab5(Rab5S34N)construct.Our study has thus pointed to that RIN3 acts through Rab5 to impact endosomal trafficking and signaling.Conclusion RIN3 is significantly upregulated and correlated with endosomal dysfunction in APP/PS1 mouse.Through interacting with BIN1 and CD2AP,increased RIN3 expression alters axonal trafficking and procession of APP.Together with our previous studies,our current work has thus provided important insights into the role of RIN3 in regulating endosomal signaling and trafficking. | Ruinan Shen Xiaobei Zhao Lu He Yongbo Ding Wei Xu Suzhen Lin Savannah Fang Wanlin Yang Kijung Sung Brian Spencer Robert A.Rissman Ming Lei Jianqing Ding Chengbiao Wu | 2020 | Translational Neurodegeneration2020,9,2: | 1 |

| 10 | Real-time hybrid simulation for structural control performance assessment显示文摘Real-time hybrid simulation is an attractive method to evaluate the response of structures under earthquake loads.The method is a variation of the pseudodynamic testing technique in which the experiment is executed in real time,thus allowing investigation of structural systems with rate-dependent components.Real-time hybrid simulation is challenging because it requires performance of all calculations,application of displacements,and acquisition of measured forces,within a very small increment of time.Furthermore,unless appropriate compensation for actuator dynamics is implemented,stability problems are likely to occur during the experiment.This paper presents an approach for real-time hybrid simulation in which compensation for actuator dynamics is implemented using a model-based feedforward compensator.The method is used to evaluate the response of a semi-active control of a structure employing an MR damper.Experimental results show good agreement with the predicted responses,demonstrating the effectiveness of the method for structural control performance assessment. | Juan E Carrion BF Spencer Jr Brian M Phillips | 2009 | Earthquake Engineering and Engineering Vibration2009,8,4: | 1 |

| 12 | Dorsal root ganglion progenitors differentiate to gamma-aminobutyric acid-and choline acetyltransferase-positive neurons显示文摘This study examined the isolation and differentiation of dorsal root ganglion progenitor cells for therapeutic use in neurodegenerative diseases. Rat embryonic dorsal root ganglia progenitors were isolated and purified using the differential adhesion method combined with cytosine arabinoside treatment. After culture in serum-free medium supplemented with B27, basic fibroblast growth factor and epidermal growth factor, these cells remained viable and survived for more than 18 months in vitro. Most cells differentiated to neurons that were immunoreactive for gamma-aminobutyric acid and choline acetyltransferase as detected by immunohistochemical staining. In addition, nerve growth factor and neurotrophic tyrosine kinase receptor expression were also observed in dorsal root ganglion progenitors and differentiated cells. | 15 | Cs_(3)Bi_(2)l_(9)-hydroxyapatite composite waste forms for cesium and iodine immobilization显示文摘Perovskite-based ceramic composites were developed as potential waste form materials for immobilizing cesium(Cs)and iodine(I)with high waste loadings and chemical durability.The perovskite Cs_(3)Bi_(2)I_(9)has high Cs(22 wt%)and I(58 wt%)content,and thus can be used as a potential host phase to immobilize these critical radionuclides.In this work,the perovskite Cs_(3)Bi_(2)I_(9)phase was synthesized by a cost effective solution-based approach,and was embedded into a highly durable hydroxyapatite matrix by spark plasma sintering to form dense ceramic composite waste forms.The chemical durabilities of the monolithic Cs_(3)Bi_(2)I_(9)and Cs_(3)Bi_(2)I_(9)-hydroxyapatite composite pellets were investigated by static and semi-dynamic leaching tests,respectively.Cs and I are incongruently released from the matrix for both pure Cs_(3)Bi_(2)I_(9)and composite structures.The normalized Cs release rate is faster than that of I,which can be explained by the difference in the strengths between Cs-I and Bi-I bonds as well as the formation of insoluble micrometer-sized BiOI precipitates.The activation energies of elemental releases based on dissolution and diffusion-controlled mechanisms are determined with significantly higher energy barriers for dissolution from the composite versus that of the monolithic Cs_(3)Bi_(2)I_(9).The ceramic-based composite waste forms exhibit excellent chemical durabilities and waste loadings,commensurate with the state-of-the-art glass-bonded perovskite composites for I and Cs immobilization. | Kun YANG Yachun WANG Junhua SHEN Spencer M.SCOTT Brian J.RILEY John D.VIENNA Jie LIAN | 2022 | Journal of Advanced Ceramics2022,11,5: | 0 |
