CCL5 deficiency promotes liver repair by improving inflammation resolution and liver regeneration through M2 macrophage polarization显示文摘Despite the diverse etiologies of drug-induced liver injury(DILI),innate immunity activation is a common feature involved in DILI progression. However, the involvement of innate immunity regulation in inflammation resolution and liver regeneration in DILI remains obscure.Herein,we identified the chemokine CCL5 as a central mediator of innate immunity regulation in the pathogenesis of DILI.First,we showed that serum and hepatic CCL5 levels are elevated in both DILI patients and an APAP-induced liver injury(AILI)mouse model.Interestingly, both nonparenchymal cells and stressed hepatocytes are cell sources of CCL5 induction in response to liver injury.Functional experiments showed that CCL5 deficiency has no effect on the early phase of AILI but promotes liver repair in the late phase mainly by promoting inflammation resolution and liver regeneration, which are associated with an increased number of hepatic M2 macrophages.Mechanistically,CCL5 can directly activate M1 polarization and impede M2 polarization through the CCR1-and CCR5-mediated activation of the MAPK and NF-κB pathways. We then showed that CCL5 inhibition mediated by either a CCL5-neutralizing antibody or the antagonist Met-CCL5 can greatly alleviate liver injury and improve survival in an AILI mouse model. Our data demonstrate CCL5 induction during DILI,identify CCL5 as a novel innate immunity regulator in macrophage polarization, and suggest that CCL5 blockage is a promising therapeutic strategy for the treatment of DILI. | Meng Li Xuehua Sun Jie Zhao Lei Xia Jichang Li Min Xu Bingrui Wang Han Guo Chang Yu Yueqiu Gao Hailong Wu Xiaoni Kong Qiang Xia | 2020 | Cellular & Molecular Immunology2020,17,7: | 13
In situ experiments on width and evolution characteristics of excavation damaged zone in deeply buried tunnels显示文摘The seven long tunnels of Jinping II hydropower station are deeply buried. The width and evolution characteristics of excavation damaged zone(EDZ) are the key problem to the design of tunnels excavation and supports. In order to study this problem, several specific experimental tunnels with different overburden and geometric sizes were excavated at this site.Digital borehole camera, sliding micrometer,cross-hole acoustic wave equipment and acoustic emission apparatus were adopted. This paper introduced the comprehensive in situ experimental methods through pre-installed facilities and pre-drilled boreholes.Typical properties of the surrounding rock mass, including cracks, deformation, elastic wave and micro fractures, were measured during the whole process of the tunnel excavation. The width and characteristics of formation and evolution of tunnels EDZ were analyzed under different construction methods involving of TBM and drilling and blasting, the test tunnels were excavated by full-face or two benches. The relationships between EDZ and tunnel geometry sizes, overburden and excavation method were described as well. The results will not only contribute a great deal to the analysis of rock mass behavior in deeply buried rock mass, but also provide direct data for support design and rockburst prediction. | LI ShaoJun 1,FENG XiaTing 1,LI ZhanHai 3,CHEN BingRui 1,JIANG Quan 1,WU ShiYong 2,HU Bin 2 &XU JinSong 2 1 State Key Laboratory of Geomechanics and Geotechinical Engineering,Institute of Rock and Soil Mechanics,Chinese Academy of Sciences,Wuhan 430071,China 2 Ertan Hydropower Development Company,Ltd.,Chengdu 610051,China 3 School of Resources and Civil Engineering,Northeastern University,Shenyang 110004,China | 2011 | Science China(Technological Sciences)2011,54,S1: | 13
Toward a general evaluation model for soil respiration (GEMSR)显示文摘Soil respiration is an important component of terrestrial carbon budget. Its accurate evaluation is es- sential to the study of terrestrial carbon source/sink. Studies on soil respiration at present mostly focus on the temporal variations and the controlling factors of soil respiration, but its spatial variations and controlling factors draw less attention. Moreover, the evaluation models for soil respiration at present include only the effects of water and heat factors, while the biological and soil factors controlling soil respiration and their interactions with water and heat factors have not been considered yet. These models are not able to accurately evaluate soil respiration in different vegetation/terrestrial ecosystems at different temporal and spatial scales. Thus, a general evaluation model for soil respiration (GEMSR) including the interacting meteorological (water and heat factors), soil nutrient and biological factors is suggested in this paper, and the basic procedure developing GEMSR and the research tasks of soil respiration in the future are also discussed. | ZHOU GuangSheng, JIA BingRui, HAN GuangXuan & ZHOU Li Key Laboratory of Vegetation and Environmental Change, Institute of Botany, Chinese Academy of Sciences, Beijing 100093, China | 2008 | Science China(Life Sciences)2008,51,3: | 12
Studies on the evolution process of rockbursts in deep tunnels显示文摘This paper focuses on the evolution processes of different types of rockbursts occurring in deep tunnels. A series of laboratory tests and in-situ monitoring in deep tunnels excavated by tunnel boring machine (TBM) and drill-and-blast (D&B) method have been conducted to understand the mechanisms and processes of the evolution of different types of rockbursts, including strain rockburst, strain-structure slip rockburst, immediate rockburst and time-delayed rockburst. Three different risk assessment methods are proposed to evaluate the intensity and potential failure depth of rockbursts. These methods can be applied before excavation and the results can be updated according to the real-time information during excavation. Two micro-seismicity based real-time warning systems have been established for predicting various intensities of rockbursts, such as slight, moderate, intensive and extremely intensive rockbursts. Meanwhile, the probability and intensity of the rockburst are also given. The strategy for excavation and support design has been suggested for various intensities of rockbursts before excavation. The strategy for dynamic control of the rockburst evolution process is also proposed according to the monitoring results. The methodology has been successfully applied to rockburst risk reduction for deep tunnels at Jinping II hydropower project. The results have illustrated the applicability of the proposed methodology and techniques concerning rockbursts. | Xiating Feng Bingrui Chen Shaojun Li Chuanqing Zhang Yaxun Xiao Guangliang Feng Hui Zhou Shili Qiu Zhouneng Zhao Yang Yu Dongfang Chen Huajun Ming | 2012 | Journal of Rock Mechanics and Geotechnical Engineering2012,4,4: | 11
Review of research on Arctic sea ice physics based on the Chinese National Arctic Research Expedition显示文摘China launched its Arctic research program and organized the first Chinese National Arctic Research Expedition(CHINARE-Arctic) in 1999. By 2016, six further expeditions had been conducted using the R/V Xuelong. The main region of the expeditions has focused on the Pacific sector of the Arctic Ocean for sea ice observations. The expeditions have used icebreaker, helicopter, boat, floe, and buoy platforms to perform these observations. Some new technologies have been developed, in particular, the underway auto-observing system for sea ice thickness using an electromagnetic instrument. The long-term measurement systems, e.g., the sea ice mass balance buoy, allow observations to extend from summer to winter. Some international cooperation projects have been involved in CHINARE-Arctic, especially the 'Developing Arctic Modeling and Observing Capabilities for Long-Term Environmental Studies' project funded by the European Union during the International Polar Year. Arctic sea ice observations have been used to verify remote sensing products, identify changes in Arctic sea ice, optimize the parameterizations of sea ice physical processes, and assess the accessibility of ice-covered waters, especially around the Northeast Passage. Recommendations are provided as guidance to future CHINARE-Arctic projects. For example, a standardized operation system of sea ice observations should be contracted, and the observations of sea ice dynamics should be enhanced. The upcoming launch of a new Chinese icebreaker will allow increased ship time in support of future CHINARE Arctic oceanographic investigations. | LEI Ruibo ZHANG Zhanhai LI Zhijun YANG Qinghua LI Bingrui LI Tao | 2017 | Advances in Polar Science2017,28,2: | 5
On the Response of Subduction in the South Pacific to an Intensification of Westerlies and Heat Flux in an Eddy Permitting Ocean Model显示文摘Based on an eddy permitting ocean general circulation model, the response of water masses to two distinct climate scenarios in the South Pacific is assessed in this paper. Under annually repeating atmospheric forcing that is characterized by different westerlies and associated heat flux, the response of Subantarctic Mode Water(SAMW) and Antarctic Intermediate Water(AAIW) is quantitatively estimated. Both SAMW and AAIW are found to be warmer, saltier and denser under intensified westerlies and increased heat loss. The increase in the subduction volume of SAMW and AAIW is about 19.8 Sv(1 Sv =10~6m^3s^(-1)). The lateral induction term plays a dominant role in the changes in the subduction volume due to the deepening of the mixed layer depth(MLD). Furthermore, analysis of the buoyancy budget is used to quantitatively diagnose the reason for the changes in the MLD. The deepening of the MLD is found to be primarily caused by the strengthening of heat loss from the ocean to the atmosphere in the formation region of SAMW and AAIW. | Chengyan LIU Zhaomin WANG Bingrui LI Chen CHENG Ruibin XIA | 2017 | Advances in Atmospheric Sciences2017,34,4: | 5
Evolution and breaking of a propagating internal wave in stratified ocean显示文摘The evolution and breaking of a propagating internal wave are directly numerically simulated using a pseudo-spectral method. The mechanism of PSI (parametric subharmonic instability) involved in the evolution is testified clearly. It dominates gradually in nonlinear resonant interactions. As a consequence, the energy cascades to a second plant wave packet which has lower frequencies and higher wavenumbers than that of the primary wave. With the growth of this wave packet, wave breaking occurs and causes strongly nonlinear regime,i.e. stratified turbulence. The strong mixing and intermittent of the turbulence can be learned from the evolution of the total energy and kurtosis of vorticity vs. time. Some statistic properties of the stratified turbulence are also analyzed, including the spectra of KE (kinetic energy) and PE (potential energy). The results show that the PE spectra display a wavenumber range scaling as 0.2 N4k-3y (N is the Brunt-Visl frequency,ky is the vertical wavenumber),which is called buoyancy sub-range. However, the KE spectra cannot satisfy the negative cubic law of vertical wavenumber, which have a much larger downtrend than that of the PE spectra, for the potential energy is transferred more efficiently toward small scales than the kinetic energy. The Cox number of diapycnal diffusivity is also calculated, and it shows a good consistency with the observations and deductions in the ocean interior, during the stage of the stratified turbulence maintaining a fairly active level. | LI Bingrui FAN Haimei TIAN Jiwei SUN Bo ZHANG Zhanhai | 2008 | Acta Oceanologica Sinica2008,27,1: | 3
Genomic landscapes of Chinese sporadic autism spectrum disordersrevealed by whole-genome sequencing显示文摘Autism spectrum disorder (ASD) is a neurodevelopmental disorder with considerable clinical and genetic heterogeneity. In this study,we identified all classes of genomic variants from whole-genome sequencing (WGS) dataset of 32 Chinese trios with ASD,including de novo mutations, inherited variants, copy number variants (CNVs) and genomic structural variants.A higher mutation rate (Poisson test,P<2.2×10^(-16)) in exonic (1.37×10^(-8)) and 3'-UTR regions (1.42×10^(-8)) was revealed in comparison with that of whole genome (1.05×10^(-8)).Using an integrated model,we identified 87 potentially risk genes (P<0.01) from 4832 genes harboring various rare deleterious variants, including CHD8 and NRXN2,implying that the disorders may be in favor to multiple-hit.In particular, frequent rare inherited mutations of several microcephaly-associated genes (ASPM,WDR62,and ZNF335)were found in ASD.In chromosomal structure analyses,we found four de novo CNVs and one de novo chromosomal rearrangement event, including a de novo duplication of UBE3A-containing region at 15q11.2-q13.1,which causes Angelman syndrome and microcephaly, and a disrupted TNR due to de novo chromosomal translocation t (1;5) (q25.1;q33.2).Taken together, our results suggest that abnormalities of centrosomal function and chromatin remodeling of the microcephaly-associated genes may be implicated in pathogenesis of ASD.Adoption of WGS as a new yet efficient technique to illustrate the full genetic spectrum in complex disorders, such as ASD,could provide novel insights into pathogenesis, diagnosis and treatment. | Jinyu Wu Ping Yu Xin Jin Xiu Xu Jinchen Li Zhongshan Li Mingbang Wang Tao Wang Xueli Wu Yi Jiang Wanshi Cai Junpu Mei Qingjie Min Qiong Xu Bingrui Zhou Hui Guo Ping Wang Wenhao Zhou Zhengmao Hu Yingrui Li Tao Cai Yi Wang Kun Xia Yong-Hui Jiang Zhong Sheng Sun | 2018 | Journal of Genetics and Genomics2018,45,10: | 3
Single-cell transcriptomic landscape of human blood cells显示文摘High throughput single-cell RNA-seq has been successfully implemented to dissect the cellular and molecular features underlying hematopoiesis. However,an elaborate and comprehensive transcriptome reference of the whole blood system is lacking. Here,we profiled the transcriptomes of 7551 human blood cells representing 32 immunophenotypic cell types, including hematopoietic stem cells, progenitors and mature blood cells derived from 21 healthy donors. With high sequencing depth and coverage,we constructed a single-cell transcriptional atlas of blood cells(ABC) on the basis of both protein-coding genes and long noncoding RNAs(lncRNAs),and showed a high consistence between them.Notably, putative lncRNAs and transcription factors regulating hematopoietic cell differentiation were identified. While common transcription factor regulatory networks were activated in neutrophils and monocytes, lymphoid cells dramatically changed their regulatory networks during differentiation.Furthermore,we showed a subset of nucleated erythrocytes actively expressing immune signals, suggesting the existence of erythroid precursors with immune functions. Finally,a web portal offering transcriptome browsing and blood cell type prediction has been established. Thus, our work provides a transcriptional map of human blood cells at single-cell resolution, thereby offering a comprehensive reference for the exploration of physiological and pathological hematopoiesis. | Xiaowei Xie Mengyao Liu Yawen Zhang Bingrui Wang Caiying Zhu Chenchen Wang Qing Li Yingying Huo Jiaojiao Guo Changlu Xu Linping Hu Aiming Pang Shihui Ma Una Wang Wenbin Cao Shulian Chen Qiuling Li Sudong Zhang Xueying Zhao Wen Zhou Hongbo Luo Guoguang Zheng Erlie Jiang Sizhou Feng Lixiang Chen Lihong Shi Hui Cheng Sha Hao Ping Zhu Tao Cheng | 2021 | National Science Review2021,8,3: | 3

Evaluation of ArcIOPS sea ice forecasting products during the ninth CHINARE-Arctic in summer 2018显示文摘Numerical sea ice forecasting products during the ninth Chinese National Arctic Research Expedition(9th CHINARE-Arctic)from Arctic Ice Ocean Prediction System(ArcIOPS)of National Marine Environmental Forecasting Center of China are evaluated against satellite-retrieved sea ice concentration data,in-situ sea ice thickness observations, and sea ice products from Pan-Arctic Ice Ocean Modeling and Assimilation System(PIOMAS).The results show that ArcIOPS forecasts reliable sea ice concentration and thickness evolution.Deviations of the 168 h sea ice concentration and thickness forecasts with respect to the observations are less than 0.2 and 0.36 m.Comparison between outputs of the latest version of ArcIOPS and that of its previous version shows that the latest version has a substantial improvement on sea ice concentration forecasts due to data assimilation of new observational component, the sea surface temperature.Meanwhile, the sea ice volume product of the latest version is more close to the PIOMAS product. In the future, with more and more kinds of observations to be assimilated, the high-resolution version of ArcIOPS will be put into operational running and benefit Chinese scientific and commercial activities in the Arctic Ocean. | LIANG Xi ZHAO Fu LI Chunhua ZHANG Lin LI Bingrui | 2020 | Advances in Polar Science2020,31,1: | 3
Response of the mixed layer depth and subduction rate in the subtropical Northeast Pacific to global warming显示文摘The response of the mixed layer depth(MLD)and subduction rate in the subtropical Northeast Pacific to global warming is investigated based on 9 CMIP5 models.Compared with the present climate in the 9 models, the response of the MLD in the subtropical Northeast Pacific to the increased radiation forcing is spatially nonuniform, with the maximum shoaling about 50 m in the ensemble mean result. The inter-model differences of MLD change are non-negligible,which depend on the various dominated mechanisms. On the north of the MLD front,MLD shallows largely and is influenced by Ekman pumping, heat flux, and upper-ocean cold advection changes. On the south of the MLD front,MLD changes a little in the warmer climate, which is mainly due to the upper-ocean warm advection change. As a result, the MLD front intensity weakens obviously from 0.24 m/km to0.15 m/km(about 33.9%)in the ensemble mean, not only due to the maximum of MLD shoaling but also dependent on the MLD non-uniform spatial variability. The spatially non-uniform decrease of the subduction rate is primarily dominated by the lateral induction reduction(about 85%in ensemble mean)due to the significant weakening of the MLD front. This research indicates that the ocean advection change impacts the MLD spatially non-uniform change greatly, and then plays an important role in the response of the MLD front and the subduction process to global warming. | Ruibin Xia Bingrui Li Chen Cheng | 2021 | Acta Oceanologica Sinica2021,40,4: | 2
Research and application on corn crop identification and positioning method based on Machine vision显示文摘Weeds that grow among crops are undesirable plants and have adversely affected crop growth and yield. Therefore, the study explores corn identification and positioning methods based on machine vision. The ultra-green feature algorithm and maximum betweenclass variance method(OTSU)were used to segment maize corn, weeds, and land;the segmentation effect was significant and can meet the following shape feature extraction requirements. Finally, the identification and positioning of corn were achieved by morphological reconstruction and pixel projection histogram method. The experiment reveals that when a weeding robot travels at a speed of 1.6 km/h,the recognition accuracy can reach 94.1%.The technique used in this study is accessible for normal cases and can make a good recognition effect;the accuracy and real-time requirements of robot recognition are improved and reduced the calculation time. | Bingrui Xu Li Chai Chunlong Zhang | 2023 | Information Processing in Agriculture2023,10,1: | 2
Updating the models and uncertainty of mechanical parameters for rock tunnels using Bayesian inference显示文摘Rock mechanical parameters and their uncertainties are critical to rock stability analysis, engineering design, and safe construction in rock mechanics and engineering. The back analysis is widely adopted in rock engineering to determine the mechanical parameters of the surrounding rock mass, but this does not consider the uncertainty. This problem is addressed here by the proposed approach by developing a system of Bayesian inferences for updating mechanical parameters and their statistical properties using monitored field data, then integrating the monitored data, prior knowledge of geotechnical parameters, and a mechanical model of a rock tunnel using Markov chain Monte Carlo(MCMC)simulation. The proposed approach is illustrated by a circular tunnel with an analytical solution, which was then applied to an experimental tunnel in Goupitan Hydropower Station,China. The mechanical properties and strength parameters of the surrounding rock mass were modeled as random variables. The displacement was predicted with the aid of the parameters updated by Bayesian inferences and agreed closely with monitored displacements. It indicates that Bayesian inferences combined the monitored data into the tunnel model to update its parameters dynamically.Further study indicated that the performance of Bayesian inferences is improved greatly by regularly supplementing field monitoring data.Bayesian inference is a significant and new approach for determining the mechanical parameters of the surrounding rock mass in a tunnel model and contributes to safe construction in rock engineering. | Hongbo Zhao Bingrui Chen Shaojun Li Zhen Li Changxing Zhu | 2021 | Geoscience Frontiers2021,12,5: | 2

Development and validation of a predictive model for endocervical curettage in patients referred for colposcopy:A multicenter retrospective diagnostic study in China显示文摘Objective:This study aimed to develop a nomogram that can predict occult high-grade squamous intraepithelial lesions or worse(HSIL+)and determine the need for endocervical curettage(ECC)in patients referred for colposcopy. Methods:This retrospective multicenter study included 4,149 patients who were referred to any one of six tertiary hospitals in China for colposcopy between January 2020 and November 2021 because of abnormal screening results.ECC data were extracted from the medical records.Univariate and multivariate logistic regression analyses were performed to identify factors that could predict HSIL+on ECC.Patients were randomly assigned to a training set or to an internal validation set for performance and comparability testing. The model was externally validated and tested in patients from two additional hospitals. The nomogram was assessed in terms of discrimination and calibration and subjected to decision curve analysis. Results:HSIL+was found on ECC in 38.8%(n=388)of cases. Our predictive nomogram included age group, cytology, human papillomavirus(HPV)status, visibility of the cervix and colposcopic impression. The nomogram had good overall discrimination, which was internally validated[area under the receiver-operator characteristic(AUC),0.839;95%confidence interval(95%CI),0.773-0.904].In terms of external validation, the AUC was 0.843(95%CI,0.773-0.912)for the consecutive sample and 0.843(95%CI,0.783-0.902)for the comparative sample.Calibration analysis suggested good consistency between predicted and observed probabilities.Decision curve analysis suggested this nomogram would be clinically useful with almost the entire range of threshold probabilities. Conclusions:This internally and externally validated nomogram can be easily applied and incorporates multiple clinically relevant variables that can be used to identify patients with occult HSIL+who need ECC. | Peng Xue Bingrui Wei Samuel Seery Qing Li Zichen Ye Yu Jiang Youlin Qiao | 2022 | Chinese Journal of Cancer Research2022,34,4: | 1
