| 1 | Evaluation of rabbit antibody response against 8 and 16 kDa recombinant subunits of antigen B from Echinococcus granulosus显示文摘Objective:To immunize rabbits with 12 and 16 kDa recombinant subunits of antigen B from Echinococcus granulosus(E.granulosus) and measuring polyclonal antibody and humoral immune response using ELISA and gel diffusion.Methods:Two mentioned antigens were cloned and expressed in expression vector and purified by affinity chromatography.Four young rabbits were selected and challenged intradermallt with yielded recombinant antigens.Rabbits’ sera were collected post infection and were tested using ELISA and gel diffusion for polyclonal antibody detection 10 days after last injection.Results:The specific antibody against the recombinant peptides was efficiently produced within 4 weeks post infection.Conclusions: Produced recombinants proteins could induce the immune response of the rabbits successfully. | 2 | Molecular cloning of virB12 gene of Brucella melitensis 16M strain in pET28a vector显示文摘Objective:To clone the virB12 gene in pET28a expression vector for production of recombinant protein to be used as antigenic component for future serological test development.Methods: Brucella melitensis(B.melitensis) 16M strain was cultured and bacterial DNA was extracted by Bioneer AccuPrep~ Genomic DNA Extraction Kit.Oligonucleotide primer pair was designed based on Brucella virB12 gene sequence with BamHI and HindIII restriction site at 5’ end of the forward and reverse primers,respectively.DNA amplification was performed using PrimSTAR~ HS DNA polymerase and the PCK product was purified by DNA AccuPrepGel Purification Kit.Purified DNA was cloned into pJET1.2 cloning vector.VirB12 gene fragment was excised from pJET1.2 asing BamHI/HindIII and subsequendy subcloned into pET28a(+).Results:Brucella virB12 gene was successfully cloned in pJET1.2 and then in pET28a(+) plasmids.PCR and restriction enzyme digestion confirms the procedure.Conclusion:We cloned and expressed the Brucella virB12 gene which could be used as antigenic component for specific serological assay development. | Shiva Mirkalantari Nour Amirmozafari Bahram Kazemi Gholamreza Irajian | 2012 | Asian Pacific Journal of Tropical Medicine2012,5,7: | 2 |

| 4 | Influence of random heterogeneity of shear wave velocity on sliding mass response and seismic deformations of earth slopes显示文摘Soil shear wave velocity has been recognized as a governing parameter in the assessment of the seismic response of slopes.The spatial variability of soil shear wave velocity can influence the seismic response of sliding mass and seismic displacements.However,most analyses of sliding mass response have been carried out by deterministic models.This paper stochastically investigates the effect of random heterogeneity of shear wave velocity of soil on the dynamic response of sliding mass using the correlation matrix decomposition method and Monte Carlo simulation(MCS).The software FLAC 7.0 along with a Matlab code has been utilized for this purpose.The influence of statistical parameters on the seismic response of sliding mass and seismic displacements in earth slopes with different inclinations and stiffnesses subject to various earthquake shakings was investigated.The results indicated that,in general,the random heterogeneity of soil shear modulus can have a notable impact on the sliding mass response and that neglecting this phenomenon could lead to underestimation of sliding deformations. | Pourya Kazemi Esfeh Bahram Nadi Nicholas Fantuzzi | 2020 | Earthquake Engineering and Engineering Vibration2020,19,2: | 2 |

| 14 | Cloning and expression of Neisseria meningitides luxS gene显示文摘Neisseria meningitides is a Gram-negative bacterium which is an important causative agent of septicemia and meningitis.Numerous pathogenic bacteria contain luxS,which is required for autoinducer-2 production.Neisseria meningitides contains a functional copy of luxS that is necessary for full meningococcal virulence.Neisseria meningitides DNA was extracted and its luxS gene was amplified by nested PCR.PCR product was purified and cloned in to pQE-30 expression vector.Recombinant plasmid was transformed and mass cultured.luxS was expressed in E.coli and confirmed by western blot analysis.In this study,N.meningitides luxS was amplified, cloned and expressed successfully.The sequencing of PCR product confirmed that amplified gene was luxS.
