2Study on coupled 3D seepage and stress fields of the complex channel project显示文摘Seepage is a vital reason that may bring many adverse consequences such as subsidence,inclination and fracture to the channel,and is harmful to the safety and stability of the channel.Thus,a 3D visualization model is established for the engineering geological information of the research channel section by using NURBS-TIN-BRep hybrid data structure.Coupled with the VOF(volume of fluid)method,the N-S(Navier-Stokes)equations are applied to seepage simulation of the research channel section.Then the stability of the channel is studied coupled with the seepage simulation results,to comprehensively analyze the stress and displacement conditions of the channel under the impact of different factors such as seepage and underground goafs.The results of this study illustrate that the channel seepage has great influence on its stability,especially on the displacement field:it will lead to a significant sedimentation to the foundation.Therefore,during the practical construction,it is suggested that the certain part of the channel should be reinforced and effective seepage control measures should be taken.ZHONG DengHua ZHANG XiaoXin AO XueFei WANG XiaoLing TONG DaWei REN BingYu 2013Science China(Technological Sciences)2013,56,8:5
3Influence of MoSi_(2) on oxidation protective ability of TaB_(2)-SiC coating in oxygen-containing environments within a broad temperature range显示文摘TaB_(2)-SiC coating modified by different content of MoSi_(2) was fabricated on graphite substrate with SiC inner coating by liquid phase sintering to elevate the anti-oxidation capability of the TaB_(2)-SiC coatings.As compared to the sample with the TaB_(2)-40wt% SiC coating,the coating sample modified with MoSi_(2) exhibited a weight gain trend at lower temperatures,the fastest weight loss rate went down by 76%,and the relative oxygen permeability value reduced from about 1% to near 0.More importantly,the large amount of SiO_(2) glass phase produced over the coating during oxidation was in contact with the modification of MoSi_(2),which was proved to be beneficial to the dispersion of Ta-oxides.A concomitantly formed continuous Ta-Si-O-B compound glass layer showed excellent capacity to prevent oxygen penetration.However,when the TaB_(2) content was sacrificed to increase the MoSi_(2) content,the relative oxygen permeability of the coating increased instead of decreased.Thus,on the basis of ample TaB_(2) content,increasing the MoSi_(2) content of the coating is conducive to reducing the relative oxygen permeability of the coatings in a broad temperature region.Xuanru REN Junshuai LV Wei LI Yuwen HU Ke SUN Can MA Hong’ao CHU Weiguang WANG Leihua XU Ziyu LI Peizhong FENG 2020Journal of Advanced Ceramics2020,9,6:4
4Effects of ketamine on mouse hippocampal inflammatory cytokines IL-6,IL-1β and TNF-α levels in acute and chronic administration models显示文摘OBJECTIVE Ketamine is an injectable anesthetic and recreational drug of abuse commonly used worldwide. Many experimental studies have shown that ketamine can impair cognitive function and induce psychotic states. Neuroinflammation has been suggested to play an important role in neurodegeneration. Meanwhile,ketamine has been showed to modulate the levels of inflammatory cytokines.Therefore,we sought to investigate whether the effects of ketamine on the central nervous system is associated with the inflammatory cytokines. METHODS We established acute(single or multiple intraperitoneal injection) and chronic(six months daily intraperitoneal injection) ketamine administration models in C57BL/6 mice,evaluated the spatial recognition memory and emotional response by applying the Y maze test and open field test. We analyzed the changes of inflammatory cytokines interleukin-6(IL-6),interleukin-1β(IL-1β) and tumor necrosis factor-α(TNF-α) levels in mouse hippocampus,employing Western blot,quantitative reverse transcriptase-polymerase chain reaction(qR T-PCR) and immunohistochemistry. RESULTS Ketamine induced spatial recognition memory deficit,reduced anxiety-like behaviors in mice after chronic administration,and it was dose-dependent. Moreover,we found that ketamine could increase the levels of mouse hippocampal inflammatory cytokines IL-6 and IL-1β after single,multiple and long-term administration in a dose-dependent manner. However,the level of TNF-α expressed differently in mouse hippocampus under different conditions. Single administration of ketamine increased the level of TNF-α,whereas multiple and long-term administration decreased it significantly. We considered that TNF-α might exist bi-directional regulatory pathway,which was associated with the dose and duration of ketamine administration. 5An Inverse Power Generation Mechanism Based Fruit Fly Algorithm for Function Optimization显示文摘As a novel population-based optimization algorithm, fruit fly optimization(FFO) algorithm is inspired by the foraging behavior of fruit flies and possesses the advantages of simple search operations and easy implementation. Just like most population-based evolutionary algorithms, the basic FFO also suffers from being trapped in local optima for function optimization due to premature convergence.In this paper, an improved FFO, named IPGS-FFO, is proposed in which two novel strategies are incorporated into the conventional FFO. Specifically, a smell sensitivity parameter together with an inverse power generation mechanism(IPGS) is introduced to enhance local exploitation. Moreover,a dynamic shrinking search radius strategy is incorporated so as to enhance the global exploration over search space by adaptively adjusting the searching area in the problem domain. The statistical performance of FFO, the proposed IPGS-FFO, three state-of-the-art FFO variants, and six metaheuristics are tested on twenty-six well-known unimodal and multimodal benchmark functions with dimension 30, respectively. 6Energy-efficient recovery of tetrahydrofuran and ethyl acetate by triple-column extractive distillation:entrainer design and process optimization显示文摘An energy-efficient triple-column extractive distillation process is developed for recovering tetrahydrofuran and ethyl acetate from industrial effluent.The process development follows a rigorous hierarchical design procedure that involves entrainer design,thermodynamic analysis,process design and optimization,and heat integration.The computer-aided molecular design method is firstly used to find promising entrainer candidates and the best one is determined via rigorous thermodynamic analysis.Subsequently,the direct and indirect triple-column extractive distillation processes are proposed in the conceptual design step.These two extractive distillation processes are then optimized by employing an improved genetic algorithm.Finally,heat integration is performed to further reduce the process energy consumption.The results indicate that the indirect extractive distillation process with heat integration shows the highest performance in terms of the process economics.Ao Yang Yang Su Tao Shi Jingzheng Ren Weifeng Shen Teng Zhou 2022Frontiers of Chemical Science and Engineering2022,16,2:3
7A chromosome-scale genome assembly of a diploid alfalfa,the progenitor of autotetraploid alfalfa显示文摘Alfalfa(Medicago sativa L.)is one of the most important and widely cultivated forage crops.It is commonly used as a vegetable and medicinal herb because of its excellent nutritional quality and significant economic value.Based on Illumina,Nanopore and Hi-C data,we assembled a chromosome-scale assembly of Medicago sativa spp.caerulea(voucher PI464715),the direct diploid progenitor of autotetraploid alfalfa.The assembled genome comprises 793.2 Mb of genomic sequence and 47,202 annotated protein-coding genes.The contig N50 length is 3.86 Mb.This genome is almost twofold larger and contains more annotated protein-coding genes than that of its close relative,Medicago truncatula(420 Mb and 44,623 genes).The more expanded gene families compared with those in M.truncatula and the expansion of repetitive elements rather than whole-genome duplication(i.e.,the two species share the ancestral Papilionoideae whole-genome duplication event)may have contributed to the large genome size of M.sativa spp.caerulea.Comparative and evolutionary analyses revealed that M.sativa spp.caerulea diverged from M.truncatula~5.2 million years ago,and the chromosomal fissions and fusions detected between the two genomes occurred during the divergence of the two species.In addition,we identified 489 resistance(R)genes and 82 and 85 candidate genes involved in the lignin and cellulose biosynthesis pathways,respectively.The near-complete and accurate diploid alfalfa reference genome obtained herein serves as an important complement to the recently assembled autotetraploid alfalfa genome and will provide valuable genomic resources for investigating the genomic architecture of autotetraploid alfalfa as well as for improving breeding strategies in alfalfa.Ao Li Ai Liu Xin Du Jin-Yuan Chen Mou Yin Hong-Yin Hu Nawal Shrestha Sheng-Dan Wu Hai-Qing Wang Quan-Wen Dou Zhi-Peng Liu Jian-Quan Liu Yong-Zhi Yang Guang-Peng Ren 2020Horticulture Research2020,7,1:3
8Maternal intake restriction programs the energy metabolism, clock circadian regulator and m TOR signals in the skeletal muscles of goat offspring probably via the protein kinase A-cAMP-responsive element-binding proteins pathway显示文摘The biological mechanism by which maternal undernutrition increases the metabolic disorder risk of skeletal muscles in offspring is not fully understood.We hypothesize that maternal intake restriction influences metabolic signals in the skeletal muscles of offspring via a glucagon-mediated pathway.Twentyfour pregnant goats were assigned to the control group(100%of the nutrients requirement,n=12)and restricted group(60%of the control feed allowance from pregnant days 45 to 100,n=12).Blood and Longissimus thoracis muscle were sampled from dams(100 d of gestation),fetuses(100 d of gestation),and kids(90 d after birth)in each group.The data were analyzed using the linear MIXED model,with the multiple comparison method of SIDAK applied.Intake restriction reduced(P<0.05)the total blood protein of dams and fetuses.Maternal restriction decreased(P<0.05)the cAMP-responsive element-binding protein 1(CREB1),CREB-binding protein(CREBBP),protein kinase A(PKA),aryl hydrocarbon receptor nuclear translocator-like protein 1(BMAL1),protein kinase B(AKT1),mammalian target of rapamycin(mTOR),and regulatory-associated protein of mTOR(RPTOR)mRNA expression in the fetuses,and reduced(P<0.05)the CREBBP,nuclear receptor subfamily 1 group H member 3(NR1 H3),D-box binding PAR bZIP transcription factor(DBP)and PKA mRNA levels in the kids,but increased(P<0.05)the peroxisome proliferator-activated receptor gamma coactivator 1 alpha(PGC1 A)and tuberous sclerosis 2(TSC2)mRNA levels in the fetuses.The mRNA expression of clock circadian regulator(CLOCK)and TSC2 genes was increased(P<0.05)in the restricted kids.The protein expression of total PKA and phosphorylated PKA in the restricted fetuses and kids were downregulated(P<0.05),and the protein expression of total mTOR and phosphorylated mTOR were reduced(P<0.05)in the restricted fetuses and kids.Maternal intake restriction regulated fat oxidation,protein synthesis,and circadian clock expression in the muscles of the offspring probably via the glucagon-mediated PKA-CREB pathway,which reveals a noteworthy molecular pathway that maternal undernutrition leads to metabolic adaptation of skeletal muscle in offspring.Xiaoling Zhou Qiongxian Yan Hong Yang Ao Ren Zhixiong He Zhiliang Tan 2021Animal Nutrition2021,,4:2
9Deficiency of Tfh Cells and Germinal Center in Deceased COVID-19 Patients显示文摘Summary:The COVID-19 pandemic caused by SARS-CoV2 is characterized by a remarkable variation in clinical severity ranging from a mild illness to a fatal multi-organ disease.Understanding the dysregulated human immune responses in the fatal subjects is critical for management of COVID-19 patients and the pandemic.In this study,we examined the immune cell compositions in the lung tissues and hilar lymph nodes using immunohistochemistry on 6 deceased COVID-19 patients and 4 focal organizing pneumonia(FOP)patients who underwent lung surgery and served as controls.We found a dominant presence of macrophages and a general deficiency of T cells and B cells in the lung tissues from deceased COVID-19 patients.In contrast to the FOP patients,Tfh cells and germinal center formation were largely absent in the draining hilar lymph nodes in the deceased COVID-19 patients.This was correlated with reduced IgM and IgG levels compared to convalescent COVID-19 patients.In summary,our data highlight a defect of germinal center structure in deceased COVID-19 patients leading to an impaired humoral immunity.Understanding the mechanisms of this deficiency will be one of the key points for the management of this epidemic.Ya-qi DUAN Ming-hui XIA Liang REN Yan-fang ZHANG Qi-lin AO San-peng XU Dong KUANG Qian LIU Bing YAN Yi-wu ZHOU Qian CHU Liang LIU Xiang-Ping YANG Guo-ping WANG 2020Current Medical Science2020,40,4:2
10Effect of Tb Doping on Structural and Electrical Properties of BiFeO_3 Thin Films Prepared by Sol-Gel Technique显示文摘Bi_(1-x)Tb_xFeO_3 thin films were prepared on SnO_2(fluorine doped tin oxide) substrates by a sol—gel method.The structural and electrical properties of the BiFeO_3 thin films were characterized and tested.The results indicated that the diffraction peak of the Tb-doped BiFeO_3 films was shifted towards right as the doping amounts were increased.The structure was transformed from the rhombohedral to tetragonal/orthorhombic phase.The Bi_(0.89)Tb_(0.11)FeO_3 thin film showed the well-developed P—E loops,which enhanced remnant polarization(Pr = 88.05 μC/cm^2) at room temperature.The dielectric constant and dielectric loss of Bi_(0.89)Tb_(0.11)FeO_3 thin film at 100 kHz were 185 and 0.018,respectively.Furthermore,the Bi_(0.89)Tb_(0.11)FeO_3 thin film showed a relatively low leakage current density of 2.07 × 10^(-5) A/cm^2 at an applied electric field of 150 kV/cm.The Xray photoelectron spectroscopy(XPS) spectra indicated that the presence of Fe^(2+) ions in the Bi_(0.89)Tb_(0.11)FeO_3thin film was less than that in the pure BiFeO_3.Guohua Dong Guoqiang Tan Wenlong Liu Ao Xia Huijun Ren 2014Journal of Materials Science & Technology2014,30,4:2
11Exosomes secreted from cardiomyocytes suppress the sensitivity of tumor ferroptosis in ischemic heart failure显示文摘Heart failure(HF)patients in general have a higher risk of developing cancer.Several animal studies have indicated that cardiac remodeling and HF remarkably accelerate tumor progression,highlighting a cause-and-effect relationship between these two disease entities.Targeting ferroptosis,a prevailing form of non-apoptotic cell death,has been considered a promising therapeutic strategy for human cancers.Exosomes critically contribute to proximal and distant organ-organ communications and play crucial roles in regulating diseases in a paracrine manner.However,whether exosomes control the sensitivity of cancer to ferroptosis via regulating the cardiomyocyte-tumor cell crosstalk in ischemic HF has not yet been explored.Here,we demonstrate that myocardial infarction(MI)decreased the sensitivity of cancer cells to the canonical ferroptosis activator erastin or imidazole ketone erastin in a mouse model of xenograft tumor.Post-MI plasma exosomes potently blunted the sensitivity of tumor cells to ferroptosis inducers both in vitro in mouse Lewis lung carcinoma cell line LLC and osteosarcoma cell line K7M2 and in vivo with xenograft tumorigenesis model.The expression of miR-22-3p in cardiomyocytes and plasma-exosomes was significantly upregulated in the failing hearts of mice with chronic MI and of HF patients as well.Incubation of tumor cells with the exosomes isolated from post-MI mouse plasma or overexpression of miR-22-3p alone abrogated erastin-induced ferroptotic cell death in vitro.Cardiomyocyte-enriched miR-22-3p was packaged in exosomes and transferred into tumor cells.Inhibition of cardiomyocyte-specific miR-22-3p by AAV9 sponge increased the sensitivity of cancer cells to ferroptosis.ACSL4,a pro-ferroptotic gene,was experimentally established as a target of miR-22-3p in tumor cells.Taken together,our findings uncovered for the first time that MI suppresses erastin-induced ferroptosis through releasing miR-22-3p-enriched exosomes derived from cardiomyocytes.Therefore,targeting exosome-mediated cardiomyocyte/tumor pathological communication may offer a novel approach for the ferroptosis-based antitumor therapy.Ye Yuan Zhongting Mei Zhezhe Qu Guanghui Li Shuting Yu Yingqi Liu Kuiwu Liu Zhihua Shen Jiaying Pu Yanquan Wang Changhao Wang Zhiyong Sun Qian Liu Xiaochen Pang Ao Wang Zijing Ren Tong Wang Ying Liu Jinhuan Hong Jiajie Xie Xin Li Zhonghua Wang Weijie Du Baofeng Yang 2023Signal Transduction and Targeted Therapy2023,8,4:2

16The Establishment of Double-Transgenic Mice that Co-Express the appA and MxA Genes Mediated by Type A Spermatogonia In vivo显示文摘Type A spermatogonial stem cells are the only immortal diploid cells in the postnatal animal that undergo self-renewal through the lifetime of an animal and transmit genes to subsequent generations. In this paper, the generation and characterization of double-transgenic mice co-expressing the Escherichia coliappA gene and human MxA gene generated via the in vivo transfection of type A spermatogonial cells were reported for the first time. The dicistronic expression vector pc DNA-app A-MxA(AMP) and ExGen500 transfection reagent were injected into the testicular tissue of 7-d-old male ICR mice. The mice that underwent testismediated gene transfer were mated with wild-type female mice, and the integration and expression of the foreign genes in the offspring were evaluated. Transgenic mice that co-expressedappA and MxA showed a gene integration rate of 8.89%(16/180).
