1Antitumor effects of vaccine consisting of dendritic cells pulsed with tumor RNA from gastric cancer显示文摘AIM:To investigate the immunotherapeutic potential of vaccine consisting of dendritic cells (DCs) pulsed with total RNA from MFC gastric cancer cells.METHODS:DCs were prepared from the spleens of strain 615 mice by magnetic cell sorting (MACS). Alter culture for 24h, DCs were pulsed with total RNA from MFC gastric cancer cells. Mice of one group were immunized with tumor RNA pulsed DC (RNA/DC) at the dosage of 1×10^6 on d 14 and 7 by s c inoculation before tumor implantation. Mice of another group were immunized with unpulsed DC (UDC) at the same dosage on days as the RNA/DC group. The third group of control mice was untreated. On d 0, all the mice were challenged with s c injections of 5×10^5 MFC gastric cancer cells.Alter inoculation, the mice were monitored closely with respect to tumor growth. Activities of NK cells in PBL and splenocytes and CTL were tested.RESULTS:On d 21 alter tumor cell inoculation, the mice of control group manifested the largest tumors with volume at a mean of 2.6323±1.1435cm^3, followed by the UDC and RNA/DC groups with mean volumes at 0.7536±0.3659cm^3 and 0.3688±0.6571cm^3, respectively. The activities of NK cells in PBL and splenocytes in RNA/DC group were 66.2% and 65.4%, respectively, higher than that in the control group. 2进展期胃癌病理和预后影响因素的关系显示文摘目的:研究机体免疫因素和肿瘤生物学行为与进展期胃癌病理之间的相关性,比较并筛选对进展期胃癌预后有影响的指标。方法:获得61例行根治性切除术的进展期胃癌预后资料,用免疫组化(SABC法)和TUNEL(原位末端标记)方法检测组织切片中胃癌细胞增生细胞核抗原标记指数(PCNA-LI)、树突状细胞(DCs,dendritic cells)浸润密度,凋亡指数(AI,apoptosis index),研究上述指标和病理之间的相关性并进行单因素、多因素生存分析。结果:PCNA-LI,DCs,AI与TNM分期、淋巴结转移、淋巴管浸润、胃癌分化有相关性。单因素生存分析表明胃癌患者的年龄、性别、病灶的大小、肿瘤分化、是否有淋巴管癌栓、AJ等与胃癌预后无关。胃癌分期、浸润深度、淋巴结转移、PCNA—LI、DCs浸润密度是胃癌预后的影响因素(Log-rank test,P<0.05)。多参数回归生存分析表明肿瘤大小(RR=2.328),UICC(1997年)TNM分期(RR=5.251)是胃癌预后的独立影响因素(P<0.05)。结论:UICC的胃癌TNM分期是判断胃癌预后有价值的指标。胃癌细胞本身生物学特性和机体的免疫状态与胃癌病理有一定相关性并且参与了对胃癌预后的影响。黄海力 吴本俨 尤纬缔 申明识 2003世界华人消化杂志2003,11,9:12
3创伤早期患者外周血树突状细胞的变化及临床意义显示文摘目的 探讨创伤后早期患者外周血树突状细胞 (DC)变化及临床意义。方法 分离创伤患者 ( 2 7例 ,创伤组 )和健康人 ( 12例 ,对照组 )外周树突状细胞 ;通过流式细胞仪检测各组的DC数量 (CMRF 44标记法 )及DC表面HLA DR、CD80、CD86表达水平以及DC诱导的T细胞反应性增殖。检测各组外周血上清中白细胞介素 6(IL 6)、IL 10的浓度。结果 创伤组DC细胞数( 7.9± 3 .2 )× 10 6/L明显低于对照组DC ( 14 .9± 5 .1)× 10 6/L(P <0 .0 1)。创伤组DC表面HLA DR及CD80、CD86的表达水平与对照组相比明显下调 (P <0 .0 1)。DC诱导的T细胞增殖能力对照组明显强于创伤组 (P <0 .0 1)。在创伤组中血清IL 6、IL 10的浓度 ( 2 .42± 0 .3 3 ) μg/L和( 1.49± 0 .2 7) μg/L显著升高 ,与对照组比较差异有非常显著性 (P <0 .0 1)。 结论 创伤早期患者外周血DC数量少 ,功能低下 ,与创伤后的免疫功能低下关系密切。白祥军 唐朝晖 邹声泉 邱文洪 2003中华实验外科杂志2003,20,11:10
4Functional changes of dendritic cells derived from allogeneic partial liver graft undergoing acute rejection in rats显示文摘AIM: To investigate functional change of dendritic cells (DCs)derived from allogeneic partial liver graft undergoing acuterejection in rats.METHODS: Allogeneic (SD rat to LEW rat) whole and 50 %partial liver transplantation were performed. DCs from livergrafts 0 hr and 4 days after transplantation were isolated andpropagated in the presence of GM-CSFin vitro. Morphologicalcharacteristics of DCs propagated for 4 days and 10 dayswere observed by electron rmicroscopy. Phenotypical featuresof DCs propagated for 10 days were analyzed by flowcytometry. Expression of IL-12 protein and IL-12 receptormRNA in DCs propagated for 10 days was also measured byWestern blotting and semiquantitative RT-PCR, respectively.Histological grading of rejection were determined.RESULTS: Allogeneic whole liver grafts showed no featuresof rejection at day 4 after transplantation. In contrast,allogeneic partial liver grafts demonstrated moderate tosevere rejection at day 4 after transplantation. DCs derivedfrom allogeneic partial liver graft 4 days after transplantationexhibited typical morphological characteristics of DC after 4days' culture in the presence of GM-CSF. DCs from allogeneicwhole liver graft 0 hr and 4 days after transplantation didnot exhibit typical morphological characteristics of DC untilafter 10 days' culture in the presence of GM-CSF. 5NF-κB activation and zinc finger protein A20 expression in mature dendritic cells derived from liver allografts undergoing acute rejection显示文摘AIM: To investigate the role of NF-κB activation and zinc finger protein A20 expression in the regulation of maturation of dendritic cells (DCs) derived from liver allografts undergoing acute rejection. METHODS: Sixty donor male SD rats and sixty recipient male LEW rats weighing 220-300 g were randomly divided into whole liver transplantation group and partial liver transplantation group. Allogeneic (SD rat to LEW rat) whole and 50 % partial liver transplantation were performed. DCs from liver grafts 0 hour and 4 days after transplantation were isolated and propagated in the presence of GM-CSF in vitro. Morphological characteristics and phenotypical features of DCs propagated for 10 days were analyzed by electron microscopy and flow cytometry, respectively. NF-κB binding activity, IL-12p70 protein and zinc finger protein A20expression in these DCs were measured by EMSA and Western blotting, respectively. Histological grading of rejection was determined. RESULTS: Allogeneic whole liver grafts showed no signs of rejection on day 4 after the transplantation. In contrast,allogeneic partial liver grafts demonstrated moderate to severe rejection on day 4 after the transplantation. After propagation for 10 days in the presence of GM-CSF in vitro,DCs from allogeneic whole liver grafts exhibited features of immature DC with absence of CD40 surface expression,these DCs were found to exhibit detectable but very low level of NF-κB activity, IL-12 p70 protein and zinc finger protein A20 expression. Whereas, DCs from allogeneic partial liver graft 4 days after transplantation displayed features of mature DC, with high level of CD40 surface expression, and as a consequence, higher expression of IL-12p70 protein, higher activities of NF-κB and higher expression of zinc finger protein A20 compared with those of DCs from whole liver grafts (P<0.001). CONCLUSION: These results suggest that A20expression is up-regulated in response to NF-κB activation in mature DCs derived from allogeneic liver grafts undergoing acute rejection. 7小鼠IL-18基因修饰瘤苗的抗白血病作用研究显示文摘我们在前期工作中成功制备了IL-18基因修饰的白血病疫苗,为了探讨IL-18基因修饰瘤苗的体内抗白血病作用,实验采用L1210小鼠淋巴细胞白血病模型,在小鼠体内接种IL-18基因修饰瘤苗,观察瘤苗对L1210细胞致瘤性的影响及免疫保护作用,并进一步对其抗白血病作用机制进行了探索。结果显示,IL-18基因修饰瘤苗能够明显延长荷瘤小鼠存活时间,大部分小鼠达到长期生存,且长生存小鼠用野生型L1210细胞二次攻击后大多数仍能长期生存,表明IL-18基因修饰瘤苗有显著的抗白血病作用,并可诱导小鼠产生免疫记忆和免疫保护。机制探讨发现,接种IL-18基因修饰瘤苗后,小鼠脾脏淋巴细胞对L1210肿瘤细胞的CTL及NK细胞杀伤活性明显高于对照组(P<0.05),提示IL-18基因修饰瘤苗能够显著增强抗肿瘤CTL和NK细胞反应。接种瘤苗可使小鼠IFN-γ水平升高,但与对照相比无统计学意义,提示IFN-γ可能在IL-18基因修饰瘤苗诱导的抗肿瘤免疫应答中作用不大。徐玢 马小彤 董成亚 石洋 林永敏 吴克复 2007现代免疫学2007,27,4:6
8β-catenin和Cyclin D1在肝癌肝内转移中的作用显示文摘目的:探讨β-catenin和 Cyclin D1与癌栓形成及临床病理参数的关系.方法:用免疫组化法检测32例肝癌原发灶、癌栓和癌旁组织中β-catenin、Cyclin D1的表达,同时分析其与临床病理参数之间的关系.结果:β-catenin在原发灶、癌栓和癌旁组织中的阳性率分别为62.5%、81.25%、9.38%,三者之间有显著差别;β-catenin与癌栓形成、HBV感染、肝硬化有关.CyclinD1在原发灶、癌栓和癌旁组织中的阳性率分别为46.9%、75%、18.8%,三者之间有显著差别;Cyclin D1与癌栓形成、组织类型、分化程度有关.结论:β-catenin和 Cyclin D1促进癌栓形成,参与HBV感染和肝硬化形成过程;Cyclin D1影响肿瘤组织类型和分化程度.苏小康 赵先明 李锦清 崔学教 谢晓华 杨海燕 徐发彬 石明 2003世界华人消化杂志2003,11,9:5
9Continuous release of interleukin 12 from microencapsulated engineered cells for colon cancer therapy显示文摘AIM: To explore the anti-tumor immunity against CT26 colon tumor of the microencapsulated cells modified with murine interleukine-12 (mIL-12) gene.METHODS: Mouse fibroblasts (NIH3T3) were stably transfected to express mIL-12 using expression plasmids carrying mIL-12 gene (p35 and p40), and NIH3T3-mIL-12cells were encapsulated in alginate microcapsules for longterm delivery of mIL-12. mIL-12 released from the microencapsulated NIH3T3-mIL-12 cells was confirmed using ELISA assay. Transplantation of the microencapsulated NIH3T3-mIL-12 cells was performed in the tumor-bearing mice with CT26 cells. The anti-tumor responses and the anti-tumor activities of the microencapsulated NIH3T3-mIL12 cells were evaluated.RESULTS: Microencapsulated NIH3T3-mIL-12 cells could release mIL-12 continuously and stably for a long time. After the microencapsulated NIH3T3-mIL-12 cells were transplanted subcutaneously into the tumor-bearing mice for 21 d, the serum concentrations of mIL-12, mIL-2 and mIFN-γ the cytotoxicity of the CTL from the splenocytes and the NK activity in the treatment group were significantly higher than those in the controls. Moreover, mIL-12 released from the microencapsulated NIH3T3-mIL-12 cells resulted in a significant inhibition of tumor proliferation and a prolonged survival of tumor-bearing mice.CONCLUSION: The microencapsulated NIH3T3-mIL-12cells have a significant therapeutic effect on the experimental colon tumor by activating anti-tumor immune responses in vivo. 10癌细胞裂解物修饰的DC疫苗体外诱导T细胞特异性抗胰癌免疫显示文摘目的 观察经胰癌细胞裂解物修饰的树突状细胞 (DC )疫苗 ,在体外诱导抗胰癌的特异性T细胞免疫应答的效果。方法 从胰腺癌患者外周血中分化、增殖出DC ,经胰癌细胞裂解物修饰 ,并与T细胞体外共培养 (分为致敏DC组、未致敏DC组、肿瘤裂解物组、对照组 ) ,检测了各组上清液中细胞因子IL 12与IFN γ的浓度和T细胞的反应性增殖 ,并评估激活的T细胞对胰腺癌肿瘤细胞的特异性细胞毒作用。结果 致敏DC组中IL 12与IFN γ的浓度 [(1161± 2 39) pg/ml和 (10 44± 312pg/ml) ]与未致敏DC组及肿瘤裂解物组相比差异有极显著性 (P <0 .0 5 ,P <0 .0 1) ;在致敏DC组中T细胞有明显的增殖 ;激活的T细胞对胰腺癌细胞有高效、特异的细胞毒作用。结论 癌细胞裂解物致敏的DC可高效的诱发机体T细胞 (Th1和CTL )唐朝晖 邹声泉 邱文洪 杨想平 裘法祖 2003中国普通外科杂志2003,12,4:4
11热休克肿瘤细胞抗原负载的树突状细胞瘤苗对原位接种结肠肿瘤的免疫预防作用显示文摘目的:研究热休克肿瘤细胞抗原负载的树突状细胞的体内抗肿瘤作用以及对原位接种结肠肿瘤小鼠的免疫保护作用. 方法:肿瘤细胞CT-26经热休克处理后制备冻融抗原,体外负载树突状细胞,免疫同源小鼠(热休克组).以IFN-γELISPOT实验、LDH释放实验检测小鼠体内肿瘤特异性T细胞免疫反应,以原位接种结肠肿瘤的体积以及荷瘤小鼠的生存时间来评价该树突状细胞瘤苗对结肠原位接种CT-26小鼠的免疫保护作用,并与非热休克肿瘤细胞抗原负载的树突状细胞的免疫效果(非热休克组)作一比较. 结果:热休克处理肿瘤细胞可提高肿瘤细胞抗原负载的树突状细胞表面MHC-Ⅱ类分子(66.3% vs 59.1%)、共刺激分子CD86的表达(39.4% vs 36.7%);热休克组小鼠脾淋巴细胞中肿瘤特异性CTL数量高于非热休克组(P=0.001),相同效靶比下前者肿瘤特异性细胞毒活性强于后者:热体克组与非热休克组小鼠原位接种结肠肿瘤14 d后的肿瘤体积均明显小于单纯DC免疫组(P=0,000),但HSCT-26DC免疫组与CT-26 DC免疫组间差异无显著性(P=0.480).单纯DC免疫组50% (3/6)可见腹膜转移,CT-26 DC免疫组以及HSCT- 26DC免疫组均未见腹腔转移;热休克组小鼠荷瘤生存时间显著长于非热休克组(P=0.0384). 结论:热休克处理肿瘤细胞可以提高肿瘤细胞抗原负载的树突状细胞的体内抗肿瘤作用,热休克肿瘤细胞抗原负载的树突状细胞瘤苗对原位接种结肠肿瘤小鼠具有良好的免疫保护作用.邱健 李国威 隋延仿 宋宏萍 司少艳 葛伟 2005世界华人消化杂志2005,13,5:4
12小鼠白介素18与白介素1β转化酶共表达载体的构建及真核表达显示文摘为了制备白介素18前体(proIL-18)和白介素1β转化酶(ICE)共表达肿瘤疫苗,从小鼠脾细胞克隆proIL-18和ICE基因,构建小鼠proIL-18和ICE共表达质粒(pIRES-proIL-18-ICE),经测序证实序列正确后,脂质体法转染小鼠淋巴细胞白血病细胞系L1210,筛选稳定表达细胞株。转染细胞RT-PCR及转染细胞培养上清IL-18ELISA检测结果表明,肿瘤细胞可以表达并分泌成熟的IL-18。培养上清的IL-18可显著诱导ConA刺激的小鼠脾细胞产生IFN-γ,表明其具有功能活性。生长曲线和细胞周期实验证实与未转染的肿瘤细胞相比,转染空载体pIRES、pIRES-proIL-18、pIRES-ICE、pIRES-proIL-18-ICE基因对肿瘤细胞生长增殖无明显影响。IL-18白血病瘤苗的成功制备表明,利用pIRES载体在同一肿瘤细胞中共表达的proIL-18和ICE能够完全模拟生理过程,在真核细胞内有效酶切,分泌出成熟并有功能活性的IL-18;构建的IL-18瘤苗为进一步评价其在小鼠体内的抗白血病效果奠定基础。徐玢 马小彤 董成亚 石洋 林永敏 宋玉华 吴克复 2005现代免疫学2005,25,3:4
13比较两类树突状细胞激活的肿瘤特异性CTL对肝癌荷瘤裸鼠的治疗作用显示文摘目的研究人外周血不同来源的树突状细胞(dendritic cells,DC)光镜形态学、细胞超微结构及体内外诱导抗肿瘤免疫反应的差异。方法联合应用粒/巨噬细胞集落刺激因子(GM-CSF)、白介素-4(IL-4)及肿瘤坏死因子(TNFα)从人外周血中培养出两类DC;人肝癌细胞系SMMC-7721细胞的可溶性肿瘤抗原刺激两类DC;两类DC激活同源的T淋巴细胞;观察两类DC激活的T淋巴细胞分别抑制已接种于裸鼠的人肝癌细胞系SMMC-7721移植瘤生长。结果细胞因子诱导的人外周血DC群可见到两类DC亚群:一类可能来自DC前体细胞,另一类是由单核细胞分化来的DC;两类DC体外激活的T淋巴细胞能分别抑制肝癌细胞系SMMC-7721裸鼠移植瘤生长。结论细胞因子诱导的人外周血DC群存在多相性,两类DC亚群体内外诱导的抗肿瘤细胞免疫反应不仅能抑制肝癌细胞系SMMC-7721裸鼠移植瘤发生,而且抑瘤率不同,提示两类DC亚群在肝癌的治疗中发挥着不同的作用。张希国 师建国 刘彦仿 阎庆国 2006肿瘤防治研究2006,33,7:3
14肝癌细胞抗原致敏树突状细胞体外诱导特异性细胞毒T淋巴细胞显示文摘目的:探讨原发性肝癌患者外周血树突状细胞(DC)体外经自体肝癌细胞抗原致敏后,对特异性细胞毒T淋巴细胞(CTL) 的诱导作用. 方法:肝癌患者外周血经梯度密度离心法分离,获得DC前体细胞,用重组人粒细胞-巨噬细胞集落刺激因子(rhGM- CSF)和重组人白介素-4(rhIL-4)联合培养,诱导扩增DC. 制备自体肝癌细胞抗原,体外脉冲DC,检测DC诱导自体T细胞增生能力及特异性CTL在体外对自体肝癌细胞的杀伤活性. 结果:经自体肝癌细胞抗原致敏的DC能诱导较强的自体T 细胞增生,且能诱导特异性CTL,该CTL,对自体肝癌细胞具有很强的杀伤活性,杀伤率明显高于DC、未经肝癌细胞抗原致敏的DC激活的CTL及T淋巴细胞的杀伤率,而对CT26细胞、BEL-7402细胞无明显的杀伤作用. 结论:肝癌患者外周血DC体外能诱导高效而特异的抗肝癌免疫,提示DC可能在肿瘤治疗及预防其复发与转移中发挥重要作用.谢裕安 罗小玲 梁安民 匡志鹏 吴继宁 2004世界华人消化杂志2004,12,6:3

17DC-SIGN:Binding receptor for HCV?显示文摘DC-SIGN, a dendritic Cell-specific adhesion receptor and a type Ⅱ transmembrane mannose-binding C-type lectin, is very important in the function of DC, both in mediating naive T cell interactions through ICAM-3 and as a rolling receptor that mediates the DC-specific ICAM-2-dependent migration processes. It can be used by viral and bacterial pathogens including Human Immunodeficiency Virus (HIV), HCV, Ebola Virus, CMV and Mycobacterium tuberculosis to facilitate infection. Both DC-SIGN and DC-SIGNR can act either in cis, by concentrating virus on target cells, or in bans, by transmission of bound virus to a target cell expressing appropropriate entry receptors. Recent work showed that DC-SIGN are highaffinity binding receptors for HCV. Besides playing a role in entry into DC, HCV E2 interaction with DC-SIGN might also be detrimental for the interaction of DC with T cells during antigen presentation. 19大鼠胰腺癌细胞裂解物修饰的树突状细胞疫苗在大鼠胰腺癌模型中的抗肿瘤作用显示文摘目的:观察经大鼠胰腺癌细胞裂解物修饰的树突状细胞(DC)疫苗对大鼠胰腺癌皮下移植瘤的免疫治疗作用方法:大鼠胰腺癌细胞注入SD大鼠胰腺被膜下,形成肿瘤,将肿瘤取出后切碎,植入同种大鼠的腋窝皮下,建立皮下移植瘤模型;将大鼠胰腺癌细胞反复冻融4次,提取出裂解物,与SD大鼠骨髓来源的DC共培养72 h制成DC疫苗。根据DC致敏情况分为DC疫苗组、DC组、裂解物组与对照组,分别注入大鼠皮下移植瘤体内,每组5只。每两天测一次肿瘤长径与横径,共8次,观察大鼠的生存期。结果:随着时间延长,DC疫苗组肿瘤大小呈递减趋势;DC组、裂解物组、对照组肿瘤大小呈递增趋势;DC疫苗组与对照组之间肿瘤大小差异有统计学意义(P<0.05);DC组、裂解物组与对照组之间肿瘤大小差异无统计学意义(P>0.05);DC疫苗组与DC组、裂解物组之间肿瘤大小差异有统计学意义(P<0.05);DC疫苗组大鼠生存期为(55.6±9.4)d,明显长于其他各组(P<0.01)。结论:大鼠胰腺癌细胞裂解物修饰的DC疫苗有可能诱导胰腺癌皮下移植瘤大鼠产生高效的免疫应答反应,发挥抗肿瘤作用。娄月红 张海龙 梁宗锁 2011中国肿瘤临床2011,38,16:2
