1三种不同乳酸菌对凡纳滨对虾生长、抗病力及肠道菌群结构的影响显示文摘本研究比较了饲料中添加粪肠球菌(Enterococcus faecalis LYB1)、嗜酸乳杆菌(Lactobacillus acidophilus W2)和凝结芽孢杆菌(Bacillus coagulans NJ17)对凡纳滨对虾(Litopenaeus vannamei)幼虾的生长、抗病力及肠道菌群结构的影响。以基础饲料中分别添加1.0×1010 cfu/kg浓度的3种不同乳酸菌为实验组,以基础饲料为对照组,以投喂方式养殖对虾42 d。养殖结束后,进行14 d的副溶血弧菌(Vibrio parahaemolyticus)攻毒实验。研究表明:投喂3种乳酸菌的实验组对虾的体重、特定生长率、饲料效率和表观消化率均显著高于对照组(P<0.05),其中以添加凝结芽孢杆菌组效果最佳。攻毒实验显示,添加粪肠球菌和嗜酸乳杆菌均可显著降低对虾的累计死亡率(P<0.05)。添加乳酸菌的各实验组对虾的肠道细菌群落结构与对照组相比均发生了明显变化,其中以添加粪肠球菌组差异最大。在门水平上,变形菌门(Proteobacteria)、厚壁菌门(Firmicutes)和拟杆菌门(Bacteroidetes)为对虾肠道优势菌群。在纲分类水平上,α-变形菌纲(Alphaproteobacteria)、丹毒丝菌纲(Erysipelotrichia)和黄杆菌纲(Flavobacteriia)为对虾肠道优势菌群。研究结果显示,凝结芽孢杆菌NJ17对凡纳滨对虾促长效果最佳,而嗜酸乳杆菌W2对对虾抗病力提高效果最佳,3种乳酸菌均明显改善了对虾肠道微生物群落的组成和丰度。李佳徽 王明阳 田相利 刘龙镇 李海东 李丽 董双林 薛玲 2021中国海洋大学学报(自然科学版)2021,51,4:12
2养殖刺参早期发育阶段体内可培养细菌的菌群特征及其与环境菌群相关性分析显示文摘采用传统细菌培养方法,对养殖刺参(Apostichopus japonicus)早期发育各阶段幼体体内及环境(投入饵料及培育用水)菌群的组成与结构展开研究,对分离的优势细菌进行分子鉴定,在此基础上,进行了刺参幼体体内菌群结构与环境菌群结构相关性分析。幼体各发育期的细菌培养结果显示,在幼体开口前的各发育时期(性腺、卵、受精卵、原肠胚)均无可培养细菌,在投饵以后,耳状幼体、樽形幼体体内可分离到可培养细菌,幼体发育到稚参以后,消化道可培养细菌总数急剧增加,并在4月龄时达到10~8 CFU/g数量级。在幼体体内可培养细菌中,弧菌(Vibrio)占比为2.2%~77.3%。对环境菌群的细菌培养结果显示,培育用水中细菌含量变化不显著,随着幼体发育期饵料的转变,不同时期饵料中细菌含量差异显著。整个养殖系统中共分离到65株优势细菌,16S rDNA鉴定结果显示,所分离的65株优势菌鉴定为14个属43种细菌。相关性分析结果显示,随着幼体的发育,生物饵料中的细菌对消化道中的菌群结构影响越来越大。本研究结果为解析刺参消化道菌群的形成过程和演替规律以及养殖用益生菌的筛选与应用奠定了基础。李靖 廖梅杰 李彬 王印庚 荣小军 张正 范瑞用 刘安然 2019渔业科学进展2019,40,2:5
3广西涠洲岛3种海参共附生细菌多样性及其延缓衰老活性分析显示文摘【目的】明确广西涠洲岛海参共附生细菌多样性,筛选出具有延缓衰老活性的菌株,为挖掘延缓衰老活性的新化合物提供菌种资源。【方法】选用9种选择性培养基对来源于广西涠洲岛的3种海参[异色哈威参(Havelockia versicolor)、玉足海参(Holothuria leucospilota)和方柱翼手参(Colochirus quadrangularis)]共10份样品进行共附生细菌分离纯化,基于16S rRNA测序分析其多样性,并以秀丽隐杆线虫(Caenorhabditis elegans)为模型评价共附生细菌发酵粗提物的延缓衰老活性。【结果】从广西涠洲岛3种海参中共分离获得63株共附生菌株,隶属于3门4纲14目25科35属,以放线菌纲(Actinobacteria)为优势菌群,共有31株(占49.21%);放线菌GXIMD LM0085和GXIMD LM0159有可能为潜在新种,与对应有效发表菌株Kocuria assamensis、K. salsicia的相似性分别为97.38%和97.00%。不同培养基分离获得的海参共附生细菌在属类水平上存在明显的数量差异,以M9培养基分离获得的菌属数量最多(19属28株),P7培养基分离获得的最少(仅6属6株);在筛选确定的35属细菌中,仅Cobetia和嗜盐单胞菌属(Halomonas)在9种培养基均有生长分布,多达13个属仅在单一的培养基中生长。不同海参分离获得的共附生细菌数量排序为1#异色哈威参>方柱翼手参>2#异色哈威参>玉足海参,除了玉足海参外,其他3只海参体内的共附生放线菌相对丰度明显高于体表共附生放线菌相对丰度。线虫寿命试验发现,有6株共附生细菌的发酵粗提物能显著延长秀丽隐杆线虫寿命(P<0.05),分别是GXIMD LM0037(寡养单胞菌属)、GXIMD LM0005(链霉菌属)、GXIMD LM0071(副球菌属)、GXIMD LM0114(寡养单胞菌属)、GXIMD LM0099(原小单孢菌属)和GXIMD LM0032(短状杆菌属)。【结论】广西涠洲岛海域海参蕴含着多样性丰富的共附生细菌资源,具有潜在的新物种,且部分菌株具有延缓衰老活性,在延缓衰老活性物质挖掘及新型抗衰老药物研发方面具有巨大潜力。银江林 李蜜 易湘茜 刘永宏 于清武 高程海 2021南方农业学报2021,52,10:2
4Development of Fatty Acid Biomarkers for the Identification of Wild and Aquacultured Sea Cucumber (Apostichopus japonicus)显示文摘In this study,the fatty acids(FAs) of the organs and tissues of sea cucumber(Apostichopus japonicus) were profiled in order to compare the FA composition of sea cucumber collected from natural habitat(wild) and cages(cultured).The differences in FA contents in dermomuscular tube,peripharyngeal annulus,gonad and intestine(with or without content) between the wild and the cultured were determined.The main fatty acids in all organs and tissues were 20:5n-3,16:1n-7,20:4n-6,22:6n-3,18:0,and 18:1n-7.The basically different FAs of body wall and digestive tube were 16:1n-7,18:1n-9 and 20:1n-11.The ratio of saturated to mono-and polyunsaturated FAs in digestive tube was independent on inside content while there was a redistribution of the total amount of n-3 and n-6 fatty acids.The comparison of FA composition of the wild and the cultured sea cucumber showed that 20:5n-3,16:1n-7 and 18:1n-7 predominated the wild while 20:4n-6 predominated the cultured.The content of branched-chain fatty acids in the wild was 3%–4% and about 9% in the cultured.The possible FAs for identifying the wild and the cultured sea cucumbers were selected.It was suggested that the indexes such as the ratio of either(n-3:n-6) to(n-7:n-6) or(n-3) +(n-7) to(n-6) may serve as the biomarkers distinguishing the wild and the cultured sea cucumber.P. 5养水机对刺参池塘理化参数和细菌丰度的影响显示文摘养水机是一套能打破温、盐跃层的水质改良设备。为评价养水机对池塘水质的改良效果,该研究于大连庄河刺参(Apostichopus japonicus)养殖场,以自然纳潮和微孔曝气池塘为对照,对养水机池塘8项理化参数[温度、盐度、pH、溶解氧(DO)、总氨氮(TAN)、亚硝态氮(NO_(2)^(−)-N)、活性磷酸盐(PO_(4)^(3+)-P)和沉积物有机质(TOM)]以及环境中活的异养菌、可培养弧菌的周年动态进行了监测,旨在对养水机净化水质效能进行评价。结果表明,与对照组相比,养水机池塘水中TAN、NO_(2)^(−)-N质量浓度在3—11月均最低(NO_(2)^(−)-N质量浓度6月除外),PO_(4)^(3+)-P质量浓度在3—8月最高,沉积物中TOM各月质量分数均最低(P<0.05),在温、盐跃层期,养水机能显著提高水体溶解氧质量浓度(P<0.05);与微孔曝气池塘相比,在8~9个月期间养水机均能促进池塘中活的异养菌生长,抑制弧菌生长。养水机在春、夏、秋季具有改善刺参养殖池塘水质和显著降低水中弧菌的良好效果,以及显著降低全年沉积物TOM的能力,在刺参养殖场具有较好的应用前景。张东升 王祖峰 周玮 2021南方水产科学2021,17,6:1
6Comparative analysis on microbial community associated with different gastrointestinal regions of wild northern snakehead Channa argus Cantor, 1842显示文摘Microbial communities in different gastrointestinal regions(stomach, foregut, midgut, and hindgut) of the northern snakehead C hanna argus(Cantor, 1842) were compared by polymerase chain reaction and partial 16 S r DNA sequencing. A total of 194, 140, 212, and 122 OTUs were detected in the stomach, foregut, midgut, and hindgut, respectively. Significant dif ferences were found in the Sobs, ACE, Shannon, and Simpson indices among samples(P <0.05). The gastrointestinal microbial community of C. argus consisted predominantly of Proteobacteria with either H alomonas, Shewanella, Plesiomonas, or Sphingomonas. Fusobacteria, Firmicutes, and Bacteroidetes also existed in the gastrointestinal tracts. However, significant diff erences were found in the compositions of microbial community among the four regions( P <0.05). Cyanobacteria and Spirochetes were significantly higher in the midgut and hindgut( P <0.05). Fusobacteria and Firmicutes were dominant in the hindgut and foregut, respectively(P <0.05). Proteobacteria was the lowest in the hindgut( P <0.05). At genus level, C etobacterium and Plesiomonas were significantly higher in the hindgut than in the other three samples( P <0.05). Clostridium and Prevotella were the highest in the midgut(P <0.05). Halomonas, Shewanella, and S phingomonas were the highest in the foregut( P <0.05). Paracoccus and Vibrio were the highest in the stomach. Several genera were only detected in certain regions, as follows: stomach, P aracoccus and Vibrio; foregut, Halomonas, Shewanella, and Sphingomonas; midgut, Clostridium and Prevotella; and hindgut, C etobacterium and Plesiomonas( P <0.05). At the species level, A cinetobacter rhizosphaerae was only detected in the stomach. P revotella copri and Clostridium perfring were not detected in the foregut and midgut, respectively, whereas Prevotella copri and Faecalibacterium pra were not detected in the hindgut. These findings provide valuable information on the microbial community in each gastrointestinal region of C. argus. 7Bacterial community composition in gut content and ambient sediment of two tropical wild sea cucumbers(Holothuria atra and H.leucospilota)显示文摘Sea cucumbers play an important role in sediment bioturbation in coral reef and rocky intertidal ecosystems,and bacteria were a commonly-reported component of holothuroid diets.Bacterial community composition in the foregut and hindgut of two common tropical sea cucumbers(Holothuria atra and H.leucospilota)and the ambient surface sediment were investigated using high throughput sequencing based on 16S rRNA gene analysis.A total of 5584 operational taxonomic units(OTUs)were obtained from 25 samples based on a 97%threshold,and more than half of the OTUs(n=3694,66.2%)were shared by the gut contents of two species of sea cucumbers and surrounding sediments.Bacterial richness and diversity in sediment samples were signifi cantly higher than those in the gut content samples(P<0.05).Proteobacteria was the predominant phylum in most samples showing 45.69%±8.61%,70.09%±4.03%,45.88%±5.38%,and 55.19%±0.79%reads in the foregut of H.leucospilota,hindgut of H.leucospilota,hindgut of H.atra,and sediment libraries,respectively,but Bacteroidetes was the predominant phylum with the relative content of 34.98%±5.52%in the foregut of H.atra.Among the dominant genera,reads related to the genera Anderseniella,Ilumatobacter,and Ruegeria were detected in all the gut contents and sediment libraries.A comparison of gut bacteria community between the two species of sea cucumbers suggested that H.atra had stronger feeding preference than H.leucospilota,and the same types of microbes escaped digestion of the two sea cucumber species.Obvious diff erent bacterial community composition in the foreguts of the two species of sea cucumbers and the surrounding sediments might result from the animal’s selective feeding for sediment patches.Bacterial community structure in hindgut contents of H.atra and H.leucospilota both diff ered clearly from adjacent sediments,which indicated feeding activity of deposit-feeding sea cucumbers could change the sedimental bacterial composition.In conclusion,from the perspective of bacteria,sea cucumber H.atra and H.leucospilota had diff erent feeding preferences,yet they could both aff ect bacterial composition in sediments by feeding activity.The motivation for selective feeding and sea cucumber-sediment interaction might be explored in the future.Fei GAO Yue ZHANG Peilin WU Mengling CHEN Linwen HE Qiang XU Aimin WANG 2022Journal of Oceanology and Limnology2022,40,1:0
