1MicroRNAs and Their Cross-Talks in Plant Development显示文摘Plant development is a complex process influenced by exogenous and endogenous elements.A series of postembryonic developmental events is involved to form the final architecture and contend with the changing environment.MicroRNA(miRNA) is one of endogenous non-coding RNAs,which plays an important role in plant developmental regulation.In this review,we summarized 34 miRNA families that are closely associated with plant development.Among these families,nine are expressed only in specific organs,whereas 20 families are expressed in at least two different organs.It is known that some miRNAs are expressed across most processes of plant growth,while some appear only at particular developmental stages or under special environmental conditions such as drought,waterlogging and short-day time.These miRNAs execute their diverse functions by regulating developmental gene expression levels,interacting with phytohormone signaling response,participating in the biogenesis of ta-siRNAs and affecting the production of miRNAs.Danfeng Jin Yue Wang Yuhua Zhao Ming Chen 2013Journal of Genetics and Genomics2013,40,4:9
2High-throughput sequencing-based genome-wide identification of micro RNAs expressed in developing cotton seeds显示文摘Micro RNAs(mi RNAs) have been shown to play critical regulatory roles in gene expression in cotton. Although a large number of mi RNAs have been identified in cotton fibers, the functions of mi RNAs in seed development remain unexplored. In this study, a small RNA library was constructed from cotton seeds sampled at 15 days post-anthesis(DPA) and was subjected to high-throughput sequencing. A total of 95 known mi RNAs were detected to be expressed in cotton seeds. The expression pattern of these identified mi RNAs was profiled and 48 known mi RNAs were differentially expressed between cotton seeds and fibers at 15 DPA. In addition, 23 novel mi RNA candidates were identified in 15-DPA seeds. Putative targets for 21 novel and 87 known mi RNAs were successfully predicted and 900 expressed sequence tag(EST) sequences were proposed to be candidate target genes, which are involved in various metabolic and biological processes, suggesting a complex regulatory network in developing cotton seeds. Furthermore, mi RNA-mediated cleavage of three important transcripts in vivo was validated by RLM-5′ RACE. 3利用高通量测序技术鉴定棉纤维发育相关miRNAs及其靶基因显示文摘miRNA(microRNA)是一类21~24个核酸长度的非编码小分子RNA(sRNA),它主要通过抑制或降解靶基因来调控植物生长发育等过程。试验利用在纤维长度上有显著差异的2个回交自交系BILs(Backcrossinbred lines)的0 DPA(Days post anthesis)、3 DPA的胚珠和10 DPA的纤维构建6个sRNA文库并进行Solexa测序。以已公布的棉花D5基因组序列和棉属其他序列为参考,经分析共发现561个miRNAs,其中包括254个已知的miRNAs(属于40个miRNA家族),75个候选的miRNAs和232个新的miRNAs,研究结果极大地丰富了棉属miRNAs。通过miRNA靶基因预测分析发现多数miRNAs负调控其对应的靶基因,少数正调控。KEGG(Kyoto encyclopedia of genes and genomes)注释结果表明miRNA的靶基因在植物激素代谢途径中显著富集。南文智 吴嫚 于霁雯 范术丽 黄双领 李兴丽 张红卫 张金发 喻树迅 2013棉花学报2013,25,4:6

5元江干热河谷木棉蒴果形成和纤维发育过程显示文摘采取石蜡切片和扫描电子显微镜等方法研究了元江干热河谷木棉蒴果形成和纤维发育过程.结果表明:木棉蒴果形成过程分为4个阶段:花后0~5 d为形成期(FS),5~35 d为质量增加期(MGS),35~50 d为缓慢生长或脱水期(DS),50 d以后为成熟爆裂期(BS).与木棉蒴果形成过程相对应的纤维发育过程为:果皮内壁细胞分化(花后0~2 d)、膨大(花后2~5 d)、突起(花后5~10 d)、纤维伸长(花后10~40 d)、纤维脱离内壁及脱水成熟(花后40~50 d).在纤维发育过程中,纤维长度和投影宽度均呈幂函数曲线增长,花后第20天日均增长率达到最大.纤维鲜质量呈先增加后下降的趋势,花后25~30 d日均鲜质量增长率达到最高,而纤维干质量呈逐渐增长趋势,且花后20~25 d纤维干质量日均增长率达到最高.花后第30天,木棉种子和纤维质量继续增加,而果皮呈现下降趋势,表明果皮开始脱水老化.木棉纤维在蒴果壳体内壁的附着力小,较棉花等种子纤维更容易分离.花后5~35 d是蒴果生长和纤维发育最关键的时期,需做好水肥管理;而花后50 d后果实开始爆裂,需做好采收准备.赵高卷 葛娈 马焕成 杨建军 黄冬 平盼 2014应用生态学报2014,25,12:4
6Two-dimensional gel electrophoresis-based analysis provides global insights into the cotton ovule and fiber proteomes显示文摘Proteomic analysis of upland cotton was performed to profile the global detectable proteomes of ovules and fibers using two-dimensional electrophoresis(2DE).A total of 1,203 independent protein spots were collected from representative 2DE gels,which were digested with trypsin and identified by matrix-assisted laser desorption and ionization-time-offlight/time-of-flight(MALDI-TOF/TOF)mass spectrometry.The mass spectrometry or tandem mass spectrometry(MS or MS/MS)data were then searched against a local database constructed from Gossypium hirsutum genome sequences,resulting in successful identification of 975 protein spots(411 for ovules and 564 for fibers).Functional annotation analysis of the 975identified proteins revealed that ovule-specific proteins were mainly enriched in functions related to fatty acid elongation,sulfur amino acid metabolism and post-replication repair,while fiber-specific proteins were enriched in functions related to root hair elongation,galactose metabolism and D-xylose metabolic processes.Further annotation analysis of the most abundant protein spots showed that 28.96%of the total proteins in the ovule were mainly located in the Golgi apparatus,endoplasmic reticulum,mitochondrion and ribosome,whereas in fibers,27.02%of the total proteins were located in the cytoskeleton,nuclear envelope and cell wall.Quantitative real-time polymerase chain reaction(q RT-PCR)analyses of the ovule-specific protein spots P61,P93 and P198 and fiber-specific protein spots 230,477 and 511 were performed to validate the proteomics data.Protein-protein interaction network analyses revealed very different network cluster patterns between ovules and fibers.This work provides the largest protein identification dataset of 2DE-detectable proteins in cotton ovules and fibers and indicates potentially important roles of tissue-specific proteins,thus providing insights into the cotton ovule and fiber proteomes on a global scale.Xiang Jin Limin Wang Liping He Weiqiang Feng Xuchu Wang 2016Science China(Life Sciences)2016,59,2:3

9A comparative analysis of small RNAs between two Upland cotton backcross inbred lines with different fiber length: Expression and distribution显示文摘The cotton fiber is the most important raw material for the textile industry and an ideal model system for studying cell elongation. However, the genetic variation of fiber elongation in relation to miRNA is poorly understood. A high-throughput comparative RNA-seq of two lines differing in fiber length(FL) from a backcross inbred line(BIL)population of G. hirsutum × G. barbadense revealed differentially expressed(DE) miRNAs and their targets in rapidly elongating fibers. A real-time quantitative PCR analysis was further performed to validate the results. A total of 463(including 47 DE) miRNAs were identified,and seven DE miRNAs were co-localized with seven FL quantitative trait loci(QTL)identified in the G. hirsutum × G. barbadense population. Of 82(including 21 DE) targets identified, nine(including one DE) were also co-localized with the seven FL QTL. The relationship between the allopolyploid and its diploid ancestral species with respect to miRNAs and their targets was also characterized.
