2遗传分化对多样性种植下主栽和间栽水稻品种抗稻瘟病效率的影响显示文摘水稻品种多样性的种植方式对控制稻瘟病非常重要,但对其作用机理的了解甚少,其中,品种间遗传阻隔效应是众多假说之一。为验证水稻多样性混栽种植模式下,品种组合主栽和间栽的品种遗传分化水平对其田间抗稻瘟病效果的影响,本研究建立了利用插入/缺失(InDel)中性分子标记快速和准确检测水稻品种间遗传分化的技术方法。该方法可以对主栽和间栽品种之间的遗传分化系数(GDI)进行检测和定量描述,其变化范围在0.00~1.00之间,GDI数值越高表示品种间遗传分化水平越高。利用GDI结合云南省在10年内广泛用于多样性种植的主栽和间栽水稻品种组合,筛选出19对品种组合进行其GDI与抗稻瘟病防治效率相关性分析的田间试验。根据对各组合品种田间稻瘟病发生情况的调查发现,不同搭配组合在田间所表现出的稻瘟病防治效果存在很大差异。进一步分析的结果表明,主栽和间栽品种的GDI与其田间的稻瘟病防效具有显著正相关性(R2〉0.92,P〈0.01),验证了在多样性混栽种植模式中,水稻组合品种间的遗传分化水平对其田间抗稻瘟病效率有显著影响的遗传假设。该发现不仅揭示了水稻多样性种植模式下,主栽和间栽水稻品种之间遗传分化影响其田间稻瘟病防效的遗传机制,还为混合间栽中不同水稻品种搭配组合的选择及对其田间生产中稻瘟病发生的预测提供了实用技术。郎杰 韩光煜 徐俊采 王静文 王云月 卢宝荣 2015云南农业大学学报(自然科学版)2015,30,3:4
3亚洲杂草稻红色果皮分子鉴定及其Rc-bHLH序列多态性分析显示文摘多数杂草稻的果皮是红色,该特征可用于稻田中杂草稻的早期的初步鉴定,也可用于水稻果皮Rc/rc基因的起源进化研究。本研究利用红色果皮Rc基因第六外显子的In Del标记RID14对来自亚洲不同稻作区的199份杂草稻(O.sativa f.spontanea)、24份栽培稻(O.sativa)和9份野生稻(O.rufipogon)进行基因型鉴定并与表型比对;同时对9份代表性杂草稻Rc-b HLH位点的DNA序列进行测序,并将其与下载的29份栽培稻(籼,粳)和14份野生稻的对应序列进行比对分析。研究结果表明:(1)亚洲杂草稻的果皮颜色的表型多样,多数(84.4%)为红色或褐色(由浅至深)、少数(15.6%)为白色,其基因型与表型鉴定结果高度一致;杂草稻红色果皮Rc基因第六外显子均为无14 bp缺失的类型,仅有一份中国东北杂草稻除外(为14 bp缺失型),揭示用RID14标记可以早期快速的鉴定稻田中红色果皮杂草稻;(2)测试的9份杂草稻其Rc-b HLH位点的DNA序列均较为保守,其中出现4个突变位点;(3)在Rc-b HLH部分区域(1-115 bp),红色果皮的籼型栽培稻、杂草稻和野生稻的DNA序列相同,但红色果皮粳型栽培稻与白色果皮籼型栽培稻和杂草稻在106 bp处存在一个A/G替换;(4)通过对比野生稻和栽培稻,杂草稻在Rc-b HLH位点核苷酸多态性多态性最高,但是野生稻在该位点Tajima'D检验值呈显著,说明Rc-b HLH位点在野生稻中有选择性;通过聚类分析发现分为2个类群,类群1由白色果皮的杂草稻、籼型栽培稻和红/白果皮的粳型栽培稻组成;类群2由红色果皮的栽培稻(籼型)、杂草稻和野生稻组成。该结果暗示:在粳稻区,白色果皮的亚洲杂草稻可能由栽培稻退化返祖而形成的;在籼稻区,红色果皮的亚洲杂草稻可能经由栽培稻及野生稻杂交(如籼籼交或栽野交)演化而来。张树林 李娟 董陈文华 熊海波 张利东 朱骞 张小玲 伍腾飞 李东宣 陈丽娟 2015分子植物育种2015,13,1:3

5黄河流域豫北粳稻区杂草稻特性及其发生机制分析显示文摘杂草稻泛指生长在稻田及其周边被视为杂草的稻株,在国内外不同生态稻区均有不同程度的发生,但有关中国河南省豫北地区杂草稻特性及其发生状况鲜有报道。本研究以河南省黄河流域豫北粳稻区采集到的56份杂草稻为试验材料,考察其生物学特征特性;利用RID14引物鉴定红色果皮Rc位点基因型并与表型比对;同时通过34对In Del分子标记检测籼粳分化,分析了该区域杂草稻的特性、遗传分化及其可能发生的机制。研究结果表明:(1)黄河流域豫北粳稻区杂草稻大部分为淡黄色和黄褐色斑点的颖壳、多数无芒和红色果皮、易落粒、千粒重低,株高、分蘖、穗粒数与栽培品种有明显差异;(2)杂草稻果皮颜色的表型多样,89.29%为红色或者褐色(由浅至深)、10.71%为白色,其基因型与表型鉴定结果高度一致,红色果皮杂草稻在Rc位点均无14 bp缺失;(3)InDel标记和籼粳分化分析表明:56份杂草稻中有44份是典型籼型杂草稻、5份为籼型杂草稻、3份为偏籼型杂草稻、4份为中间型杂草稻,它们之间的遗传差异小;通过聚类分析,56份杂草稻中的92.86%的属于籼型杂草稻,7.14%属于中间型杂草稻,与当地粳型栽培稻有着较远的亲缘关系。根据研究结果推断,黄河流域豫北粳稻区杂草稻的发生,可能主要经由曾经种植过的籼粳栽培品种杂交而产生,或者是种植的杂交品种收获后落粒的后代去驯化而产生。本研究为黄河流域豫北地区杂草稻的防控提供了理论依据。熊海波 白怀瑾 董陈文华 李娟 文建成 许红云 朱骞 张小玲 张慧鑫 汪琨璧 刘思琪 付亚雄 郭效琼 Sadia Nadir 李东宣 陈丽娟 2017分子植物育种2017,15,8:2
6低血糖生成指数水稻南粳丝苗的选育及应用显示文摘以籼稻高产品种扬稻6号、低血糖生成指数品种Basmati、粳稻香味品种R405为亲本,进行杂交和复交,历经多代系谱选择,获得多份丰产性、抗倒性等综合农艺性状优良的新品系;通过模拟精米人体外消化试验,获得消化速度慢的品系若干份;再测定直链淀粉含量以及抗性淀粉含量,同时进行食味品质评价,筛选到生育期适中、丰产性好的偏粳型新品系南粳丝苗。南粳丝苗具有优质、低血糖生成指数(GI)等特性,多次检测结果表明,其稻米主要品质指标达国标一级优质米标准,且适口性较好,血糖生成指数为55,明显低于普通稻米,是一个适合糖尿病、高血糖、肥胖等特殊人群的低升糖指数稻米品种。南粳丝苗已在江苏宜兴江南米道等企业进行规模化生产,具有较大的市场前景。下一步拟通过育种技术进一步提高低GI水稻品种的稻谷产量潜力,并强化低GI水稻配套栽培技术的研究。蒋彦婕 杨杰 王芳权 许扬 李霞 陈智慧 李文奇 陶亚军 范方军 王军 2022江苏农业科学2022,50,18:1
7Development and validation of InDel markers for identification of QTL underlying flowering time in soybean显示文摘Soybean [Glycine max(L.) Merrill] is a major plant source of protein and oil. An accurate and well-saturated molecular linkage map is a prerequisite for forward genetic studies of gene function and for modern breeding for many useful agronomic traits. Next-generation sequence data available in public databases provides valuable information and offers new insights for rapid and efficient development of molecular markers. In this study, we attempted to show the feasibility and facility of using genomic resequencing data as raw material for identifying putative In Del markers. First, we identified 17,613 In Del sites among 56 soybean accessions and obtained 12,619 primer pairs. Second, we constructed a genetic map with a random subset of 2841 primer pairs and aligned 300 polymorphic markers with the 20 consensus linkage groups(LG). The total genetic distance was 2347.3 c M and the number of mapped markers per LG ranged from 10 to 23 with an average of 15 markers. The largest and smallest genetic distances between adjacent markers were 52.3 c M and 0.1 cM, respectively. Finally, we validated the genetic map constructed by newly developed In Del markers by QTL analysis of days to flowering(DTF) under different environments. One major QTL(qDTF4) and four minor QTL(qDTF20, qDTF13, qDTF12,and q DTF11) on 5 LGs were detected. 8Enhanced genetic diversity of weedy rice populations associated with latitude decreases revealed by simple sequence repeat fingerprints显示文摘Weedy rice (WR,Oryza sativa L.f.spontanea)is a noxious agricultural weed,infesting rice fields worldwide and causing tremendous yield losses of cultivated rice.However,little is known about the relationship between genetic diversity and distribution of WR populations across a wide latitudinal gradient,in addition to its reasons for genetic differentiation.To determine the distribution of genetic diversity and differentiation,we analyzed 20 WR populations collected from wide geographic ranges of rice-planting regions across Northeast,Jiangsu,and Guangdong provinces of China,and Sri Lanka,based on 20 simple sequence repeat loci.Our results indicated a significant negative correlation (R =0.84,P <0.01)between genetic diversity and latitudinal locations of WR populations.The Mantel test (R2 =0.49,P <0.01)showed distinct groupings of WR populations from different rice-planting regions,fitting an isolation-by-distance pattern.In addition,the STRUCTURE analysis and principal coordinates (PCoA)analysis indicated considerable genetic differentiation of WR from different rice-planting regions,which was associated with the types of co-occurring rice cultivars.We conclude,based on the above results,that WR genetic diversity is affected by the latitudes where WR populations are located.The genetic differentiation of WR populations is determined by their spatial distances and co-occurring rice cultivars.Such a pattern of genetic diversity and differentiation across different regions might facilitate the design of effective WR control,in addition to understanding the adaptive evolution of this weed.Zhe Wang Min-Jie Fu Hai-Ge Zhu Yue Zhu Xiang-Xiang Zhao Bao-Rong Lu 2019Journal of Systematics and Evolution2019,57,1:0

10Origins of weedy rice revealed by polymorphisms of chloroplast DNA sequences and nuclear microsatellites显示文摘Conspecific weeds that permanently infest worldwide agroecosystems are evolved from their crop species.These weeds cause substantial problems for crop production by competing for resources in agricultural fields.Weedy rice represents such a conspecific weed infesting rice ecosystems,and causing tremendous rice yield losses owing to its strong competitiveness and abundant genetic diversity,likely resulted from its complex origins.Here,we report the use of chloroplast DNA(cpDNA)fingerprints to determine whether weedy rice is evolved from its wild(exo-feral)or cultivated(endo-feral)rice progenitor as the maternal donor in recent hybridization events.In addition,we also applied nuclear simple sequence repeat(SSR)markers to confirm the exo-feral or endo-feral origins of weedy rice accessions determined by the cpDNA fingerprints.We found that the studied weedy rice accessions evolved either from their wild or cultivated rice progenitor,as the maternal donor,based on the cpDNA network and structure analyses.Combined analyses of cpDNA and nuclear SSR markers indicated that a much greater proportion of weedy rice accessions had the endo-feral origin.In addition,results from the genetic structure of nuclear SSR markers indicated that weedy rice accessions from the endo-feral pathway are distinctly associated with either indica or japonica rice cultivars,suggesting their complex origins through crop-weed introgression.The complex pathways of origin and evolution could greatly promote genetic diversity of weedy rice.Therefore,innovative methods should be developed for effective weedy rice control.Nan Yao Zhe Wang Zhuo-Jun Song Lei Wang Yong-Sheng Liu Ying Bao Bao-Rong Lu 2021Journal of Systematics and Evolution2021,59,2:0
11Genetic Characterization of Indigenous Rice Varieties in Eastern Himalayan Region of Northeast India显示文摘The eastern Himalayan region of Northeast(NE) India is home to a large number of indigenous rice varieties, which are traditionally classified as Oryza sativa subspecies indica, japonica or intermediate types. The classification based on traditional Cheng's index is often inconclusive due to phenotypic plasticity of morphological characters, which are influenced by environmental conditions. We used molecular markers specific for indica and japonica subspecies to assess the degree of genetic relatedness of indigenous rice varieties in NE India. The results revealed that majority of upland(jum) and glutinous rice varieties, traditionally considered as japonica, were genetically close to the subspecies indica. All varieties of boro ecotype were found to be indica type, and only a few varieties cultivated in lowland and upland areas were japonica type. 12基于叶绿体DNA的籼-粳稻谷(米)SYBR-Green实时荧光PCR鉴定方法的建立显示文摘亚洲栽培稻分为籼稻(hsien或indica)和粳稻(keng或japonica)两个亚种。基于籼-粳稻叶绿体DNA(cpDNA)的PstⅠ-12片段ORF100区域的序列差异,即籼稻cpDNA的ORF100区域缺失69 bp的碱基片段,建立籼-粳稻亚种鉴定方法。本文选择典型的籼稻谷(米)样品(台籼11号)和粳稻谷(米)(东北珍珠大米)进行叶绿体DNA(cpDNA)PstⅠ-12片段的ORF100区域DNA的SYBR-Green实时荧光PCR扩增和测序,在籼-粳稻2个PCR产物序列比对的基础上,设计正向引物4条(F1-F4)、反向引物3条(R1-R3),组合成12对引物进行优化筛选,选出F4R1引物对。在粳稻样品中,检出cpDNA PCR产物片段长度为204 bp(非缺失型),荧光PCR扩增熔解温度Tm值为78.00;在籼稻样品中,检出cpDNA PCR产物片段长度为135 bp(cpDNA缺失型),荧光PCR扩增熔解温度Tm值为76.50。建立了籼-粳稻亚种SYBR-Green实时荧光PCR鉴定方法。基于上述实验结果,选择国内多个省份的典型籼/粳稻品种与籼/粳型杂交稻组合共538个,对该籼粳稻米亚种鉴定方法进行验证。在177个粳稻及粳型杂交组合样品中,cpDNA荧光PCR熔解温度Tm值为78.00的样品170个,与粳稻符合率为96.05%,Tm值为76.50的7个样品,不符合率为3.95%;在361个籼稻及籼型杂交稻样品中,Tm值为76.50的样品331个,与籼稻符合率为91.69%,Tm值为78.00的30个样品,不符合率为8.31%。本方法只检测一个遗传稳定的细胞质叶绿体DNA位点,用于籼-粳稻米的亚种鉴定,简便高效,准确率高。陈文炳 邵碧英 林阳武 缪婷玉 彭娟 郭菁 陈彬 彭华毅 2020中国口岸科学技术2020,2,10:0
13超高产杂交粳稻的遗传特征的初步分析显示文摘2012年江苏省杂交晚粳区试11份材料的平均亩产为683.24kg,仅甬优杂交组合E240(Z06)在6个试点的平均亩产达到796.1kg的超高产水平,较对照甬优8号增产15.13%.为探索该杂交组合的超高产遗传结构,利用在籼粳有多态性的分布于12条染色体的36对InDel标记检测,发现该组合的籼型基因频率为0.47,属于中间型,其他组合的籼型基因频率都低于0.4,属于粳型或偏粳型.分子标记检测杂交组合Z06在第5和第12染色体的基因型为纯合籼型和杂合型,与其他组合明显不同.广亲和基因的功能标记检测仅发现Z03、Z05和Z06这3个杂交粳稻携带广亲和基因S5n,其中Z05为纯合基因型,Z03和Z06为杂合基因型.该研究表明杂交粳稻超高产组合应该接近一半籼型一半粳型,第5和12染色体片段为籼型基因型,并携带广亲和基因.范方军 王芳权 黄胜东 许明 王军 朱金燕 李文奇 沈文飚 仲维功 杨杰 2014安徽大学学报(自然科学版)2014,38,6:0
