| 2 | Humulus lupulus L. Extract Prevents Ovariectomy-Induced Osteoporosis in Mice and Regulates Activities of Osteoblasts and Osteoclasts显示文摘Objective:To systematically evaluate the protective effects of Humulus lupulus L.extract(HLE)on osteoporosis mice.Methods:In vivo experiment,a total of 3512-week-old female ICR mice were equally divided into 5 groups:the sham control group(sham);the ovariectomy with vehicle group(OVX);the OVX with estradiol valerate[EV,0.2 mg/(kg·d)];the OVX with low-or high-dose HLE groups[HLE,1 g/(kg·d)and 3 g/(kg·d)],7 in each group.Treatment began 1 week after the ovariectomized surgery and lasted for 12 weeks.Bone mass and trabecular bone mircoarchitecture were evaluated by micro computed tomography,and bone turnover markers in serum were evaluated using enzyme-linked immunosorbent assay(ELISA)kits.In vitro experiment,osteoblasts and osteoclasts were treated with HLE at doses of 0,4,20 and 100μg/mL.Biomarkers for bone formation in osteoblasts and bone resorption in osteoclasts were analyzed.Results:Compared with the OVX group,HLE exerted bone protective effects by the increase of estradiol(P<0.05),the improvement of cancellous bone structure,bone mineral density(P<0.01)and the reduction of serum alkaline phosphatase(ALP),tartrate resistant acid phosphatase(TRAP),bone gla-protein,c-terminal telopeptides of typeⅠcollagen(CTX-Ⅰ)and deoxypyridinoline levels(P<0.01 for all).In vitro experiment,compared with the control group,HLE at 20μg/mL promoted the cell proliferation(P<0.01),and increased the expression of bone morphogenetic protein-2 and osteopontin levels in osteoblasts(both P<0.05).HLE at 100μg/mL increased the osteoblastic ALP activities,and HLE at all dose enhanced the extracellular matrix mineralization(both P<0.01).Furthermore,compared with the control group,HLE at 20μg/m L and 100μg/m L inhibited osteoclastic TRAP activity(P<0.01),and reduced the expression of matrix metalloproteinase-9 and cathepsin K(both P<0.05).Conclusion:HLE may protect against bone loss,and have potentials in the treatment of osteoporosis. | XIA Tian-shuang LIN Liu-yue ZHANG Qiao-yan JIANG Yi-ping LI Chang-hui LIU Xiao-yan QIN Lu-ping XIN Hai-liang | 2021 | Chinese Journal of Integrative Medicine2021,27,1: | 4 |
| 3 | Regulation of cholesterol homeostasis in health and diseases:from mechanisms to targeted therapeutics显示文摘Disturbed cholesterol homeostasis plays critical roles in the development of multiple diseases,such as cardiovascular diseases(CVD),neurodegenerative diseases and cancers,particularly the CVD in which the accumulation of lipids(mainly the cholesteryl esters)within macrophage/foam cells underneath the endothelial layer drives the formation of atherosclerotic lesions eventually.More and more studies have shown that lowering cholesterol level,especially low-density lipoprotein cholesterol level,protects cardiovascular system and prevents cardiovascular events effectively.Maintaining cholesterol homeostasis is determined by cholesterol biosynthesis,uptake,efflux,transport,storage,utilization,and/or excretion.All the processes should be precisely controlled by the multiple regulatory pathways.Based on the regulation of cholesterol homeostasis,many interventions have been developed to lower cholesterol by inhibiting cholesterol biosynthesis and uptake or enhancing cholesterol utilization and excretion.Herein,we summarize the historical review and research events,the current understandings of the molecular pathways playing key roles in regulating cholesterol homeostasis,and the cholesterol-lowering interventions in clinics or in preclinical studies as well as new cholesterol-lowering targets and their clinical advances.More importantly,we review and discuss the benefits of those interventions for the treatment of multiple diseases including atherosclerotic cardiovascular diseases,obesity,diabetes,nonalcoholic fatty liver disease,cancer,neurodegenerative diseases,osteoporosis and virus infection. | Yajun Duan Ke Gong Suowen Xu Feng Zhang Xianshe Meng Jihong Han | 2022 | Signal Transduction and Targeted Therapy2022,7,9: | 3 |
| 4 | 高胆固醇对小鼠原代成骨细胞Akt/NF-κB信号通路的影响显示文摘目的观察高胆固醇对原代成骨细胞Akt/NF-κB信号通路的影响。方法将小鼠原代成骨细胞按1×105接种于6孔板上,待细胞融合至80%左右,分别加入0μg/ml、12.5μg/ml、25μg/ml和50μg/ml的胆固醇溶液培养基继续培养24 h。采用蛋白质免疫印迹法(Western blot)的方法,检测成骨细胞核转录因子kappa B(NF-κB)和蛋白激酶B(Akt)的蛋白表达;采用实时荧光定量PCR方法,检测成骨细胞中人白细胞介素1α(IL-1α)、人白细胞介素6(IL-6)和肿瘤坏死因子α(TNF-α)的信使RNA(m RNA)表达;采用酶联免疫吸附法,检测细胞培养上清中IL-1α、IL-6和TNF-α的蛋白水平。结果胆固醇诱导成骨细胞中NF-κB的增加和Akt磷酸化水平降低,增加IL-1α、IL-6与TNF-α的m RNA和蛋白表达,且呈一定的剂量依赖性。结论高胆固醇可以通过Akt/NF-κB信号通路及其介导的炎症因子表达参与成骨细胞的调节。 | 邓莉 宋丹 | 2016 | 中国现代医学杂志2016,26,10: | 2 |
| 5 | 长链非编码RNA-AF289591调控SOST表达对成骨细胞分化的影响显示文摘目的探讨长链非编码RNA(long noncoding RNA,lncRNA)-AF289591对间充质干细胞(human mesenchymal stem cells,hMSC)成骨分化的影响及其分子机制。方法收集骨质疏松症(osteoporosis,OP)患者以及健康人群的骨髓标本,通过lncRNA芯片分析hMSC中lncRNAs的表达,并通过实时荧光定量聚合酶链式反应(real-time polymerase chain reaction,qPCR)验证lncRNA-AF289591在两类人群中的表达,ELISA检测骨髓组织中硬骨抑素(sclerostin)水平。分离hMSC并通过流式细胞技术进行细胞鉴定,qPCR检测hMSC骨形成相关基因COL、OPN、OCN表达以及SOST基因表达水平,试剂盒检测碱性磷酸酶(alkaline phosphatase,ALP)活性,免疫印迹法(Western blot)检测硬骨抑素和RUNX2蛋白表达水平,荧光素酶报告基因检测Wnt-1启动子活性。结果相比健康人群,OP患者骨髓组织中lncRNA-AF289591表达显著升高,伴随着SOST基因表达水平增加(P<0.05)。分离hMSC并通过流式细胞技术确定为阳性细胞(CD105、CD44和CD29表达百分率分别为99.9%、100%和99.8%),过表达lncRNA-AF289591后,抑制骨形成相关分子COL、OPN、OCN基因表达(P<0.05),降低ALP活性(P<0.05),促进SOST基因表达(P<0.05),抑制Wnt-1启动子活性以及减少RUNX2蛋白表达水平(P<0.05)。干扰lncRNA-AF289591表达后,结果与上述情况相反(P<0.05)。结论lncRNA-AF289591表达通过调控SOST基因表达而抑制hMSC成骨分化。 | 曾越茜 杨明 陈琳 李思雨 李丽 游利 | 2021 | 中华骨质疏松和骨矿盐疾病杂志2021,14,2: | 2 |

| 11 | 绝经后骨质疏松症对颈后路椎板成形术的影响显示文摘目的:探讨绝经后骨质疏松症(PMOP)对于颈后路椎板成形术治疗效果的影响。方法:回顾性分析2013年1月—2017年6月我院行颈后路椎板成形术并完成两年随访的173例绝经女性患者临床资料,年龄52~80岁,平均(63.23±10.73)岁。根据是否存在PMOP,将患者分为绝经后骨质疏松组(PMOP,n=62)、非绝经后骨质疏松组(non-PMOP,n=111)。收集并分析两组术前基本资料和并存疾病的情况,并通过平片与CT评估患者锚钉松动以及钛板位移情况,采用视觉模拟量表(VAS)、颈椎功能障碍指数(NDI)、颈椎活动度(ROM)以及日本骨科协会评估治疗分数(JOA),评估两组患者的手术预后情况。结果:PMOP组血镁及25-(OH)D浓度较non-PMOP组低(P<0.001),β-CTX、TPINP与PTH较non-PMOP组升高(均P<0.001)。PMOP患者在术前更容易存在合并症,以高血压(72.58%,χ^2=15.471,P<0.001)、高血脂(61.29%,χ^2=15.39,P<0.001)以及糖尿病(59.68%,χ^2=31.25,P<0.001)最为显著。术中,PMOP患者手术时间更短[(94.76±36.26)min vs.(122.73±25.4)min,P=0.022]。术后,PMOP患者更容易出现锚钉松动(P=0.029)以及钛板移位(P=0.036),然而在临床效果评价(VAS、JOA、NDI、ROM以及C5神经根麻痹)方面,两组差异无统计学意义。结论:PMOP患者在术前更可能合并其他系统疾病,行颈后路椎板成形术的时间更短,术后锚钉松动与钛板移位率增加,在治疗效果方面没有明显差异。 | 樊清馨 贾宇涛 刘洋 贺超 李玉乔 孙天威 | 2020 | 天津医科大学学报2020,26,4: | 0 |
